Abstract

Kenikir (Cosmos caudatus), often consumed as a vegetable in Southeast Asia, possesses therapeutic benefits for several diseases, including diabetes mellitus. However, there has been limited investigation of the aqueous extract of C. caudatus for this disease model. This study aimed to examine the glucose-lowering effect of C. caudatus aqueous extract in an alloxan-induced diabetes model in rats. Ultrasound-assisted extraction was performed to obtain an aqueous extract of C. caudatus leaves. Qualitative phytochemical screening was carried out to identify the presence of bioactive compounds. Male Wistar rats were allocated to the following experimental groups: non-diabetic, diabetic without treatment, and diabetic treated with glibenclamide or aqueous C. caudatus extract at concentrations of 25%, 50%, or 100%. Diabetes was induced by intraperitoneal injection of 150 mg/kg alloxan. Random blood glucose and body weight were monitored before (Day 0) and after treatment (Days 3 and 7). There was a trend of weight loss in diabetic rats compared to non-diabetic rats, though the difference was not statistically significant. After 7 days of treatment, there was a comparable decrease in the blood glucose of diabetic rats treated with 50% or 100% of aqueous C. caudatus extract and those treated with glibenclamide. Qualitative phytochemical screening indicated the presence of steroid, saponin, phenol, and flavonoid compounds. The total phenolic content was 38.48 mg GAE/g and IC50 DPPH antioxidant activity was 375.64 ppm. This study demonstrated that an aqueous extract of C. caudatus exhibits a blood glucose-lowering effect in an alloxan-induced diabetic rat model.

Abstract

Cherry mistletoe (Dendrophthoe pentandra (L). Miq) have an inhibitory effect on the α-glucosidase enzyme, but research related to solvent optimization to find active compound is unclear. Secondary metabolites that can be identified depend on the level of solvent polarity. This study aims to determine the effect of solvent polarity on the secondary metabolite content and the α-glucosidase enzyme by using the polar and nonpolar fractions of the ethyl acetate extract of cherry mistletoe leaves. Cherry mistletoe leaves were extracted using ethyl acetate and were followed by a liquid-liquid fractionation. The polar fraction used ethanol and ethyl acetate as solvents, whereas the nonpolar fraction used n-hexane and ethyl acetate as solvents. Secondary metabolites present in polar fractions were alkaloids, flavonoids, steroids, tannins, and terpenoids, whereas those present in nonpolar fractions were flavonoids and steroids. The IC50 value of the polar fraction (54.8 ppm) was lower than that of the nonpolar fraction (192.0 ppm). The polar fraction of the ethyl acetate extract of cherry mistletoe leaves could inhibit the α-glucosidase enzyme and, therefore, is classified as active. On the other hand, the nonpolar fraction of the ethyl acetate extract of cherry mistletoe leaves could not inhibit the α-glucosidase enzyme and, therefore, is classified as inactive in general.

Abstract

Pendahuluan: Hiperurisemia pada pasien kanker meningkatkan angka kematian terutama pada stadium lanjut. Metode: Studi observasional analitik korelatif dengan rancangan cross-sectional, bertujuan untuk mengetahui korelasi kadar asam urat dan derajat keganasan kanker kolorektal. Subjek penelitian 35 orang penderita kanker kolorektal yang dirawat di RSUP Mohammad Hoesin (RSMH), terdiri atas 15 laki-laki dan 20 perempuan. Stadium kanker kolorektal berdasarkan pemeriksaan histopatologi di Laboratorium Patologi Anatomi RSMH/FK Unsri Palembang. Pemeriksaan kadar asam urat serum dilakukan di Laboratorium Biokimia FK Unsri. Hasil: Stadium klinis penderita terdiri dari stadium II (17,1%), stadium III (48,6%), dan stadium IV (34,3%). Rata-rata kadar asam urat penderita adalah 11,8±3,5 mg/dL, dengan 88,6%-nya mengalami hiperurisemia. Kadar asam urat pada pasien kanker kolorektal stadium awal adalah 10,6 mg/dL, pada pasien kanker kolorektal stadium akhir adalah 12,3 mg/dL. Korelasi antara kadar asam urat dan stadium kanker kolorektal sangat lemah (r=0,072) dan tidak bermakna (p=0,647). Simpulan: Hiperurisemia terjadi pada hampir semua penderita kanker kolorektal. Kadar asam urat tidak berkorelasi dengan stadium kanker kolorektal. Introduction: Hyperuricemia in cancer patients increases mortality, especially in advanced phase. Methods: An observational analytic correlative study with cross-sectional design on correlation between uric acid level and cancer stage in colorectal cancer patients. Subjects were 35 colorectal cancer patient, which are 15 male and 20 female colorectal cancer patients treated at RSUP Mohammad Hoesin (RSMH). Stage of colorectal cancer based on histopathology examination in Anatomy Pathology Laboratory of RSMH/Medical Faculty of Sriwijaya University, Palembang. Serum uric acid level was measured in Biochemistry Laboratory of Medical Faculty of Sriwijaya University. Results: The cancer stage of the patients were stage II (17.1%), stage III (48.6%), and stage IV(34.3%). The mean uric acid levels were 11.8±3.5 mg/dL. Approximately 88.6% of colorectal cancer patients have hyperuricemia. The mean level of uric acid in patients with early stage colorectal cancer was 10.6 mg/dL, in patients with end-stage colorectal cancer was 12.3 mg/dL. The correlation between uric acid levels and colorectal cancer stage was very weak (r=0.072) and was not significant (p=0.647). Conclusion: Hyperuricemia occurs in almost all colorectal cancer patients. Uric acid level did not correlate with colorectal cancer stage.

Abstract

Siswa sekolah dasar (SD) laki-laki prapubertas berada pada fase perkembangan yang penting, di mana mereka mulai mengalami perubahan fisik, emosional, dan psikologis yang signifikan. Untuk membantu siswa SD laki-laki prapubertas memahami dan mengelola perubahan yang terjadi dalam tubuh mereka, pendekatan yang dapat digunakan adalah dengan memberikan pengajaran tentang pendidikan seks dan hormon seks. Penerapan case-based method dapat membantu siswa memahami perubahan fisik dan emosional yang terjadi pada masa prapubertas secara konkret. Kegiatan pengabdian ini diikuti oleh 43 siswa laki-laki dari kelas empat, kelas lima, dan kelas enam. Sebagian besar siswa berumur 11 tahun. Pembicara mengambil tema “Saleh di Masa Pubertas” dengan tiga subtopik yakni tahapan proses penciptaan manusia, proses pertumbuhan dan perkembangan manusia, dan masa dan tanda pubertas pada laki-laki. Pada pengabdian ini, didapatkan ada dua anak (4,7%) yang sudah mengalami pubertas. Sebagian besar siswa dapat memahami topik dengan baik (55,8%). Kegiatan pengabdian kepada masyarakat dilaksanakan dengan baik. Siswa senang dengan metode berbasis kasus yang digunakan pada kegiatan ini. Mereka berharap topik-topik edukasi dapat diberikan dengan metode tersebut. Peran utama dari penerapan metode ini adalah untuk mendorong siswa berpikir kritis, memecahkan masalah, dan memahami konsep-konsep yang kompleks melalui penerapan pengetahuan dalam situasi nyata.

Abstract

Drug-resistant tuberculosis (DR-TB) is difficult to eradicate due to several factors, including insufficient treatment and medication side effects. Linezolid is among the medications with serious adverse effects. Linezolid toxicity is suspected to be related to the drug's binding to mitochondrial 16s rRNA. Some studies indicate that polymorphisms in patients' mtDNA may increase vulnerability to the development of toxic optic neuropathy. This study aims to identify a genetic influence on the vulnerability to the occurrence of toxic optic neuropathy side effects in drug-resistant tuberculosis patients receiving linezolid treatment. This research was conducted at the Faculty of Medicine, Universitas Sriwijaya, Palembang, from September to October 2023. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was undertaken to detect mutations in the G3010A mtDNA of the patients. Two patients experiencing toxic optic neuropathy side effects during drug-resistant tuberculosis treatment underwent blood sample collection. Blood samples were examined for mutations in the G3010A and A2706G genes using the PCR-RFLP method. The PCR-RFLP examination results indicated the presence of mutations in G3010A. In conclusion, polymorphism of G3010A mtDNA may contribute to the vulnerability to toxic optic neuropathy side effects in drug-resistant tuberculosis patients receiving linezolid. Further research with a larger population is needed to prove the involvement of mtDNA polymorphisms in the vulnerability to toxic optic neuropathy.

Abstract

Abstract: Exclusive breastfeeding is the best nutrition for babies to grow and develop. Currently, urban communities tend to stop breastfeeding at earlier age because of working mother. To overcome this, mothers can give expressed breast milk to babies by paying attention to the duration and temperature of storage, because it can affect the nutrients contained in breast milk. This study aims to determine the effect of the duration of storage of breast milk in the refrigerator on the protein contained in the breast milk. This research is a pure experimental research study with one group pre-test and post-test research design. The research was conducted at the Basic Medical Chemistry Laboratory, Faculty of Medicine, Universitas Sriwijaya. The number of expressed breastmilk samples in this study was 30 samples. Samples were divided into 5 groups, with variations in the duration of storage in the refrigerator (4-8°C) for 0 hour (control), 24 hours, 72 hours, 120 hours, and 192 hours. Protein level was measured by using Lowry method. Data were analyzed by repeated anova and paired t-test. The average protein content of ASIP at 0, 24, 72, 120, and 192 hours, respectively, was 2.93, 2.64, 2.61, 2.35, and 2.14 mg/dL (p=0,000 for repeated anova). The p values of the paired t-test storage durations of 24, 72, 120, and 192 hours, respectively, were 0.231, 0.137, 0.047 and 0.001. The duration of storage in the refrigerator that affects the protein levels of expressed breatmilk is 120 and 192 hours. Expressed breast milk protein levels significantly decrease after being stored in the refrigerator for 120 hours.Abstrak: ASI eksklusif adalah nutrisi terbaik bagi bayi untuk tumbuh dan berkembang. Saat ini masyarakat perkotaan cenderung menghentikan pemberian ASI pada usia yang lebih dini karena ibu bekerja. Untuk mengatasinya, ibu dapat memberikan ASI Perah (ASIP) pada bayi dengan memperhatikan durasi dan suhu penyimpanannya karena dapat mempengaruhi nutrisi yang dikandung ASI. Penelitian ini bertujuan untuk mengetahui efek durasi penyimpanan di lemari pendingin terhadap kadar protein ASI perah. Studi ini merupakan penelitian eksperimental murni (true ekseperimental) dengan desain penelitian one group pretest and posttest. Pemeriksaan kadar protein dilakukan di Laboratorium Kimia Dasar Kedokteran Fakultas Kedokteran Universitas Sriwijaya, dengan total sampel 30 sampel ASIP. Sampel dibagi menjadi 5 kelompok, dengan variasi durasi penyimpanan dalam lemari pendingin (4-8°C) selama 0 jam (kontrol), 24 jam, 72 jam, 120 jam, 192 jam. Uji kadar protein dengan metode Lowry. Data dianalisis dengan repeated annova dan paired t-test. Rata-rata kadar protein ASIP secara berturut-turut pada 0, 24, 72, 120, dan 192 jam adalah 2,93, 2,64, 2,61, 2,35, dan 2,14 mg/dL dengan hasil uji repeated anova p=0,000. Nilai p dari paired t-test masing-masing durasi penyimpanan 24, 72, 120, dan 192 jam adalah 0,231, 0,137, 0,047 dan 0,001. Lama penyimpanan dalam lemari es yang mempengaruhi kadar protein ASI perah adalah 120 dan 192 jam. Kadar protein ASI perah mengalami penurunan signifikan setelah disimpan dalam lemari pendingin selama 120 jam.

Abstract

ABSTRACT. Folate binding protein (FBP) is a protein in breast milk that plays a role in the regulation and bioavailability of folic acid. In contrast to cow's milk FBP, information about breast milk FBP is still limited. This research aims to determine the isolation and purification methods of breast milk FBP and the molecular weight of breast milk FBP. The sample in this study was 1000 mL of breast milk. Breast milk was prepared in several stages to yield whey. Isolation and purification of FBP from whey were carried out in stages, salting-out, ion exchange chromatography, and affinity chromatography. Whey salting-out with 95% saturation of ammonium sulfate could precipitate folate-binding proteins. This precipitate showed three peaks on DEAE chromatography. Peak II DEAE 95% was thought to be a negatively charged folate-binding protein. Peak II DEAE 95% also showed the presence of two peaks on affinity chromatography. It was believed that Peak II AF 95% was a pure folate-binding protein. Peak II AF 95% showed the presence of a single band on SDS-PAGE and western blot. This indicated that the folate-binding protein was 100% pure. FBP can be isolated from breast milk by the salting-out method using 95% ammonium sulfate, DEAE chromatography, and affinity chromatography. FBP from breast milk has a molecular weight of approximately 37 kDa. The final level of FBP isolated from breast milk is approximately 0.022 mg/mL. The successful isolation of FBP from breast milk provides an opportunity to use it to understand the clinical role of FBP in increasing folic acid levels in both breast milk and infant serum, as well as to develop methods for determining folic acid levels in these fluids. Abstract

Salah satu komponen makronutrien ASI ialah protein yang sangat bermanfaat dalam pertumbuhan dan perkembangan bayi. Faktor ibu, bayi, dan fisiologis memengaruhi kadar protein dalam ASI. Salah satu faktor ibu, yaitu indeks massa tubuh (IMT). Penelitian ini dilakukan dengan tujuan untuk mengetahui pengaruh IMT terhadap kadar protein pada ASI perah. Desain penelitian ini adalah analitik observasional dengan pendekatan cross sectional yang dilakukan di Laboratorium Kimia Dasar Kedokteran, Fakultas Kedokteran, Universitas Sriwijaya. Penelitian ini menggunakan 63 sampel ASI perah dari ibu menyusui di kota Palembang berusia 20-35 tahun dengan bayi berusia 1-6 bulan. Sampel ASI perah berjumlah 63 tersebut terbagi menjadi 2 kategori, yaitu 30 sampel ASI ibu dengan IMT normal dan 33 sampel ASI ibu dengan IMT gemuk dan obesitas. Pengukuran protein menggunakan metode Lowry dan data dianalisis dengan uji Mann-Whitney. Rata-rata kadar protein ASI perah pada ibu dengan IMT normal yaitu 6,35±5,78 mg/mL dan rata-rata kadar protein ASI perah pada ibu dengan IMT gemuk dan obesitas yaitu 8,64±3,70 mg/mL. Uji Mann-Whitney menunjukkan nilai p=0,03 (p< 0,05). Pada ibu dengan IMT gemuk dan obesitas, peningkatan porsi dan frekuensi makan, meningkatkan asupan asam amino sehingga meningkatkan kadar protein dalam ASI perah. Kesimpulannya, IMT ibu berpengaruh terhadap kadar protein pada ASI perah.

Abstract

Abstract:Thalassemia is a genetic disease that is inherited through marriage. At present, the incidence of thalassemia can only be reduced by reducing the birth of thalassemia children. Giving a correct understanding to the community is the right way to prevent the increase in new thalassemia sufferers. This community service activity is carried out in Lubuklinggau City, South Sumatra by involving lecturers who focus on the prevention of thalassemia. Activities carried out in the form of counseling and demonstration about thalassemia to 56 participants. In this activity, the demonstration was carried out using tools such as brochures / leaflets. An evaluation of the success of the activity is carried out using random questions to the participants before and after the activity. After counseling, most participants understood the simple way to recognize and prevent thalassemia. From these community service activities, it can be concluded that counseling and demonstration, can be used to improve people's knowledge and simple skills in recognizing and preventing thalassemia. Keywords:  Counseling, prevent, recognize, thalassemia Abstrak: Thalassemia merupakan penyakit genetik yang diturunkan melalui perkawinan. Saat ini, angka kejadian penyakit thalassemia hanya dapat turunkan dengan cara mengurangi kelahiran anak thalassemia. Pemberian pemahaman yang benar kepada masyarakat merupakan cara yang tepat guna mencegah peningkatan penderita thalassemia baru. Kegiatan pengabdian masyarakat ini dilakukan di Kota Lubuklinggau, Sumatera Selatan dengan melibatkan dosen yang fokus pada usaha pencegahan thalassemia. Kegiatan yang dilakukan berupa penyuluhan dan demonstrasi tentang thalassemia terhadap 56 orang peserta. Dalam kegiatan ini, demontrasi dilakukan dengan menggunakan alat bantu berupa brosur/leaflet. Evaluasi terhadap keberhasilan kegiatan dilakukan dengan menggunakan pertanyaan acak kepada peserta sebelum dan sesudah kegiatan. Setelah penyuluhan, sebagian besar peserta memahami cara sederhana mengenali dan mencegah thalassemia. Dari kegiatan pengabdian masyarakat ini, dapat simpulkan bahwa penyuluhan dan demonstrasi, dapat digunakan untuk meningkatkan pengetahuan dan keterampilan sederhana masyarakat dalam mengenali dan mencegah thalassemia.
