Abstract

Hyperpigmentation is a condition of darkening of the skin which is generally caused by an increase in melanin production. Melanin is produced in melanocytes by the enzyme tyrosinase. Piper crocatum contains flavonoid compounds that are known from previous research to inhibit tyrosinase. The goals of this study were to determine the tyrosinase inhibitory activity and total flavonoid content of seven accessions, as well as look at the Pearson’s correlation and clustering PCA (principal component analysis). The method used was water content analysis, extraction yield measurement, total flavonoids analysis, and in vitro tyrosinase inhibition. Based on the results, P. crocatum from Kendari had the best yield and total flavonoid content of 24.07% and 5.10 mg QE g-1, while P. crocatum from Bogor had the lowest water content with a value of 6.21% and the best in tyrosinase inhibition of 13.77. The correlation between total flavonoid content and percent inhibition showed a very weak correlation. The results of clustering formed four clusters of seven accessions based on total flavonoids and percent inhibition. The cluster was divided into Malang (506mDPL) and Jayapura (287mDPL), Banda Aceh (0.80mDPL) and Bandung (670mDPL), Samarinda (8mDPL) and Bogor (190-350m DPL), and Kendari (14mDPL). In conclusion, the correlation between total flavonoid levels and percent inhibition is very weak and regional diversity had a significant effect on total flavonoids and total inhibition.

Abstract

Melasma adalah kelainan pigmentasi yang diakibatkan sintesis melanin berlebih berupa bercak cokelat disekitar wajah atau leher. Melasma dapat mengenai semua kelompok ras, terutama daerah dengan radiasi ultraviolet tinggi seperti suku Hispanik/Latin dan Asia. Zat yang sering digunakan untuk mencegah hiperpigmentasi adalah asam kojat, asam azelat dan merkuri, akan tetapi zat tersebut bersifat karsinogen jika digunakan terus menerus. Sirih merah (Piper crocatum) memiliki senyawa aktif yang berperan sebagai antioksidan seperti flavonoid dan fenol. Penelitian ini bertujuan untuk mengetahui potensi senyawa aktif daun sirih merah terhadap penghambatan tirosinase. Penelitian ini dilakukan dengan metode in silico dan in vitro. Reseptor yang digunakan adalah tirosinase dengan kode PDB 5M8O, tropolon sebagai native ligand, dan asam kojat sebagai ligan pembanding. Sebanyak 34 senyawa aktif daun sirih merah hasil analisis LC-MS dan GC-MS.digunakan sebagai ligan uji. Penambatan molekuler dilakukan dengan menggunakan AutoDock Vina dan AutoDock Tools, serta divisualisasikan menggunakan Ligplot+ dan PyMOL. Penghambatan tirosinase secara in vitro menggunakan kit tyrosinase activity assay ab252899 dan asam kojat sebagai kontrol positif. Senyawa katekin memiliki energi bebas pengikatan (ΔG) terkecil, yaitu −6.7 kkal/mol, dan kinetika inhibisi sebesar 12.12 µM. Hasil pengujian in vitro, fraksi air memiliki aktivitas inhibisi tirosinase terbesar yaitu 84.84% dan asam kojat memiliki aktivitas sebesar 82.47%.

Abstract

Cinnamon (Cinnamomum burmannii) is an herbal plant that has a function, one of which is lowering glucose in the blood and warding off free radicals that cause oxidative stress. This study aims to determine the inhibitory activity of the enzyme compound α-glucosidase and the suppression of DPPH free radicals using the first, second, and third replications of cinnamon bark water extract (C. burmannii) as a natural source of antidiabetic drug candidates. The extraction method uses the infundation technique and is carried out using a spectronometer. The highest yield was obtained in the extraction of the first repeat, which is with an average of 0.674%, the yield did not differ markedly (p>0.05) with the second and third repeats. The highest results of total phenolic testing were obtained in the second repeat extraction of 100.374 GAE g-1, the results differed markedly (p<0.05) with the first and third repeats. The highest results of testing antioxidant activity were obtained at the extraction of the first repeat which was 9.012 mg of AAE g-1, the results differed markedly (p<0.05) with the second and third repeats. The highest result of testing of enzyme inhibition activity α-glucosidase was found in the extraction of the first repeat which was 99.650%, the results did not differ markedly (p>0.05) with the second and third repeats.

Abstract

Kesehatan masyarakat di pedesaan Indonesia seringkali terhambat oleh pola konsumsi pangan yang tidak seimbang, dengan ketergantungan tinggi pada karbohidrat dan asupan protein nabati yang kurang optimal. Salah satu solusi potensial adalah peningkatan konsumsi sari kedelai, yang kaya akan protein, vitamin, dan mineral. Sari kedelai dapat membantu memperbaiki gizi dan mengurangi risiko penyakit degeneratif. Desa Sukamaju di Kecamatan Cigudeg, Kabupaten Bogor, memiliki potensi untuk mengembangkan produksi sari kedelai, tetapi pemanfaatannya belum maksimal. Program ini bertujuan untuk meningkatkan gizi masyarakat dan memberdayakan ekonomi lokal melalui pendirian rumah produksi sari kedelai. Kegiatan melibatkan kader Posyandu dan PKK melalui sosialisasi dan pelatihan. Metode yang digunakan meliputi pengumpulan data melalui kuisioner pre-test dan post-test untuk menilai pengetahuan sebelum dan setelah pelatihan. Hasil pelatihan menunjukkan peningkatan pemahaman masyarakat tentang sari kedelai, meskipun awalnya pengetahuan mereka terbatas. Program ini berhasil meningkatkan minat dan pengetahuan mengenai sari kedelai, serta berpotensi untuk menjadi model bagi desa lain. Kesimpulannya, program ini memberikan manfaat ganda dengan meningkatkan gizi masyarakat dan mendukung ekonomi lokal, meskipun masih ada tantangan dalam mendorong produksi dan konsumsi sari kedelai secara mandiri.

Abstract

Daun sirih merah (Piper crocatum) dan kulit kayu manis (Cinnamomum burmanii) berkhasiat sebagai tanaman obat. Tanaman tersebut memiliki kandungan senyawa yang berperan sebagai antioksidan. Penelitian ini bertujuan menentukan aktivitas antioksidasi formula campuran ekstrak daun sirih merah dan kulit kayu manis dengan metode Thiobarbituric acid (TBA) dan metode 1,1- diphenyl-2-picrylhydrazyl (DPPH). Proses ekstraksi daun sirih merah dan kulit kayu manis dilakukan secara terpisah dengan pelarut air. Ekstrak air daun sirih merah dicampur dengan kulit kayu manis dengan perbandingan 5:0, 4:1, 3:2, 2.5:2.5, 0:5. Hasil penelitian menunjukkan formula 3:2 konsentrasi 200 ppm (61%) memiliki daya hambat yang sama dengan vitamin E terhadap pembentukan malondialdehida. Sementara itu, Formula 2.5:2.5 konsentrasi 25 ppm (59%) memiliki daya hambat yang sama dengan vitamin E terhadap penghambatan radikal DPPH.

Abstract

Coronary heart disease is a serious health problem and it is known to be lethal. The disease can be triggered by the accumulation of cholesterol plaques in the walls of blood vessels This cholesterol formation is assisted by methylglutaril-CoA reductase (HMG-CoA reductase) enzyme. Red betel contains active compounds that have the potential to act as HMG-CoA reductase inhibitor. This study aimed to predict the potential of ethanol extract compounds, water fraction, ethyl acetate fraction, and red betel n-hexane fraction as HMG-CoA reductase inhibitors by in vitro method. The in vitro method which consisted of extraction by maceration, graded fractionation with n-hexane, ethyl acetate, and water, continued by thiobarbituric acid (TBA) method to see the potential antioxidant activity of the sample. The Result showed that the highest inhibition of HMG-CoA reductase is 94% was found in the concentration 200 ppm ethyl acetate fraction. Also antioxidant activity of red betel showed by the inhibition of malondialdehyde formation had the highest activity in the ethyl acetate fraction than other fractions used.

Abstract

Red ginger (Zingiber officinale var. rubrum) is traditionally recognized for its medicinal properties, largely due to its antioxidant potential attributed to secondary metabolites such as flavonoids. This study aimed to evaluate the total flavonoid content and antioxidant capacity of ethanol and water extracts of red ginger from Sukabumi, specifically the Jahira 1 variety, as well as identify bioactive compounds. Extraction was carried out using the maceration method. The total flavonoid content was determined using the aluminium chloride colorimetric method, while the antioxidant capacity was assessed via the ferric reducing antioxidant power (FRAP) assay. Results showed that the ethanol extract had higher total flavonoid content and antioxidant capacity, with values of 5.99 mg quercetin equivalent (QE)/g and 473.93 µM Trolox equivalent (TE)/g, respectively. In contrast, the water extract exhibited 3.73 mg QE/g and 206.75 µM TE/g. Further LC-MS/MS analysis of the ethanol extract identified 26 compounds, including phenolics and terpenoids, which contribute to the antioxidant capacity of red ginger from Sukabumi.

Abstract

Colorectal cancer remains a leading cause of mortality worldwide and is closely linked to the immune system, particularly lymphocytes. This study evaluated the ethyl acetate fraction of red betel (Piper crocatum Ruiz & Pav) leaf ethanol extract for its anticancer activity against WiDr colon cancer cells and its effect on lymphocyte cell proliferation. Using MTT assays, the fraction showed significant WiDr cell inhibition (98.18% ± 1.21% at 500 ppm) and enhanced lymphocyte viability (143.3% ± 19.46% at 20 ppm). These results suggest the ethyl acetate fraction serves both as an effective anticancer agent against WiDr cells and as a promoter of lymphocyte proliferation, offering potential for dual therapeutic application.

Abstract

This study aimed to examine the antihyperglycemia activity of ethanol extract of 70% on surian bark and leaves on diabetic rats induced with streptozotocin based on body weight, and fasting blood glucose level. Consist of 28 rats are grouped into 7 groups namely normal, negative control, positive control which given glybenclamide with a dose of 3.22 mg/kg bw, bark extract with dose of 150 mg/kg bw, bark extract with a dose of 300 mg/kg bw, leaf extract with dose 150 mg/kg bw and leaf extract with dose 300 mg/kg bw. Diabetogenic mechanisms was conducted with intraperitoneal injection of streptozotocin at  dose of 50 mg / kg bw. This research was conducted experimentally using complete randomized design (RAL) to analyze the results obtained. Bark extract with a dose of 150 mg/kg bw had a better antihyperglycemia activity compared to the other groups which was showed by the decrease of blood glucose level percentage of 72.83% and this group also had ability to maintain body weight was better than the other groups as well as stable feed consumption during research process.

Abstract

Spirulina platensis is a commercially cultivated cyanobacterium known for its rich bioactive compounds, including phenolics, flavonoids, and phycocyanin, which contribute to its antioxidant and anticancer properties. While previous studies focused on laboratory-grown strains, this study evaluates the total flavonoid and phenolic content, antioxidant activity, and in vitro anticancer effects of ethanol extracts from two commercial manufacturers (MA and MB). The antioxidant potential was assessed using DPPH and FRAP assays, while cytotoxicity against A549 lung and HeLa cervical cancer cells was determined via MTT assays. Results indicated that MB exhibited higher flavonoid (12.52 ± 0.23 mg QE.g-1 DW) and phenolic content (19.10 ± 0.66 mg GAE.g-1 DW) than MA, correlating with stronger antioxidant activity (FRAP: 16.55 ± 0.1 µmol TE.g-1 DW vs. 11.1 ± 0.29 µmol TE.g-1 DW). DPPH inhibition reached 80.33% (MA) and 77.77% (MB). MB also demonstrated greater anticancer activity, with 28.31% inhibition in A549 cells at 100 ppm, compared to MA’s 20.6%, and 33.85% inhibition in HeLa cells at 50 ppm. Both extracts displayed preferential toxicity toward cancer cells over normal Chang cells, supporting their potential as safer alternatives to chemotherapy. This study confirms the dual therapeutic potential of commercially cultivated S. platensis as an antioxidant and anticancer agent, highlighting its applications in pharmaceutical and nutraceutical industries. Further research is recommended to isolate and identify the bioactive compounds responsible for its cytotoxic effects.
