Abstract

Escherichia coli (E. coli) is an indicator bacteria that is often used to assess the level of microbial contamination in food and plants. This study aims to isolate and identify the presence of E. coli from Coleus amboinicus plants which are known to have antibacterial activity. Fresh leaf samples of C. amboinicus were taken from the home yard environment. Isolation was carried out using the serial dilution method and planting on eosin methylene blue (EMB) placement media. Identification was carried out based on colony morphology, Gram staining, and biochemical tests including IMViC (Indole, Methyl Red, Voges-Proskauer, Citrate). The results showed the presence of metallic green colonies on EMB, which showed the characteristics of E. coli. Biochemical tests showed Indole (+), Methyl Red (+), Voges-Proskauer (-), and Citrate (-), which confirmed the isolate as E. coli. The presence of E. coli in C. amboinicus indicates the possibility of environmental fecal contamination, and is a concern in the processing and consumption of this medicinal plant.

Abstract

Agar is the main medium used in microbiology laboratories to grow bacteria and fungi because of its ability to form stable gels that are not easily degraded by microorganisms. However, the availability of expensive commercial agar and dependence on imports is a challenge for small-scale laboratories. This literature study aims to review the potential of natural materials such as local seaweed (Gracilaria sp.), banana peels, cassava starch, and coconut pulp as alternative media for microorganism growth. The results of the study indicate that Gracilaria has the ability to form gels similar to commercial agar, while banana peels and cassava starch are rich in nutrients but require the addition of thickeners. The main challenge lies in the sterilization process and media consistency. The use of these materials can be a cheap and sustainable solution in the development of microbiology media based on local resources.

Abstract

This study aims to isolate and identify microorganisms in the form of bacteria and fungi in tilapia fish waste and to demonstrate the effectiveness of turmeric extract (Curcuma longa Linn.) as an antimicrobial agent. Tilapia fish waste samples were fermented for seven days and then microorganisms were isolated using Nutrient Agar (NA) media for bacteria and Potato Dextrose Agar (PDA) for fungi. Turmeric extract treatment was carried out using the disc diffusion method. The results showed the presence of several species of Gram-positive and Gram-negative bacteria, as well as fungal colonies with dominant characteristics of Aspergillus sp. and Fusarium sp. Turmeric extract with a concentration of 20% showed a significant inhibition zone against the growth of pathogenic bacteria and fungi. The content of secondary metabolites such as alkaloids, tannins, and phenolics in turmeric extract is thought to be the main factor in antimicrobial activity.

Abstract

White glutinous rice fermentation (tape ketan putih) is a traditional food product containing various microorganisms, including Gram-positive bacteria that may act as contaminants and reduce product quality. Ginger (Zingiber officinale) contains active compounds such as gingerol and shogaol, known for their antibacterial properties. This study aimed to evaluate the antibacterial activity of ginger extract against microorganisms isolated from white fermented glutinous rice. Methods: Ginger extract was obtained by maceration using ethanol as a solvent and tested by disk diffusion method on Nutrient Agar against isolated microorganisms. Results: An inhibition zone was observed around the ginger extract disk, indicating moderate antibacterial activity against Gram-positive bacteria. Conclusion: Ginger (Zingiber officinale) extract shows potential as a natural antibacterial agent against Gram-positive bacteria in tape ketan putih and may be developed as a natural preservative for fermented products.

Abstract

Staphylococcus Aereus is a common us of skin and soft tissue infection. The aim of study was to evaluate the effectivaness of Betadine solution, Sos, and hand sanitizer as antimicrobial agent against S. Aereus. The study used a laboratory based experimental design, Where S. aereus was exposed to paper discs soaked in betadine, SOS, and hand sanitizer, the betadine disc showed a more significant antimicrobial effect than other This study shows that betadine is more effective than the others. Testing is carried out using the diffusion method where paper discs that have been soaked in antimicrobial substances are attached to the surface of agar media that has grown bacteria. In this way, a clear zone will be visible surrounding the disc paper as a sign of the microbe's sensitivity to the antimicrobial.

Abstract

Breast cancer is the second leading cause of cancer-related deaths in women, after cervical cancer. To date, breast cancer treatment still faces various challenges, particularly in terms of effectiveness and therapy-related side effects. Therefore, alternative treatments that are safer and more effective are needed, one of which is through the use of natural ingredients such as soursop leaves (Annona muricata). This review article aims to examine the potential of soursop leaf extract in slowing the growth of breast cancer cells. Several studies have shown that soursop leaf extract contains active compounds such as acetogenin, which are known to inhibit cancer cell proliferation, induce apoptosis, and block the formation of new blood vessels (anti-angiogenesis) in cancer tissues. These bioactive compounds make soursop leaves a promising candidate for natural anticancer agents. Thus, soursop leaf extract may be considered as a complementary therapy in the management of breast cancer.

Abstract

Microorganisms are microscopic organisms that cannot be observed directly without the aid of optical instruments. They play an important role in various biological and environmental processes, both beneficial and detrimental. Most microorganisms are unicellular, but some are multicellular. Some microorganisms are also known to have benefits in the fields of food and health. Bamboo shoots or young bamboo are one type of vegetable that is widely consumed by the community, especially in Central Java. Bamboo shoots have a low nutritional content such as water, protein, carbohydrates, minerals, and fat, making them a healthy low-calorie food. In addition, bamboo shoots also contain bioactive compounds such as vitamins, essential amino acids, and antioxidants that are beneficial for body health.Fermentation of bamboo shoots is one method to increase their nutritional value and probiotic content.

Abstract

Cinnamon bark (Cinnamomum burmannii) is known to contain antibacterial active compounds such as cinnamaldehyde and eugenol. However, as a natural material collected from open environments, it can become a habitat for various microorganisms, including bacteria. This study aimed to isolate and observe bacterial growth found on cinnamon bark originating from Kerinci. Samples were collected from a local market and processed using serial dilution, then cultured on Nutrient Agar (NA) and incubated for 24–48 hours at room temperature. The results revealed the growth of bacterial colonies with diverse morphological characteristics, such as round shape, smooth edges, and cream to cloudy white colors. Gram staining indicated the presence of both Gram-positive and Gram-negative bacteria. Preliminary biochemical tests showed both catalase-positive and catalase-negative isolates. These findings suggest that despite the antibacterial properties of cinnamon bark, certain microorganisms are capable of surviving. The detected bacteria are likely to be either endophytic or environmental contaminants that have adapted. These results provide an initial insight into the local microbial diversity and open opportunities for further exploration of bacteria with biotechnological potential, while also supporting post-harvest quality control of spice products.

Abstract

Buah nanas (Ananas comosus) merupakan komoditas tropis yang umum dijual di pasar tradisional dan sering dikonsumsi dalam keadaan segar, sehingga rentan terhadap kontaminasi mikroba. Penelitian ini bertujuan untuk mengidentifikasi jenis mikroorganisme yang terdapat pada permukaan buah nanas yang dijual di tiga pasar tradisional di Kota Jambi, yaitu Pasar Simpang Pulai, Pasar Keluarga, dan Pasar Mama. Sampel buah diambil secara purposive dan dianalisis di Laboratorium Universitas Adiwangsa Jambi menggunakan metode isolasi pada media Nutrient Agar (NA) dan Potato Dextrose Agar (PDA), serta dilakukan uji Gram dan uji biokimia. Hasil penelitian menunjukkan adanya kontaminasi mikroba berupa bakteri Staphylococcus aureus, Escherichia coli, Bacillus subtilis, dan Klebsiella pneumoniae, serta jamur Aspergillus niger, A. flavus, dan Penicillium sp.. Tingkat kontaminasi tertinggi ditemukan pada buah dari Pasar Mama. Temuan ini menunjukkan pentingnya peningkatan sanitasi dan edukasi penanganan pangan di pasar tradisional guna menjamin keamanan konsumsi buah segar.

Abstract

Duruan (Durio zibethunus L.) merupakan buah tropis musiman yang hanya berbuah satu kali dalam setahun dan termasuk golongan buah klimaterik, yaitu buah yang mengalami proses pematangan lebih cepat. Untuk memperpanjang daya simpan yang lebih lama daging buah durian diolah melalui fermentasi menjadi tempoyak. Tempoyak merupakan makananan khas Sumatra terutama jambi, tempoyak adalah makanan fermentasi dari daging durian (Durio zibenthinus L.) dengan cara daging buah durian yang sudah matang yang dilepas dari bijinya, ditambah dengan garam dan disimpan dalam toples/wadah selama kurang lebih 3 hari.. Penelitian ini bertujuan menganalisis mikroorganisme yang terkandung didalam fermentase tempoyak khas jambi, dengan menggunakan metode cawan sebar (Spread Plate Method) yaitu penanaman bakteri pada media pertumbuhan yang sesui seperti agar (NA), proses ini meliputi sterilisasi, pengenceran, inokulasi menggunakan metode Spread Plate pada media Nutrient Agar (NA), inkubasi pada suhu 30–37°C selama 24–48 jam, dan pengamatan morfologi koloni serta karakteristik mikroskopis isolat. Hasil penelitian menunjukkan bahwa tempoyak mengandung BAL dengan karakteristik koloni berwarna putih susu, berbentuk bulat, tepi rata hingga bergelombang, dan permukaan cembung. Secara mikroskopis, BAL yang diisolasi berbentuk batang (basil) dan bulat (coccus), bersifat gram positif, tidak membentuk endospora, nonmotil, dan katalase negatif. Berdasarkan karakteristik tersebut, BAL dalam tempoyak berpotensi sebagai probiotik yang mendukung kesehatan pencernaan dan keseimbangan mikroflora usus.
