Abstract

Omah Iwak Badher Bank ialah salah satu destinasi wisata di Kabupaten Blitar yang dikelola oleh POKMASWAS Fajar Bengawan. Penelusuran reviewer pada google.co.id mendapatkan total 77 orang pemberi ulasan dengan rata-rata rating 4.5. Penelitian ini bertujuan untuk menilai rating destinasi terhadap responden terpilih, ialah mahasiswa S1 dan S3 Fakultas Perikanan dan Ilmu kelautan UB Malang. Total responden berjumlah 46 mahasiswa, 33 ialah mahasiswa S1 dan 13 mahasiswa S3. Kedua kelompok responden mendapat kesempatan yang sama untuk mengunjungi destinasi wisata. Rating dan penilaian terhadap destinasi dilakukan dengan memberikan 12 jenis pertanyaan kepada masing-masing resonden. Hasil penelitian mendapatkan bahwa rating responden mahasiswa S1 (3,7±0,57) dan S3 (3,7±0,48) lebih rendah dibandingkan dengan penilaian publik (4,5). Hal ini menunjukkan kualitas destinasi yang menjadi perhatian mahasiswa lebih tinggi dibandingkan dengan umum. Kesediaan membayar mahasiswa S1 (Rp. 150.000 Trip-1) pada destinasi wisata relatif lebih rendah dibandingkan dengan mahasiswa S3 (Rp. 250.000 Trip-1). Sebagai destinasi wisata baru, Omah Iwak Badher Bank perlu melakukan peningkatan jasa untuk meningkatkan kualitas destinasi.Kata Kunci: kesediaan membayar, jasa wisata, wisata alam, POKMASWAS Willingness-To-Pay To Omah Iwak Badher Bank Tourism DestinationABSTRACT Omah Iwak Badher Bank is one of the tourist destinations in Blitar Regency which is managed by POKMASWAS Fajar Bengawan. A reviewer search on google.co.id found a total of 77 reviewers with an average rating of 4.5. This study aimed to assess the destination rating of selected respondents, are undergraduate and doctoral students of the UB’s Faculty of Fisheries and Marine Science Malang. The total respondents were 46 students, 33 were S1 students and 13 were S3 students. Both groups of respondents received the same opportunity to visit tourist destinations. Rating and assessment of destinations was done by giving 12 types of questions to each respondent. The results found that the average rating of undergraduate student respondents was 3.7±0.57, and 3.7±0.48 for S3 students, respectively. These were lower than that of public assessment (4.5). This indivated that the quality of destination that are of concern to students is higher than the public. Willingness-t-pay of undergraduate students (Rp. 150,000 Trip-1) at tourist destinations was relatifly lower compared to doctoral students (Rp. 250,000 Trip-1). As a new tourist destination, Omah Iwak Badher Bank needs to improve its services to improve the quality of the destination.Keywords: willingness-to-pay, tourism services, nature tourism, POKMASWAS

Abstract

Efforts to cultivate snakehead fish (Chanidae) still experience many problems, especially the availability of high-quality fry. To improve the quality of fry, improving the quality of broodstock is important. Genetic factors play an important role in the selection and breeding of snakehead fish parents. Therefore, it is necessary to explore sources of superior parents with good genetic characteristics. To achieve this goal requires studies from various fields such as ecology, biology, and genetics. Genetic studies can be started by examining the identity and relationships of snakehead fish from Merauke, as a first step in finding a source of germplasm that has high diversity. The aim of this research is to analyze the cytochrome oxidase subunit 1 (COI) gene fragment of snakehead fish from Merauke, Papua. DNA isolation was carried out using the Promega kit with standard protocols, amplification of the COI gene with primers F2 and R2 with the Go Taq Green amplification kit. A total of 653 base pairs of the COI gene fragment were obtained from the sequencing process, with a nucleotide composition of T (29.9%), C (28.8%), A (24.0%), G (17.3%). BLAST results on the NCBI database show that this sequence has 100% similarity to the Channa striata sequence from Papua with sequence ID OQ405388.1. This shows that the identification results of the snakehead fish from Merauke are Channa striata. Phylogenetic tree analysis also showed consistent results, namely the formation of a clade that was the same as the Channa striata fish group derived from GenBank data.

Abstract

Nitrogen plays a significant role in the growth and metabolism of microalgae. The purpose of this study was to investigate the influences of different sodium nitrate concentrations on growth, biomass, and biochemical profile in Tetraselmis chuii. Four different nitrate concentrations, 0.5, 1.0, 1.5, and 2.0 g/L NaNO3 were applied in T. chuii culture under a batch system. It was found that a low nitrate concentration of 0.5 g/L NaNO3 produced the highest specific growth rate and biomass concentration of T. chuii. On the other hand, increasing nitrate concentration led to an increase in chlorophyll a+b and carotenoid in T. chuii, with the optimum nitrate concentration found at 1.5 g/L NaNO3. Under the nitrogen limitation condition, protein content was significantly decreased, but lipid and carbohydrate content were highly accumulated in the cells. This study provides a unique phenomenon that low nitrogen concentrations not only produce higher biomass but also accumulate high lipid and carbohydrate content.

Abstract

Mastacembelus sp., a member of the Mastacembelidae family, is distributed across Java, Sumatera, and Kalimantan. Mastacembelus, locally named berod fish in the Brantas River lacks population’s descriptions and morphological analyses. Morphological analyses, including morphometric and meristic descriptions, are important for precise identification and classification of kinship relationships, serving as a cornerstone in the fields of biology, taxonomy, and conservation. This study aimed to conduct a comprehensive morphometric and meristic identification of the berod fish population in the Brantas River, East Java. A total of 37 samples were collected from 6 different locations: Blitar, Tulungagung, Kediri1, Kediri2, Nganjuk, and Mojokerto. The morphometric measurements were based on 25 characters, while the meristic calculations were based on 6 characters. The data analyzed with PCA and CA using SPSS and PAST programs. The results revealed that berod fish in the Brantas River had morphological characters belongs to the Mastacembelus unicolor. However, notable disparities were observed in meristic characters, particularly in the Dorsal Fin Spines (DFS). PCA analysis showed significant differences in morphometric characters but not supported by meristic characters. These results underscore the importance of integration both morphological and meristic analyses for a comprehensive understanding of the berod fish population.

Abstract

Nilem fish (Osteochilus vittatus) is a species of freshwater fish from the Cyprinidae family which has ecological and economic importance in Indonesia, with its natural habitat in rivers, swamps and stagnant waters. Accurate species identification is very important in biological research and fisheries resource management, but morphological identification methods have several limitations. DNA barcoding is a revolutionary molecular technique that uses short DNA sequences to identify and differentiate species with high accuracy. The use of DNA barcoding in identifying freshwater fish in Indonesia has shown significant development, including in a comprehensive study documenting 1,172 native freshwater fish species. The research "Genetic Identification of Nilem Fish using the DNA Barcoding Approach" aims to characterize the genetics of nilem fish, compare the genetic profiles of various populations, and evaluate the effectiveness of DNA barcoding in distinguishing nilem fish from other Osteochilus species, with the hope of contributing to fish conservation and sustainable management efforts nilem in Indonesia. The DNA sequencing results obtained a sequence with a length of 619 base pairs with the composition of Nucleotides A (27.2%), G (17.6%), C (26.4%), T (28.8%). The BLAST results with the NCBI database showed a similarity of 99.84% with the O. vittatus sequence and 99.83% with O. hasseltii. Both species are synonyms with the official name O. vittatus. Phylogenetic analysis is also in line with the BLAST results which show that the sample sequence is in the same clade as the O. vittatus and O. hasseltii sequences. Identification with molecular markers of the COI gene was successfully used in determining the species of the Nilem fish (O. vittatus).

Abstract

The Sentani snakehead fish (Oxyeleotris heterodon) is an endemic fish species that inhabits Lake Sentani and several other lakes in Papua Province, Indonesia. This species has important ecological and economic value, but is vulnerable to environmental change and overexploitation. Genetic identification, especially using the Cytochrome Oxidase 1 (COI) gene, is very important for conservation efforts and understanding the genetic diversity of Sentani Snakehead Fish. DNA barcoding techniques with the COI gene have become the standard method in molecular identification of species, allowing rapid and accurate identification. Unfortunately, the Sentani Snakehead Fish faces various threats such as habitat degradation, pollution, overfishing and the introduction of invasive foreign species. Therefore, genetic research is becoming increasingly important to support conservation efforts, providing in-depth understanding of the population status and adaptive potential of species. The results of DNA amplification and sequencing of COI gene fragments showed a DNA sequence with a length of 619 base pairs with the composition A (23.9%), G (18.1%), C (29.1%), T (28.9%). The results of identification by homology analysis in the NCBI and BOLD databases showed the highest similarity at 97.58% and 97.84% with the O. lineolate sequence. This is because there is no O. heterodon sequence from the COI gene fragment found in Genbank or the BOLD system. Phylogenetic tree analysis with several sequences that have close similarities shows a difference in clade between the O. heterodon sequence from this study with the O. lineolate sequence and other species.

Abstract

Intensive cultivation of Tilapia with high stock density and continuous artificial feeding will decrease the water quality and increase the growth of pathogenic bacteria. That situation led to the fish stressed so that the fish become the target of bacterial pathogen attacks. Alternatively, the environmental friendly treatment using herbal immunostimulant to solve that problem is required. The herbal in this study used neem leaf (Azadirachta indica A. Juss). The aim of this study was to evaluate ethanol fraction of neem leaf on immunostimulant activity of Oreochromis niloticus. In this study, the extraction method used maceration. Fractionation methods were using Thin Layer Chromatography (TLC) and Column Chromatography (CC). The biological component was analyzed using Phytochemical, FTIR and GC-MS methods. TLC test showed that ethyl acetate: n-hexane (1:1, v/v) result in brightest luminescence colors on the plates of silica gel under UV light (365 nm). The chromatography column from the ethanol extract of neem leaf (A. indica A. Juss) showed that separation of the fraction was done based on the formation of color, which obtained 5 (five) types of fractions. The results of the phytochemical test for the fraction 4 of neem leaf contains two compounds, i.e. flavonoids (+) and triterpenoids (++). The FTIR test showed that fraction 4 of neem leaves contain a class of terpenoid and flavonoid compounds. GCMS test results on fraction 4 of the neem leaves (A. indica A. Juss) obtained the high peak, which has 4 terpenoid compounds, 1 compound of flavonoid, and 1 type of lauric acid. The total leukocyte of the fish test increased after treated with fraction 4 of neem leaf (A. indica A. Juss) compared with control on day 7 after injection, i.e. 25.29 x 104 cells.mL-1.Keywords: fraction, leukocyte, Neem Leaves, Tilapia, Phytochemicals.

Abstract

Medicinal plants as an antimicrobial agent may provide an alternative way to replace the use of antibiotics to control disease agents in aquaculture. Roselle flowers (Hibiscus sabdariffa L.) has been used in many sectors as a source of functional food, natural coloring agents as well as antimicrobial agents. The objectives of this study were to evaluate the phytochemical compound in methanolic extract of roselle flower and their antimicrobial activity against Aeromonas hydrophila. The phytochemical composition of roselle flower was evaluated using phytochemical screening and FTIR. While the antimicrobial activity was performed by using the disc diffusion agar and co-culture with A. hydrophila. The results of phytochemical screening confirmed the presence of alkaloids, flavonoids, saponins, steroids, triterpenoids, and tannins. The results of FTIR revealed that Roselle flower extract had the main phenolic compounds. The result of disc diffusion and co-culture method indicated that the roselle flower extract had antibacterial activity against A. hydrophila. Abstract

Aeromonas hydrophila is a type of gram-negative bacteria that can harm humans and major animals that have poikilotherm properties such as fish and shrimp. These bacteria can cause haemorrhages in fish until death and causing fish farming production to failure. The use of medicinal plants has been trusted by people all over the world to overcome various disease problems, one of which is a disease caused by bacteria. Plantago major L. is known that able to inhibit the growth of both gram-positive and gram-negative bacteria. The purposes of this study were to identify the extracted compounds contained in Plantago major L., using FTIR and UV-VIS spectrophotometer and to evaluate antibacterial activity against A. hydrophila. The results of this study showed that crude extracts of Plantago major L. contained polar compounds such as phenols, flavonoids, saponins, and tannins. The presence of phenols and flavonoids was confirmed by the results of FTIR and UV-VIS. Furthermore, the crude extract of Plantago major L. significantly inhibited the growth of A. hydrophila (P <0.05).Keywords: Antibacterial, Co-culture, Extraction, FTIR, UV-VIS.

Abstract

The limitations of fish in digesting food depend on the presence of enzymes protease, amylase, and lipase that react with substrates in the digestive channel of fish. Supplementation methods can be performed to increase fish growth by adding prebiotics and probiotics to the feed. This study aims to determine the influence of the difference in the length of time fermented prebiotic feed (sweet potato extract) and probiotics (Bacillus megaterium) on the quality of feed. The results of the physical analysis of fish feed showed the color looks brown, fishy smell, texture, and general conditions of feed seemed completely normal. Chemical analysis showed that the proteins were best improved in B3 treatment, with the period of fermentation approximately 72 hours. Biological analysis suggests that the longer the fermentation time taken, the higher the abundance of bacteria obtained.Keywords: B. megaterium, chemistry and biology of fish feed, physical analysis, sweet potatoes (Ipomoea batatas L)
