Abstract

Background: Holothuria atra is a type of sea cucumber found in the waters of Lemukutan Island, West Kalimantan, which has economic and ecological benefits. Personal experience found the presence of a water-soluble pigment when isolating sea cucumbers, which was bright red, thus drawing attention. Research on the water-soluble pigment of the ethanol extract of H. atra from Lemukutan Island waters has not been carried out yet. Natural pigments in sea cucumbers have a specific role as part of secondary metabolite compounds with the potential of having antimicrobial, antifungal, anticancer, antitumor, antiviral, anticoagulant, and antioxidant activities.Objective: This study aimed to determine the natural pigments, secondary metabolites, and cytotoxic compounds in the ethanolic extract of H. atra.Methods: Samples of H. atra were extracted with 96% ethanol. The pigment analysis used UV-Vis spectrophotometry while the secondary metabolites were tested by the phytochemical screening method, and the cytotoxicity testing involved the Brine Shrimp Lethal Toxicity (BSLT) method.Results: The obtained optimum absorption was 230 nm, which indicated a melanin pigment. The phytochemical test on the ethanolic extract of H. atra found flavonoid, phenolic, alkaloid, saponin, and triterpenoid compounds. The cytotoxicity test based on LC50 was 6.985 ppm, indicating the extract was categorized as extremely toxic because it was <30 ppm.Conclusion: The pigment in H. atra is melanin, the secondary metabolites consist of flavonoids, phenolics, alkaloids, saponins, and triterpenoids, and it has a highly toxic activity.Keywords: Holothuria atra, melanin, phytochemical test, BSLT, cytotoxic IntisariLatar belakang: Holothuria atra adalah salah satu jenis teripang yang ditemukan di Perairan Pulau Lemukutan Kalimantan Barat, memiliki manfaat ekonomi maupun ekologi. Pengalaman saat pengambilan sampel dijumpai adanya pigmen larut air dari tempat sampel, berwarna merah cerah dan menarik perhatian. Penelitian tentang pigmen larut air pada ekstrak etanol H. atra dari perairan Pulau Lemukutan belum pernah dilakukan. Pigmen alami pada teripang memiliki peranan tertentu, sebagai bagian dari senyawa metabolit sekunder berpotensi sebagai antimikroba, antijamur, antikanker, antitumor, antivirus, antikoagulan dan antioksidan.Tujuan: Penelitian ini bertujuan mengetahui pigmen alami, senyawa metabolit sekunder dan potensi sitotoksik pada ekstrak etanol H. atra.Metode: Sampel H. atra diekstraksi dengan etanol 96%. Analisis pigmen menggunakan spektrofotometri UV-Vis, pengujian senyawa metabolit sekunder dengan metode skrining fitokimia dan uji sitotoksik menggunakan metode Brine Shrimp Lethal Toxicity (BSLT).Hasil: Serapan optimum yang dihasilkan oleh ekstrak etanol adalah 230 nm yang merupakan pigmen melanin. Hasil uji fitokimia pada ekstrak etanol H. atra ditemukan senyawa flavonoid, fenolik, alkaloid, saponin dan triterpenoid. Uji sitotoksik berdasarkan LC50 adalah 6,985 ppm menunjukkan ekstrak dengan kategori sangat toksik karena nilainya <30 ppm.Kesimpulan: Pigmen pada H.atra yaitu melanin. Kadungan ekstrak etanol metabolit sekunder teridiri dari flavonoid, fenolik, alkaloid, saponin dan triterpenoid serta memiliki aktivitas toksistas yang sangat toksik.Kata kunci: Holothuria atra, melanin, uji fitokmia, BSLT, sitotoksik

Abstract

The waters of Sedanau, Natuna Regency are congested with traffic activities of inter-island transport ships and the construction of docks that have the potential to cause pollution to the aquatic environment. Determination of the levels of heavy metals (Pb) and (Cd) in feather shells (Anadara antiquata) in these waters has been carried out. The purpose of this study was to determine the concentration of heavy metals Pb, Cd in feather shells (Anadara antiquata) which was carried out for 2 time periods, 2020 and 2021 at 2 different sampling stations. Station 1 is close to fishing activities, fishing boat traffic docks and inter-island passenger ships, station 2 is close to fishing boat docks and floating net cages but far from fishing activities and ship traffic. Measurement of the concentration of heavy metals Pb and Cd using the Atomic Absorption Spectrophotometer (SSA) method with reference to the Indonesian National Standard (SNI) Number 06-6992.2-2004 heavy metals Pb and Cd in feather shells (Anadara antiquata). The results of the measurement of heavy metal concentrations of Pb and Cd in feather shells (Anadara antiquata) in the first year were the content of (Cd) at station 1 of 0.0214 g/g, station 2 of 0.0236 g/g and (Pb) of station 1 of 0. 0.0109 g/g, station 2 was not detected, while in the second year the content (Cd) of station 1 was 1.3472 g/g, station 2 was 0.9980 g/g and (Pb) stations 1 and 2 were not detected. heavy metal content. Abstract

Fish nugget is a type of fishery product processing that has a long shelf life at cold or frozen temperatures. The presentation is more practical, tastes more savory and is liked by children and adolescents, so it can be used to meet the nutritional needs of the family. The implementation of this PKM activity aims to provide training to the people of Setapuk Besar, Singkawang City, in processing trash fish fisheries waste into nuggets, with the addition of Eucheuma cottonii pulp as a partial substitute for wheat flour. This activity is carried out offline while still paying attention to and strictly implementing health protocols such as wearing masks and using hand sanitizers. The activity was attended by 20 participants consisting of members of the Setapuk Besar Village Family Welfare Development (PKK) divided into 3 working groups. This PKM activity was carried out using the lecture method followed by a demonstration of making nuggets. Monitoring and evaluation of the activities carried out during the activity showed the ability of the 3 groups of PKK mothers to process trash fish waste into frozen food nuggets based on variations in the concentration of 0, 25 and 50 g E. cottoni pulp. The results of the nugget-making activity for PKK women in Setapuk Besar village, showed that the addition of 0.25 g of E. cottoni pulp provided a better consistency of the dough and texture of processed nuggets, as well as a more savory taste.

Abstract

Astaxanthin is a pigment from the carotenoid group found in algae, shrimp, and crabs. Due to its chemical structure, astaxanthin has many health benefits but low stability against direct exposure to light and oxygen. In this study, the Zn-astaxanthin complex was synthesized using a reflux reactor at 37o and 60oC. Complex compounds were analyzed using a UV-Vis spectrophotometer and FTIR. The UV-Vis spectrophotometer analysis showed a bathochromic shift in acetone (475 to 477 nm). In comparison, in dimethyl sulfoxide, a hypsochromic shift (493 to 475 nm) was observed, and a new absorption peak was observed at 330 nm. FTIR analysis shows a decrease in the intensity of the C=O stretching vibration and -OH group vibration at 1712 and 1219 cm-1, respectively. This indicated an interaction between the metal ion Zn2+ and astaxanthin. Zn-astaxanthin has better stability than astaxanthin during irradiation, using a halogen lamp at a light intensity of 300 W/m2 for 6 hours. Based on the zero-order degradation kinetics model, the degradation rate constant of the Zn-astaxanthin complex was 0.0621, smaller than that of astaxanthin (0.0880).

Abstract

Latar Belakang: Sargassum dan Padina mengandung senyawa bioaktif yang potensial untuk dikembangkan dalam industri farmasi. Ekstrak etanol Sargassum dan Padina mengandung metabolit sekunder yang memiliki efek sebagai antiseptik, antibiotik, antioksidan, antiinflamasi, antibakteri, antivirus dan astringent yang berguna dalam proses penyembuhan luka. Tujuan: Penelitian ini bertujuan untuk membandingkan efektivitas plester luka ekstrak etanol Sargassum polycystum dan Padina australis  terhadap waktu penutupan luka sayat pada mencit (Mus musculus) Metode: Penelitian ini merupakan penelitian eksperimen jenis post-test only controlled group design untuk membandingkan efektivitas plester luka ekstrak etanol Sargassum polycstum dan Padina australis pada lama penyembuhan luka sayat pada mencit (Mus musculus). Hasil: Hasil penelitian menunjukkan rata-rata kecepatan penyembuhan luka sayat, plester luka ekstrak Padina australis memiliki waktu tercepat yaitu 6,6 hari diikuti dengan plester luka ekstrak Sargassum polycstum 7,8 hari dan kontrol 12 hari. Kesimpulan: Pemberian ekstrak etanol Sargassum polycstum dan Padina australis dapat dijadikan sebagai salah satu alternatif pada penyembuhan luka.

Abstract

Telah dilakukan penelitian studi karakteristik hidro-oseanografi di perairan muara Sungai Duri. Karakteristik yang diteliti adalah pasang surut, gelombang, dan arus, serta nilai kedalaman di muara Sungai Duri Kabupaten Bengkayang. Tujuan dari penelitian ini adalah untuk menganalisis karakteristik pasang surut, gelombang, dan arus, serta pengukuran kedalaman di Muara Sungai Duri Kabupaten Bengkayang. Perhitungan yang digunakan untuk perhitungan pasang surut menggunakan metode admiralty, untuk perhitungan tinggi gelombang dan periode gelombang menggunakan metode SMB (Sverdrup Munk tahun 1947 dan Bretschneider 1985). Perhitungan pasang surut perairan muara Sungai Duri didapat nilai bilangan formzahl 1,3 dengan rata-rata arus 0,07 m/s. Hasil peramalan gelombang pada musim barat, musim peralihan I, musim timur, dan musim peralihan II memiliki tinggi gelombang signifikan rata-rata   0,23 m, 0,07 m, 0,19 m dan 0,08 m, dengan periode gelombang 1,60 detik, 0,89 detik, 1,44 detik, dan 0,91 detik. Gelombang signifikan rata-rata pengukuran lapangan 0,18 m. Pengukuran batimetri di lokasi studi tersebut didapatkan nilai kedalaman dengan rentang 0,65 m hingga 2,95 m setelah dikoreki dengan pasang surut.   Berdasarkan data tersebut bahwa kondisi pasang surut di daerah penelitian ini bertipe pasang surut campuran condong ke harian ganda yaitu pasang surut dalam satu hari terjadi dua kali pasang dan dua kali surut, tapi mempunyai tinggi dan periode yang berbeda.

Abstract

Teripang merupakan salah satu hewan invertebrata yang memiliki potensi kolagen cukup besar karena penyusun dinding tubuhnya adalah kolagen. Kandungan kolagen yang terdapat dalam teripang mencapai 86% dari total protein pada tubuh teripang. Penelitian ini bertujuan untuk mengetahui rendemen terbaik dan mengkaji karakteristik fisikokimia kolagen teripang Acaudina molpadioides. Proses pre-treatment dilakukan dengan perendaman daging teripang menggunakan NaOH 0,1 N selama 24 jam. Tahap ekstraksi kolagen dilakukan menggunakan asam asetat 0,25 M, 0,50 M, dan 0,75 M selama 48 jam. Rendemen kolagen teripang Acaudina molpadioides yang dihasilkan sebesar 6,8%. Karakteristik fisikokimia kolagen yang dihasilkan yaitu memiliki pH 7,96, kadar air 9,40%, kadar abu 9,97%, serta gugus fungsi FTIR memiliki puncak serapan amida A, amida B, amida I, amida II, dan amida III pada bilangan gelombang berturut-turut 3446,79 cm-1, 2926,01 cm-1, 1641,42 cm-1, 1566,2 cm-1, dan 1257,59 cm-1. Gugus fungsi protein kolagen yang diperoleh memiliki struktur triple helix yang menunjukkan bahwa karakteristik kolagen sudah sesuai dan kolagen belum terdegradasi menjadi gelatin.

Abstract

The coastal area of ​​Kabung Island is an area of ​​Karimunting Village, Bengkayang Regency, which has a population of around 300 families, whose main livelihood is fishing. Water conditions are very risky due to bad weather, such as strong waves, making it impossible to go to sea, causing new activities for people to plant gardens by utilizing makeshift land and plantation commodities along the hills on the island.Apart from coconuts and cloves, nutmeg is a plantation commodity that is widely available in this region. The training for the processing of syrup from the flesh of the nutmeg for the people of Kabung Island is to improve the skills and welfare of the people of Kabung Island who have previously carried out simple processing of nutmeg by drying the seeds and making candied nutmeg. The training is carried out by direct processing of nutmeg in front of the Kabung island community who are also participants in this activity.This activity was attended by 50 participants consisting of 15 men and 35 women. Based on the questionnaire before the activity started, it showed that 40% had already known and read about the processing of nutmeg syrup, while 30% of the participants had made simple syrup preparations from nutmeg pulp. Then the evaluation conducted after the activity showed that 80% of the participants were able to make good nutmeg syrup, including packaging and labeling of the resulting products.

Abstract

Seaweed E. cottonii is carragaenophytes or carrageenan-producing plant. Carrageenan has a functions stabilizer, thickener and gel former, so it can be used as edible coating that can maintain fish quality. Tuna has nutritional content per 100 grams such protein around 25-26.2%; water 69.40%; fat 1.50%; and carbohydrates 0.03%. Tuna filets quickly decompose if proper handling is not carried out, it is necessary to apply edible coating because this edible coating can protect the product from air contamination and can prevent microbial growth. This study aims to determine the effect of extraction time characteristics of carrageenan and the appropriate concentration of carrageenan for edilbe coating tuna filet. The extraction process was carried out by heating dried seaweed samples with 8% KOH solution for 1 hour, 2 hours and 3 hours at a temperature of 50˚C. The best characteristics of carrageenan resulted from 2 hours of extraction time with 40% yield, 14.95% water content, 63 dPas viscosity and 12.396 ppm sulfate content. The best carrageenan was applied as edible coating on tuna filets with 3 concentrations, namely 1.5%, 2% and 2.5%, each of which was added with 2 mL of glycerol. Based on the Total Plate Count (TPC), it was found that a carrageenan concentration of 2.5% could prevent bacterial growth in the sample as indicated by the TPC number obtained at 2.7×106, which was lower than the control, which was 1.7×107.

Abstract

Minuman teh Sargassum polycystum merupakan salah satu produk olahan rumput laut berbentuk serbuk kasar dalam kemasan kantong teh celup.. preparasi bahan melalui proses pengeringan dan pemotongan kasar untuk menghasilkan formulasi teh terbaik yang tidak beraroma amis, disukai konsumen dan memiliki aktivitas antioksidan. Formulasi yang digunakan pada penelitian ini terdiri dari Sargassum polycystum, daun kesum, kulit lemon, kulit buah naga dengan komposisi kontrol 1:1, formula 1 (Sargassum polycystum : kulit lemon), formula 2 (Sargassum polycystum : daun kesum), formula 3 (Sargassum polycystum : kulit buah naga). Hasil penelitian menunjukkan bahwa teh Sargassum mengandung senyawa fukoidan, aktivitas antioksidan dengan nilai IC50 135 µg/mL, dengan formulasi terbaik yang disukai konsumen terdapat pada formulasi F2 (Sargassum polycystum : daun kesum) dengan nilai hedonik 3,5 (tidak suka-suka).
