Abstract

Selenium is a trace element that has antioxidant properties and is a component of selenoproteins. In certain circumstances, such as injury, selenium supplementation is required to regulate the inflammatory response. Contusion injury increases the inflammatory response to a muscle or tissue that begins with vasodilation, increases blood flow, increases vascular permeability, and leads to edema formation at the site of injury. Objectives: To determine the effect of selenium supplementation on changes in blood markers in rats treated with contusion injury. Methods: Experimental design, using experimental male rats (Rattus norwegius), eight weeks old, weighing 200-220 grams, divided into three groups, namely: The Control group, the contusion group, the contusion + Selenium group. Selenium supplementation was given orally on days 1-3 after injury at a dose of 0.0153 mg into 2% PGA. Examination of markers such as White Blood cells (WBC), lymphocytes, monocytes, and neutrophils was carried out on day three after treatment. Results: This study found that the administration of post-muscular selenium supplementation based on examination of blood markers such as WBC, Lymphocytes, Monocytes was higher in the contusion group and lower in the selenium group. Neutrophils were higher in the contusion and selenium groups. This was not significantly different from the control group. The result of blood marker examination tended to change, but it was not statistically significant. Conclusion: Selenium supplementation in post-contusive injury mice did not provide significant changes in blood markers such as WBC, lymphocytes, monocytes, and neutrophils.Keywords:  antioxidant, blood marker, contusion, selenium

Abstract

The immunosurveillance system in the human body can detect and eradicate abnormal cells. However, cancer cells can avoid immune destruction by exploiting the immunosuppression system. Extensive research has been done to find therapeutic agents that could reverse this condition. Novel drugs called checkpoint inhibitors have been developed and showed a promising result. Besides, “living’’ drug, an engineered T-cells which can accurately target cancer cells, has increased public attention in immunotherapy. These immunotherapies might bring a new era of cancer management, a highly effective and specific treatment with fewer adverse effects. Abstract

Background Protein is essential for skin integrity, while exercise modulates cellular and molecular processes that regulate autophagy, mitochondrial biogenesis, and collagen synthesis. This study evaluates the combined effects of dietary protein restriction and varying exercise intensities on epidermal thickness, autophagy markers (LC3, p62), mitochondrial biogenesis (PGC1α), and collagen synthesis (Col1a1, Col3a1) in male Wistar rats.    Methods An experimental laboratory study was conducted involving 32 male Wistar rats. They were randomized into 8 groups: control, low protein, low intensity, group IV, moderate intensity, moderate intensity low protein, high intensity, and high intensity low protein. Low protein diet (5%) compared to the control (24%) was given for 60 days, while the exercise regimen was eight weeks of treadmill exercise (low/10 m per minute, moderate/20 m per minute, and high intensity/30 m per minute) for 30 minutes a day, five days a week. Epidermal thickness, LC3, p62, PGC1α, Col1a1, and Col3a1 mRNA expression were measured at the end of the study. One-way ANOVA/Kruskal-Wallis followed by Tukey HSD/Mann–Whitney test was used to analyze the data.   Results A low-protein diet significantly reduced epidermal thickness (p=0.048), increased LC3 (p=0.043) and p62 (p=0.011), indicating impaired skin regeneration and autophagy. However, exercise, especially at high intensities with low protein, mitigated this effect by increasing both LC3 (p=0.011) and PGC1α (p=0.011).   Conclusions These findings highlight the compensatory role of exercise in maintaining tissue integrity under protein-deprived conditions in male rats. High-intensity exercise potentially enhances autophagy and mitochondrial biogenesis.

Abstract

High intensity exercise (HI) induces oxidative stress. Running for 2400 meters is one of the simple exercise form which can be counted as HI. During HI, increase of oxygen consumption happened in muscle cell resulted in an increase of oxidant level. The aim of the study was to measure the effects of Indonesian green tea supplementation, before and after HI to the level of lipid peroxidation (TBARSMalonyldialdehyde (MDA)). The subjects of study were 42 male students from Faculty of Medicine, Universitas Padjadjaran (FMUP) and Faculty of Physical Exercise, Indonesia University of Education (FPOK-UPI). The subjects were divided into trained and untrained subject. Subjects were divided into groups using Astrand Harvard step test. The purpose of this test was measuring the level of VO2 max. Blood samples for MDA level were collected at 3 hours before and after high intensity aerobic  exercise. Oxidative stress was induced by 2400 meters run. The result showed that green tea supplement-ation was effective to decrease MDA plasma level especially after exercise. Green tea could serve as natural antioxidant resources, which potentially can be used as potential sport supplemental drink.Key words: Aerobic exercise, Malonyldialdehyde (MDA), pre and post exerciseStudi Pra dan Pasca Pemberian Suplemen Ekstrak Teh Hijauterhadap Kadar MDAHigh intensity exercise (HI) dapat menginduksi stres oksidatif. Berlari sejauh 2400 meter merupakan salah satu bentuk latihan sederhana yang dapat digolongkan sebagai HI. Selama HI, konsumsi oksigen pada sel otot meningkat yang dapat menyebabkan peningkatan kadar oksidan. Penelitian ini bertujuanuntuk mengukur efek dari suplemen teh hijau Indonesia sebelum dan setelah HI terhadap kadar lipid peroksida (TBARS-Malonyldialdehyde (MDA)). Subjek penelitian ini adalah 42 mahasiswa laki-laki dari Fakultas Kedokteran Universitas Padjadjaran dan Fakultas Pendidikan Olahraga dan Kesehatan,Universitas Pendidikan Indonesia. Subjek penelitian dibagi ke dalam kelompok terlatih dan tidak terlatih dengan menggunakan cara pengujian Astrand Harvard. Tes ini bertujuan untuk mengukur kadar VO2 max. Sampel darah untuk kadar MDA diambil 3 jam sebelum dan setelah high intensity aerobic exercise. Stres oksidatif diinduksi dengan berlari sejauh 2400 meter. Hasil penelitian menunjukkan bahwa suplemen teh hijau efektif dalam menurunkan kadar MDA dalam plasma khususnya setelah berlari. Teh hijau dapat menjadi sumber antioksidan alami yang berpotensi sebagai minuman suplemen olahraga.Kata kunci: Aerobic exercise, Malonyldialdehyde (MDA), sebelum dan setelah berlari

Abstract

Citarum is the longest river in West Java and one of the most polluted rivers in Indonesia and the world. Heavy metals from agriculture, industry, and household waste pollute the Citarum river. Heavy metals enter humans and animals by consuming contaminated food and beverages. Several studies have identified mitochondria as a primary target for heavy metal poisoning, resulting in impaired mitochondrial energy production, induction of oxidative stress, apoptosis, and mitophagy. This study aims to investigate the impact of heavy metals from the upstream and downstream areas of the Citarum river on mitochondrial gene expression of cox4i1, sod2, baxa, mfn1b, and ppargc1a in the skeletal muscle of zebrafish (Danio rerio) using in vivo models for biological monitoring of early detection of environmental heavy metal pollution. This experimental study was conducted from December 2019 to March 2020. The study involved four treatment groups and one control group of zebrafish, which were exposed to river water and sediments collected from two sites in the Citarum river area in December 2019. The biomarker levels were analyzed using multivariate analysis. Although all heavy metal levels except mercury were below the WHO threshold in all samples, this study's results showed that the RT-PCR results indicated that the levels of cox4i1, baxa, and ppargc1a in all samples were generally higher than those in the control. There were significant differences (p<0.05) using multivariate analysis in sod2 in Cibeureum water compared to Cibeureum sediment and ppgarc1a compared to control and Balekambang sediment. In conclusion, heavy metals from the upstream and downstream areas of the Citarum river had an impact on mitochondrial gene expression of cox4i1, sod2, baxa, mfn1b, and ppargc1a in the skeletal muscle of zebrafish. Biomarkers pparc1gc and sod2 could be further studied to identify the most valid and reliable parameters for biological monitoring, which may aid in the early detection of environmental damage to humans and animals.

Abstract

Stroke patients could get sequelae, and it will decrease the patient’s quality of life. Stroke could be caused by comorbid diseases such as hypertension, diabetes mellitus, and dyslipidemia. Thus, stroke could be prevented by reducing the comorbid diseases. The prevention of stroke is necessary to be done in order to reduce stroke cases, especially in Bandung’s rural area. Villagers have limited access to health information and health services. In this community service, the methods we used were to screen the two risk factors of stroke (blood pressure and blood glucose ad random measurement) in Cigugur Girang Village, West Bandung area. The aim of this community service is to find comorbid factors of stroke and give education to people who have one or more stroke risk factors. The results of the blood pressure measurement screening showed that more than 40% of the residents of Cigugur Girang village had blood pressure above normal, and 5% of the population had blood sugar levels higher than normal. The risk factor for stroke in residents of Cigugur Girang Village mainly is high blood pressure. In this community service, people with high blood pressure were educated on how to maintain normal blood pressure and how to change their lifestyle. On the other hand, some participant has high blood glucose level. High blood glucose levels will develop into diabetes mellitus. In this education program, we gave information about the complications of high blood glucose, including stroke. Thus, this education program could increase community awareness to prevent stroke.

Abstract

This study aims to determine the appropriate training program and portion in order to improve the physical performance of athletes.This literature review inquiry was obtained from various PubMed and Google Scholar databases by entering keywords, namely: Sports, Yoga, Basic Physical Components, MDA and IL6. This inquiry reviewed 33 articles published in national and international online journals and have been accredited. The results literature review done on prior studies show basic physical components can be optimally improved through exercises based on the FITT formula, namely interval training, fartlek, weight training, circuit training, plyometrix, rope jump and PNF. In addition, showed that aerobic and anaerobic exercises decreased MDA levels. However, the control group experienced a very significant increase in MDA levels. High-intensity exercise was found able to trigger releasing inflammatory mediators such as TNF-ά and IL-6. Furthermore, some literature showed that long-term yoga practice can reduce MDA levels and IL-6 levels as it as it increases antioxidant levels and anti imflamatory levels. This literature review is expected to provide comprehensive explanation of good and measurable training programs for athletes, coaches and sports institutions. Thereby, athletes’ physical performance can be optimally improved and injuries can be avoided so that athletes get sports achievements.
