Abstract

ABSTRAKTeknik komputasi virtual screening dapat digunakan untuk memprediksi dan menjelaskan aktivitas senyawa apa yang berperan dalam penghambatan DPP-4. Lima tanaman Indonesia seperti Caesalpinia sappan, Mutingia calabura, Rheum palmatum, Ficus religiosa dan Tinospora crispa telah terbukti memiliki aktivitas penghambat dipeptidyl-peptidase-4 (DPP-4) secara in-vitro akan tetapi belum ada penjelasan senyawa apa yang berperan terhadap aktivitas tersebut. Penelitian ini bertujuan untuk memprediksi dan menjelaskan senyawa-senyawa dari lima tanaman yang memiliki aktivitas penghambatan DPP-4 secara In-silico. Metode docking menggunakan perangkat lunak Molegro Virtual Docker dengan 16 protokol kombinasi pencarian algortima dan fungsi nilai. Sebelum Docking dilakukan validasi metode dengan cara redocking native ligand protein penghambat DPP-4 dengan kode PDB 3G0B, kemudian dilanjutkan docking secara bersamaan 151 senyawa dari lima tanaman Indonesia dan 1 ligan standar alogliptin dengan reseptor dipeptidy lpeptidase-4 (kode PDB: 3G0B) dengan metode docking yang valid. Hasil validasi metode menunjukkan 16 protokol kombinasi memberikan nilai RMSD < 1 Å. Hasil docking berdasarkan rerank score paling rendah dan kemiripan interaksi residu asam amino dibandingkan ligan standar Alogliptin menunjukkan tiga senyawa 6'-O-Lactoyl Borapetoside B (rerank score -99.78 kcal/mol) dan Borapoteside A (rerank score -98.76 kcal/mol) dari tanaman Tinospora crispa dan Rhaponticin (rerank score -99.37) dari tanaman Rheum Palmatum  sedangkan ligan standar Alogliptin memiliki rerank score -93.14 kcal/mol dengan interaksi residu asam amino Glu206, Glu206, Tyr547, Tyr632. Metode docking dengan mvd terbukti valid dan Physcion 8-gentiobioside, Luteolin 4'-metil eter 7-glukosida, 6'-O-Lactoyl borapetoside B, Rhaponticin dan Tinocrispol A diprediksi memiliki aktivitas penghambatan DPP-4.Kata Kunci: docking, penghambat DPP-4, Tinospora crispa, Rheum palmatum

Abstract

The main bitter constituents of sambiloto (Androgaphis paniculata (Burm., F) Ness) are diterpene lactones, namely andrographolide and deoxyandrographolide which have been reported to have antidiabetic, cytotoxic, antiatherosclerosis, anti-inflammatory, and antimicrobial activity. There are many studies that performed the isolation of deoxyandrographolide and andrographolide from A. paniculata herbs, but most of them included several steps that make them not efficient. This research was conducted to do an isolation of deoxyandrographolide and andrographolide through liquid-liquid fractionation (LLF) due to its simplicity, low cost, and time efficient. The extraction of deoxyandrographolide and andrographolide from the herbs was carried out using chloroform as the solvent by using Soxhlet apparatus, and LLF was performed to isolate the compounds. The identities of the compounds were confirmed by TLC scanner compared to its standard references. Hence, these present methods were successfully isolated and determined deoxyandrographolide and andrographolide of A. paniculata. The compounds were also showed relatively moderate cytotoxicity on 3T3-L1 cell lines using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt (MTS) assay, with LC50 of deoxyandrographolide and andrographolide; 29.3173 µg/mL and 37.7011 µg/mL, respectively.

Abstract

Melanoma is the deadliest type of skin cancer, having a high mortality rate. This cancer has an aggressive nature, is highly invasive, and has the tendency to metastasize. Matrix metalloproteinases (MMPs) are essential in that process, especially MMP2 and MMP9. Their expression is upregulated during metastasis progression. Myristicin is one example of a compound that can be utilized to target MMP 2 and MMP 9 in melanoma. This research concerns the activity of myristicin to inhibit melanoma cell invasion and migration by suppressing MMP2 and MMP9 gene expression. The MTT assay in this study demonstrated that myristicin exhibited strong cytotoxic activity against melanoma cells. This compound works in a dose-dependent manner by inhibiting cell migration and invasion. The invasion test was performed using the transwell assay, whereas the migration test was performed using the wound healing assay. The invasion assay results were consistent with MMP2 and MMP9 gene expression. These two genes were analyzed using the RT-qPCR technique. It has been demonstrated that low gene expression in melanoma cells inhibits cell invasion. In contrast, higher MMP2 and MMP9 gene expression was associated with an increase in the number of invasive cells on average. However, MMP2 and MMP9 in excessive expression and uncontrolled activity impair the ability of melanoma cells to form a monolayer sheet to cover wound gaps. This condition significantly reduced the migration rate and percentage of wound closure.

Abstract

American Heart Association (AHA) mempublikasikan fenomena kesehatan baru yang disebut sindrom kardiovaskular-ginjal-metabolik (cardiovascular-kidney-metabolic/CKM syndrome). Terdapat beban berat dari kesehatan kardiovaskular-ginjal-metabolik yang buruk di masyarakat, yang memengaruhi hampir semua sistem organ dan memiliki dampak yang sangat kuat terhadap kejadian penyakit kardiovaskular. Sindrom CKM, yang dikenal juga sebagai sindrom ginjal kardiometabolik (cardiometabolic renal syndrome/CMR), melibatkan interkoneksi antara penyakit ginjal kronis (chronic kidney disease/CKD), faktor risiko metabolik seperti obesitas dan diabetes tipe 2 (type 2 diabetes/T2D), dan penyakit kardiovaskular (cardiovasular disease/CVD).

Abstract

Dalam dua tahun belakangan ini, kita telah mendapatkan banyak perubahan dalam bidang genomik, termasuk diagnosis berbasis pengurutan DNA yang sangat cepat untuk mendeteksi penyakit langka dan munculnya teknologi yang menjanjikan pengurutan (sequencing) seluruh genom seharga sekitar 1,5 juta rupiah ($100). Pengurutan DNA kemungkinan akan menjadi komoditas dalam waktu dekat karena kombinasi kecepatan dan biaya yang lebih rendah. Perkiraan menunjukkan bahwa penggunaan kecerdasan buatan (artificial intelligence/AI) pada proyek genomik akan menghasilkan hingga 40 exabyte data pada tahun 2030, atau lima hingga sepuluh miliar data. Satu exabyte terdiri dari sekitar satu triliun byte atau satu miliar gigabyte.

Abstract

Sirkulasi otak yang optimal sangat penting untuk memastikan bahwa otak menerima pasokan oksigen dan nutrisi yang cukup, yang merupakan dasar bagi semua aktivitas neurologis. Gangguan sirkulasi darah ke otak dapat menyebabkan berbagai kondisi serius, termasuk strok, demensia, dan gangguan kognitif lain. Penelitian terkini telah mengungkap berbagai aspek baru tentang bagaimana otak beradaptasi dan berfungsi dalam kondisi normal maupun patologis. Dengan meningkatnya prevalensi penyakit neurodegeneratif dan adanya fenomena penuaan populasi, penelitian tentang sirkulasi otak menjadi semakin mendesak. Temuan-temuan terbaru dalam bidang ini tidak hanya memberikan pemahaman yang lebih baik tentang bagaimana otak berfungsi, tetapi juga membuka jalan bagi pengembangan terapi baru yang dapat membantu menjaga kesehatan otak sepanjang hidup seseorang.

Abstract

Synthetic biology represents a groundbreaking frontier in biotechnological innovation, empowering researchers to design and construct genetic and protein networks with a level of precision previously unattainable. At the heart of this transformative field lies enzymatic DNA synthesis, a cutting-edge technology revolutionizing how genetic information is assembled and applied. By facilitating the rapid and accurate creation of DNA sequences, enzymatic DNA synthesis has significantly advanced our ability to engineer compact protein circuits, enabling precise regulation of protein expression and cellular communication. This review explores the multifaceted impact of enzymatic DNA synthesis, highlighting its pivotal role in addressing complex biological challenges. The technology's ability to support the construction of highly efficient and compact protein circuits has unlocked new possibilities in therapeutic applications, such as personalized medicine and gene therapy, as well as industrial innovations in biotechnology and synthetic manufacturing. Furthermore, the integration of enzymatic DNA synthesis with other synthetic biology tools has opened avenues for creating synthetic organisms and programmable cellular systems, paving the way for novel applications in agriculture, environmental sustainability, and biopharmaceuticals. Despite its transformative potential, enzymatic DNA synthesis is not without challenges. Issues related to scalability, cost-effectiveness, error rates in synthesis, and integration with existing biotechnological frameworks remain areas requiring further refinement. Emerging trends in automation, machine learning, and integration with high-throughput screening platforms, however, offer promising solutions to these hurdles. By synthesizing recent advancements, this review underscores the disruptive potential of enzymatic DNA synthesis in reshaping our understanding of cellular communication and protein regulation. As the technology continues to mature, its contributions are poised to redefine synthetic biology's role in solving pressing global challenges across medicine, industry, and beyond.

Abstract

Indonesia has great potential in the development of biowealth-based phytopharmaceutical products, considering its rich natural wealth and traditional medicine traditions. This study uses theories of legal utilitarianism, distributive justice, and legal positivism to explore the challenges facing Indonesia, as well as opportunities for more inclusive and adaptive reforms. This analysis employs a normative juridical approach, focusing on the analysis of written laws, regulations, doctrines and legal interpretations related to patent protection for phytopharmaceutic products. This approach enables a thorough understanding of the patent legal framework in Indonesia and its practical implementation. The case study method is used to evaluate several patents registered with the Directorate General of Intellectual Property (DJKI). The results show that Indonesia is facing a number of complex legal challenges in the patent registration process, ranging from barriers to novelty criteria and inventive steps to lengthy administrative processes. In addition, Indonesia needs to strengthen its national patent system and intellectual property protection strategies to ensure that local communities benefit fairly from the use of their traditional knowledge. This research could investigate how these mechanisms influence innovation within local communities and their role in fostering sustainable practices.

Abstract

The fragmented global patent landscape poses significant challenges for biotechnology, one of the most innovative sectors of the 21st century. Despite the potential to revolutionize healthcare and agriculture, differences in patent regulation between jurisdictions impede progress, limit equitable access to therapies, and complicate international collaboration. This paper examines the key regulatory frameworks in the United States, the European Union, Japan, and China, focusing on the inherent strengths and challenges of each, by analyzing case studies, such as CRISPR gene editing and CAR-T therapy. The article identifies strategies to deal with ethical dilemmas, intellectual property rights differences, and cross-border enforcement issues. The analysis in this publication encourages harmonization initiatives through regional agreements, international dialogue, and open innovation models to promote global access and sustainable innovation.

Abstract

Since 1997, hand, foot, and mouth disease (HFMD) has become a common health problem in Southeast Asia. Various types of research have been conducted and published to handle HFMD. However, until now, many children, especially in the Asia-Pacific region, including Indonesia, still have HFMD-causing enterovirus (EV) infection. By conducting a bibliometric analysis of the literature published over the last 27 years (1997–2024), the direction of HFMD research in children can be predicted, thus research areas that have the potential and still need to be developed for future better HFMD treatment can be known. The important HFMD research topics predicted to continue to develop were identified through keyword analysis, which was subsequently mapped using a network approach. Based on this study, it can be concluded that HFMD research is predicted to lead to the development of synbiotic supplements, which can reduce HFMD severity, especially in children, by utilizing a genome-wide association study (GWAS) and machine learning.
