Abstract

Obat Pahit is a potion that has been long commonly consumed by Lingga Malay society for generations as stamina keeper. The most dominant plants found in the packaging of the Obat Pahit were namely Bauhunia semibifida, Cnestis palala and Penawa Root (3 species). This research aimed to investigate and determine activity of antioxidant contents in Obat Pahit from five Traditional Medicine Practitioners (TMPs) in the district of Lingga. The tested samples were mashed then being soaked into 2 types of solvent: distilled water and methanol, containing HCl 1%. DPPH method was also used in this research. Quantitatively antioxidant activity test of Obat Pahit from the five TMPs by using methanol solvent had extremely highest activity compared to the distilled water solvent. The test, using TLC plate by spraying the extract from three dominant plants with 0.1 mM of DPPH solution, produced a pale-yellow spots at a wavelength of 366 nm. On the other hand, the test using HPLC at wavelengths of 230 nm and 280 nm showed the presence of two dominant secondary metabolites contents: flavonoid and phenolic. IC50 (ppm) of Bauhinia semibifida (6.6247), Penawa Root (5.0124) and Cnestis palala (5.9968) were much lower than IC50 of mangosteen’s rind (41.7675), vitamin C (6.6612) and Stimuno drug (8.333). This antioxidant analysis has not been reported previously. This proof contributed greatly to uncovering potentially native natural resources as an indigenous Indonesian drug which is expected to decrease dependence on imported drugs especially imunomodulator, antihypertensive, antidiabet etc. This research would be beneficial and excellent manifestation for the development of natural antioxidant-based medicines from traditional knowledge of Indonesia’s local ethnicities.

Abstract

Depression is a psychological disorder caused by an imbalance of neurotransmitters that can be managed with antidepressants. One of the local plants with the potential as an antidepressant is areca nut. However, its’ antidepressant effects in nonpolar solvents have not been studied recently. Equipping the extract to animals sub-chronically could mimic the clinical antidepressant treatment. Thus, the current investigation studied the antidepressant action of the hexane extract of old areca nut by utilizing acute and sub-chronic FST. This research started with extracted areca nut using n-hexane and continued with phytochemical tests. During FST, the animals were treated with n-hexane extract (50 and 100 mg/kg), fluoxetine (20 mg/kg), and saline (0.1 mL/20 g). The phytochemical test of the extract showed positive results from the content of secondary metabolites, namely saponins and steroids. n-Hexane extract at a dose of 50 mg/kg gave the best action in decreasing the immobility period. After sub-chronic medication, the secondary metabolites of the extract did not induce any toxic effects. This study’s findings imply that depression may be treated with conventional medicine, such as old areca nut extract.

Abstract

Depression is a common neuropsychiatric disorder that occurs in humans regardless of gender and age. Areca catechu, as a local plant, can be utilized as an antidepressant to treat depression. The current investigation aimed to determine the antidepressant action from an n-hexane extract of young areca nut using the Forced Swim Test (FST) as a standard method in screening antidepressants validated by the behavior of animals. Short-term medication or acute FST was performed by injecting mice with extract at doses 10 and 50 mg/kg, while the mice were treated with the best dose in long-term medication or sub-chronic FST. A day after the re-test on sub-chronic FST, toxicity test was performed. The extract at the dose of 50 mg/kg possessed antidepressant action during acute and sub-chronic FST. The results of the phytochemical analysis stated that the saponins and steroids present in the young areca nut n-hexane extract may be the active constituents that act as antidepressants. No toxic effects were detected from the young areca nut n-hexane extract in mice that had been injected for 7 consecutive days. n-Hexane extract from young areca nut presents antidepressant potential.
