Abstract

Bakteri asam laktat (BAL) merupakan mikroorganisme yang aman ditambahkan ke dalam pangan. Bakteri asam laktat mampu menghambat bakteri patogen. Salah satu dari kelompok bakteri asam laktat yang menghasilkan antibakteri adalah genus Lactobacillus. Penelitian ini bertujuan untuk mengetahui isolat bakteri asam laktat Lactobacillus sp. (KG61) berpotensi dalam menghambat pertumbuhan bakteri Escherichia coli dan Staphylococcus aureus dan lama waktu inkubasi yang baik isolat bakteri asam laktat Lactobacillus sp. (KG61) dalam menghambat pertumbuhan E. coli dan S. aureus. Pengujian uji daya hambat menggunakan metode sumuran pada media Nutrient Agar (NA) dengan perlakuan inkubasi 7 hari dan 14 hari serta ciprofloxacin (kontrol positif) dan aquades steril (kontrol negatif) dengan waktu inkubasi 24 jam dan 48 jam. Hasil penelitian menunjukkan bahwa bakteri asam laktat berpotensi dalam menghambat E. coli dan S. aureus yang baik pada inkubasi 14 hari dengan rata-rata zona hambat 7,76 mm dan 31,17 mm dengan kategori sedang dan sangat kuat

Abstract

White rice flour (Oryza sativa L.) is the main ingredient used by the Dayak community in the manufacture of tape yeast, the tape yeast is one of the substrates for the growth of microorganisms, one of which is yeast. Yeast belongs to the group of fungi that are unicellular in shape and have been isolated from various kinds of traditional fermented foods that are consumed such as tape. The purpose of this study was to determine the genus of yeast isolated from tape yeast made from white rice flour. This research was conducted in December 2021 – April 2022. Yeast isolates were identified based on macroscopic and microscopic observations of colonies from yeast cells, cell reproduction, and physiological tests with reference to the book The Yeast a Taxonomic Study. Based on the results of this study, 12 yeast isolates were obtained consisting of three genera, namely Kluyveromyces, Pichia and Saccharomyces.

Abstract

Simpur (Dillenia sp.) is a plant that can be used as traditional antibacterial medicine. This study aims to determine the inhibitory activity of Staphylococcus aureus bacteria when exposed to a concentration of methanol extract from Simpur leaves (Dillenia sp.). This study utilized a completely randomized design with five concentration levels of Dillenia sp. specifically, the study included a control group and methanol extracts of Simpur leaves at concentrations of 5%, 10%, 15%, and 20%. The methods used included total plate count, hemocytometer, and UV-Vis spectrophotometry, which were then analyzed using ANOVA (Analysis of Variance). The results showed that the greatest inhibition occurred in the methanol extract of Simpur leaves (Dillenia sp.) at a concentration of 20%, there was a decrease in the number of colonies ranging from 90% to 94%, while the decrease in the number of cells ranged from 17% to 18%. The lowest inhibition was found in the methanol extract of Simpur leaves (Dillenia sp.) with a concentration of 10%, the proportion of decreasing the number of colonies ranged from 63% to 70%, while the proportion of decreasing the number of cells ranged from 6% to 7%. The highest optical density (OD) value at a 20% concentration ranged from 0.482 to 0.547, while the lowest value at a 5% concentration ranged from 0.127 to 0.131. The conclusion of this study is that the methanol extract of Simpur leaves (Dillenia sp.) Bacteriostatic antibiotics only inhibit bacterial growth and cannot kill bacteria.

Abstract

Nypa palm worms (Namalycastis rhodochorde: Polychaetes) play important ecological roles as detritivores of organic matter in mangrove ecosystems, bioindicators of water pollution, and sediment stabilizers in estuarine environments. N. rhodochorde also shows potential for use in fermented feed-based aquaculture due to its high nutritional value as a natural food source for aquatic organisms. Cellulolytic lactic acid bacteria isolated from the coelomic fluid and gastrointestinal tract of N. rhodochorde, identified as isolates NrLtC4 and NrLtG2, have potential as starter cultures for fermenting niypa palm fronds, which are used as feed in nypa palm worm cultivation. This study aimed to evaluate the growth of juvenile N. rhodochorde fed nypa palm fronds fermented by the cellulolytic bacteria NrLtC4 and NrLtG2. Juveniles were maintained under consistent stocking density, container size, and water volume, with three treatments arranged in a completely randomized design: unfermented nypa palm fronds (control), nypa palm fronds fermented with isolate NrLtC4, and nypa palm fronds fermented with isolate NrLtG2. The results demonstrated that fermented feed significantly influenced both the number of body segments [p = 0.00021 and 0.0028 (p < 0.05)] and body length [p = 0.0020 and 0.0035 (p < 0.05)] of the juveniles. Juveniles fed nypa palm fronds fermented with strain NrLtC4 showed increases of 29.6% in segment number and 58.5% in body length, while those fed fronds fermented with strain NrLtG2 exhibited increases of 36.6% and 63.9%, respectively. The highest juvenile survival rate was observed in the NrLtC4 fermented feed group at 72%, followed by 69.33% in the NrLtG2 group. These findings suggest that bacterial isolates NrLtC4 and NrLtG2 have promising potential as a consortium feed to enhance the growth of nypa palm worms in aquaculture.

Abstract

Desa Rasau Jaya II merupakan kawasan yang didominasi oleh masyarakat petani. Pengetahuan masyarakat tentang cara mengatasi rendahnya kesuburan tanah melalui penggunaan pupuk organik sangat perlu ditingkatkan. Di Rasau Jaya II, sampah organik dan limbah Tandan Kosong Kelapa Sawit (tankos) tersedia dalam jumlah melimpah, mencapai 21–23%, sehingga berpotensi diolah menjadi kompos untuk meningkatkan kesuburan tanah. Melalui program PKM dilakukan pemberdayaan masyarakat melalui pemberian pengetahuan dan keterampilan dalam mengolah sampah serta limbah organik menjadi kompos. Tujuan dari pelatihan ini adalah meningkatkan pengetahuan masyarakat mengenai teknik pembuatan kompos dari limbah organik. Metode pelaksanaan PKM dilakukan melalui  pelatihan ketrampilan singkat. Pelatihan dilaksanakan melalui metode ceramah, demonstrasi penggunaan biodekomposer serta praktik langsung pembuatan kompos. Evaluasi kegiatan pengabdian masyarakat dilakukan melalui penyebaran kuesioner pada akhir sesi pelatihan Kegiatan telah berhasil meningkatkan pengetahuan dan keterampilan masyaraka, terlihat dari keberhasilan pembuatan kompos pengurangan penggunaan pupuk kimia, dan kebersihan lingkungan akibat pengurangan sampah.
