Abstract

ABSTRACTBreast cancer is the second mostly found disease after cervix cancer to Indonesian woman. Research on the effect of Ulva fasciata Delile and Turbinaria decurrensBory ethyl acetate extract feeding on the Adenocarcinoma mammaecell proliferation on C3H mice has beenconducted. Thirty experimental mice were devided into 6 treatment groups, i.e.: K-I, negative control; K-II, solvent control; K-III, given U. fasciata ethyl acetate extract dose of 41.32 mg/20 g BW (body weight); K-IV, given U. fasciataethyl acetate extract dose of 82.64 mg/20 g BW; K-V, given T. decurrensethyl acetate extract dose of 24.29 mg/20 g BW; and K-VI, given T. decurrens ethyl acetate extract dose of 48.59 mg/20 g BW. All treatments were given during 21 days. The mice body weight and tumor volume were measured and collected subsequently at every two days. Tumor doubling time was calculated at the end of research. Results showed that there wasa highly tumor growth inhibition on the K-IV shown by the high doubling time value. Tumor growth inhibition decreased according to the sequence as follows: K-IV (U. fasciata dose of 82.64 mg/20g BW), K-V (T. decurrensdose of 24.29 mg/20 g BW), K-VI (T. decurrensdose of 48.59 mg/20 g BW) which equal to K-II (solvent control), K-III (U. fasciatadose of 41.32 mg/20 g BW), K-I(negative control).

Abstract

ABSTRAKPenelitian tentang pengaruh pemberian κ-karagenan dan ί-karagenan terhadap penurunan kadar glukosa darah tikus yang mengalami kerusakan pankreas akibat induksi aloksan monohidrat telah dilaksanakan. Dibuat 6 kelompok perlakuan hewan percobaan, masing-masing kelompok terdiri dari 5 ekor. Kelompok I kontrol normal (tanpa perlakuan), kelompok II (kontrol negatif) diberikan air suling (1 mL/200 g bobot badan), kelompok III (kontrol positif) diberikan injeksi insulin (0,756 UI/200 g bobot badan), kelompok IV (kontrol positif) diberikan suspensi klorpropamida (2,52 mg/200 g bobot badan), kelompok V diberikan κ-karagenan (0,01 g/200 g bobot badan), kelompok VI diberikan ί-karagenan (0,01 g/200 g bobot badan). Kelompok I, II, IV, V, dan VI, diberikan secara oral, sedangkan kelompok III diberikan secara subkutan. Hasil riset menunjukkan bahwa к-karagenan dan ί-karagenan dapat menurunkan kadar glukosa darah selama 15 hari pemberian pada tikus diabetes. Pemberian κ-karagenan dosis 0,01 g/200 g bobot badan dapat menurunkan kadar glukosa darah sebesar 37,55% dan pemberian ί-karagenan dosis 0,01 g/200 g bobot badan dapat menurunkan kadar glukosa darah sebesar 42,87%.

Abstract

Telah dilakukan kajian awal tentang potensi bioaktivitas ekstrak etanol dari spons Callyspongia sp. dan fraksinya terhadap sel lestari tumor HeLa. Spons dimaserasi dalam etanol, kemudian ekstrak diuji toksisitasnya tehadap Artemia salina menggunakan metode Brine Shrimp Lethality Test (BSLT). Hasil uji memberikan nilai LC50 sebesar 3,30 μg/mL. Selanjutnya ekstrak diuji sitotoksisitasnya terhadap sel lestari tumor HeLa dengan metoda MTT (3-(4,5-dimetiltiazolil-2)-2,5-difenil tetrazolium bromida), dan memberikan nilai IC50 sebesar 30,71 μg/mL. Ekstrak etanol lalu difraksinasi dengan pelarut heksan (non polar), etil asetat (semi polar), dan n-butanol (semi polar). Tiap ekstrak fraksi diuji sitotoksisitasnya terhadap sel lestari tumor HeLa. Hasil uji menunjukkan bahwa fraksi etil asetat memiliki nilai sitotoksisitas (IC50=6,06 μg/mL) paling tinggi terhadap sel lestari HeLa dibandingkan dengan fraksi heksan (IC50=50,06 μg/mL) dan fraksi nbutanol (IC50=43,75 μg/mL).

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas sitotoksik dua jenis algae coklat (Sargassum cinerum dan Sargassum polycystum) dan kandungan fukosantin kedua jenis rumput laut tersebut. Rumput laut diperoleh dari pantai Binuangeun, Lebak, Banten, Indonesia. Ekstrak aseton yang diperoleh dipekatkan dengan menggunakan konsentrator vakum. Aktivitas sitotoksik terhadap sel HeLa dan T47D dievaluasi dengan menggunakan uji MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide). Kandungan fukosantin pada ekstrak aseton S. cinerum dan S. polycystum dianalisis dengan menggunakan Kromatografi Cair Kinerja Tinggi (KCKT). Hasil uji sitotoksik memperlihatkan bahwa ekstrak S. cinerum dan S. polycystum memiliki aktivitas sitotoksik yang lebih tinggi terhadap sel T47D dibanding terhadap sel HeLa. Nilai IC50 S. cinerum dan S. polycystum terhadap sel T47D masing-masing sebesar 79,2 dan 52,2 µg/ml. Analisis KCKT memperlihatkan kadar fukosantin pada ekstrak aseton S. cinerum dan S. polycystum masing-masing sebesar 0,179 dan 0,318 mg/g ekstrak.

Abstract

Antioxidants are substances that may inhibit free radical reactions in the body. The purpose of this research is to identify lethal dose, in vitro antioxidant activity and effect of kemboja merah flowers decoction (Plumeria rubra L.) on MDA level. Lethal dose was measured by Weil method. Antioxidant activity assays was performed in vitro by DPPH method and in vivo by measuring MDA plasma level. The level of MDA was conducted using the thiobarbituric acid reactivity test. In the test, 30 mices were divided into 6 groups: normal, negative control, positive control and 3 group treated with variation dose of kemboja merah flowers decoction. LD50 of decoction kemboja merah flowers is > 15 g/kg of body weight. In vitro, antioxidant activity showed the IC50 value of decoction kemboja merah flowers is 36.96 μg/mL.The antioxidant activity of decoction kemboja merah flowers is smaller than vitamin C (IC50 5.89 μg/mL). In vivo antioxidant activity showed levels of MDA on normal group is 1.66 nmol/mL, negative control group is 5.67 nmol/mL, positive control group is 1.45 nmol/mL, treated group which was given by decoction kemboja merah flowers at dose 0.39 g/kg of body weight is 4.32

Abstract

Black pepper (Piperis nigri fructus) has been empirically used to eliminate fatigue and to increase stamina or as stimulant. The aim of this study was to determine the efficacy of black pepper infuse as stimulant on 30 male DDY mice using swimming and rotarod methods. The mice were divided into five groups. First group was given distilled water as control, second group was given caffeine of 0.36 mg/20 g bw. The other three groups were given orally with black pepper infuse of 0.32 mg/20 g bw, 0.65 mg/20 g bw, or 1.3 mg/20 g bw. Observations on mice swimming endurance and mice endurance on a rotarod, were carried out 60 minutes after oral administration of the solutions. Analysis using Kruskal-Wallis showed that there was significant difference in the stimulant efficacy between the black pepper infuse and the negative control (p<0.05). There was no significant difference in stimulant effect between the 0.32 mg/20 g bw of black pepper infuse and the caffeine positive control, however, the infuse dose of 0.65 mg/20 g bw and 1.3 mg/20 g bw have demonstrated stronger stimulant effects (p>0.05) as compared to the caffeine positive control.

Abstract

The 1,5-bis(3’-ethoxy-4’-hydroxyphenyl)-1,4-pentadiene -3-one (EHP) is a curcumin analogue. The structure has also been identified before. EHP is obtained, which has been pharmacologically proven to have higher antioxidant activity than curcumin. The objectives of this research were to determine the acute toxicity (LD50) of EHP compound by Weil C.S. method using male mice as experimental animals and to determine its antiinflammatory activity. The LD50 value of the synthesized EHP compound is 6,8675 g/kg body weight (bw) with the criteria of mild toxic, so it is assumed to be safe as drug substance candidate. The anti-inflammatory activity of the compound was determined by obeserved legs swelling index of the mouse. Anti-inflammatory activity of EHP with doses of 137.35 mg, 274.70 mg and 549.40 mg were equal to aspirin with the dose of 90 mg/200 g bw

Abstract

Bioanalysis method is needed to pharmacokinetic study of INH as antituberculosis. The main problem is INH structure is similar to that of acetyl isoniazid (AcINH) as its metabolite. Therefore, a selective separation method is needed to separate the INH from its metabolite and matrix. The aim of this study was to test the selectivity of separation method of INH and AcINH in human plasma in-vitro by high performance liquid chromatography (HPLC). The HPLC system used a reversed phase with UV detector and hexane sulphonate as an ion counter. The optimum conditions was obtained by using C18 as stationary phase, hexane sulphonate (pH 2.47)-methanol (65:35) as mobile phase, with flow rate of 1mL/min, and UV detector at wavelength of 265nm. The selectivity of method separation was indicated by a resolution value of ≥ 1.5. The tailing factor for INH and AcINH were 1.297, dan 1.912, respectively. The k values ware ​​less than 10 and N values ​​were greater than 5000 indicate good separation efficiency. The developing of HPLC was a selective for separating of INH and AcINH in human plasma in-vitro.

Abstract

Diabetes mellitus characterized by hyperglycemia whereas the condition for high blood glucose. InIndonesia, there are a lot of medicinal plants that used as antidiabetic treatment. Medicinal plants are known have relatively few side eff ects compared to chemical based drugs. Asparagus (Asparagus offi cinalis L.) was reported to have fl avonoids and saponins compounds that could be functioned as antidiabetic. The aim of this research was to fi nd out the eff ect of boiled asparagus on blood glucose level in mice induced by Alloxan. This research used 30 male DDY strained mice, which were classifi ed into 6 groups, and each group consisted of 5 mice. These 6 groups were: normal control, negative control, positive control (Acarbose), the boiled asparagus with dose of 280, 560, and 840 mg/kgBW for 14 days. Blood glucose level of mice were observed on day-0, 8, 15, and 22. The results showed that Alloxan injection increased the level of blood glucose. Another result showed that boiled asparagus with dose of 280, 560 and 840 mg/kgBW reduced the level of blood glucose and the eff ect showed treatment groups doses of 560 dan 840 mg/kgBW had no signifi cantly diff erent with positive control group (Acarbose).

Abstract

Secara empiris penggunaan daun senggani mempunyai kemampuan sebagai anti-inflamasi dan analgesik. Penelitian ini bertujuan menguji efek anti-inflamasi dan analgesik dari infusa daun senggani. Pada uji anti-inflamasi menggunakan metode Winter, tikus dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif diberi air suling, kontrol positif diberi natrium diklofenak 1,8 mg/200 g bb, kelompok uji diberi infusa daun senggani 2,8 mL/200 g bb dengan konsentrasi 10%, 20% dan 40%. Setelah 30 menit diinduksi karagenan 1%, 0,2 mL/200 g bb. Pada pengujian analgesik menggunakan metode Sigmund, mencit dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif diberi air suling, kontrol positif diberi natrium diklofenak 0,26 mg/20 g bb, kelompok uji diberi infusa daun senggani 0,4 mL/20 g bb dengan konsentrasi 10%, 20% dan 40%. Setelah 30 menit diinduksi asam asetat 3%, 0,2 mL/20 g bb. Dari hasil ANOVA satu arah infusa daun senggani dengan dosis 2,8 mL/200 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek sebagai anti-inflamasi dan infusa daun senggani dosis 0,4 mL/20 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek sebagai analgesik. Infusa daun senggani dengan dosis 2,8 mL/200 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek anti-inflamasi yang lebih rendah dibandingkan natrium diklofenak dan infusa daun senggani dosis 0,4 mL/20 g bb dengan konsentrasi 10%, 20% dan 40% mempunyai efek analgesik yang lebih rendah dibandingkan natrium diklofenak. Peningkatan dosis infusa daun senggani tidak memberikan peningkatan efek sebagai anti-inflamasi maupun analgesik.
