Abstract

This study aimed to determine the antibacterial activity of soursop flower extract (Annona muricata L.) and hematological profile of rats (Rattus norvegicus) due to administration of soursop flower ethanol extract and Salmonella enteritidis infection. The concentrations of soursop flower ethanol extract used for the antibacterial activity test were 25%, 50%, 75%, 100%, with the antibiotic ampicillin 10 g/disk was used s a positive control (PC) and dimethyl sulfoxide (DMSO) 10% as a negative control (NC). For examination of hematological features, 15 male rat aged two months were used. All rats were divided into 5 treatment groups, each consisting of three rats. The NC group was not given soursop flower ethanol extract and S. enteritidis infection. The PC group was not given soursop flower extract but was given S. enteritidis infection. Groups P1, P2, and P3 were given ethanol extracts of soursop flower at a dose of 0.18, 0.36, and 0.72 g/rat/day peroral for a week using gastric sonde. On the following day (after administration of soursop flower extract) the first blood drawing was performed. All rats, except NC group, were then infected with S. enteritidis intraperitonially at dose of 3x108 CFU/mL dose (0.5 mL McFarland). One week after being infected with S. enteritidis, a second blood drawing was performed. The results of the antibacterial activity test showed that there was no antibacterial activity was observed since no inhibition at various concentrations was formed. The administration of soursop flower extract at various dosage levels was able to maintain the number of leukocytes but reduced the number of erythrocytes, hemoglobin concentration, hematocrit value and the number of platelets in rats; whereas S. enteritidis infection decreased all the hematologic features of lab rats.

Abstract

The objective of this study is to observe the relationship of development of adrenocorticotropic hormone (ACTH) cells of adenohypophysisadrenal cortex axis during pre and postnatal period of longtailed monkey (Macaca fascicularis). Five fetuses aged 70, 85, 100, 120, and 150 days of gestation and 2new born aged 10 and 105 days old were used as research materials. The sections of the pituitary gland stained immunohistochemically using human ACTH antiserum and the adrenal glands stained by Hematoxylin-Eosin procedures. At the fetus 120 days old, the ACTH cells and blood vessel of fetaladenohypophysis developed well and its similar that found in older age. This condition suggested distributing the ACTH to whole body optimally. At the same time, the adrenal cortex cells of fetus 120 days old were active in proliferation and differentiation processes to form transition zone that lies between definitive and fetal zones, and central vein also developed in adrenal gland. In fetus aged 150 days, the cortex adrenal structure was similar with found in older fetus. The adenohypophysis ACTH cells developed earlier than adrenal cortex and their secretion suggested influencing the development of adrenal cortex cells of long-tailed monkey.

Abstract

Penelitian ini bertujuan mendiagnosis kasus newcastle disease (ND) lapangan pada ayam bukan ras yang bersifat akut melalui hasil pemeriksaan di Laboratorium Patologi dan Laboratorium Biomedik Fakultas Kedokteran Hewan Universitas Udayana tahun 2008-2009. Sebanyak sepuluh ekor sampel ayam buras telah diperiksa. Gejala klinis yang teramati meliputi: anoreksia, lesu, bersin, batuk, dan diare putih kehijauan dengan diagnosis sementara sebagai penyakit ND yang bersifat akut. Sampel diambil dari organ yang mengalami perubahan patognomonis seperti pada proventrikulus, ventrikulus, seka tonsil, paru-paru dan otak. Perbanyakan virus menggunakan telur ayam bertunas (TAB) umur 10 hari melalui ruang alantois dan diinkubasikan pada inkubator telur suhu 37 C selama 3 hari. Koleksi cairan alantois dilakukan pada hari ke-3, selanjutnya diidentifikasi dengan uji serologi hemaglutinasi (Haemaglutination-Inhibition Test/HA/HI) dengan teknik mikrotiter baku dan dikonfirmasi dengan uji reverse transcriptase-polymerase chain reaction (RT-PCR) menggunakan primer FNDIFP (5-CCCCGTTGGAGGCATAC-3) dan FNDIBP (5-TGTTGGCAGCATTTTGATTG-3). Hasil penelitian menunjukkan bahwa semua sampel kasus ayam bukan ras yang diperiksa positif terinfeksi oleh virus penyakit ND akut. Kajian ini mengindikasikan bahwa penyakit ND di Bali masih bersifat endemis.

Abstract

The research was conducted after tsunami destroyed mangrove in Aceh Besar coastal area, whichserves as waterfowl habitat. This study was aimed to find out bird diversity and abundant after tsunamidisaster. The study was conducted in Kecamatan Mesjid Raya, Aceh Besar, focusing on bird biodiversityand its number based on habitat type, using count dot survey technique. The study revealed that as manyas eight waterfowl species from four families were found. The variety of bird species in the researchlocation was low, ie; 1.2196 for fish pond, 1.7257 for muddy land, and 1.2528 for vegetation area. The lowlevel of biodiversity indicated fragmented habitat in the mangrove area.Keywords: mangrove conservation, bird biodiversity index, bird conservation

Abstract

Penelitian ini bertujuan mengetahui distribusi glycoconjugate yang terekspresi pada sel epitelium oviduk kancil (Tragulus javanicus). Dalam penelitian ini digunakan satu oviduk kancil yang berasal dari satu ekor kancil b etina dewasa berumur lebih dari satu tahun. Sampel difiksasi dengan larutan Bouin dan diproses menurut standar histologi sampai menjadi blok parafin dan dipotong dengan ketebalan 5 m. Jenis lektin yang digunakan adalah biotinylated (Con A, PNA, RCA, UEA I, dan WGA) dengan dosis masing-masing sebanyak 15 g/ml. Hasil penelitian diketahui bahwa glycoconjugate dengan residu gula galaktosa, glukosa, manosa, N-asetilgalaktosamin, N-asetilglukosamin, fukosa, dan asam sialat ditemukan pada bagian apikal sel epitel dan di dalam sitoplasma. Glycoconjugate dengan residu gula N-asetilgalaktosamin merupakan glycoconjugate yang paling banyak ditemukan di bagian apikal sel epitel dan di dalam sitoplasma dibandingkan dengan glycoconjugate dengan residu gula lainnya.

Abstract

Penelitian ini bertujuan mengetahui kemampuan perkembangan awal embrio sapi in vitro menggunakan media tunggal untuk maturasi, fertilisasi, dan kultur berbahan dasar tissue culture medium (TCM) 199. Oosit sapi dikumpulkan dari rumah potong hewan dengan teknik aspirasi dan diklasifikasikan berdasarkan kekompakan sel kumulus dan sitoplasma yang homogen. Oosit dimaturasi pada medium TCM 199 yang disuplementasi dengan 10 IU/ml pregnant mares serum gonadotropin (PMSG), 10 IU/ml human chorionic gonadotropin (hCG), dan 10% fetal bovine serum (FBS), dilakukan selama 24 jam pada inkubator 5% CO2, 39 C. Fertilisasi dilakukan pada dua media yang berbeda yaitu media rutin fertilisasi dan media berbahan dasar TCM 199 dengan suplemen bovine serum albumin (BSA) dan heparin. Setelah fertilisasi, kumulus sel dihilangkan (denudasi), kemudian dikultur pada media TCM 199 yang disuplementasi dengan asam amino esensial dan non-esensial serta 10% FBS selama 3 hari. Hasil penelitian menunjukkan tingkat maturasi oosit pada sistem yang digunakan mampu mendukung 81,5% oosit mencapai tahap metafase II (M-II). Tingkat pembelahan embrio lebih tinggi pada media rutin dibandingkan dengan media TCM 199 yakni masing-masing 44,4 dan 23,2%. Jumlah embrio tahap 4-8 sel pada kedua perlakuan tidak berbeda nyata. Dapat disimpulkan media tunggal berbasis TCM dapat digunakan untuk produksi embrio in vitro.

Abstract

The aim of this research was to investigate the potential effect of methanolic extract of Scurrula atropurpurea (MESA) in reducing oxidative stress through observation of superoxide dismutase (SOD) activity and malondialdehyde (MDA) concentration in mice. In this research, 25 male mice were divided into four groups with five replications. Group I (K1, without MESA) was considered as a control group, while Group II (K2), III (K3), and IV (K4) were given 1000, 2000, and 4000 mg MESA/kg BW, respectively. After 14 days, SOD activity and MDA concentration were measured spectrophotometrically. The SOD activity did not significantly increased (P0.05) in group K1, K2, K3, and K4 with values were 19.4692.072, 17.41.373, 20.533.411, and 23.872.528 U/ml, respectively. In contrast, MDA concentration was decreased (P0.05) with values of 492.75019.418, 492.75019.418, 457.12530.095, 454.62519.080, and 475.87537.659 ng/mL in groups K1, K2, K3, and K4, respectively. It can be concluded that the administration of MESA tend to increase SOD activity and decrease MDA concentration in this study, thus might be able to reduce oxidative stress in mice.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak epididimis (EE) terhadap peningkatan kualitas spermatozoa kambing jantan lokal. Dalam penelitian ini digunakan 12 ekor kambing jantan lokal, berumur 1,5 tahun dengan bobot badan 10-15 kg dan dibagi atas empat kelompok (K0, KP1, KP2, dan KP3). Kelompok K0, hanya diinjeksi dengan NaCl fisiologis sedangkan kelompok KP1, KP2, dan KP3 diinjeksi EE masing-masing 1, 2, dan 3 ml/ekor selama 13 hari berturut-turut. Pada hari ke-14, dilakukan pengambilan semen kambing dengan elektroejakulator dan selanjutnya dilakukan pemeriksaan kualitas spermatozoa. Hasil penelitian menunjukkan bahwa pemberian EE dengan dosis 1 dan 3 ml/ekor EE selama 13 hari berturut-turut menyebabkan peningkatan kualitas spermatozoa dibanding kelompok kontrol. Berdasarkan hal tersebut dapat disimpulkan bahwa EE berpotensi meningkatkan kualitas spermatozoa pada kambing jantan lokal.

Abstract

This study investigated the influences of rainy and dry seasons on semen characteristics of fat-tailed and Garut rams under tropical conditions. A total of 4 healthy rams (2 fat-tailed rams and 2 Garut rams) aged 3 to 4 years old were used in this study. The semen was collected from each ram during November 2016 to February 2017 (rainy season) and May to August 2017 (dry season). The measurement of semen characteristics including semen volume (SV), sperm concentration (SpC), total sperm (TSp), individual sperm motility (ISM), post-thawing sperm motility (PTSM), and frozen semen production (FSP). The rainy season resulted higher SV (P= 0.021), TSp (P= 0.005), ISM (P= 0.028), and FSP (P= 0.005) than the dry season. On the other hand, fat-tailed rams had higher (P0.001) SV, TSp, and FSP compared to Garut rams. There was no interaction (P0.05) between season and breed on all semen characteristic parameters found in the present study. It can be concluded that the semen collection during the rainy season had a beneficial impact to produce high-quality semen for artificial insemination program than in the dry season. Fat-tailed rams have better semen characteristics compared to Garut rams.

Abstract

The purpose of this research was to determine the effect of decoction of pulutan leaves simplicia(Urenalobata L.) on the development of the uterus of Balb C mice (Musmusculus). Female mice aged 10-12 weeks were given decoction of pulutan leaves simplicia with concentration of 5%, 7.5%, 10%, 12.5%, 15%, and control. The administration of pulutan leaves was performed by gavage for 11 consecutive days. On day 12, mice were dissected, then, the uterus was taken for histological preparations using paraffin method. Data such as uterine diameter and myometrium and epimetrium thickness were analyzed using one way analysis of variance followed by least significant different. The results showed that decoction of pulutan leaves simplicia decrease the uterine diameter and epimetrium and myometrium thickness compared to control. The decrease of uterine diameter and uterine myometrium and epimetrium thickness was started at the concentration of 7.5%. Decoction of pulutan leaves simplicia could reduce uterine diameter and thickness of uterine myometrium and epimetrium, thus, it might decrease the uterine function as the site of implantation. The decline in the function of the uterus might decrease the number of born children, thus decoction of pulutan leaves simplicia is potential as antifertility agent.
