Abstract

ABSTRAKSusu mempunyai kandungan yang berperan sebagai imunomodulator respon imun seluler dan humoral yang melindungi tubuh dari infeksi bakteri pathogen intrasel Salmonella Thyphimurium melalui peningkatan aktifitas fagositosi dan mekanisme mikrobiosida makrofag. Studi dilakukan untuk membuktikan efek susu kuda Sumbawa terhadap peningkatan respon imun seluler yang di amati dari indeks aktifitas fagositosi, sekresi Reactive Oxygen Intermediates (ROI) dan aktivitas mikrobiosida dari sel makrofag tikus yang diinduksi dengan S. Thyphimurium dengan desain posttest-only control group. Studi dilakukan menggunakan 24 tikus usia 8-12 minggu yang di bagi dalam 1 kelompok kontrol dan 3 perlakuan pemberian susu kuda Sumbawa dengan 3 dosis 0.5 m;/day ; 1 ml/hari selama 14 hari. Sel makrofag peritoneal diisolasi dan di beri perlakuan dengan S. Thyphimurium. Terdapat perbedaan respon imun selular antar kelompok pada semua indikator. Indeks aktifitas fagositosis makrofag paling tinggi ( 51.23 ± 9.72) dan sekresi ROI paling tinggi (67.00±9.16%) didapatkan pada dosis 1.5 ml/hari. Pada dosis 0.5 ml/hari (77.67±15.83)  pemberian susu kuda Sumbawa dapat menghambat pertumbuhan bakteri. Dapat di simpulkan pemberian susu kuda Sumbawa dapat meningkatkan imun seluler terhadap infeksi S. Abstract

There is considerable evidence, suggest that, consumption of food additives monosodium glutamate (MSG), a flavor enhancer was unhealthy. Herbal medicine Nigella sativa (NS) has antioxidant properties able to cure the toxic induced by MSG. This study aimed to evaluate the risks of excessive use of MSG and to study the role of NS to inhibit inflammation and renal damage. Treated rats (twenty four male wistar rats) were divided into six group and analyzed by measuring the cells in blood, interleukin-10, interferon-γ serum levels by ELISA method and remove kidneys for histological examination. Histological of kidney for all groups except control, were showed different abnormalities include congestion of some blood vessels, hemorrhage  between tubules, widening in the renal tubules, revealed severe dilatation of Bowman's capsule and shrinkage of glomeruli, and areas of huge vacuole, were observed compared with control. Interleukin-10 was reduced in Groups 2,3,4 and 5, whereas increase in NS group compared with control. Interferon-γ was increased in groups 2,3,4 and reduced in groups 5,6 compared with control.  Eosinophil was increased in groups 2,5 and reduced in groups 3,4, 6 compared with control. This present study showed that administration of MSG to rats induced many changes effects on inflammatory cells, cytokines and histological of kidneys. NS has benefit in blood parameters, whereas harmful on kidney at these doses.

Abstract

Type 1 diabetes mellitus (T1DM) is an autoimmune disease characterized by pancreatic β-cell destruction and considered to be correlated with oxidative stress. This study aimed to investigate the association of oxidative stress [superoxide dismutase (SOD) and malondialdehyde (MDA) levels], inflammation [interleukin 10 (IL-10)], and glycemic control (HbA1c) in pediatric T1DM patients. This study included 25 T1DM subjects and 25 healthy control subjects and was designed as a cross- sectional study. SOD, MDA, and IL-10 levels were measured by ELISA. We obtained that that IL-10 and SOD levels were significantly decreased in the T1DM group, but MDA and HbA1c levels were significantly elevated in the T1DM group. IL-10 levels were positively correlated with SOD levels and negatively correlated with MDA and HbA1c. SOD levels were negatively correlated with HbA1c levels. MDA was positively correlated with HbA1c levels. IL-10 and SOD levels were significantly decreased, but MDA and HbA1c levels were significantly elevated in the T1DM group.

Abstract

Faktor prilaku dan pola makan yang tidak sehat dapat menyebabkan penyakit kanker. Penyakit ini menjadi penyebab utama kematian seluruh dunia.  Kanker dapat terjadi akibat paparan radikal bebas atau disebut ROS salah satu produknya adalah hydrogen peroksida. Ketidakseimbangan ROS dengan antioksidan menyebabkan oxidative stress dan oxidative damage sel tubuh. Untuk itu dibutuhkan antioksidan pencegah terjadinya kanker. Karboksimetil Kitosan dapat menurunkan kadar hydrogen peroksida pada tikus uji setelah dipapar dengan allergen,  melalui gugus aktifnya. Efektivitas Karboksimetil Kitosan ini sangat berpotensi sebagai antikanker.

Abstract

Prevalence of tooth extraction or dental surgery was 48.5% of all dental care in Indonesia. Tooth extraction carries potential health risks and side effects such as pain, swelling, trismus and dysfunction of the oral cavity during recovery. Secretory leukocyte protease inhibitor (SLPI) is one of innate immunity proteins that can inhibit the activation of macrophages. We are expecting the provision of SLPI can decrease excessive inflammatory response in healing after tooth extraction. This study was to investigate the administration of SLPI on cellular expression of NF-Kβ and IL-1β on wound macrophages of Rattus novergicus post tooth extraction. The research design is in vivo experimental study. In total, 20 rats were randomly divided into four groups (each group n=5) and underwent tooth extraction on left incisor teeth of mandible. One of the groups did not receive SLPI administration (control group) and the socket was stitched after tooth extraction. Meanwhile, the remaining three groups (experimental groups) were given SLPI administration after tooth extraction with three different doses (0.1 µM, 0.5 µM and 2.5 µM, respectively). After SLPI administration, the socket of experimental groups was stitched. The effects of SLPI administration were evaluated by counting at the percentage of NF-Kβ translocation and the expression of expression of IL-1 in macrophages cells of the rat socket using immunohistochemistry analysis. The cellular expression of NF-Kβ and IL-1β were significantly decreased (p < 0.05) on groups with SLPI may decrease cellular expression of NF-Kβ and IL-1β on wound macrophages cells of rats post tooth extraction in a dosedependent manner.

Abstract

Moringa oleifera is a popular plant for combating malnutrition that grows well and is commonly consumed by Indonesian as a vegetable, but local varieties' nutrient composition never was studied before. Methods: Proximate, amino acids, minerals, vitamin C, and total phenolic composition of red and white cultivars of Moringa oleifera leaves from Probolinggo were determined on a dry weight basis based on AOAC methods. Results: Crude protein and fat were higher in white cultivar (35.36%, 6.25%). Total Amino Acid in both cultivars (83.06 g/100g for red and 82.86g/100g for white) were higher than in previous studies. Total Essential Amino Acid (TEAA) percentage of both of the cultivars was 46-49% and met the ideal protein needs of children (26%) and adults (11%). Iron, Zinc, Copper, Calcium, and Manganese was higher in white cultivar (36.21±0.08 mg/kg, 16.32±0.02%, 6.76±0.08mg/kg, 1.56±0.00 mg/kg, 7.11±0.00 mg/kg), while Magnesium, Potassium, Sodium, Phosphorus was higher in red cultivar (0.61±0.00%, 0.89±0.00%, 0.65±0.00%, 0.99±0.00). The content of vitamin C and total phenols was higher in the red cultivar (4162.8mg/kg and 239.8mg/kg). Conclusions: The red cultivar were best in 10/22 parameters (45%), white cultivar was best in 14/22 (55%). Those characteristics can contribute significantly to the nutrient requirements for protein and energy malnutrition treatment.

Abstract

Endothelial dysfunction after high-intensity exercise occurs through various mechanisms characterized by decreased nitric oxide (NO) bioavailability. This study aims to determine the relationship between circulating endothelial cells (CEC), endothelial progenitor cells (EPC), and endothelial dysfunction after high-intensity exercise through an experiment using male Wistar strain rats. This study uses the randomized control group posttest-only design. A total of 32 male Wistar rats aged 20-22 weeks with a body weight of 140±20 g were taken randomly and divided into two groups. Group 1 (n=16), the control group, was given low-intensity exercise. Meanwhile, group 2 (n=16), named the treatment group, was given high-intensity exercise. The groups were run on a special treadmill where light-intensity exercise is done at a speed of 5-8 meters.minute-1 for 30 minutes and high intensity at 25-30 meters.minute-1 for 30 minutes. The treatment was performed at 5 to 10 pm, 3 times weekly for 12 weeks. Blood and aortic tissue samples were taken after the mice were rested for 12 hours after the last exercise. ELISA and Flow cytometry measured NO, CEC, and EPC levels. This research showed that CEC and EPC expression and high-intensity exercise have a positive relationship, yet not significant to the occurrence of endothelial dysfunction (NO), with a value of Sig. (0.111); p > 0.05, and the relationship value is r = 0.414 (R²= 17.15%). In conclusion, endothelial dysfunction after high-intensity exercise is not associated with CEC and EPC expression, which indicates endothelial damage.  Keywords: exercise, circulating endothelial cells, endothelial dysfunction, endothelial progenitor cells, nitric oxide

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh induksi EGF terhadap lokalisasi Cx43 dan gap junction selama ekspansi sel kumulus. Hasilpenelitian ini menunjukkan bahwa ekspansi sel kumulus mempunyai signifikansi dengan meningkatnya konsentrasi EGF dan waktu kultur. Ekspansi sel kumulus diindikasikan dengan perubahan bentuk sel menjadi memanjang dan menyebar. Ekspresi Cx43 meningkat pada kultur jam ke-5 dan semakin meningkat pada kultur jam ke-10 dengan semakin padatnya protein pada badan sel. Pada kultur jam ke-15 terjadi penurunan ekspresi Cx43 pada badan sel. EGF berpengaruh terhadap peningkatan ekspansi sel kumulus tetapi tidak pada ekspresi Cx43 denganbertambahnya konsentrasi dan waktu kultur. Hal ini menunjukkan bahwa komunikasi antar sel kumulus menurun, karena jarak antar sel yang semakin jauh.
