Abstract

The aim of this study was to investigate the influence of intermittent hypobaric hypoxia on the expression hypoxia adaptation proteins, namely hypoxia inducibla factor-1a (HIF-1a) and myoglobin (Mb). Twenty five male Sprague-Dawley rats were exposed to intermittent hypobaric hypoxia in a hypobaric chamber in Indonesian Air Force Institute of Aviation Medicine, for 49.5 minutes at various low pressure, 1 week interval for 4 times (day 1, 8, 15 and 22). HIF-1α and Mb protein were measured with ELISA. mRNA expression of Mb was measured with one step real time RT-PCR. HIF-1α protein levels increased after induction of hypobaric hypoxia and continues to decrease after induction of intermittent hypobaric hypoxia 3 times (ANOVA, p = 0.0437). mRNA expression and protein of Mb increased after induction of hypobaric hypoxia and continues to decrease after induction of intermittent hypobaric hypoxia 3 times (ANOVA, p = 0.0283; 0.0170), and both are strongly correlated (Pearson, r = 0.6307). The heart of rats adapted to intermittent hypoxia conditions by upregulation the expression of HIF-1a and myoglobin and then both return to normal level.

Abstract

Abstract

BACKGROUND: In our previous study, we found higher cytoglobin (Cygb) expression in keloid than normal tissue. Cytoglobin is a new globin family protein which function is still being studied to date. The purpose of this research is to elucidate the function of Cygb in human skin keloid fibroblasts (KFs), especially its role in intracellular reactive oxygen species (ROS) levels.METHODS: The study was conducted on human skin KFs obtained from primary culture. Inhibition of Cygb expression was achieved by using siRNA targeting Cygb. We compared the relative expression of Cygb between treatment and control group, and its effect on intracellular ROS levels. Gene expression was measured using quantitative real-time polymerase chain reaction (qRT-PCR) while the ROS level counted by dichlorodihydrofluorescein diacetate (DCFHDA) assay.RESULTS: There was an increase in intracellular ROS levels in the small interfering RNA (siRNA) (+) Cygb group compared to control group (1.673 vs. 1.260; 1.773 vs. 1.393; 1.710 vs. 1.360; respectively). There is a negative correlation between Cygb expression and ROS level (p<0.05; r=-0.651).CONCLUSION: There is a negative correlation between Cygb expression and intracellular ROS levels, we suggest Cygb acts as a ROS scavenger in human skin KFs.KEYWORDS: skin keloid fibroblasts, cytoglobin, siRNA, ROS

Abstract

BACKGROUND: Cancer-associated fibroblast (CAF) is the most abundant tumor stroma. Our previous study has demonstrated that the secretomes of CAF isolated from colorectal carcinoma (CRC) patients could induce epithelial-mesenchymal transition in the HT-29 CRC cell line. However, the role of CAF secretomes in CRC stemness is needed to be further investigated. Therefore, the present study aimed to investigate the effect of CAF secretomes from CRC patients on the expression of stemness markers in HT-29 CRC cells in comparison with the secretomes from normal fibroblasts.METHODS: Fibroblasts were isolated from tumor (CAF) and their counterpart non-tumor (NF) areas of three CRC patients undergone surgical resection. Normal preputium fibroblasts (PF) were isolated during circumcision of three healthy boys aged 8 years. All fibroblasts were grown in free-serum culture medium for 24 hours to collect 50% (v/v) conditioned medium (CM). Then, CM was supplemented to HT-29 CRC cells for 72 hours. The effects of CAF- and NF-CM on the mRNA expression of CD44, CD133, OCT4, and ALDH1A1 were analysed using qRT-PCR. Cells proliferation was measured using the trypan blue exclusion assay.RESULTS: Supplementation of CAF-CM (50% v/v) significantly increased CD44, CD133, OCT4, and ALDH1A1 mRNA expressions compared to that of NF-CM and control without supplementation but had no effect on the proliferation of HT-29 cells.CONCLUSION: CAF secretomes from CRC patients upregulate the expression of CRC stemness.KEYWORDS: cancer-associated fibroblasts, ALDH1A1, OCT4, CD44, CD133, colorectal carcinoma

Abstract

Background: There is now increasing evidence that HIF-1 is also responsive to a variety of non-hypoxic stimuli. However, the mechanisms by which these non-hypoxic stimuli induce HIF-1α are not completely known, yet, although some evidence points to a role of ROS as messengers regulating HIF activity. Objective: To determine the expression of HIF-1α in liver rat tissue induced by carbon tetrachloride under normoxic conditions, with or without N-acetylcysteine protection. Methods: Twenty-five male Sprague-Dawley rats were divided into 5 groups: normal control rats, normal rats orally administered with coconut oil (1 mL/200 g body weight) for 1 day, rats orally administered with CCl4 (0.55 mg/g body weight) for 1 day, rats injected i.v. with NAC (0.15 mg/g body weight) for 8 days and then orally administered with CCl4 (0.55 mg/g body weight) for 1 day, rats orally administered with CCl4 (0.55 mg/g body weight) for 1 day and then injected i.v. with NAC (0.15 mg/g body weight) for 2 days. The expression of HIF-1α mRNA was measured by real-time RT-PCR using the Livak method. The expression of HIF-1α protein was measured by ELISA assay. Results: The highest HIF-1α mRNA and protein expression found in the group treated by CCl4 and then was gradually lowered in the pre-NAC group, post-NAC group, control group, and last, in the oil group. Conclusion: Our study shows the effect of CCl4-treated rats under normoxic conditions increased the mRNA and protein HIF-1α. NAC post-treatment provide a better protective effect compared with NAC pre-treatment

Abstract

Background: Chronic systemic hypoxia is severe environmental stress for the heart and might lead to the development of heart failure. Apelin is an endogenous peptide that has been shown to have various beneficial effects on cardiac function. Apelin appears to have a role to play in the ventricular dysfunction and maintaining the performance of the heart. Objectives: In the present study we want to investigate the adaptive response of heart tissue to chronic systemic hypoxia and the correlation with apelin expression and oxidative stress in rat. Methods: An experimental study was performed using 28 Sprague-Dawley male rats, 8 weeks of age. Rats were divided into 7 groups 4 each, namely control group; normoxia (O2 atmosphere) and the treatment group of hypoxia (8% O2) for 6 hours; 1;3;5;7 and 14 days respectively. Body weight and heart weight were measured at each treatment. Ventricular thickness was measured by caliper, Apelin mRNA was measured using real-time qRT-PCR with Livak formula and malondialdehyde (MDA) level was used to assess oxidative stress due to cardiac tissue hypoxia. Results: Macroscopic exams showed hypertrophy at day 7th. The relative expression of Apelin mRNA in hypoxic heart is decreased at the beginning and then increased, starting from day-7 to day-14. The MDA levels were significantly increased from day-7 and were strongly correlated with relative expression Apelin. Conclusion: It is concluded that the increase of Apelin expression is related to oxidative stress in heart tissue of rats during chronic systemic hypoxia.

Abstract

Background: The kidneys in physiological conditions are always in a state of relative hypoxia. Cytoglobin (Cygb) is the newest globin protein found of the globin family. One of the functions of Cygb is in oxygen supply. Cygb expression is found to increase in hypoxic conditions, which are thought to be an adaptation response to hypoxia. Objective: This study aimed to analyze the expression of Cygb in rat kidneys which were exposed to chronic systemic hypoxia. Methods: Twenty five male Sprague-Dawley rats, weighing 150-200 g were used in this experiment. Rats were divided into 5 groups: The control group was exposed to normoxia; the hypoxia groups (10% oxygen / 90% nitrogen) for 1 day; 3 days; 7 days and 14 days. After treatment, rats were sacrificed and their kidneys were taken. Cygb mRNA expression was measured by qRT-PCR, while Cygb protein expression was measured by the ELISA method. Results: The expressions of Cygb mRNA and protein were found to be highest on day 3 of hypoxia and was correlated very strongly and significantly (r2 = 0.96; p <0.05). Conclusion: The highest expression of Cygb on day 3 of chronic systemic hypoxia exposure is suggested as an attempt to restore oxygen supply to the kidneys.
