Abstract

Undur-undur laut (Emerita spp.) telah diteliti mempunyai kandungan gizi yang tinggi. Peptida dari hewan laut berpotensi besar dalam bidang kesehatan dan aplikasi biomedis, berkat aktivitas biologisnya yang beragam, seperti antioksidan, anti-hipertensi, antimikroba, dan perlindungan terhadap penuaan kulit. Teknik analisis in silico digunakan untuk mengidentifikasi peptida bioaktif dari protein sekuen dengan metode komputasi, seperti: database, aplikasi online dan software. Penelitian ini dilakukan untuk memprediksi peptida yang berasal dari undur-undur laut (Emerita spp.) dengan analisis in silico. Tahapan analisis in silico dilakukan menggunakan database NCBI dan dilanjutkan dengan analisis melalui software BIOPEP-UWM dan Peptide Ranker. Berdasarkan hasil analisis in silico pada hidrolisat protein undur-undur laut menggunakan BIOPEP, terdapat 16 peptida dengan potensi anti-hipertensi dan 14 peptida dengan potensi anti-diabetes. Berdasarkan hasil Peptide Ranker dapat dilihat urutan nilai probabilitas peptida yaitu kategori tinggi (WL, YF, AF, AW, IF, SF, ML, WT), kategori sedang (PR, AG, IG), dan kategori rendah (DP, HL, VPL, SL, NL, DR, AH, AI, VG, AEL, NT). Sekuen peptida yang tertinggi yaitu WL (Triptofan-Leusin) yang memiliki skor 0,99 dengan fungsi sebagai anti-diabetes. Sementara itu, nilai terkecil yaitu NT (skor 0,05) yang berfungsi anti-diabetes. Berdasarkan analisis sensori menggunakan BIOPEP, sekuen peptida yang dihasilkan tersusun atas asam amino yang memiliki rasa pahit dan manis. Peptida dan asam amino yang memiliki rasa pahit di antaranya : P, F, VF, IF, VY, V, VL, VI, L, IL, dan W, sedangkan yang memilki rasa manis di antaranya : P, G, dan V.

Abstract

Lactid Acid bacteria are capable of fermenting sugars or carbohydrates to produce lactic acid in large numbers. The characteristics of lactic acid bacteria in general are cells reacted positively to the Gram  stain, catalase react negatively and do not form spores. And fermentation of glucose would result Iaktat acid. Isolation of lactic acid bacteria carried in the intestines of three species of shrimp that Tiger Shrimp (Penaeus monodon), shrimp Seker (Metapenaopsis sp.), Guts White shrimp (Penaeus merguensis). Selection of bacteriocin -producing lactic acid bacteria were in a qualitative and semi-quantitative (pitting) in MRS medium. Selected isolates were identified by morphological characteristics, biochemical, physiological, A number of 209 isolates were isolated from three species of shrimp. The results of the qualitative selection there were 54 isolates produced bacteriocins. The results of the quantitative selection of 54 isolates produced 24 fine isolates.  From the 24 isolates there are 2 isolates seed SFE-7 (33) and P12A (25). Characteristics of seed isolates had morphological characteristics of spherical shape, cell arrangement tetrad, gram positive, negative motility. While the results of the biochemical approach to gram-negative, catalase negative,  homofermentative (no gas in the fermentation of glucose), can ferment D-Galactose, D-Glucose, D-mannose, D-lactose. Physiological approaches to isolate growth SFE-7 (33) and P12A (25) can be generated that both isolates able to live in the range of 4 ° C to 50 ° C, in the range of pH between 4-10, and able to grow at levels of 5-10% NaCl . Abstract

Sea urchins (Echinoidea) are one of the marine biota found in seagrass ecosystems in West Kupang District, East Nusa Tenggara. Sea urchins have an important role in the food chain cycle in seagrass ecosystems and are known as healthy food due to their high nutritional content. Sea urchin gonads contain protein, fat, vitamins and essential fatty acids, which are beneficial for human health. In addition to economic value, sea urchins also have ecological value in balancing and controlling macroalgae in coral reef and seagrass ecosystems. The aim of this research is to determine the bioecology of sea urchins in four research locations, namely Bolok, Lalendo, Baliana, and Tablolong beaches. The research method used was direct observation with Megabenthos and Benthos Belt Transect (BBT) techniques. The results showed that there were five types of sea urchins in the study site, namely Diadema setosum, Tripneustes gratilla, Echinometra calamaris, Echinothrix diadema, and Echinometra mathaei.   The results showed that the abundance of Diadema setosum individuals on Tablolong beach (0.46 ind/m²) while Tripneustes gratilla was dominant on Baliana beach (0.22 ind/m²). The highest species diversity index (D) was found in Lalendo (0,7) and the lowest in Tablolong (0,5), indicating moderate diversity. The uniformity index (E) was highest in Baliana (0.8) indicating a more even distribution of species than Tablolong (0.6), the dominance index (C) was highest in Lalendo (1.0), indicating a strong dominance of species at the site.   The highest Index of Gonadal Maturity (IKG) value for Diadema setosum was found in Tablolong (26.9), while Echinometra mathaei in Lalendo (68.6). The food consumption pattern of sea urchins showed that Tripneustes gratilla consumed more seagrass, especially at Baliana beach (82,08%), while Echinothrix diadema predominantly consumed seagrass at Lalendo and Baliana beaches.  Bulu babi (Echinoidea) merupakan salah satu biota laut yang banyak ditemukan pada ekosistem padang lamun di Kecamatan Kupang Barat, Nusa Tenggara Timur. Bulu babi memiliki peran penting dalam siklus rantai makanan di ekosistem lamun dan dikenal sebagai makanan yang menyehatkan karena kandungan nutrisinya yang tinggi. Gonad bulu babi mengandung protein, lemak, vitamin, dan asam lemak esensial, yang bermanfaat bagi kesehatan manusia. Selain nilai ekonomi, bulu babi juga memiliki nilai ekologi dalam menyeimbangkan dan mengendalikan makroalga di ekosistem terumbu karang dan lamun. Tujuan dari penelitian ini adalah untuk mengetahui bioekologi bulu babi di empat lokasi penelitian, yaitu pantai Bolok, Lalendo, Baliana, dan Tablolong. Metode penelitian yang digunakan adalah pengamatan langsung dengan teknik Megabentos dan Benthos Belt Transect (BBT). Hasil penelitian menunjukkan bahwa terdapat lima jenis bulu babi di lokasi penelitian yaitu Diadema setosum, Tripneustes gratilla, Echinometra calamaris, Echinothrix diadema, dan Echinometra mathaei. Hasil menunjukkan bahwa  kelimpahan individu Diadema setosum terrtinggi di pantai Tablolong (0,46 ind/m²) sedangkan Tripneustes gratilla dominan di pantai Baliana (0,22 ind/m²). Indeks keanekaragaman jenis (D) tertinggi ditemukan di Lalendo (0,7) dan terendah di Tablolong (0,2), menunjukkan keanekaragaman sedang. Indeks keseragaman (E) tertinggi di Baliana (0,8) menandakan distribusi spesies lebih merata dibandingkan Tablolong (0,6), Indeks dominansi (C) tertinggi di Lalendo (1,0), menunjukkan dominasi spesies yang kuat di lokasi tersebut.   Nilai Indeks Kematangan Gonad (IKG) tertinggi untuk Diadema setosum ditemukan di Tablolong (26,9), sedangkan jenis Echinometra mathaei tertinggi di Lalendo (68,6). Pola konsumsi jenis makanan bulu babi menunjukkan bahwa Tripneustes gratilla lebih banyak mengonsumsi lamun, terutama di pantai Baliana (82,08%), sementara Echinothrix diadema lebih dominan mengonsumsi lamun di pantai Lalendo dan Baliana.

Abstract

Village-Owned Enterprises, hereinafter referred to as BUMDes, are a village economic institution/entity that is a legal entity formed and owned by the Village Government, managed independently and professionally with capital that is wholly or mostly separated village assets. Ultimately, BUMDes was formed with the aim of increasing business opportunities in order to strengthen village autonomy and reduce unemployment. With Law No. 6 of 2014, the village government is given broader authority in terms of developing and empowering the economy of the village community in accordance with the potential, opportunities and capabilities of the village. However, many economic potentials of villages cannot be optimally utilized by the village to improve the economy and welfare of the community due to limited management strategies in managing BUMDes businesses and limited Human Resource capabilities in managing BUMDes. To achieve this goal, the Lecturers together with Students of the LP3I Jakarta Polytechnic held incidental training for the community and BUMDes Managers of Tugu Selatan Tourism Village in order to provide knowledge and understanding of SWOT analysis and Management Strategy so that it is hoped that the community and BUMDes Managers can further optimize the efforts that are currently being carried out and analyze how to make Tugu Selatan Tourism Village more advanced and developed and known by the wider community.

Abstract

This study aims to analyze the effect of various operational factors on the weight of dust collected by a dust collector machine. Using a multilevel factorial design, this study evaluates the interaction between frequency (Hz), hose length (m), and tube height (m). Analysis of variance shows that all three factors have a significant effect on the dust weight response (P-Value <0.05). The regression model with an R² of 99.15% shows a very high prediction accuracy. Parameter optimization was carried out to maximize the weight of dust collected, resulting in optimal conditions at a frequency of 50 Hz, a hose length of 2 m, and a tube height of 0.3 m with a dust weight of 72.75 g.

Abstract

Pantai Pok Tunggal merupakan salah satu destinasi wisata alam di Kabupaten Gunungkidul yang memiliki potensi berupa hamparan pasir putih, tebing karst, dan ekosistem pantai yang masih alami. Namun, pemanfaatan kawasan ini belum sepenuhnya optimal dan belum didasarkan pada kajian kesesuaian wisata yang mempertimbangkan karakteristik fisik pantai. Penelitian ini bertujuan untuk mengidentifikasi karakteristik fisik kawasan, menganalisis tingkat kesesuaian wisata, dan mengevaluasi pemanfaatan ruang eksisting sebagai dasar pengembangan kawasan. Penelitian dilakukan di tiga stasiun pengamatan dengan menggunakan sepuluh parameter fisik pantai, seperti lebar pantai, kemiringan, kedalaman, arus, dan kecerahan air. Analisis dilakukan menggunakan metode Indeks Kesesuaian Wisata (IKW). Hasil menunjukkan bahwa karakteristik fisik Pantai Pok Tunggal umumnya mendukung aktivitas wisata, ditandai dengan pantai landai dan perairan yang relatif jernih. Nilai IKW berkisar antara 80,56% hingga 97,22%, dengan dua stasiun termasuk kategori sangat sesuai (S1) dan satu stasiun tergolong sesuai (S2). Meskipun potensi fisik kawasan tinggi, pemanfaatan ruang saat ini belum sepenuhnya selaras dengan karakteristik lingkungan. Oleh karena itu, pengembangan kawasan perlu diarahkan secara berkelanjutan dan berbasis daya dukung fisik pantai.

Abstract

Coloring of aluminum oxide on the Aluminum type AA1100, which is the typical type for household appliances and purposes, through anodizing coloring process by the blue color of commercial printer ink had been successfully performed. The blue color was well adhered and was not easy to be removed by physical treatment such as wiping. The anodize coloring process includes chemical cleaning of aluminum surface, the formation of pores in the sulfuric acid solution at a specific current and a specified duration of time, coloring and closing the pores via sealing. It also shows the anodizing process increases aluminum oxide surface hardness increased by applying higher current density, and longer duration time of the process. The aluminum oxide hardness was in harmony to the duration of time processing, and current density; however, not in line to a concentration of the solution. The maximum hardness of the aluminum oxide layer obtained is 50 kg/mm2. The relation of current density and voltage, the effect of current density and sulfuric acid concentration toward aluminum oxide hardness were analyzed. Moreover, the difference of aluminum oxide hardness of the front side and backside to cathode also discussed.

Abstract

Penelitian ini bertujuan untuk menganalisis kelimpahan dan karakteristik mikroplastik (MP) yang terakumulasi pada kerang hijau (Perna viridis) dari empat lokasi di Kota Semarang, serta mengevaluasi efektivitas depurasi dalam mengurangi kandungan MP. Penelitian dilakukan menggunakan metode deskriptif-analitik dan eksperimental. Sampel kerang dibedakan berdasarkan ukuran (kecil 2–5 cm dan besar >5 cm), masing-masing sebanyak 15 ekor dari setiap lokasi. Mikroplastik dianalisis menggunakan mikroskop stereo melalui teknik pencacahan visual, berdasarkan bentuk, warna, dan ukuran partikel. Depurasi dilakukan terhadap 15 ekor kerang dari lokasi budidaya Tambak Lorok selama 96 jam menggunakan sistem sirkulasi air laut yang telah didesinfeksi, dengan pengamatan setiap 24 jam. Hasil penelitian menunjukkan bahwa kelimpahan MP tertinggi pada kerang kecil ditemukan di BTL ST.3 (13.892,97 ± 6700,45 partikel/kg) dan pada kerang besar di PTL (14.469,49 ± 746,38 partikel/kg). Bentuk MP yang teridentifikasi meliputi fiber, film, fragmen, dan pelet, dengan warna dominan merah, cokelat, biru, dan hitam. Proses depurasi menunjukkan penurunan jumlah partikel MP dari 44 menjadi 6,8 partikel/kerang, dengan efektivitas sebesar 88,28% pada jam ke-96. Hasil ini menunjukkan bahwa kerang hijau di seluruh lokasi telah terkontaminasi MP, namun kandungannya dapat dikurangi secara signifikan melalui proses depurasi

Abstract

Bacteria found in the digestive tract of shrimp mostly play a major role in nutrient absorption. One of the nutritional contents that is quite difficult for shrimp to absorb is fiber. Bacteria that can degrade fiber content are known as cellulolytic bacteria. This study aimed to determine the activity index of cellulolytic bacteria found in the digestive tract of shrimp. The research method includes sampling shrimp with three different categories based on average length and weight, divided into three categories such as large (22.2 g ± 87.3 cm), medium (17.1 g ± 36.7 cm), and small (13.4 g ± 18.8 cm) taken from the north coast waters of Pemalang and the south coast waters of Cilacap. Furthermore, bacterial isolation, morphological observation, calculation of bacterial abundance, and testing of cellulolytic activity were carried out. The results showed that 150 bacteria were successfully isolated from both locations at different sizes and a total of 68 bacterial isolates had cellulolytic activity. There were 22 potential isolates from each coastal water that had a great cellulolytic activity index, ranging from 1.4 to 7.0. Moreover, the most potential activities were performed by 8 bacterial isolates encoded with GM.1, GM.2, GM.3, GM.4, GM.5, GM.6, GM.7, and GM.8 with a range of cellulolytic index obtained ranging from 3.0-7.0. The high activity index of the cellulolytic bacteria obtained is a fairly promising potential as a probiotic candidate that plays a role in degrading fiber in food found in the digestive tract of shrimp so that it can accelerate its growth.

Abstract

Digital transformation in parking systems is essential to improving operational efficiency, staff integrity, and customer satisfaction. PT Reska Multi Usaha (KAI Services) has implemented various technology-based parking systems such as Manless, Full Manless, Combo, and LPR. However, several locations still rely on manual supervision, which increases the risk of fraud and inaccurate overnight vehicle data. This study developed an Internet of Things (IoT)-based prototype system to monitor gate status and manage customer surveys automatically, with real-time data transmission to a centralized dashboard. The system utilizes gate position sensors, microcontrollers, and digital survey forms integrated with an online server. Test results show that the system operates effectively without manual input, with an average data latency of 1.57 seconds and sensor detection accuracy of up to 50 cm. The system has the potential to be integrated with the Early Warning System (EWS) Command Center to support safer and more responsive parking services.
