Abstract

This study aims to evaluate the use of machine learning technology, particularly Teachable Machine, in identifying seaweed genus in Indonesia. The limitations of identification databases and the lack of previous research were the main drivers of this study. The research focused on Panjang Island with three representative field data collection stations. This field data became the basis for training machine learning models for identification. In addition to field data, information from the literature on seaweed visual characteristics was also taken to support the identification process. The machine learning model developed achieved 99.42% accuracy in identifying 13 classes of 9 seaweed genus. The implementation of the model on the web application showed satisfactory responsive performance, including in the speed test on Google PageSpeed. Overall, the integration of machine learning technology in a web application platform provides a practical solution for accurate and accessible seaweed identification. This invention has great potential in supporting research, conservation, and sustainable utilization of marine resources in Indonesia.

Abstract

Natural ingredients from animals and plants generally have potential in pharmaceutical applications. However, there are still relatively little exploration made on the active ingredients from marine life. Sea cucumbers inhabit seabed and are filter feeders, consuming all microorganisms within their vicinity, which opens the prospect of medicinal applications of sea cucumber intestinal symbiont microbes. This study aims to obtain sea cucumber intestinal symbiont microbes with potential pharmaceutical application as probiotic microbe candidate. The study was carried out by collecting samples, microbe isolation, antimicrobial screening, identification of potential probiotic microbe candidate. Characteristics of the microbe were studied by biochemical screening, and molecular identification. The study found 21 microbial isolates from Holothuria atra and 30 microbial isolates from Holothuria leucospilota. Antimicrobial activity screening results against pathogens of Bacillus cereus dan Pseudomonas aeruginosa found 13 microbial isolates with positive activity. Four species were identified by molecular identification, namely Bacillus aquimaris, Bacillus maritimus, Bacillus toyonensis, and Virgibacillus chiguensis, whereas the other nine were identified by biochemical screening. Four microbe genus, namely Rothia sp., Listeria sp., Micrococcus sp., and Staphylococcus sp, were found to be the most viable candidate for probiotics. It was concluded that Bacillus sp., Rothia sp., Micrococcus sp., and Staphylococcus sp. Exhibited the most potential as probiotic microbes.

Abstract

Gastropod association bacteria from magrove ecosystem have a great potency as antibacteria. The contamination of Escherichia coli and Bacillus cereus can cause damage in fish product. The aims of these research are to determine gastropod association bacteria that can inhibit E. coli and B. cereus growth, as well as the contaminantion bacteria in Belanak (Mugil subviridis). Gastropods sample Certhideopsilla alata, Cerithidea quoyii, Cassidula aurisfelis, Cassidula nucleus, Telescopium telescopium were collected at Mangrove Education Park, Tugu, Semarang. The resaerch procedure were isolation, antibacteria test, biochemistrically identification, and then application in Belanak fish (Mugil subviridis). There were 61 bacteria isolated. Thirteen isolates were able to inhibit E. coli and eight to B. cereus. Supernatant of GMT 3.2.5 and GMT 4.1.3 have the most widely inhibition zone, (8,48 mm and 7,84 mm). Biochemistry identification shown that GMT 3.2.5 and GMT 4.1.3 has identic characteristic with genus Micrococcus sp. and Bacillus sp. Total plate count (cfu/ml) of bacteria from Belanak fish flesh that soaked by Micrococcus sp. GMT 3.2.5 and Bacillus sp. GMT 4.1.3 supernatant were 115 x 104 and 265 x 103colony. The colony were less than negative control  (2312 x 103 colony). The association bacteria can inhibit contaminant bacteria in Belanak fish.

Abstract

The Pacific white shrimp (Penaeus vannamei) is a key species in global aquaculture, particularly under intensive farming systems where high stocking densities often lead to deteriorating water quality, disease outbreaks, and reliance on antibiotics. While probiotics are increasingly applied to enhance shrimp health and performance, comparative evidence on the efficacy of different commercial probiotic formulations remains limited. This study aimed to evaluate the effects of three commercial probiotic products on growth performance, water quality, microbial populations, and immune response in intensively reared Pacific white shrimp. Shrimp were fed diets supplemented with one of three probiotics (PB, PL, and PMB) or a control diet for 40 days. Growth parameters, nitrogenous waste levels, bacterial counts in water and organs, and total hemocyte count (THC) were analyzed. The PB treatment (multi-strain Bacillus) significantly enhanced specific growth rate (2.99±0.0027% day⁻¹), feed efficiency (57.5±0.09%), weight gain, and THC (3.80 × 10⁵ cells mm⁻³) compared to control (P<0.05). The PB and PMB treatments also reduced total ammonia nitrogen, nitrite, nitrate, and Vibrio populations in water and shrimp tissues. Intestinal colonization by beneficial bacteria was highest in PB and PL groups. These results demonstrate that specific probiotic formulations, particularly PB, can improve growth, immunity, and water quality while reducing reliance on antibiotics and water exchange. The findings support the integration of targeted probiotics in sustainable shrimp aquaculture, including biofloc and recirculating aquaculture systems (RAS).

Abstract

Salah satu faktor penurunan produktivitas udang disebabkan oleh faktor lingkungan, termasuk salinitas dan kualitas pakan, sehingga perlu ditingkatkan. Tujuan penelitian ini untuk mengetahui pengaruh pemberian Artemia yang diperkaya alginat terhadap tingkat kelangsungan hidup, pertumbuhan dan ketahanan terhadap stres salinitas pada Post-Larva (PL)1 Litopenaeus vannamei. Pengkayaan Artemia dilakukan dengan proses perendaman selama satu jam. Metode yang digunakan pada penelitian ini adalah Rancangan Acak Kelompok, dengan Kontrol (tanpa pengkayaan alginat) dan 3 perlakuan (pengkayaan Artemia sebanyak 10 ekor/mL/hari dengan alginat pada dosis 0 (kontrol), 400, 600, 800 ppm). Setiap perlakuan diulang sebanyak tiga kali. Pemeliharaan dengan pemberian pakan pengkayaan Artemia dilakukan selama 14 hari. Pertumbuhan panjang mutlak tertinggi diperoleh perlakuan yang diberi Artemia yang diperkaya alginat 600 ppm. Pertumbuhan mutlak berat diperoleh dari perlakuan Artemia dengan pengkayaan alginat 600 dan 800 ppm. Ketahanan stres salinitas tertinggi dengan kematian terlama diperoleh pada perlakuan yang diberi Artemia dengan pengkayaan alginat 400 ppm. Pemberian Artemia yang diperkaya alginat mampu meningkatkan pertumbuhan dan ketahanan terhadap stres salinitas PL L. vannamei, namun tidak menunjukan pengaruh terhadap tingkat kelangsungan hidup.

Abstract

Litopenaeus vannamei has emerged in the aquaculture industry. Production consistency, nutrition, and disease management play critical roles, particularly the digestive enzymes such as trypsin. This study assesses Ulvan, an immunostimulant from Ulva lactuca, on shrimp trypsin activity. Trypsin has been found to significantly enhance the activity of hemocyanin phenoloxidase, a crucial component of humoral immunity. This study aims to evaluate the potency of ulvan related to trypsin as an immunostimulant agent, extracted from Ulva sp.  Ulvan, extracted using various methods (P-HWE, O-HWE, P-A-HWE, and O-A-HWE), was evaluated using different doses of (0 g kg-1 (Control), 0.75 g kg-1 (ULV-0.75), 1.50 g kg-1 (ULV-1.50), and 3.00 g kg-1 (ULV-3.00) of feed). The O-A-HWE exhibited the fastest and highest increase in trypsin activity on day 4, surpassing the control on days 2, 3, 7, and 8. The P-HWE, O-HWE, and P-A-HWE also showed significant changes in trypsin activity compared to the control on specific days. Meanwhile, trypsin activity in Ulvan-fed shrimp did not significantly differ from the control on days 0 and 1. The differences emerged on day 2 and 3, notably between ULV-1.50 g kg−1 and ULV-0.75 g kg−1. The ULV-3.00 g kg−1 showed no significant difference from ULV-1.50 g kg−1. O-A-HWE demonstrated significant differences in trypsin activity compared to other Ulvan extracts, suggesting its potential to enhance shrimp health.Litopenaeus vannamei memiliki peran yang besar dalam industri akuakultur. Konsistensi dalam produksi, nutrisi, dan pengelolaan terhadap penyakit merupakan bagian yang sangat penting, terutama enzim pencernaan, di antaranya tripsin. Tripsin berfungsi untuk meningkatkan fenoloksidase hemosianin, yang peranannya penting untuk kekebalan. Studi ini mengevaluasi Ulvan, yaitu bahan yang bersifat imunostimulan dari Ulva sp., terhadap aktivitas tripsin udang. Ulvan, diekstraksi menggunakan berbagai metode (P-HWE, O-HWE, P-A-HWE, dan O-A-HWE), dievaluasi menggunakan perlakuan dosis yang berbeda (0 g kg-1 (Kontrol), 0,75 g kg-1 (ULV-0,75), 1,50 g kg-1 (ULV-1,50), dan 3,00 g kg-1 (ULV-3,00) pada pemeliharaan udang vaname selama 10 hari. O-A-HWE menunjukkan peningkatan aktivitas tripsin tercepat dan tertinggi pada hari ke-4, melebihi kontrol pada hari ke-2, 3, 7, dan 8. P-HWE, O-HWE, dan P-A-HWE juga menunjukkan perubahan signifikan dalam aktivitas tripsin dibandingkan dengan kontrol pada hari-hari tertentu. Sementara aktivitas tripsin dalam udang yang diberi Ulvan tidak berbeda secara signifikan dari kontrol pada hari ke-0 dan 1, perbedaan mulai terlihat pada hari ke-2 dan 3, terutama antara ULV-1,50 g kg−1 dan ULV-0,75 g kg−1. ULV-3,00 g kg−1 tidak menunjukkan perbedaan yang signifikan dari ULV-1,50 g kg−1. O-A-HWE menunjukkan perbedaan signifikan dalam aktivitas tripsin dibandingkan dengan ekstrak Ulvan lainnya, sehingga berpotensi dalam meningkatkan kesehatan udang.

Abstract

The whiteleg shrimp (Litopenaeus vannamei) is a highly valued aquaculture species globally, yet its production faces challenges due to disease outbreaks, notably acute hepatopancreatic necrosis disease (AHPND). This study aimed to evaluate the growth and survival of L. vannamei juveniles fed with synbiotic bio-encapsulated Artemia and their resilience against AHPND-causing Vibrio parahaemolyticus and salinity stress. The experiment employed a completely randomized design with two treatments: one with synbiotic-enriched Artemia (600 ppm alginate and Lactobacillus bulgaricus) and a control without synbiotic. Each treatment was replicated five times, using 600 juveniles at a density of 30 post-larvae per L over a 14-day rearing period. Growth and survival metrics were recorded, followed by challenge tests for AHPND and salinity shock. The juveniles' survival rate was recorded 54 hours post-infection with VpAHPND and every 10 minutes for 230 minutes after salinity exposure until 100% mortality. Results indicated higher survival (92.0 ± 9%), length gain (243.33 ± 18.80 mm), specific growth rate (18.44±2.01%), and stress tolerance in juveniles fed synbiotic encapsulated Artemia compared to the control. The survival rates for the challenge test with AHPND and salinity shock were similarly improved under synbiotic treatment, suggesting that synbiotics significantly benefit nursery production of L. vannamei. This study highlights the potential of synbiotic application in enhancing the resilience and growth of L. vannamei against common stressors in aquaculture, indicating its potential to support more sustainable shrimp farming practices.Udang vaname (Litopenaeus vannamei) adalah komoditas budidaya bernilai tinggi di seluruh dunia, namun produksinya menghadapi tantangan akibat wabah penyakit, terutama serangan acute hepatopancreatic necrosis disease (AHPND). Penelitian ini bertujuan untuk mengevaluasi pertumbuhan dan kelangsungan hidup benih L. vannamei yang diberi pakan Artemia yang dibioenkapsulasi dengan sinbiotik serta ketahanannya terhadap Vibrio parahaemolyticus penyebab AHPND dan stres salinitas. Eksperimen ini menggunakan rancangan acak lengkap dengan dua perlakuan: satu dengan Artemia diperkaya sinbiotik (600 ppm alginat dan Lactobacillus bulgaricus) dan kontrol tanpa sinbiotik. Setiap perlakuan diulang lima kali, dengan menggunakan 600 ekor benih udang pada kepadatan 30 ekor pascalarva per L selama 14 hari periode pemeliharaan. Parameter pertumbuhan dan kelangsungan hidup dicatat, diikuti dengan uji tantang terhadap AHPND dan kejutan salinitas. Tingkat kelangsungan hidup benih dicatat 54 jam pasca-infeksi dengan VpAHPND dan setiap 10 menit selama 230 menit setelah paparan salinitas hingga mortalitas mencapai 100%. Hasil menunjukkan kelangsungan hidup yang lebih tinggi (92,0 ± 9%), peningkatan panjang (243,33 ± 18,80 mm), laju pertumbuhan spesifik (18,44 ± 2,01%), dan toleransi stres yang lebih baik pada benih udang yang diberi pakan Artemia berenkapsulasi sinbiotik dibanding kontrol. Tingkat kelangsungan hidup pada uji tantang dengan AHPND dan kejutan salinitas juga meningkat dengan perlakuan sinbiotik, menunjukkan bahwa sinbiotik memberikan manfaat signifikan pada produksi L. vannamei fase pendederan. Penelitian ini menunjukkan adanya potensi aplikasi sinbiotik dalam meningkatkan ketahanan dan pertumbuhan L. vannamei terhadap stresor umum dalam akuakultur, serta potensinya untuk mendukung kegiatan budidaya udang yang lebih berkelanjutan.

Abstract

Litopenaeus vannamei merupakan komoditas yang bernilai ekonomi tinggi pada sektor perikanan dan budidaya. Spirulina sp. merupakan mikroalga yang mengandung nutrisi tinggi dan senyawa potensial yang memiliki efek terapeutik. Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi ekstrak Spirulina sp. dalam pakan terhadap tingkat kelangsungan hidup, pertumbuhan, dan tingkat kelangsungan hidup L. vannamei pada uji stres salinitas. Hewan uji yang digunakan adalah post-larvae (PL 8) L. vannamei dengan bobot rata-rata 0,002 g/ekor. Penelitian ini menggunakan metode Rancangan Acak Lengkap (RAL) yang terdiri atas 3 konsentrasi, yaitu 3 mg/kg (SP 3), 6 mg/kg (SP 6), dan 9 mg/kg (SP 9) serta satu kontrol dengan masing-masing 4 pengulangan. L. vannamei dipelihara dan diberi pakan suplementasi Spirulina sp. selama 14 hari. Pada akhir penelitian dilakukan uji stres salinitas dengan menggunakan tabung falcon yang masing-masing diisi 20 ekor PL dan dipaparkan pada akuades (45 mL) dan ditutup rapat. PL diamati dan dihitung jumlah kelulushidupannya setiap 10 menit. Waktu pengamatan dihentikan ketika kematian udang mencapai 50%.Hasil penelitian menunjukkan bahwa pemberian suplementasi ekstrak Spirulina sp. pada pakan tidak berpengaruh nyata (p>0,05) terhadap pertumbuhan dan tingkat kelangsungan hidup udang. Namun, perlakuan SP 9 menunjukkan pengaruh nyata (p<0,05) terhadap tingkat kelangsungan pada uji stres salinitas. Suplementasi ekstrak Spirulina sp. mampu meningkatkan toleransi L. vannamei terhadap stres salinitas.  Litopenaeus vannamei is high economically viable cultivate in the aquaculture sectors. Spirulina sp. is high nutrients microalgae and potential compound that have therapeutic effects. This study aims to determine the effect of dietary supplementation of Spirulina sp. on feed towards survival rate, growth, and resistance of L. vannamei in the salinity stress test. This experiment was using post-larvae (PL 8) (0.002 g/ind). This study used a completely randomized design (CRD) consisting of 3 concentrations, namely 3 mg/kg (SP 3), 6 mg/kg (SP 6), and 9 mg/kg (SP 9), and one control with 4 repetitions for each treatment.  L. vannamei was reared and fed with Spirulina sp. supplementation for 14 days. On stress salinity test, PL were put in the falcon tube filled with aqua dest with 20 PL each and exposed until 50% mortality. The results showed that the supplementation of Spirulina sp. on feed had no significant effect (p>0.05) on the growth and survival rate of shrimp. However, the SP 9 treatment showed a significant effect (p<0.05) on the survival rate in the salinity stress test. Is concluded that supplementation of Spirulina sp. in the diet managed to increase the resistance of L. vannamei toward stress salinity.
