Abstract

Background: Bagendit Lake is a tourist lake located in Garut (West Java) and has experienced a decline in water quality. This research was conducted in settlements around Bagendit Lake with the aim of studying human activities and the level of concern for the existence of Bagendit Lake. Method: Mix method (Quantitative and Qualitative) was used in this study to collect data about the activities, knowledge education and environmental ethics of the community. The data collection technique was carried out by purposive sampling involving 40 respondents aged between 18-60 years. Data were obtained by conducting field observations, interviews, questionnaires and document recording. Findings: The results showed that the community received environmental knowledge education from schools (54%) and parents (46%). Surprisingly, as many as 93% of respondents stated that they played an active role in preserving the Bagendit Lake. However, various   activities (such as tourism, fisheries, agriculture and settlements) have resulted in environmental damage to Bagendit Lake. It can be concluded that the level of knowledge and public awareness about the impact of activities on the environment has not been maximized. It is necessary to support the local government's role in implementing environmental improvement and conservation.

Abstract

Situ Bagendit merupakan danau yang dimanfaatkan sebagai objek wisata alam sekaligus kawasan lindung. Revitalisasi kawasan wisata ini berpotensi memengaruhi komunitas tumbuhan air yang berperan penting dalam menjaga stabilitas ekosistem dan berfungsi sebagai indikator pencemaran air. Penelitian ini bertujuan untuk mengevaluasi kualitas air dan indeks pencemaran menggunakan indikator tumbuhan air pasca-revitalisasi. Data dikumpulkan melalui metode purposive sampling pada lima stasiun yang mewakili berbagai tingkat aktivitas manusia. Parameter yang dikaji meliputi data biologi (komposisi tumbuhan air), serta parameter fisika dan kimia perairan. Analisis keanekaragaman tumbuhan dilakukan menggunakan Indeks Dominansi (C), Indeks Keanekaragaman Shannon-Wiener (H’), dan Indeks Kemerataan (E). Ditemukan 10 jenis tumbuhan air, dengan empat spesies dominan yang berfungsi sebagai bioindikator pencemaran, yaitu Eichhornia crassipes (Eceng Gondok), Salvinia molesta (Kiambang), Nelumbo nucifera (Teratai), dan Ceratophyllum demersum (Hornwort). Nilai rata-rata keanekaragaman tumbuhan berada pada kategori sedang (H’ = 1,11), dengan tingkat kemerataan komunitas juga sedang (E = 0,481). Berdasarkan parameter nitrogen total, kecerahan, kadar klorofil-a, serta jenis tumbuhan air yang ditemukan, status trofik Danau Situ Bagendit dikategorikan sebagai eutrofik hingga hipertrofik. Hasil ini menunjukkan bahwa revitalisasi kawasan dapat berdampak terhadap kondisi ekologis perairan yang perlu dimonitor secara berkelanjutan.

Abstract

This study aims to optimize the production of cellulase enzyme by the cellulolytic fungus Aspergillus niger using rice husk powder (Oryza sativa Linn.) as substrate. Cellulase enzymes have important roles in various industrial applications, including the bioconversion of lignocellulosic waste into high-value products such as bioethanol, pulp, paper, and animal feed. The utilization of rice husk as a substrate offers economic and environmental advantages due to its abundant availability, low cost, and ability to reduce the volume of agricultural waste. The research method used a quantitative experimental approach to evaluate the effect of fermentation parameters, such as pH, temperature, and incubation time, on enzyme production. A. niger isolates were developed on carboxymethylcellulose (CMC) media to ascertain their cellulolytic capacity, with measurements of biomass, pH, temperature, and enzyme activity. The results showed that the biomass of A. niger increased from hour 0 to hour 96 at pH 4.5 to 5.5, and temperature 29.5°C to 30.5°C. The optimum condition for enzyme production was found at pH 4.5 and temperature 30.5°C, with the highest enzyme activity reaching 0.736 U/mL. These findings suggest that optimization of cellulase enzyme production from rice husk not only improves enzyme production efficiency, but also contributes to more sustainable waste management.

Abstract

Penelitian ini bertujuan untuk menganalisis pengembangan keterampilan kolaborasi mahasiswa Pendidikan IPA melalui penerapan strategi BuPAcE berbasis pendekatan Citizen Science project dalam mata kuliah Mitigasi Bencana. Strategi BuPAcE (Building Community, Project Expedition, Action, Evaluate) dikembangkan untuk menciptakan pembelajaran berbasis proyek yang kontekstual, kolaboratif, dan berpusat pada mahasiswa. Metode penelitian yang digunakan adalah deskriptif kuantitatif, dengan subjek sebanyak 27 mahasiswa. Instrumen penelitian berupa angket yang menilai keterampilan kolaborasi berdasarkan aspek interpersonal dan intrapersonal. Hasil penelitian menunjukkan bahwa mayoritas mahasiswa (74%) berada dalam kategori keterampilan kolaborasi sangat kuat, dengan kontribusi tertinggi ditunjukkan pada indikator kontribusi tim dan dukungan terhadap kelompok. Analisis korelasi Pearson menunjukkan hubungan yang sangat kuat dan signifikan antara keterampilan interpersonal dan intrapersonal (r = 0.981, p < 0.001). Selain itu, analisis dampak menunjukkan bahwa setiap fase BuPAcE berkontribusi terhadap penguatan kolaborasi. Pendekatan Citizen Science project memperkuat keterlibatan mahasiswa dalam konteks sosial nyata, memungkinkan mereka membangun solusi berbasis data dan pengalaman lapangan. Temuan ini menegaskan bahwa pembelajaran berbasis BuPAcE dan Citizen Science project mampu membentuk pengalaman belajar yang bermakna dan transformatif, sekaligus memperkuat keterampilan abad ke-21 terutama keterampilan kolaborasi mahasiswa calon guru IPA.

Abstract

Corncob is an agricultural waste rich in cellulose, making it a promising alternative substrate for cellulase enzyme production, which is widely used in various industrial applications such as bioenergy, food, and textile industries. Aspergillus niger is a well-known cellulolytic fungus capable of efficiently producing cellulase enzymes. This study aimed to optimize cellulase production by A. niger using corncob powder (Zea mays) as a substrate through submerged fermentation (SmF), with variations in temperature (29.5°C and 30.5°C) and pH (4.5 and 5.5). The cellulolytic activity of A. niger was tested on CMC agar medium. Cellulase activity was analyzed based on the concentration of reducing sugars using the DNS method. The highest enzymatic activity was recorded at 1.125 IU/mL with a biomass yield of 0.754 mg/mL at 30.5°C and pH 4.5. These findings suggest that optimizing environmental conditions and utilizing corncob waste as a carbon source can enhance cellulase production by A. niger, offering a sustainable and cost-effective approach for enzyme-based industries.

Abstract

Ecoenzymes are fermentation products from organic waste in the form of vegetables or fruit. Many people produce organic waste in households every day, including residents in Cilingga village, Purwakarta Regency. The aim of this community service activity (PkM) is to provide education in waste/waste management as additional information. Apart from that, it also provides knowledge and awareness of community groups in making ecoenzymes as a multi-functional product, which can overcome the problem of waste or organic waste, especially in households. The service method used is Participation Action Research (PAR) through certain stages. This PkM activity is carried out in several stages, namely discussion, socialization of activities and providing education through lectures and discussions, direct practice and filling out questionnaires. The results of this training in making ecoenzymes can help the community in maintaining environmental cleanliness, managing organic waste, both agricultural and livestock waste, as well as household waste. With this training, villagers can utilize their own organic waste into ecoenzymes. Apart from reducing organic waste entering the environment, this training can also create additional sources of income and increase awareness of the importance of maintaining environmental sustainability.

Abstract

Physalis angulata and Moringa oleifera have long been the key ingredients of traditional medicine. This study aimed to obtain the metabolite profiles of the ethanolic leaf extracts of P. angulata (ELP) and M. oleifera (ELM), evaluate their antioxidant activity, and assess their toxicity against normal and cancer cells. The leaves were sourced from Balittro, West Java. The extraction used 70% ethanol and continued for 3 days. The metabolites were identified using Gas Chromatography-Mass Spectrometry (GC-MS), antioxidant activity was ascertained via the DPPH assay, and cytotoxicity was evaluated employing the PrestoBlue assay. GC-MS identified 23 compounds in ELP and 15 in ELM. The DPPH assay showed that ELM, ELP, or their combination exhibited marked antioxidant activity, with inhibition ranging from 50% to 90%. The toxicity assay against MCF-7 cancer cells demonstrated that ELP at 1,500 µg/mL and ELM at 1,200 µg/mL induced cytotoxicity. The ELP + ELM formulation exhibited stronger cytotoxic effects than the positive control. In contrast, ELM or ELP did not induce toxicity in normal CV-1 cells and, stimulated viability com-pared to the control. Thus, the phytochemical contents of ELM and ELP exhibited enhanced antioxidant activity, in-duced toxicity in cancer cells, and were safe for normal cells.

Abstract

The success of learning process can be seen from students' achievements and critical thinking skills. One approach that can be used to improve learning achievement and CTS is STEM. Many researchers have conducted research on STEM, but have not been specific about the aspects that play a role on it. This research aims to determine the effect of STEM from the facility aspect, teacher aspect and environmental aspect on learning achievement and CTS. This survey research used purposive sampling involving 152 students. The research instrument consists of a questionnaire to see students' perceptions of STEM from three aspects and a test instrument to determine students' learning achievement and CTS. Data analysis was carried out using partial least square. The research results show that (1) the facility aspect has the greatest influence on learning achievement; (2) environmental aspects have the greatest influence on CTS; (3) the three aspects of STEM learning have a big influence on learning achievement and CTS; and (4) learning achievement greatly influences CTS. These results show that STEM is very feasible to be implemented in the classroom to improve learning achievement and CTS

Abstract

This study aims to describe a disaster mitigation lecture strategy based on the Citizen Science Project (CSP) through a combination of indoor and outdoor learning methods. The study employs an action research approach by implementing a lecture strategy consisting of four phases: BuPAcE (Building Community, Project Expedition, Action, and Evaluate). This study involved 22 fifth-semester students from the Science Education Study Program in the 2023-2024 academic year. The Building Community, Action, and Evaluate phases are conducted using indoor learning methods, while the Project Expedition phase is carried out using outdoor methods. The implementation of the BuPAcE strategy is evaluated using observation sheets. The results indicate that the implementation percentage of the disaster mitigation lecture strategy based on CSP is 80% for the Building Community phase, 70% for the Project Expedition phase, 80% for the Action phase, and 75% for the Evaluate phase. Based on these results, it can be concluded that the disaster mitigation lecture strategy using a combination of indoor and outdoor methods is effective for use in lectures.

Abstract

Sugarcane has been widely used to treat various diseases due to its metabolite content. Tissue culture is one method that can be used to produce metabolites and is an alternative biotechnology as a conservation strategy. This study aims to analyze the differences in metabolite content in sugarcane plantlets cultured with different media and subculture frequencies. Plantlets were extracted using the maceration method with 70% p.a. ethanol solvent. Metabolite analysis was carried out using a Gas Chromatography-Mass Spectrometry (GC-MS) instrument. Metabolite identification was carried out based on the NIST library. The results showed that the 10th subculture of sugarcane plantlets cultured on MS+Kinetin+Coconut Water media contained 5 metabolites, while the 11th subculture of plantlets cultured on MS+Kinetin+Tidiazuron media contained 7 metabolites. Differences in the type and number of metabolites indicate the influence of culture conditions on the metabolite profile of the plantlets. This study provides an initial overview of the potential of sugarcane tissue culture in the production of secondary metabolites.
