Abstract

ABSTRACT Soft Coral Sinularia sp.has been shown to have antibacterial activity. This research aims to determine the antibacterial activity of marine  organisms obtained from Manado bay. The method in this study is the diffusion method with positive control chloramphenicol and negative control of methanol in Escerichia coli and Sthaphylococcus aureus. The result showed that extracts, chloroform fraction and methanol-water fraction were obtained inhibiting category of Sthapylococcus aureus bacteria, but in the chloroform fraction and ethanol extract can inhibit Escerichia coli bacteria categorized as strong. With it can be conlueded that the extract and fraction of soft Sinularia sp. have bioactive compounds with a broth spectrum of antibacterial activity.Keywords: Sinularia sp., Chloramphenicol, Escerichia coli, Sthaphylococcus aureus, and antibacterial. ABSTRAK Karang lunak Sinularia sp. telah terbukti memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri pada organisme biota laut yang diperoleh dari teluk Manado. Metode dalam penelitian ini adalah metode difusi agar dengan kontrol positif kloramfenikol dan kontrol negatit metanol pada Escherichia coli dan Staphylococcus aureus. Hasil penelitian menunjukkan bahwa ekstrak, fraksi kloroform, dan fraksi metanol-air dapat menghambat bakteri Staphylococcus aereus dikategorikan sedang, namun pada fraksi kloroform dan ekstrak etanol dapat menghambat bakteri Escerichia coli dikategorikan kuat. Dengan demikian dapat disimpulkan bahwa ekstrak dan fraksi karang lunak Sinularia sp.memiliki senyawa bioaktif dengan spektrum yang luas terhadap aktivitas antibakteri.Kata kunci: Sinularia sp, Kloramfenikol, Escherichia coli, Staphylococcus aureus, dan antibakteri

Abstract

ABSTRACTGood service quality will lead to satisfaction in patients. Pharmaceutical services especially in hospitals are sued to realization the expansion of the pharmaceutical service paradigm from product orientation to patient orientation. This study aims to determine the level of outpatient satisfaction to quality of drug service in the Pharmacy Installation of Liun Kendage Tahuna Hospital based on five dimensions of service quality namely tangibles, empathy, reliability, responsiveness, and assurance. This research is a descriptive study with prospective data collection. Number of samples is 347 people, the process of collecting primary data is done using research instruments in the form of questionnaires and data processing to determine the validity and reliability of the questionnaire using SPSS 17 for Windows program. The results showed that the overall satisfaction index value was -0.63. The satisfaction index for each dimension is; tangible -0.62; empathy -0.58; reliability -0.59; responsiveness -0.59; and assurance -0.70. This shows that the worth satisfaction index is negative, which means that outpatients at the Liun Kendage Tahuna Hospital Pharmacy Installation are not yet feel satisfied with the services provided. Keywords: patient satisfaction, quality service, RSUD Liun Kendage Tahuna.ABSTRAKKualitas pelayanan yang baik akan menimbulkan kepuasan pada pasien. Pelayanan kefarmasian khususnya di rumah sakit dituntut untuk merealisasikan perluasan paradigma pelayanan kefarmasian dari orientasi produk menjadi orientasi kepada pasien. Penelitian ini bertujuan untuk mengetahui tingkat kepuasan pasien rawat jalan terhadap kualitas pelayanan obat di Instalasi Farmasi RSUD Liun Kendage Tahuna berdasarkan lima dimensi kualitas pelayanan yakni tangibles, emphaty, reliability, responsiveness, dan assurance. Penelitian ini merupakan penelitian deskriptif dengan pengambilan data secara prospektif. Jumlah sampel sebanyak 347 orang, proses pengumpulan data primer dilakukan dengan menggunakan instrumen penelitian dalam bentuk kuesioner dan pengolahan data untuk mengetahui validitas dan reliabilitas kuesioner menggunakan program SPSS 17 for windows. Hasil penelitian diperoleh nilai indeks kepuasan secara keseluruhan sebesar terbesar -0,63. Indeks kepuasan pada setiap dimensi yaitu; tangible -0,62; emphaty -0,58; reliability -0,59; responsiveness -0,59; dan assurance -0,70; Hal ini menunjukkan indeks kepuasan bernilai negatif yang berarti pasien rawat jalan di Instalasi Farmasi RSUD Liun Kendage Tahuna belum merasa puas terhadap pelayanan yang diberikan. Kata Kunci : kepuasan pasien, kualitas pelayanan, RSUD Liun Kendage Tahuna.

Abstract

ABSTRACT This study aims to determine the validation of IR spectroscopy method in determining the total flavonoid level in Abelmoschus manihot L., can meet the requirements and can be applied. The method for determining the total flavonoid content model using a combination of IR Spectroscopy and Chemometrics Partial Least Square Regression (PLSR). The calorimetrics method was used to determine the total flavonoid content in the green gedi leaves axtracts onn eight samples of growth where Bitung Was 2.64 mg QE/g extract  ± 0.035, Minahasa Selatan is  1.91 mg QE/g extract ± 0.027, Kotamobagu is 4.84 mg QE/g extract  ± 0.03, Minahasa Utara is  4.40 mg QE/g extract ± 0.091, Manado is 3.45 mg QE/g extract  ± 0.012, Minahasa Tenggara is  1.72 mg QE/g extract ± 0.006, Minahasa is 3.67 mg QE/g extract ± 0.033, Tomohon is 3.40 mg QE/g extract ± 0.003. This combination is involves involving x-variables (FTIR measurement results) and y-variables (data from the results of the calorimetric method analysis). Metode Kalorimetrik digunakan untuk mengetahui kandungan Flavonoid Total pada pada Ekstra Daun Gedi Hijau pada 8 sampel tempat tumbuh yaitu Bitung sebesar 2.64 mg QE/g ekstrak  ± 0.035, Minahasa Selatan sebesar  1.91 mg QE/g ekstrak ± 0.027, Kotamobagu sebesar 4.84 mg QE/g ekstrak ± 0.03, Minahasa Utara sebesar  4.40 mg QE/g ekstrak ± 0.091, Manado sebesar 3.45 mg QE/g ekstrak  ± 0.012, Minahasa Tenggara sebesar  1.72 mg QE/g ekstrak ± 0.006, Minahasa sebesar 3.67 mg QE/g ekstrak ± 0.033, Tomohon sebesar 3.40 mg QE/g ekstrak ± 0.003. Abstract

One chemichal component that is often reported to have pharmacological activity against treatment or prevention of disease is flavonoids which can be found in gedi hijau leaves. Staphylococcus aureus is the main cause of infection in humans which can cause various severe infections. This study aims to determine the total flavonoids content, minimum inhibitory concentration (MIC). To determine the correlation between total flavonoids and antibacterial activity of extract and fractions (Abelmoschus manihot L.), The method of this study was done by obtaining the results of total flavonoid content, testing the antibacterial activity by dilution using a positive control of ciprofloxacin. The results showed the highest total flavonoid content in ethyl acetate fraction, and the best antibacterial activity in ethyl acetate fraction. Correlation between total flavonoid content from extract and fraction (Abelmoschus manihot L.) with antibacterial activity against Staphylococcus aureus bacteria on ethyl acetate fraction, n-hexane fraction, and ethanol extract at a concentration of 1000 µg / mL had good values with kneeling significance values according to 0.012 (p <0.05), 0.034 (p <0.05), and 0.023 (p <0.05) with total flavonoid content affecting how much antibacterial activity is 40% for ethyl acetate fraction, 35% fraction n -heksane and 33% ethanol extract. Keywords         : Antibacterial, Total Flavonoids, Staphylococcus aureus, Abelmoschus manihot L. ABSTRAK Salah satu komponen kimia yang sering dilaporkan memiliki aktivitas farmakologi terhadap pengobatan atau pencegahan penyakit adalah flavonoid yang dapat ditemukan pada daun gedi hijau. Staphylococcus aureus merupakan kuman penyebab utama infeksi pada manusia yang dapat menyebabkan berbagai infeksi berat. Penelitian ini bertujuan menentukan kandungan flavonoid total, aktivitas antibakteri dan korelasi antara flavonoid total dengan aktivitas antibakteri dari ekstrak dan fraksi (Abelmoschus manihot L.). Metode penelitian ini dilakukan dengan mendapatkan hasil kandungan flavonoid total, uji aktivitas antibakteri dengan cara dilusi menggunakan kontrol positif ciprofloxacin. Hasil penelitian didapatkan kandungan flavonoid total tertinggi pada fraksi etil asetat, dan aktivitas antibakteri paling bagus pada fraksi etil asetat. Korelasi antara kandungan flavonoid total dari ekstrak dan fraksi (Abelmoschus manihot L.) dengan aktivitas antibakteri terhadap bakteri Staphylococcus aureus pada fraksi etil asetat, fraksi n-heksane, dan ekstrak etanol pada konsentrasi 1000 µg/mL memiliki nilai yang baik baik dengan nilai signifikansi bertutut-turut 0,012 (p <0,05), 0,034  (p <0,05), dan 0,023 (p <0,05) dengan kandungan flavonoid total mempengaruhi seberapa besar aktivitas antibakteri yaitu 40% untuk fraksi etil asetat, 35% fraksi n-heksane dan 33% ekstrak etanol.Kata kunci          : Antibakteri, Flavonoid Total, Staphylococcus aureus, Abelmoschus manihot L.

Abstract

ABSTRACTRhodamin B is a red synthetic dye commonly used as paper dyes, textiles or inks. Food products that are often added by Rhodamin B by irresponsible producers is sauce. The purpose of this study was to determine the presence and levels of Rhodamin B found in skewers meatballs sold in several elementary schools in Manado City using Thin Layer Chromatography and UV-Vis Spectrophotometer. The results of the study obtained that the results of the qualitative analysis using TLC obtained 5 sampels identified based on Rf value parameters and color visualization. Quantitative analysis of UV-Vis spectrophotometer produced Rhodamin B level values for each sample which was calculated using regression formula y = 0,0726 x + 0,0432 and R2 = 0,9957. The sample code A1 was 3,443 ± 0,080 μg/mL, B1 was 3,057 ± 0,150 μg/mL, B2 was 5,881 ± 0,170 μg/mL, C1 was 3,884 ± 0,130 μg/mL, and C2 was 4,517 ± 0,140 μg/mL. The conclusion in this study is that skewer meatballs circulating in several elementary schools in Manado City contains Rhodamin B. Keywords: Rhodamin B, Skewer Meatballs Sauce, Thin Layer Chromatography, UV-Vis Spectrophotometer. ABSTRAKRhodamin B merupakan pewarna sintetik berwarna merah yang umum digunakan sebagai pewarna kertas, tekstil atau tinta. Produk pangan yang sering di tambahkan Rhodamin B oleh produsen yang tidak bertanggung jawab adalah saos. Tujuan penelitian ini adalah untuk mengetahui keberadaan dan kadar Rhodamin B yang terdapat pada saos bakso tusuk yang dijajakan pada beberapa Sekolah Dasar di Kota Manado menggunakan metode Kromatografi Lapis Tipis dan Spektrofotometer UV-Vis. Hasil penelitian yang diperoleh bahwa hasil analisis secara  kualitatif menggunakan KLT diperoleh  5 sampel teridentifikasi berdasarkan parameter nilai Rf dan visualisasi warna. Analisis kuantitatif spektrofotometer UV-Vis menghasilkan nilai kadar Rhodamin B tiap sampel masing-masing yang dihitung dengan menggunakan rumus regresi y= 0,0726 x + 0,0432 dan nilai R2 = 0,9957.  Sampel kode A1 sebesar 3,443 ± 0,080 μg/mL , B1 sebesar 3,057 ± 0,150 μg/mL, B2 sebesar 5,881 ± 0,170 μg/mL, C1 sebesar 3,884 ± 0,130 μg/mL, dan C2 sebesar 4,517 ± 0,140 μg/mL. Abstract

ABSTRACT This study aims to determine the total phenol content and Minimum Bactericidal Concentration (MBC) and the correlation of antibacterial activity with the total phonel content of the extract and the fraction of Abemoschus manihot L. against Escherichia coli. Gallic acid was used as a comparison standard in measuring the total phenol content using a UV-Vis spectrophotometer. The total phenolic conten of gedi leaf extract and fraction from Southeast Minahasa were ethanol extract 230.23 ± 0.008 µg GAE / g, n-hexane fraction 238.37 ± 0,024 µg GAE / g, ethyl acetade fraction 269.77 ± 0.016 µg GAE / g, and ethanol fraction 221.51 µg GAE / g. The extracts and fractions were tested for antibacterial activity against E. coli  by dilution method using Nutrient Broth (NB) media at concentrations of 1000 µg / mL,500 µg / mL, and 250 µg / mL. MBC test data (Minimum Bactericidal Concentration) were analyzed using Kolmogrov-smirnov, then continued with pearson correlation test. The result of the normality test showed that the extract and the A.manihot fraction had significantly distributed normal vlue (p> 0.05). Analysis Correlation of Pearson of extracts and fraction of A. manihot with antibacterial activity against E.coli showed significant result (p <0.05). 71% of antibacterial activity is influenced by total phenolic and the rest is contribution of other compounds that have the potential as antibacterial. Keywords: Phenol Total, Correlation, Escherichia coli, Green Gedi ABSTRAK Penelitian ini bertujuan untuk mengetahui kandungan fenol total dan Kadar Bunuh Minimum (KBM) serta korelasi aktivitas antibakteri dengan kandungan fenol total ekstrak dan fraksi Abelmoschus manihot L. terhadap Escherichia coli. Asam galat digunakan sebagai standar pembanding dalam mengukur kandungan fenol total menggunakan Spektrofotometer UV-Vis. Kandungan total fenolik pada ekstrak dan fraksi daun gedi hijau dari Minahasa Tenggara yaitu ekstrak etanol sebesar 230,23 ± 0,008 µg GAE/g, fraksi n-heksan 238,37 ± 0,024 µg GAE/g, fraksi etil asetat 269,77 ± 0,016 µg GAE/g, dan fraksi etanol 221,51 µg GAE/g. Ekstrak dan fraksi diuji aktivitas antibakteri terhadap E.coli dengan metode dilusi menggunakan media Nutrient Broth (NB) pada konsentrasi 1000 µg/mL, 500 µg/mL, dan 250 µg/mL. Data uji KBM (Konsentrasi Bunuh Minimum) dianalisis menggunakan Kolmogrov-smirnov, kemudian dilanjutkan dengan uji korelasi pearson. Hasil uji normalitas menunjukkan bahwa ekstrak dan fraksi A.manihot memiliki nilai signifikan terdistribusi normal (p > 0,05). Analisis korelasi pearson ekstrak dan fraksi A.manihot dengan aktivitas antibakteri terhadap E.coli  menunjukkan hasil signifikan (p <0,05). 71% aktivitas antibakteri dipengaruhi oleh fenolik total dan sisanya merupakan kontribusi senyawa lain yang berpotensi sebagai antibakteri. Kata kunci : Fenol Total, Korelasi, Escherichia coli, Gedi Hijau

Abstract

Formalin is a solution which made from 37% formaldehyde in water which is commonly used as preservative for cosmetic products and various types of non-food industrial materials. The use of formalin as a preservative in food is very dangerous for health. The purpose of this study was to determine the presence of formalin found in skewered meatballs circulating in several elementary schools in the city of Manado. Sampling was done in Malalayang Elementary School (A), State Elementary School 6 Manado (B), State Elementary School 9 Manado (C) and State Elementary School 29 Manado (D). In a qualitative test, Nash reagents were used to determine the presence of formalin in the sample. Nash reagent was used to indicate the presence of Formalin content in a sample marked with a change in color to yellow. The results of this study showed that twelve samples of skewers from four elementary schools in Manado City did not contain formalin or safe from formalin use.   Keywords: Formalin, Formaldehyde, Skewered Meatballs, Nash Reagent, Qualitative Analysis  ABSTRAK Formalin merupakan larutan yang dibuat dari 37% formaldehida dalam air yang umumnya digunakan sebagai bahan pengawet produk kosmetik dan berbagai jenis bahan industri non makanan. Penggunaan formalin sebagai bahan pengawet pada makanan sangat berbahaya bagi kesehatan. Tujuan penelitian ini yaitu untuk mengetahui keberadaan formalin yang terdapat pada bakso tusuk yang beredar di beberapa Sekolah Dasar di Kota Manado. Pengambilan sampel dilakukan di SD Negeri Malalayang (A), SD Negeri 6 Manado (B), SD Negeri 9 Manado (C) dan SD Negeri 29 Manado (D).  Pada pengujian kualitatif digunakan pereaksi Nash untuk mengetahui adanya formalin pada sampel. Pereaksi Nash ini digunakan untuk menunjukkan adanya kandungan Formalin pada sampel yang ditandai dengan perubahan warna menjadi kuning. Hasil penelitian ini menunjukkan bahwa dua belas sampel bakso tusuk dari empat Sekolah Dasar di Kota Manado tidak mengandung formalin atau aman dari penggunaan formalin. Kata Kunci: Formalin, Formaldehida, Bakso Tusuk, Pereaksi Nash, Analisis Kualitatif

Abstract

The presence of free radicals produced inside the cells is thought to cause a range of diseases such as degenerative diseases. Antioxidant properties of carrot root (Daucus carota L.) have been reported elsewhere to inhibit the formation of these free radicals, but the compounds responsible for antioxidant activity inside its leaves have not yet determined. In Indonesia, Daucus carota L. leaves are regarded as waste product after harvesting the root. This study was conducted to isolate and identify the compound having antioxidant activity from the leaves of D.carota. n-Hexane, ethyl acetate, and methanol extracts were examined for free radical scavenger activity by using DPPH method. The n-hexane extract showing free radical scavenger activity was solid-solvent partitioned, fractionated using vacuum liquid chromatography and tested for free radical scavenger activities. The active compound was isolated by using preparative thin layer chromatography and its purity was determined by TLC with three mobile phase systems and two dimensional TLC. Based on spectroscopy UV-Vis, IR as well as 1H-NMR (CDC13) data, the active compound was suggested to be xanthophyll derivate, lutein. The compound at concentration of 0.616, 1.025, and 2.05 ppm had antioxidant activities of 0.94 ± 0.05; 18.53 ± 0.15; and 49.07 ± 0.86, respectively.

Abstract

Sago Baruk (Arenga microcarpha) is one of endemic crop type of Archipelago of Sangihe Talaud, North Sulawesi and potential as source bioactivities including antioxidant. The objectives of this research were to determine the chemical characteristics and antioxidant activity of starch sago baruk which was sequentially extracted with water and filtrate. The sago trunks pith was sequentially extracted with water and filtrate at room temparature for 1 hour. After that, starch of sago baruk were analyzed for their chemical composition (moisture, fat, protein, ash crude fiber and charbohydrate) and total phenolic and tannin condensed content. Antioxidant activity of each sago flour were evaluated in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and total antioxidant capacity. After that, the starch was characterized by IR spectroscopic technique. The sequential extraction result indicated percentage yield of starch I and starch II were 41.39% and 38.21%. The chemical characteristics of starch I revealed that percentage protein, ash, crude fibre and fat were below 1% than starch II especially protein and fiber crude. The starch I had highest total phenolic and tannin condensed content than starch II and starch K. The starch I also showed the highest free radical scavenging activity in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical than starch II and starch K. The result also showed that starch I has strongest total antioxidant capacity than starch II and starch K at all concentration level. Antioxidant activity of three starch increased with the increase in concentration of the samples. The three starch showed of hidroxy (OH), carbonyl (C=O), ether (C-O) and aliphatic group (C-C). It is concluded that starch sago baruk especially the starch I are very rich in antioxidant substituens.

Abstract

Purslane is a plant that thrives in open areas and has a fairly high water content. Hereditary purslane plant has been used for skin protection from the sun. The aim of this study is to determine the value of Sun Protecting Factor (SPF) of ethanol extracts, soluble and insoluble fractions of ethyl acetate from purslane herb.Extraction using maceration method by ethanol 90%. Fractionation process is the partition obtained soluble and insoluble fractions of ethyl acetate. Absorbance values read using UV-VIS spectrophotometry at a wavelength of 290-320 nm. SPF value is obtained for the ethanol extract 19.495, 20.829 for the soluble fraction of ethyl acetate and 30.055 for an insoluble fraction of ethyl acetate.
