Abstract

Penelitian ini bertujuan mengetahui pengaruh getah jarak cina (Jatropha multifida L.) terhadap pertumbuhan bakteri Staphylococcus aureus. Sampel yang digunakan dalam penelitian ini adalah Stapylococcus aureus dengan no Cat ATCC 25923 diperoleh dari Laboratorium Mikrobiologi Fakultas Kedokteran Hewan Universitas Syiah Kuala. Getah  jarak cina diperoleh dari Desa Rukoh Kecamatan Syiah Kuala,diambil dengan cara menyayat batang jarak dan ditampung dalam tabung reaksi (Pyrex TM) sebanyak 50 ml. Penelitian ini dilakukan dengan 3 kali ulangan menggunakan konsentrasi 25; 50; 75; dan 100% v/v getah jarak cina. Kontrol positif digunakan kertas cakram antibiotik Vankomisin 30 µg. Daya hambat getah jarak cina menggunakan metode Kirby Bauer, dengan jumlah bakteri sesuai dengan standar Mc Farland I. Data yang diperoleh dianalisis secara deskriptif. Zona hambat yang terbentuk diukur dengan jangka sorong (Caliver TM). Hasil penelitian pada konsentrasi 25; 50; 75; dan 100% v/v zona hambat yang terbentuk masing-masing adalah 13; 13,5; 14,6;  dan 15,7 mm. Zona hambat yang terbentuk dariantibiotik vankomisin 30 µg adalah 21 mm. Dari hasil penelitian dapat di simpulkan bahwa getah jarak cina (Jatropha multifida L.) dapat menghambat pertumbuhan Staphylococcus aureus.

Abstract

Newcastle Disease (ND) is a highly contagious disease with a high mortality rate, infecting poultry, and chickens are very susceptible to ND infection. The purpose of this case report is to find out how to diagnose ND disease in chickens. A 30-day-old white male broiler cadaver obtained from Pasar Lambaro, Aceh Besar was found dead in a holding cage with conjunctival hemorrhage and hair loss. Based on the physical examination of the external condition of the cadaver, it can be seen that the eye membranes are experiencing hemorrhage. There was nasal mucosa hemorrhage and mucous exudate in the nasal cavity. The oral mucosa was also found to have mucous exudate, while the cloaca was bleeding and there was residual green feces. The results of anatomic pathology examination found ptechie in the proventriculus and ventricles, swollen lungs with blackish red color and hemorrhage,with the presence of cheese. There was hemorrhage in other organs including trachea, intestines and tonsils, whilst the brain experienced hyperemia. The spleen was swollen and there were foci of necrosis. Based on the anamnesis, external examination, pathological anatomical examination, and histopathology, the diagnosis of the cause of death of broiler chickens was Newcastle Disease (ND).

Abstract

The aim of this study was to see the antimicrobial activities of oxytetracycline to the inhibition of Staphylococcus aureus biofilm of Aceh’s cattle preputium isolates. The parameters measured in this study were the percentage of antimicrobial and biofilm inhibitory power. The antimicrobial activities were tested on Nutrient Broth using U-bottom polystyrene microplate 96 wells. The Biofilm stained using 1% of crystalviolet. Then, Optical Density (OD) read at λ 595 nm. The results showed that the oxytetracycline of 10%, 20% and 30% (μg / μL) had the antimicrobial abilities of 28.60% ± 13.23%; 36.30% ± 5.37%; and 57.70% ± 2.23%. In addition, The biofilm inhibitory capacity of oxytetracycline 10%, 20% and 30% (μg / μL) were 2.20% ± 8.77%; 2.70% ± 4.17%; and 3.73% ± 14.14%. Therefore, it can be concluded that the oxytetracycline  had the ability to inhibit the biofilm of Staphylococcus aureus.

Abstract

The aim of this study was to find out the information about sumatran orangutan urine leucocyte as a medical record of them at Cagar Alam Pinus Jantho Reintroduction Center, Aceh Besar. Urine samples were collected from ten sumatran orangutans which come from forest, around cages, and in the cage. The urine was collected in the morning when orangutan woke up and left the nest or before first feeding for orangutans which were in the cages. The replication test was done six times within one month. Urine examination was carried out directly after urine collection by soaking the strip test in 5 ml urine for 120 second. The data was analyzed by qualitative descriptive method and the result was semiquantitative based on the discoloration scale: negative, trace (±15 leucocyte/μL), +1 (70 leucocyte/μL), +2 (125 leucocyte/μL), and +3 (500 leucocyte/μL). The result showed that the average of leucocyte from 4 orangutans which were in the cages (Ego, Wenda, Miky, and Ontong) was ±15 leucocyte/μL, while Ayu Ting Ting for the first test had the highest level of leucocyte (500 leucocyte/μL) then decreased on the next test. Coty, Ruben, Yusniar (arround the cages) had variative leucocyte values every week. Two orangutans in the forest (Kamelia and Brokoli) had negative value for urine leucoyte.Key words: leucocyte, sumatran orangutan, strip test, urine

Abstract

Penelitian ini bertujuan mengetahui perbandingan jumlah mikroba antara ayam normal dengan ayam yang mengalami bruises. Sampel ayam broiler diperoleh di Pasar Lambaro Aceh Besar dengan memilih lima musculus pectoralis ayam normal dan lima musculus pectoralis ayam yang mengalami bruises untuk dilakukan uji mikrobiologis. Bagian musculus pectoralis ditimbang dengan berat 25 g, kemudian dilakukan stomaker untuk mendapatkan ekstrak. Selanjutnya dilakukan uji mikrobiologis, perhitungan jumlah koloni dilakukan selama 24 jam. Jumlah mikroba pada sampelayam normal masing-masing adalah Sn1 (1,6x102), Sn2 (2,6x103), Sn3 (3,5x102), Sn4 (1,3x103), dan Sn5 (1,2x102) Cfu/ml, sedangkan pada sampel ayam yang mengalami bruises masing-masing adalah Sb1 (3,8x104), Sb2 (3,0x104), Sb3 (1,3x105), Sb4 (1,9x105), dan Sb5 (1,6x105) Cfu/ml. Berdasarkan hasil penelitian dapat disimpulkan bahwa ayam yang mengalami bruises memiliki jumlah mikroba yang lebih tinggi dibandingkan dengan ayam normal.

Abstract

Penelitian ini bertujuan menentukan dan membandingkan persentase spermatozoa hidup dari kauda epididimis pada tikus strain Wistar dan Sprague-Dawley. Dua puluh ekor tikus jantan (Rattus norvegicus) yang terdiri dari 10 ekor strain Wistar dan 10 ekor strain Sprague-Dawley berumur 5 bulan digunakan dalam penelitian ini. Pada saat berumur 2,5 bulan seluruh tikus diadaptasikan selama 75 hari di dalam kandang dan diberikan pakan Turbo Feed T.79-4 (PT. Central Protein Prima, Medan) dan minuman secara ad libitum. Setelah masa adaptasi selesai, setiap hari diambil masing-masing dua ekor strain Wistar dan Sprague-Dawley untuk dibedah dan diambil kauda epididimisnya. Selanjutnya dibuat preparat hapus semen dengan menggunakan pewarnaan eosin untuk menghitung jumlah spermatozoa hidup. Preparat diamati di bawah mikroskop dengan 5 lapangan pandang. Data yang diperoleh dianalisis menggunakan uji T. Hasil penelitian menunjukkan bahwa terdapat perbedaan rata-rata persentase spermatozoa yang sangat nyata (P0,01) antara strain tikus Wistar (92,00±2,83%) dan Sprague-Dawley (90,70±4,37%). Disimpulkan bahwa persentase spermatozoa hidup pada tikus Wistar lebih tinggi daripada tikus Sprague-Dawley

Abstract

Cats are pets which part or all of their lives depend on humans for certain things. The period of pregnancy and post partus are very crucial periods for cats. It required certain management approach which is different from general maintenance. This study aimed to find out the perception of cat owners about the cat's management during the pregnancy and post partus period. The research data were obtained by filling out the online questionnaires distributed to the member of cat lover’s Banda Aceh. Data were analyzed descriptively. This study found that the perception of cat owners during pregnancy and post partus was positive.

Abstract

This study aimed to determine the cellulase activity in broilers digestive tract after the treatment of soya wastes and palm kernel fermented by A.niger (AKBIS Prob) in the ration. This study used a complete randomized factorial design, which consists of two factors, namely the AKBIS Prob composition factor and the duration of AKBIS Prob. Twenty four broilers were divided into four groups by randomized and three replications of each. The feed supplement combinations were P0 (commercial), P1 (commercial and AKBIS Prob 2%), P2 (commercial and AKBIS Prob 4%), and P3 (commercial and AKBIS Prob 6%). The intestine preparation was taken at the 22 and 36 days to extraction and detection the cellulase activity concentration. Data were analyzed using variance analysis of factorial patterns. AKBIS Prob 2,4 and 6% was no effect (P 0.05) to the cellulase activity in the small and large intestine, while the treatment duration was significant (P0,05) in the small intestine but neither in the large intestine (P0,05) to the cellulase activity. The addition of AKBIS Prob 2.4 and 6% had no effect (P 0.05) on the concentration of cellulase activity in the small intestine and large intestine, while the duration the treatment was very significant (P 0.05) in the small intestine and neither in the large intestine (P 0.05) on the concentration of cellulase activity. The AKBIS administration in the ration for 22 days can increase the activity and concentration of cellulase activity in the small intestine of the digestive tract of broiler chickens.

Abstract

This research was aimed to isolate the Gram-positive bacteria that contaminated milk processed with ultra high temperature (UHT) at 6 and 3 months before the date expires. A number of 3 different UHT milk products at 6 and 3 months before expire date were used as sample. Milk samples were taken from several supermarket in Banda Aceh. The identification was conducted using Gram staining method. One ml milk sample was transferred into 18 tubes containing nutrient broth (NB) media and incubated at 37 C for 24 hrs. The sample was then culture on 18 petri discs containing manitol salt agar (MSA) and 18 petri discs containing trypticase soy sucrose bacitracin (TYS20B) using streak plate method. After 2 days, bacterial colonies that grow were identified by Gram staining. The results showed that in UHT processed milk at 6 and 3 prior to expiration contaminated with Gram positive cocci bacteria, which comprised of Staphylococcus aureus and Streptococcus mutant. As conclusion, UHT processed milk at 6 and 3 prior to expiration is contaminated by Gram positive cocci bacteria.

Abstract

This study aimed to determine the effect of using Moringa leaves fermented flour in feed stock toward the increasing level of hemoglobin and hematocrit value on broiler chickens. This study used a completely randomized design that consists of 4 treatments and 5 replications. The study group was divided into group P0 which was given with 100% of commercial feed, group P1 was given 95% of commercial feed and Moringa leaves fermented flour 5%, group P2 was given 92.5% of commercial feed and Moringa leaves fermented flour 7.5%, and group P3 was given 90% of commercial feed and Moringa leaves fermented flour 10%. Fermentation using EM4 (Effective Microorganism-4) were able to decrease the crude fiber of Moringa leaves flour. The treatment was conducted for 15 days, then hemoglobin levels and hematocrit values were counted. Data were analyzed using ANOVA completely randomized design with SPSS for Windows 16.0. This research showed that giving Moringa leaves fermented flour for 15 days did not have a significally effect (P0.05). The given Moringa leaves fermented flour in  feed for 15 days with 5%, 7.5% and 10% concentration was not show any increase in the levels of hemoglobin and hematocrit values.
