Abstract

The purpose of this study was to analyze the effects of age on the biochemistry profiles of aceh cattle, which included examinations of: total protein (TP), albumin, globulin, albumin/globulin ratio (A/G), aspartate aminotransferase (AST), gamma-glutamyltransferase (GGT), alkaline phosphatase (ALP), creatine kinase (CK), blood urea nitrogen (BUN), calcium (Ca), phosphorus (P) and magnesium (Mg) . Sixteen clinically healthy aceh cattle aged 2-4 years old, and 16 aceh cattle aged 4-6 years old were purposively selected for the study. Blood samples were drawn from the jugular vein and subjected to blood biochemistry measurements using commercial kits. The results showed that serum concentrations of TP, globulin, AST and Ca of aceh cattle were significantly (P0.05) higher in age group 4-6 years old, while ALP activity was significantly (P0.05) higher in age group 2-4 years old. In conclusion, age must be considered as a factor when interpreting the blood biochemistry profiles of aceh cattle.

Abstract

The aim of this study was to determine the best combination of cryoprotectant (Ethylene glycol, EG), dimethyl sulfoxide (DMSO) and propanediol (PrOH) and carrier system (hemistraw and cryotop) in improving ewe oocytes viability during cryopreservation. Oocytes with multi layers of compact cumulus cells were colleted from abbatoir and matured in TCM 199 medium supplemented with 10% fetal calf serum for 24-26 h at 38.5 C under 5% CO2 in the air. Matured oocyte was divided into six parts and vitrified in three different vitrification solutions; (i) 17% EG+17% DMSO with hemistraw as carrier system, (ii) 34% EG with hemistraw as carrier system, (iii) 17% EG+17% PrOH in hemistraw (iv), 17% EG+17% DMSO with cryotop as carrier system (v), 34% EG with cryotop as carrier system (vi), and 17% EG+17% PrOH in cryotop. Oocytes were cryopreserved for one week before revived and evaluated for viability. The result showed that oocytes vitrified in media containing EG and DMSO in cryotop had the highest viability (88.16%) compared to media containing EG only or EG and PrOH (70.95% and 68.76%, respectively) (P0.05). Moreover, oocytes viability that vitrified using cryotop and hemistraw as carrier system were not significantly different. The present results indicated that vitrification using combination of EG and DMSO as permeable cryoprotectant and cryotop as carrier system was the best system to maintain oocyte viability after vitrification-warming.

Abstract

Penelitian ini bertujuan mengetahui efektivitas enrofloksasin terhadap infeksi bakteri pada saluran pencernaan ular sanca batik (Python reticulatus). Ular yang digunakan berjumlah 10 ekor dan terindikasi klinis mengalami gangguan pencernaan berupa keradangan pada mulut. Sampel yang diambil adalah swab mulut dan kloaka untuk pemeriksaan mikrobiologi berupa isolasi dan identifikasi bakteri pada media brilliant green agar, Mc Conkay agar, triple sugar iron, dan pembiakan isolat murni. Setelah pengambilan sampel semua ular diinjeksi dengan enrofloksasin 5 mg/kg bobot badan, dosis tunggal secara intramuskular anterior. Pengamatan klinis dilakukan hingga semua ular dinyatakan sembuh dari keradangan mulut. Hasil pemeriksaan mikrobiologi menunjukkan adanya bakteri Salmonella sp., E. coli, dan Proteus sp. pada saluran pencernaan ular. Enrofloksasin yang diberikan secara injeksi intramuskular anterior mampu memberikan kesembuhan dalam rentang waktu 4-16 hari setelah pemberian.

Abstract

Research was conducted to detect APMV-1 infection by examining microscopic lesions of chicken suspected ND and confirming the causative agent with serological and molecular assay. Samples obtained from commercial and back yard farm in 9 regencies and city of Bali Province were tested by rapid test for AIV antigen detection. AI negative samples were necropsied, then brain, lungs, and intestines were collected for histopathological examination. Samples tissue of brain, lung, spleen, and intestine were taken aseptically for viral isolation and amplification. Infected allantoic fluid was collected and tested by hemagglutination assay (HA) and hemagglutination inhibition (HI) test to prove APMV-1 serologically. Viral ribonucleic acid was isolated and subsequently reverse transcribed by reverse trasncription reaction followed by amplification by polymerase chain reaction to multiply the cDNA. Microscopically, perivascular cuffing (20%), endoteliosis (75%), and gliosis (75%) were found in the brain. In the lung, an interstitialis pneumonia (50%), lobar pneumonia (5%), and proliferation of pneumosit type 2 (100%) were observed. The most prominent intestinal lesions were catarrhal enteritis (75%) and hemorrhagic necrotizing enteritis (10%). Confirmation of the 20 isolates obtained in this study both serologically and molecularly were positive APMV-1. Moreover PCR results showed that 80% of its amplicon showed a single band and 20% still require some optimizations to get single good bands.

Abstract

Penelitian ini bertujuan mengetahui peningkatan tumor necrosis factor-alpha (TNF-) dan indeks apoptosis pada tulang mencit yang diinfeksi Toxoplasma gondii (T. gondii). Tiga puluh dua ekor mencit dibagi menjadi dua kelompok. Kelompok 1 (K1), merupakan kelompok kontrol, tidak diinfeksi sedangkan Kelompok 2 (K2) diinfeksi dengan 10 takizoit T. gondii secara intraperitoneal. Enam hari setelah infeksi mencit dikorbankan, diambil tulang femur dan dilakukan pembuatan preparat histologis dengan pengecatan immunohistochemistry (IHC) dan Tunel assay. Hasil penelitian menunjukkan jumlah sel tulang yang mengekspresikan TNF- pada K2 (27,046,92) berbeda sangat nyata dibandingkan dengan K1 (11,423,92). Indeks apoptosis pada K1 dan K2 masing-masing adalah 9,173,04 dan 16,283,37. Dari hasil penelitian disimpulkan bahwa infeksi T. gondii meningkatkan TNF- dan indeks apoptosis sel tulang femur.

Abstract

This study determines effects of raw material feed (derived from corn, bran, soybean pods, concentrates and Indigofera zollingeriana local to Magelang) on the performance of native chicks. Performance was measured by feed consumption, body weight gain, Feed Conversion Ratio (FCR), mortality, and morbidity. This experiment used Completely Randomized Design (CRD) with three different feed treatments, and each treatment consisted of 30 free-range female chicks as replicants, from 0-4 weeks old. Super native chick feed was formulated by trial and error by mixing all ingredients: corn, bran, soybean pods, Indigofera zollingeriana, and premix. After mixing, a proximate test determined the nutritional content of the feed. Feed was then administered to chicks. Quantity of feed was administered according to age. The results obtained in the study were analyzed descriptively and statistically using the Variance Test (ANOVA) for the variables of body weight gain, feed consumption, and FCR. The Kruskal Wallis test was used via SPSS 26 program for mortality and morbidity variables with three treatments (T1, T2, and T3) and three replications. Each treatment consisted of 30 research individuals. Results showed that the super native chicks on varied protein diets had a significant difference in performance (P0.05), with feed 3 performing the best feed 3 produced a feed consumption of 430.20 g/chick and body weight gain of 174.67 g/chick, along with 2.46 FCR, 0% mortality, and 3.33% morbidity. The conclusion this study is holistic feed made from local ingredients with the addition of Indigofera zolliengeriana had a significant effect on increasing average daily gain, increasing feed consumption, and reducing feed conversion ratio (FCR) in native super chick.

Abstract

This study aimed to determine the occurences of mice brain ventricles dilatation that congenitally infected with Toxoplasma gondii (T. gondii) as a marker of hydrocephalus and cellular changes in the brain. A total of twenty pregnant mice (11.5 days pregnacy) were divided into 2 groups, which were control (P1) group and treatment (P2) group. The mice in the treatment group were infected with 101 tachyzoites of T. gondii. All mice were maintained until delivery. The newborn mice were sacrificed and their brain were removed and fixed in 10% buffered formalin to prepare histology slides with HE staining for observation of ventricular width, TUNEL assay for apoptosis observation, and immunohistochemistry for the expression of transforming growth factor beta (TGF-) observations. The data were analyzed using t test and linear regression. The results showed that ventricular width and apoptosis index significantly increased (P0.01) in the treatment group compared to control group, but there was no difference in the expression of TGF- (P0.05) in both groups. Dilatation of ventricle correlated with the apoptotic index of brain cells but did not correlated with the expression of TGF-.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak vesikula seminalis terhadap penurunan konsentrasi progesteron sertadiameter korpus luteum pada tikus putih. Dalam penelitian ini digunakan 30 ekor tikus putih (Rattus norvegicus) betina dewasa, galur Wistar, berumur 3-4 bulan dengan berat badan antara 200-250 g dan dibagi atas dua kelompok (K1 dan K2) masing-masing diberi 25 g cloprostenol dan 0,2 cc ekstrak vesikula seminalis secara intravaginal pada hari ke-7 kebuntingan. Tiga ekor tikus masing-masing kelompok dibunuh pada jam ke-0, 3, 6, 12, dan 26. Pemeriksaan progesteron dilakukan menggunakan metode enzymelinkedimmunosorbantassay (ELISA). Konsentrasi progesteron pada kelompok perlakuan PGF2 dan ekstrak vesikula seminalis pada lima periode waktu pengukuran yakni jam ke-0, 3, 6, 12, dan 26 memperlihatkan perbedaan yang signifikan (P0,05). Ekstrak vesikula seminalis mempunyai kemampuan yang sama dengan PGF2 komersial dalam menurunkan diameter korpus luteum yang ditandai secara mikroskopis dengan berkurangnya vaskularisasi darah menuju ovarium (P0,05). Disimpulkan bahwa ekstrak vesikula seminalis mempunyai kemampuan yang lebih baik dibandingkan dengan PGF2 komersial dalam menurunkan konsentrasi progesteron tikus putih dan mempunyai kemampuan yang sama dengan PGF2 komersial dalam menurunkan diameter CL.

Abstract

This study aimed to determine the presence of Salmonella spp. in duck meat and to identify the resistance of Salmonella spp. against several antibiotics in duck meat associated with food safety. Total of 52 meat samples of ducks was collected from 5 subdistricts in Bogor District, i.e., Cariu-Jonggol, Gunung Sindur, Klapanunggal, Parung Panjang, and Ciomas based on the assumption of 95% confidence level, 50% predicted prevalence, and 10% standard error. The results showed that three samples (5.8%) were positive Salmonella spp. The majority of Salmonella spp. isolated from duck meat showed resistance against erythromycin (66.7%), streptomycin (33.3%), and chloramphenicol (33.3%). Nevertheless, Salmonella spp. was still sensitive againts enrofloxacin, trimethoprim sulfamethoxazole, cephalothin, ampicillin, nalidixid acid, tetracycline, and gentamicin.

Abstract

Penelitian ini bertujuan mengetahui efek antikolesterol fraksi n-heksana ekstrak rumput kebar pada hewan model hiperlipidaemia. Delapan kelinci dewasa dibagi dalam dua kelompok. Kelompok kontrol (K1) hanya diberikan diet tinggi lemak selama 1 bulan dan pada kelompok perlakuan (K2) diberikan diet tinggi lemak ditambah fraksi n-heksana ekstrak rumput kebar. Uji keberhasilan penelitian dilakukan dengan pemeriksaan kolesterol total serum darah. Hasil penelitian menunjukkan konsentrasi kolesterol total serum darah pada K1 dan K2 masing-masing adalah 100,0018,28 dan 47,7512,53 mg/dl. Dari hasil penelitian disimpulkan bahwa fraksi n-heksanaa mampu menurunkan tingkat kolesterol total dalam serum darah.
