Abstract

One disease that often attacts fish is a group of parasites or parasitic diseases, parasites are not only dangerous because they are related to their activities, but also trigger the emergence of other pathogenic organisms and cause infections. Therefore, it is necessary to take fish quarantine measures to prevent the spread of diseases carried out by fish quarantine centers. This study was conducted to determine the type of parasite that infested fish samples and determine the prevalence and intensity of fish samples. A total of 48 sample fish consisting of freshwater and seawater fish were analyzed in the laboratory of Balai Karantina Ikan, Pengendalian Mutu dan Keamanan Hasil Perikanan Surabaya II during December - January 2019, either directly or directly (ectoparasites and endoparasites). Parasitic identification was carried out visually and microscopically with an approach based on identification books. The types of parasites that we found during the study consisted of Argulus sp, Trichodina sp, Anisakis sp., Gnathostoma spinigerum, and Gnathostoma spinigerum. The highest prevalence in Scomber Scombrus with Anisakis sp. Amounting  to 77.8%, mean while, the lowest prevalence and the intensity is found in Thunnus sp. The prevalence and intensity of parasites are influenced by water quality and pollution.

Abstract

The potential of freshwater fish cultivation in Indonesia is very large and is increasing every year. The high potential of freshwater and ornamental fisheries commodities and consumption must be developed through domestic and export marketing. The distribution carries the risk of entering and spreading fish pests and diseases, so is it required identification or quarantine is needed to avoid negative impacts from the distribution of fishery commodities. One of the infectious diseases which is known to attack fishery commodities in Indonesia is bacterial disease. Bacteria can cause high mortality in fish and loss for farmers. The purpose of this study is to identification bacteria in freshwater fish commodities. The study was conducted in December 2018 - January 2019 at the Fish Quarantine Center, Quality Control and Safety of Fisheries Products in Surabaya I. Identification of bacteria in this study using conventional methods. The parameters observed were external clinical symptoms that appeared on the body of the fish and internal pathology. The material examined in this study were The samples examined during Field Work Practices were Goldfish, Catfish fry, Common carp, and Koi fish with a total of 64 samples. This research was observative, then the data obtained were analyzed descriptively. The Result bacteria identified in freshwater fish commodities from 64 samples were dominated by Aeromonas hydrophila, Pseudomonas anguilliseptica, Streptococcus sp., Plesiomonas shigelloides, and Micrococcus sp.Keywords : Freshwater fisheries commodities, Bacteria, The Conventional Method

Abstract

The study aimed to determine the type of culture container for increasing the cell density of Nitzschia and producing the hightly cell biomass. The stages of this study were sterilization of containers and media, making diatom fertilizers and agar media, culture of Nitzschia in agar medium, culture of Nitzschia in test tube, and culture of Nitzschia in glass and plastic containers. The result showed that the cell density of Nitzschia cultured in the glass container was higher than plastic containers. Further study was needed to determine the nutrient composition of Nitzschia that culture in glass containers.Keyword : Nitzschia culture, Laboratory scale, Glass, Plastic container.

Abstract

      Microorganisms that often contaminate fishery products were from bacteria groups. Some spesies of bacteria were harmful to human health because they can cause disease and even death in humans, such as Escherichia coli and Salmonella. Therefore its were needed for quarantine and microbiological examination of fishery products as an effort to prevent the spread of disease in humans. The aims of this study were to identify the contamination of E. coli and Salmonella bacteria in fishery products. A total eight samples of fishery products tested consist of canned dan frozen products using conventional methods based on ISO 16649-3: 2015 for E. coli and ISO 01-2332.2-2006 for Salmonella. The results showed that one from the six fishery products tested were frozen boiled tuna that positively E. coli and no fishery product sample was identified with Salmonella.

Abstract

The purpose of this research is to find out the abalone separating technique (H. squamata) and know the problems during separating at the Marine Aquaculture Development Centre, West Nusa Tenggara Province. Research are carried out on December 23, 2019 to January 21, 2020. The method used is descriptive method with data collection including primary data and secondary data. Data is taken by observing, active participation, interviewing, and literature review. Abalone separating techniques (H. squamata) begin with container preparation, seed preparation, seed stocking, seeding, feeding, water quality management, and growth sampling. Problems encountered during the separating stage are the uncertain availability of seaweed feed.Keyword :  Abalone, H. squamata, separating technique.

Abstract

The aim of this study was to evaluate the addition of a probiotic Bacillus on culture medium through growth rate and histopathological change in African Catfish who was infected by Aeromonas hydrophila. The study consisted of five treatments such as the addition of probiotic Bacillus ND2, Bacillus P4I2, Bacillus ND2 + Bacillus P4I2 (Kom), positive control (K+) and negative control (K-) (without probiotic addition). African Catfish (13.354±2.8 g) was maintained in 15 aquariums (40 L in volume) with 30 fishes each for 30 days. Probiotic bacteria was applied in water once a day, whereas pathogenic bacteria A. hydrophila  (103 CFU/mL) were added once in earlier treatment (except for the negative control) as assumption that A. hydrophila will shape up quorum sensing in unfortunate condition. The result showed that the highest spesific growth rate in the treatment of Bacillus ND2 probiotics (1,708 ± 0.114%). Histopathology showed that damaged of liver dan kidney in probiotics treatment were lightner than positive control (K+).  The addition of Bacillus P4I1 (104 CFU/mL) efective to increased the spesific growth rate of African Catfish and prevent the damage of liver and kidney of African Catfish was infected by Aeromoniasis.

Abstract

The freshwater aquaculture fishery sector in Indonesia has the potential to be developed through extensification and intensification. The problem in aquaculture is diseases that infects cultivated organisms, one of which is caused by bacterial infection. Bacterial infection can cause huge losses to cultivation activities, including death which results in economic losses. The purpose of this study is to identify the A. salmonicida and E. ictalury in live fish that will be exported from DI Yogyakarta. The method of examining bacteria used is conventional which consists of isolation, purification, presumptive test, biochemical test, and identification. The results showed that all live fish samples (koi, gouramy, goldfish, catfish, and eel) were not infected with A. salmonicida and E. ictalury. This shows that all live fish are in a healthy condition and suitable to be exported of DI Yogyakarta.

Abstract

Penelitian ini bertujuan untuk membandingkan pola pertumbuhan Spirulina sp. yang dikultur pada wadah yang berbeda. Tahapan dalam penelitian ini yaitu persiapan dan sterilisasi wadah dan media, pembuatan media kultur Spirulina sp. dan vitamin, pemilihan inokulan Spirulina sp., dan kultur Spirulina sp. Hasil penelitian menunjukkan bahwa fase eksponensial pada wadah kaca terjadi pada hari ke-9 dengan total biomass sebesar 53,375 x 105 sel/ml, berbeda dengan wadah plastik yang terjadi pada hari ke-8 dengan total biomass sebesar 16,993 x 105 sel/ml.

Abstract

Kematian ikan akibat serangan parasit Argulus dikarenakan ikan mengalami emasiasi sehingga dapat menurunkan sistem kekebalan tubuh ikan, selain itu munculnya luka pada kulit ikan akan dimanfaatkan sebagai port of entry bagi agen infeksi sekunder seperti jamur, bakteri dan virus. Pengendalian penyakit argulosis dengan bahan kimia yang tidak tepat dosis dapat mencemari lingkungan, sehingga perlu dicari alternatif bahan kimia yang aman terhadap lingkungan, yaitu abate. Tujuan dari penelitian ini yaitu untuk mengetahui efektivitas perendaman abate untuk menurunkan derajat infestasi dan merusak organ parasit Argulus yang menginfestasi ikan mas (Cyprinus carpio). Perlakuan yang diberikan terdiri dari lima perlakuan dengan empat kali ulangan yaitu pemberian abate dengan konsentrasi 0 ppm (kontrol), 25 ppm, 50 ppm, 75 ppm, dan 100 ppm yang direndam selama 30 menit. Parameter yang diamati pada penelitian ini yaitu derajat infestasi Argulus yang menempel pada ikan mas, kerusakan organ Argulus dan nilai kualitas air pada awal dan akhir perendaman sebagai parameter pendukung. Data derajat infestasi Argulus pada ikan mas dianalisis secara statistik menggunakan ANOVA 5% yang dilanjutkan dengan uji DMRT 5% sedangkan kerusakan organ Argulus dianalisis secara deskriptif dan disajikan dalam bentuk gambar dan tabel serta dihubungkan dengan parameter pendukung. Hasil penelitian menujukkan bahwa perendaman abate tidak efektif untuk menurunkan derajat infestasi menginfestasi ikan mas (C. carpio), akan tetapi dapat menimbulkan kerusakan organ Argulus pada perlakuan 100 ppm. Kerusakan organ Argulus yang ditimbulkan yaitu rontoknya kaki ke-3 dan 4, abdomen yang menyatu serta produksi telur prematur. Perlu dilakukan penelitian lanjutan dengan meningkatkan dosis abate (lebih dari 100 ppm) serta waktu inkubasi yang lebih lama (lebih dari 30 menit). Selain itu perlu diamati perubahan fisiologis ikan selama dilakukan perendaman dalam abate.

Abstract

Utilization of Porphyridium sp. began to be developed as an anticancer, anti-inflammatory, antibacterial, antifungal, and production of biogas and biodiesel so that necessary to carry out culture activities for ensure continuity of Porphyridium sp. This study aims to determine the type of culture container that showed the best growth response of Porphyridium sp. with the highest biomass. The stages in this research were preparation of containers and media sterilization, making diatom fertilizers and agar media, Culture of Porphyridium sp. on agar media, Culture of Porphyridium sp. in the test tube, and Culture of Porphyridium sp. in glass and plastic containers. The results showed that the growth response of Porphyridium sp. that cultured in glass containers (18,9 ± 0,21 x 105 cells/mL) was higher than plastic (15.57 ± 0,03 x 105 cells/ml).
