Abstract

Continuous exposure to ultraviolet B (UVB) radiation can induce photoaging and accelerate premature skin aging. Turmeric (Curcuma longa) is a medicinal plant that contains curcumin, an active compound known for its anti-inflammatory properties and its role in accelerating wound healing. This study aimed to evaluate the effect of turmeric ethanol extract on TNF-α levels and wound lesion areas in Wistar rats exposed to UVB radiation. A post-test-only control group design was employed, involving 24 Wistar rats divided into four groups: a healthy control group (K1), a negative control group (K2), and two treatment groups receiving oral turmeric ethanol extract at doses of 100 mg/kg BW (K3) and 200 mg/kg BW (K4) for 14 days, with concurrent UVB exposure. TNF-α levels were assessed using one-way ANOVA followed by Tukey's post hoc test, while wound lesion areas were analyzed using the Kruskal–Wallis test followed by the Mann–Whitney U test. The results showed that the highest TNF-α levels were observed in the healthy control group (129.14 ng/L), although no statistically significant differences were found between groups (p = 0.538). In contrast, a significant difference was observed in wound lesion areas (p < 0.001), with the largest lesions occurring in the negative control group. In conclusion, oral administration of turmeric ethanol extract did not significantly affect TNF-α levels but was effective in reducing the area of wound lesions caused by UVB exposure.

Abstract

Xerosis cutis is a chronic dry skin condition aggravated by impaired skin barrier function and increased transepidermal water loss (TEWL). Decreased levels of interleukin-10 (IL-10) and vascular endothelial growth factor (VEGF) contribute to prolonged inflammation and hinder tissue regeneration. Cucumber (Cucumis sativus) contains flavonoids and vitamin C, which possess anti-inflammatory and angiogenic properties, making it a promising candidate for natural topical therapy. This study aimed to determine the effect of cucumber extract cream on IL-10 and VEGF levels in the skin of female Wistar rats with grade II xerosis cutis induced by acetone and ethanol. This experimental research employed a post-test only control group design. A total of 30 Wistar rats were randomly divided into five groups: normal control, negative control, positive control (10% ceramide cream), and two treatment groups receiving cucumber extract cream at concentrations of 3% and 5%. Following the induction of xerosis, the respective creams were applied topically for 14 consecutive days. Results showed that administration of 5% cucumber extract cream significantly increased VEGF levels (715.75 ± 152.65 ng/L) and IL-10 levels (200.35 ± 43.49 pg/mL) compared to the negative control (VEGF 494.29 ± 95.05 ng/L; IL-10 120.62 ± 22.11 pg/mL, p<0.05). The 3% cucumber extract cream also elevated VEGF (522.96 ± 115.48 ng/L) and IL-10 (197.86 ± 31.32 pg/mL), though less effectively than the 5% formulation. Meanwhile, the group treated with ceramide 10% cream showed increases in VEGF (694.87 ± 180.52 ng/L) and IL-10 (140.85 ± 32.42 pg/mL), indicating that cucumber extract at 5% comparable to ceramide therapy. In conclusion, cucumber extract cream, particularly at 5% concentration, significantly enhances IL-10 and VEGF levels in the skin of xerosis cutis model rats. These findings indicate its potential as an effective natural topical therapy for reducing inflammation and promoting skin regeneration in xerosis cutis caused by chemical exposure.

Abstract

Abstract The situational analysis showed that the success indicator of the Healthy Indonesia Program with a Family Approach (PIS-PK) in RW 02, Bangetayu Kulon Subdistrict, Semarang City, for the criterion of having no family members who smoke has only reached 36.7% of the 100% target. The high number of smokers in this area is partly due to the low level of public knowledge about the dangers of smoking. This community service (PkM) aims to assess the level of knowledge regarding the health hazards of smoking before and after education and demonstrations using simple tools. The implementation of the PkM consists of three stages: preparation (including situational analysis using the PIS-PK questionnaire and self-assessment), education, and evaluation using pretest and posttest questionnaires. The education activity carried the theme "Lepaskan Rokok, Gapai Awet Sehat (LEGA)" or “Let Go of Cigarettes, Embrace Lasting Health.” It used leaflets, PowerPoint presentations, videos, and simple demonstration tools. The participants’ knowledge score increased from a pretest average of 6.00±1.75 to a posttest average of 8.34±1.45, indicating a 28.05% improvement. The Wilcoxon test results showed a significant difference (p<0.05) between participants' knowledge levels before and after the educational session. Education and demonstrations using simple tools proved effective in increasing knowledge about quitting smoking among residents of RW 02, Bangetayu Kulon Subdistrict, Semarang City. Efforts to reduce the dangers of smoking in the community require support from all parties, including families, community leaders, and health centers. A recommended follow-up action is to install anti-smoking posters in smoking corners to continuously educate the public about the dangers of smoking. Keywords: : smoking; education; leaflet; video; teaching aids.

Abstract

Background: Xerosis cutis is characterized by impaired barrier function and reduced hydration markers, including Aquaporin-3 (AQP3) and hyaluronic acid (HA).  Cucumber extract (Cucumis sativus) contains antioxidants and humectants compounds that may improve skin hydration. This study investigated the effects of cucumber extract cream on AQP3 and HA levels in female Wistar rats (Rattus norvegicus) with grade II xerosis cutis. Methods:  This in vivo experimental study used a post-test-only control group design. Twenty-five female Wistar rats were randomly assigned into five groups: healthy control, xerosis control, positive control (10% ceramide), and treatment groups receiving 3% and 5% cucumber extract cream. Xerosis was induced by topical application of 70% acetone followed by 100% ethanol for 5–7 days until grade II xerosis developed.  The creams were then applied topically for 14 days. AQP3 and HA levels were measured using enzyme-linked immunosorbent assay (ELISA).Results: HA levels differed significantly among groups (p=0.016). The 5% cucumber extract cream maintained HA levels comparable to healthy controls (611.25±248.61 vs. 685.26±194.95 ng/L; p=0.394). In contrast, ceramide and 3% cucumber treatments showed lower HA levels relative to healthy controls. AQP3 levels showed no statistically significant differences among groups (p=0.131), although descriptively higher expression was observed in the 5% cucumber-treated group. Conclusion: Cucumber extract cream at 5% concentration helps maintain HA levels and shows a tendency to increase AQP3 expression, suggesting potential benefits for improving skin hydration in xerosis cutis and supporting skin health associated with aging.
