Abstract

This study are determines to: 1) the effect of ND vaccine dose to the number of white blood cells produced in the female ducks; 2) the effect of ND vaccine doses to the amount of antibody titers produced in the female ducks. This research was conducted in December 2015 until January 2016, located in the Sabah Balau, District of Tanjung Bintang, South Lampung regency. Analysis of white blood cells (WBC) conducted in the Central Veterinary of Lampung, while the number of antibody titer was conducted in PT. Vaksindo, Jakarta. The experimental design used in this study is completely randomized design with three replications and 6 treatments (P0: injected 0.5 ml of distilled water, P1: 0.1 ml of ND inactivated vaccine doses, P2: 0.2 ml of ND inactivated vaccine doses, P3: 0.3 ml of ND inactivated vaccine doses, P4: 0.4 ml of ND inactivated vaccine doses, P5: 0.5 ml of ND inactivated vaccine doses). The treatment given to 5 days old female ducks with inactivated ND vaccine.  White blood cells are tested by using the WBC test while antibody titers were tested by using the HI test.  The data result were analyzed by ANOVA at 5% significance level and will be continued by Least Significant Difference (LSD) in white blood cells because the value of analysis of variance showed tangible results. The results of this study showed that giving of inactivated ND vaccine doses significantly (P <0.05) to the number of white blood cells, but did not significantly affect (P> 0.05) titer of antibodies. Abstract

The research was conducted at the Technical Service Unit Area - Regional Artificial Insemenation Center of Lampung, Terbanggi Besar District, Central Lampung Regency, Lampung Province on July 25th to 30th 2014, aims to: 1) determine the effect of different glycerol doses in yolk tris-sitrat diluent on Brahman’s frozen semen quality; 2) determine the best glycerol dose which can improve the Brahman bull’s frozen semen quality. The design used a completely randomized design with 5 treatments with glycerol doses in the yolk tris-sitrat diluent (5%, 6%, 7%, 8%, 9%) and 3 repetitions. The measured parameters were the percentage of sperm motility and live spermatozoa. The data were analyzed by analysis of variance with the real level 5% or 1% and continued with Orthogonal Polynomials test at significance level 5% or 1%. The results showed the different glycerol doses did not significantly effect (P>0.01) against the percentage of sperm motility and live spermatozoa post equilibration, but provides a significant influence (P<0.01)  against  the  percentage of  sperm  motility and  live  spermatozoa post  thawing.  Orthogonal polynomials test also showed the different doses of glycerol gives a significant influence (P<0.01) against the percentage of sperm motility and live spermatozoa post-thawing. The Effect of different glycerol doses  (5%, 6%, 7%, 8%, 9%) of the percentage sperm motility are 35.00%, 37.32%, 32.54%, 20.66%, 1.68% and the percentage of live spermatozoa post-thawing are 44.37%, 55.52%, 50.32%, 38.07%, 9.84%. Based on research results, it proved that 6% glycerol dose is the best dose to maintain the percentage of motility and live spermatozoa post-thawing.

Abstract

Thawing is a method by melting back of semen that had been frozen prior to artificialinsemination (AI). The temperature and duration of thawing has a major influence on the state of thespermatozoa, especially the integrity of spermatozoa in the semen. The combination of temperatureand duration of thawing that will either be able to prevent damage to sperm , so it still has a highfertility rate. In common, lowland is an area that has low temperature and humidity, so it will have aneffect of thawing’s temperature. The purpose of this study was to determine the temperature andduration of thawing of Brahman’s frozen semen which have the most optimal to get used inartificial inseminations in lowlands area.Research was conducted in March 2014 using a completely randomized design (CRD) with a3x3 factorial. The first factor is the temperature (34°C, 37°C, and 40°C) and Factor II is duration ofthawing (10 seconds, 15 seconds, and 20 seconds) with 3 replications. Parameters observed in thisstudy is the percentage of sperm motility and live spermatozoa. The results were analyzed usingANOVA and Duncan's test at the advanced level of 5 % and 1%. The results showed that temperatureand duration of thawing give effects to the quality of the frozen semen of Brahman cattle, but has nointeraction between them. The most good quality spermatozoa at a temperature of 40ºC spermmotility is 37,77% and the percentage of live spermatozoa is a 40,83%. Duration of thawing at 15seconds sperm motility is 38,33% and the percentage of live spermatozoa is a 39,77%.
