Abstract

The number of tuberculosis (TB) cases worldwide reached 1.5 million in 2018; thus, TB is considered a deadly disease. TB is caused by Mycobacterium tuberculosis and involves lipid synthesis. Considering the importance of lipid metabolism in bacteria, FabH may be an essential protein target for repressing lipid synthesis. Capsaicinoids from Capsicum annuum demonstrate potent antibacterial activity. This study predicted the ability of capsaicinoid compounds to inhibit FabH. In silico analysis was performed by retrieving the structure of FabH from PDB and those of selected capsaicinoid derivatives from PubChem. The compounds were docked using AutoDock Vina in PyRx 0.8 software. The interactions of FabH and different capsaicinoid derivatives showed identical binding characteristics. The bonding type most frequently observed was hydrogen bonds. In conclusion, capsaicinoid derivatives could block lipid synthesis through FabH. The relevant mechanism and biological processes should be studied further.

Abstract

This study aims to explore the chemical compounds of the coffee pulp as an anti-covid-19 (anti-3C-like protease). In silico modeling was carried out by structure retrieval, protein preparation, and docking simulation. Specific Grid docking was used for ligands – protein interacting by using Molegro Virtual Docker 5, followed by visualizing Discovery studio version 21.1.1. Compounds contained in coffee pulp, quinic acid, gluconic acid, and ferulic acid separately bind to the same amino acid residues of 3CL Protease or each other. Furthermore, three compounds inhibited 3CLPro activity by catalytic site blocking of 3CLPro, indicating that coffee compounds have potential as molecular therapeutic materials for the coronavirus by inhibiting SARS-CoV-2 genome replication.

Abstract

Learning development is one of the innovations in improving student achievement, one of which is by adopting the Problem Based Learning model combined with the Student Worksheet (LKPD). This model of developments is adapted to the material on biodiversity. The purpose of this study is to validate and analyze the feasibility of the resulting LKPD. The development research method adopted is the research and development of the Four-D (4D) model. The results of the LKPD research were valid by material experts (80%), media experts (87.78%), and linguists (81.25%) with a very valid category and obtained a very decent feasibility score. The LKPD developed is expected to be a solution to classroom learning.

Abstract

Community service activities at SD Negeri Gembira, Sikka Regency are to create a reading corner in the library. The purpose of creating a reading corner is to increase students' interest and motivation to read, encourage better library management, and help students to be able to take advantage of free time by reading. The activity of making a reading corner is an effort made as an answer to the problems found in the learning process in the classroom, namely that there are still students who have not been able to manage smoothly even though they have been in class VI. The use of the reading corner is made to target specifically for class VI students but does not close the opportunity to all students of SD Negeri Gembira. A series of PkM activities have been carried out from August to November 2022. All components at SD Negeri Gembira strongly support the creation of a reading corner. Learners have shown progress in learning especially reading proficiency.

Abstract

Tuberculosis (TB) is a disease with the second number of cases in Indonesia. The therapy process is carried out by utilizing one of the waste materials, namely squid ink bags, which are predicted to have active compounds. In the TB cycle, the enzyme FabH is catalyzed as a mediator of lipid synthesis. Inhibition of FabH became the main target through the role of squid ink compounds. The squid ink has a potential as bactery inhibitor. The purpose of this study was to analyze the potential of squid ink as an alternative to TB therapy by inhibiting lipid synthesis (FabH). In silico approach was used in this research. Bioactive compound 5,6-dihydroxyindole-2-carboxylic acid (DHICA) (CID: 119405), cinnamic acid (CID: 444539), betaine (CID: 247) were downloaded from PubChem, and FabH protein (PDB ID 2QO1) was downloaded from the protein data bank database. (GDP). Docking used HeX 8.0.0.0, visualization and analysis using Discovery studio ver 21.1.1. Results from the analysis, we found compounds 5,6-dihydroxyindole-2-carboxylic acid (DHICA), cinnamic acid, betaine contained in squid ink molecularly have potential as an alternative TB therapy by inhibiting FabH, the interaction lead to treatment the TB. In vitro and in vivo studies are needed to analyze further before human studies.

Abstract

An interesting teaching and learning process will further motivate students to be active in learning. Processes that are standard and tend to be rigid make students bored and disinterested in learning, moreover science learning materials usually keep students motivated. The concept of contextual science learning needs to be done, so that they experience real learning more. One of them is learning with a contextual approach is outing class. Learning that is directly in contact with nature. This PKM activity involved 25 grade 4 students at SDN Napungbiri Pante, Waigete District, and Sikka Regency. PKM is carried out by involving students to be actively involved in learning with the outing class model. The results of this PKM activity show that the level of student participation reaches 100% with very good motivation, namely 94%. These results indicate that outing class activities have a good impact on increasing student motivation, which in the end will have implications for increasing students' cognitive scores.
