Abstract

Abstrak. Paraquat adalah herbisida yang digunakan secara luas untuk mengontrol pertumbuhan gulma. Penggunaan paraquat dalam jangka panjang dapat mengganggu keseimbangan ekosistem termasuk mikroorganisme tanah karena keberadaan residunya di dalam tanah. Penelitian ini bertujuan untuk mengevaluasi pengaruh paparan herbisida paraquat terhadap populasi bakteri yang berperan sebagai dekomposer, pelarut fosfat, dan nitrifikasi pada tanah perkebunan. Penelitian ini merupakan penelitian observasional. Pengaruh paparan paraquat terhadap populasi mikroba dievaluasi dari nilai Total Plate Count (CFU/g Tanah). Bakteri yang berperan sebagai dekomposer, pelarut fosfat, dan nitrifikasi pada tanah yang terpapar dan yang tidak terpapar herbisida paraquat. Data penelitian dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa nilai TPC populasi bakteri yang berperan sebagai dekomposer, pelarut fosfat dan nitrifikasi diantaranya bakteri amilolitik, selulolitik, proteolitik, lipolitik, pelarut fosfat, dan penambat nitrogen pada tanah yang tidak terpapar herbisida paraquat masing-masing sebesar 7.34, 7.04, 7.48, 7.04, 7.77, dan 5.00 (CFU/g). Sedangkan pada tanah yang terpapar herbisida paraquat masing-masing sebesar 6.42, 6.04, 6.25, 6.41, 6.29, dan 4.85 (CFU/g). Kata kunci: paraquat, populasi bakteri, dekomposer, pelarut fosfat, nitrifikasi.

Abstract

Abtract. This study was aimed at studying changes in the morphology of Salmonella enteritidis bacterial cells after the crude extract of Pheretima sp. using a scanning electron microscope (SEM). Earthworm extraction uses 96% ethanol, and binds to the appropriate bacteria in the exponential phase with extract concentrations of 5% and 10%. Observations using a microscope scanner were carried out after agreeing to a 6-hour, 12-hour, and 24-hour extract. Obtained from the results showing the crude extract of Pheretima sp. to changes in cell morphology of Salmonella enteritidis bacteria. Salmonella enteritidis bacterial cells are damaged consisting of: formed holes in the surface of bacterial cells, bumps on the surface of bacterial cells, shrinkage of bacterial cells, elongation of bacterial cells, and increase in bacterial cells. In addition, as well as longer contributions and free time, incubation, after being added, crude extract of Pheretima sp. against Salmonella enteritidis bacteria, the greater the damage caused. However, 5% extract concentration is the optimal concentration in the effect of damage to the morphology of the cells of Salmonella enteritidis bacteria.Keywords: Coarse Extract, Earthworms, Pheretima sp., Morphological Cells, Salmonella enteritidis, Scanning Electron Microscope (SEM).

Abstract

Abstract. The growth rate of Hermetia illucens larvae in the bioconversion of organic waste is influenced by the media and environmental conditions where the BSF lives. This study aims to determine the effect of the feed formula on the growth of Hermetia illucens larvae in the bioconversion of organic waste. This research method was carried out by rearing BSF larvae, making BSF feed formulas, determining larval length, determining the weight and weight of BSF cassava. The results showed that the feeding formula from fish waste, tapioca waste with a chopped diameter of 10,20,30 mm gave a significant effect on the growth of BSF larvae (larval length, larvae weight) and BSF maggots. The provision of 20 and 30 percent fish waste has a higher weight and length with a chopped diameter of 20 and 30 mm..Keywords: feed formula, hermetia illucens, bioconversion, organic waste
