Abstract

The aim of this research was to study pasteurized milk quality produced by UD. Gading Mas on pH, Acidity, alcohol test and TPC during 5 days storage in refrigerator. On first day showed that  weight mass 1,06 (w/w); fat content 2,00 (g/100g),  protein content 3,02  (g/100g), Zn 1,67 ppm, Cu<0,005 ppm, Pb 0,02 ppm,  As  0,0120 ppm, Hg< 0,0002 ppm, Sn 1,60 ppm dan Cd < 0,001 ppm and organoleptic test included color, flavor and taste were normal. Pasteurized milk characteristic during 5 days refrigeration for pH were 6,57;6,58;6,73;6,60 and 6,50 respectively. Acidity were 1,147 %, 0,145 %, 0,145%, 0,157 %, 0,156 % and 0,175  % respectively. TPC were 6,16 x 101, 1,2. 104; 3,15.104, 0,42.106 and 3,5.108 respectively. It concluded that pasteurized milk produced by UD Gading Mas fulfilled SNI quality standard based on weight mass, pH, acidity, fat, protein, heavy metal content, organoleptic test and TPC.   Keywords: pasteurized milk, quality, refrigerator

Abstract

ABSTRACT This research was conducted from January to February 2017 at Microbiology Laboratory, Faculty Science and Technology, Islamic Maulana Maliki Ibrahim University, Malang and at Microbiology Laboratory, Faculty of Science, Muhammadiyah University, Malang. The purpose of this research was to determine the effectiveness of Piper betle L. leave extract on antibacterial activity the growth of Streptococcus agalactiae. The method used was Complete Randomized Design (CRD) with 8 treatments and 6 replications. Piper betle L. leaves extract was used 20% for P0 (0-day), P1 (first day), P2 (second day), P3 (third day), P4 (fourth day), P5 (fiveth day), P6 (sixth days) and P7 (seventh day). The results showed that Piper betle L. leaves extract had a highly significant effect (P<0.01) on inhibiting the growth of the bacteria so the results were continued with LSD test analysys because there were differences among variables. The results showed that P0 was the highest (3.93±0.50); then followed by P1 (3.83±0.69); P2 (3.47±0.21); P3 (3.13±0.22); P4 (3.12±0.58); P5 (3.04±0.24); P6 (3.00±0.44) include at medium categories, and P7 (2.97±0.19) include at low category, where inhibition zone which formed on each treatment have non significant decreased. In conclusion, the storage of Piper betle L. extract have effect on antibacterial activities (Streptococcus agalactiae), which the highest at day-0.

Abstract

This research was aimed to determine the antibacterial activity of mahkota dewa extract and the effective concentration of mahkota dewa extract against Streptococcus agalactiae. This research was conducted from 2Mayth to 2Juneth 2016. The material ware Streptococcus agalactiae bacteria which isolated from mastitis milk, mahkota dewa which extracted using Aquades and ethanol. The method design was experiment research using Nested Design with 6 treatments and 5 replications. The concentrations of the treatments were P1 (10%), P2 (20%), P3 (30%), P4 (40%) and P5 (50%) and positive control P0 (iodips 10%). The collected data were analyzed using the analysis of variance indicated the significant effects, the least significant different test (LSD) was then employed. Mahkota dewa extract using ethanol (50%) result showed that significantly (P<0.01) on growth of Streptococcus agalactiae was 20.34± 0.92mm. Mahkota dewa extract using aquades (50%) result showed that significantly (P<0.01) on growth of Streptococcus agalactiae was 10.0 ± 0.50 mm. The conclusion of this research is Mahkota dewa fruits extract with ethanol had inhibitory higher in habiting the Streptococcus agalactiae bacteria compared to aquades solvent. The best of treatment of Mahkota dewa fruits extract with ethanol and Aquades solvent in concentration 50% had a high ability to inhibit the growth of Streptococcus agalactiae.

Abstract

The study aimed to evaluate the production performance of male Limousine Crossbreed cattle with different feed levels with the addition of RD flour. It used nine male Limousine crossbreed cattle, aged <1-3 years, with an average initial weight of 335±29,09kg. The method used is an experimental method using CRD with 3x3: T0= Ad libitum corn straw + concentrate without RD powder, T1= Ad libitum corn straw + concentrate containing 7,5% RD, T2= Ad libitum corn straw + concentrate containing 15% RD flour. The variables measured included Daily gain, Average daily gain, feed conversion ratio, and Income over feed cost. Data were analyzed by ANCOVA. Cattle were fed for eight weeks from (March 26 to June 8, 2021), in the Livestock Group of Kucur Village, Dau District, Malang Regency. The result showed that using feed with the addition of RD at different levels of male Limousine crossbreed cattle had a significant effect on FCR (P>0,05) and on Daily gain, Average daily gain, and IOFC (P<0,01) had a different significant effect. This study concluded that the treatment of T1 feed (7.5% "RD" flour) showed the best results for Daily gain (70,67±5,43 kg), Average daily gain (1.26±0.09 kg), FCR (5.85±0.19),, IOFC Rp. 36586.50.

Abstract

GDFP is a standard guideline for producing good milk quality and efficient production system. GDFP implementation has been less attention among the dairy smallholder farmers. The smallholder farmers are classified into three categories depending on animal ownership; strata 1 (1-3 AU), strata 2 (4-6 AU), and strata 3 (>7 AU). This research determines the score of GDFP implementation on smallholder farmers in East Java. The total respondents were 56, with 325 dairy cattle. A questionnaire and field observations were used to collect data using the survey method. The farmers were interviewed using a prepared questionnaire with general information about their farms and questions about GDFP implementation. The result showed that the score of GDFP implementation in all strata of smallholder farmers in East Java Indonesia in 6 aspects as follows: animal health was  “good enough” (score  2.68-2.70), milking hygiene was “good” (score 3.19-3.42), nutrition was “good enough” (score 2.86-2.97) in strata I and III and “good” for strata II (score 3.03), animal welfare was “good enough” (2.56-2.60), the environment was "good enough" (2.34-2.50) and socio-economic management is “not good” (score GDFP = 1.60-1.92).

Abstract

Research with aim to estimate genetic polymorphism at ACACA (Acetyl-coenzyme A carboxylase ) locus in Ettawa Crossbred goat wan its relationship with production traits was done at goat population in Batu, Lawang and Ampel Gading. 46 female goats were taken it’s blood sample to isolate the DNA and continue with PCR (Polymerase Chain Reaction) and RFLP (Restricted Fragment Length Polymorphism). PCR was used to amplify ACACA gene fragment in intron 3’ about 200 bp with primer F : 5’ – AGT GTA GAA GGG ACA GCC CAG C – 3’ and R : 5’ – GTG GAA TGA CAC ATG GAG AGG G – 3’; RFLP was used to test mutation of that fragment in particular place (point) using restriction enzyme RSA1. Variables were alelles and genotypes composition in population, milk and fat content, and birth weight of kid. Result showed that (a) genetic polymorphism at locus ACACA in three location was high that is 44,22 %, with allele frequency of G (p) = 33 % and allele T (q) = 67 %; (b) no relationship between the high polymorphism with productive performance of goat in fat and protein content, and birth weight of kid. It was concluded that in goat population there was a high polymorphism at ACACA gene, and that polymorphism was not related to production.

Abstract

A total of 106 late gestation goats (age 4-5 months of pregnancy) records each of Etawah Crosbred goats kept on small farmer over a period of 10 years were assessed to determine the effects of environmental factors on them. Body Condition Score is non-genetic factor and in dairy goats to predicted milk production, fertility, and general health of the animal because BCS is visualisasi from feeding manajement. The objective of this study was to investigated the relationship between Body Condition Score (BCS) of late gestation with litter size and birth weight on etawah crossbred goats. The results showed that the average of litter size and birth weight (kg) were 1.89±0.66 and 3.84±0.73 respectively. The relationship between BCS with litter size were 0.13 (very low) and the relationship between BCS with birth weight were 0.11 (very low). The conclusion of this research is BCS of late gestation had positive correlation on litter size and birth weight, however this was low and birth weight as first indicator of future growth rate.

Abstract

Although Etawah Crossbred (PE) goat is considered to be dual purpose (meat and milk) goat, it is mainly raised for meat production. Since early 1990, there has been a growing interest of the farmer in some places to raise PE goat for milk production without sacrificing its role to produce kids for meat. Although milk yield of PE goat was not as high as milk yield of some other dairy goats, the ability of PE goat to cope with harsh local environment, particularly climate and feed conditions, was an advantage. Therefore, raising PE goat would still be an important part of farmer activities in the rural areas in Indonesia. Identification of the genes underlying livestock production traits leads to more efficient breeding programs and it is a promising way to improve production traits of farm animals. Growth hormone is a polypeptide hormone which is the major regulator of the metabolic procedures of growth and development and it is encoded by GH gene. The objective of this study was to detect the genetic polymorphism of GH gene in major Etawah Crossbred (PE) goat using PCR-RFLP. The PCR amplified fragment were digested with HaeIII endonuclease and the result showed the presence of two genotype CC and CD. The total frequency were 47.0% and 53.0% for CC and CD genotype respectively in 94 tested goats. Statistical analysis showed that in the fragment amplified by the pair of  primer, CD genotype had significant higher birth weight and weight of 100 days old (weaning weight) than CC genotype (P<0.01). In conclusion that GH gene may be a mayor gene or linked to the mayor gene to affect the weight traits and the polymorphic site could be used to select the goat weight in marker-assisted selection program.

Abstract

The purpose of this research was determined effect of inhibitory the Phaleria macrocarpa  L. fruits extract with ethanol and aquades solvents againts Staphylococcus aureus. Materials used was Staphylococcus aureus which isolated from mastitis milk. Phaleria macrocarpa  L. fruits powder were extracted using ethanol and aquades with concentration were 10%, 20%, 30% and 40%. Iodips was used as control. Inhibitory of bacteria effect test was done by well diffusion methods. Variable was inhibition zone of each concentration, both of ethanol and aquades solvent. Data was analyzed by using two way nested ANOVA and continued by Duncan Multiple Range Test (DMRT). Result showed that highly significantly (P<0.01) on inhibition zone of Staphylococcus aures. Diameters of inhibitory was the optimum inhibition with ethanol solvent (17.46±0.67)mm and aquades solvent (11.14±0.30) mm. The best of  treatment of  Phaleria macrocarpa  L. fruits extract againts Staphylococcus aureus with ethanol and aquades solvent was 40%. The conclusion of this research is that mahkota dewa (Phaleria macrocarpa  L.) fruits extract with ethanol and aquades solvent in concentration 40% had a high ability to inhibit the growth of Staphylococcus aureus. Phaleria macrocarpa  L. fruits extract with ethanol higher in inhibiting capability the Staphylococcus aureus bacteria compared to aquades solvent.

Abstract

Penelitian ini bertujuan untuk mengetahui efek steaming up selama kebuntingan tua terhadap profil protein darah induk kambing Peranakan Etawah. 12 ekor induk kambing dipilih berdasarkan periode laktasi kedua dan ketiga. Perlakuan pertama sebagai kontrol (P0), perlakuan kedua dan ketiga adalah steaming up menggunakan Gliricidia sepium 0,4% BK (P1) dan 0,8% BK (P2). Seluruh perlakuan diberikan pakan basal konsentrat 0,9% BK dan Pennisetum purpureum secara ad-libitum. Data dianalisis menggunakan Anova dengan Rancangan Acak Kelompok. Data profil protein darah induk kambing PE bunting tua dianalisis secara deskriptif eksploratif. Perlakuan Steaming up memberikan perbedaan yang nyata pada konsumsi BK, BO, dan PK (P<0,05). Praperlakuan steaming up menghasilkan 18 pita protein dengan berat molekul 15-158 kDa. Pasca perlakuan P0 menghasilkan 18 pita protein dengan berat molekul 15-158 kDa, P1 menghasilkan 19 pita protein dengan berat molekul 15-158 kDa. P2 menghasilkan 22 pita protein dengan berat molekul 15-158 kDa. Kesimpulan dari penelitian yaitu steaming up Gliricidia sepium sebesar 0,8% menghasilkan 22 pita protein dengan berat molekul yang berbeda. Banyaknya pita yang muncul dikaitkan dengan keberhasilan perlakuan steaming up pada kebuntingan tua.
