Abstract

BACKGROUND Prematurity refers to live births before 37 weeks of gestation, wherein the baby is born before the body and its organ systems achieve perfect maturity, and this disorder is still a global problem. The high incidence of prematurity is a problem in developing and also in developed countries. Certain conditions accompanying pregnancies like preeclampsia, infection, and placental insufficiency, may trigger uterine hypoxia, causing premature birth. The placental condition is related to the intra-uterine fetal condition. In prolonged placental hypoxia, there occurs a compensatory mechanism, i.e. an increase in placental angiogenesis. This study aimed to evaluate the effect of hypoxia on fetal blood vessel count as compensatory mechanism for tissue hypoxia. METHODS An observational-analytical cross-sectional design using paraffin blocks of conserved premature placentas, comprising 31 samples of hypoxic premature placentas and 28 samples of non-hypoxic premature placentas, selected using non-random consecutive sampling. The samples were made into slides and stained with hematoxylin-eosin for assessment of histological structure, including fetal blood vessel count and integrity, villus conditions, syncytiotrophoblastic nuclear changes, and syncytiotrophoblastic nuclear aggregation. Mann-Whitney test was used to compare the difference of blood vessel count between groups. RESULTS Assessment of histological structure showed a significant increase in fetal blood vessel count in the hypoxic group [8.00 (5-15)] as compared with the non-hypoxic group [7.50 (3-15)]. CONCLUSION The hypoxia in premature placentas caused an increase in the number of fetal blood vessels as a form of compensation for disturbed oxygen homeostasis.

Abstract

Mitochondria are highly dynamic organelles that can adapt to different environmental stimuli. This study aims to analyze differences in the function and biogenesis of mitochondria and anaerobic glycolysis activity between obese adult rats that were undernourished in childhood (LCD28) and those which were normal (SD28) or already fat (HFD28). Sprague-dawley rats were assigned groups and given diet treatments from December 2017 until August 2018 in the Laboratory Animal Management Unit, Faculty of Veterinary Medicine, Bogor Agricultural Institute. To observe mitochondrial function and biogenesis, MnSOD activity and PGC1α levels were analyzed using ELISA. To investigate the existence of anaerobic glycolysis, LDH activity was analyzed using a spectrophotometry method. The results showed that MnSOD activity and PGC1α levels of LCD28 increased compared to the other groups, signifying higher mitochondrial function and biogenesis. Meanwhile, no significant difference in LDH activity was found, signifying similar levels of anaerobic glycolysis.

Abstract

Background: Maternal exposure to cigarette smoke during pregnancy and lactation might harmful for the fetus. The smoke contains many free radicals that could be eliminated by antioxidant. This study aimed to investigate the effect of Spirulina platensis ethanol extract as antioxidant against cigarette smoke exposure during pregnancy until lactation by assessing oxidative stress markers in neonatal brain tissues. Methods: The experimental study used 26 offspring divided into four groups: (C) = offspring of maternal control group; (Cg) = offspring of maternal exposed to cigarette smoke; (CgSp) = offspring of maternal given spirulina and exposed to cigarette smoke; and (Sp) = offspring of maternal given spirulina only group, during gestation and 9 days lactation (30 days). Each group consisted of 6 offspring obtained from 2 adult females mated with male Sprague-Dawley rats. The exposure of cigarette smoke was 4 burn cigarettes/day for 30 days. The dose of extract was 200 mg/kg BW. The offspring were sacrificed, and the brain tissues were taken for MDA, MnSOD activity, as well as catalase activity, carbonyl, and GSH. Results: There was no significant differences in MDA level between groups. The carbonyl, SOD, and catalase activity did not differ between the control and smoked group. Conclusion: Exposure of four burned cigarettes smoke per day during pregnancy, and 9 days of lactation did not trigger oxidative stress. However, the effect of Spirulina platensis administration on rat offspring brain could not be analyzed.

Abstract

Cytoglobin (Cygb) is a vertebrate globin protein that is synthesized by fibroblasts and expressed in a variety of organs. Cygb’s function is not fully understood. In contrast, activated fibroblasts (myofibroblasts) are known to produce collagen, which is the major component of the extracellular matrix (ECM) and contributes to abnormal wound healing. There are three phases in wound healing: (1) injury; (2) inflammation; and (3) tissue regeneration. Myofibroblasts will apoptosis throughout the healing phase. If myofibroblasts fail to undergo apoptosis, this might end in fibrosis. This condition induces relative hypoxia in fibrosis. Several investigations have found that overexpression of Cygb serves as an antifibrosis agent by reducing collagen production. The potential involvement of Cygb in avoiding fibrosis in diverse tissues is discussed in this article.  Keywords: Cytoglobin (Cygb), wound healing, anti-fibrosis, collagen

Abstract

Heme is a compound consisting of an iron (Fe) atom bound to a pyrrole ring forming protoporphyrin IX (PPIX). Protoporphyrin combines with a protein-forming hemoprotein compound that plays an essential role in oxygen-binding and transport as well as in the process of energy production in the mitochondria. Some cancer cells have more heme biosynthesis than normal cells, which is thought to be linked to more cancer cell growth. Inhibition of heme biosynthesis in some cancer cells leads to decreased cell proliferation. This review article discusses the synthesis of heme, the role of heme in energy metabolism, which is needed for cell proliferation, the inhibition of heme synthesis and its effect on cancer cell proliferation, and the possibility of the inhibition of heme biosynthesis as an approach in therapy of cancer in the future.

Abstract

Purpose: to assess the levels of Hypoxia-inducible factor-1α (HIF-1α) and intercellular adhesion molecule-1 (ICAM-1) in vitreous of proliferative diabetic retinopathy patients which were given intravitreal bevacizumab (IVB), as well as its relation to the central macular thickness (CMT) measured prior to vitrectomy. Methods: thirty-two eyes were randomized into two groups, one that received an IVB injection at 1-2 weeks previtrectomy and the control group which did not receive any injection. Measurement of HIF-1α and ICAM-1 was conducted using enzyme-linked immunosorbent assay (ELISA). The CMT were measured at the initial visit, prior to vitrectomy, and at follow up time (2, 4, and 12 weeks postoperatively) using Stratus OCT. Results: The mean levels of HIF-1α vitreous (ng/mg protein) in the control group and IVB respectively 0.020 (0.006; 0.077) and 0.029 (0.016; 0.21). Vitreous levels of ICAM-1 (ng /mL) in control group and IVB group were 20.10 (3.41; 40.16) and 23.33 (0.63; 68.5). The mean levels of HIF-1α and ICAM-1 vitreous obtained did not differ significantly between the two groups. Conclusion: The levels of HIF-1α and ICAM-1 in PDR patients do not decrease after one injection of intravitreal Bevacizumab 1-2 weeks prior to vitrectomy. The concentration of vitreous HIF-1α and ICAM-1 are not directly related to the CMT.

Abstract

Hypobaric hypoxia is situation that might occur to helicopter pilots in Indonesia who must fly at an altitude of more than 3,048 m such as in Papua. It can be dangerous because hypoxic condition can affect person's performance. So far, the heart is known as an aerobic organ and very sensitive to hypoxic conditions. Hitherto, the effects of hypobaric hypoxia exposure on biomolecular aspects of the heart are still unclear. Therefore, this study assessed cardiac response in rats exposed to intermittent hypobaric hypoxia (IHH) (equivalent to 3,048 meters/10,000 feet). Sprague-Dawley rats were divided into six groups: control; acute hypobaric hypoxia (AHH); and IHH, for 7; 14; 21; and 28 days. We measured super oxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) activities, reduced glutathione (GSH), malondialdehyde (MDA), cytoglobin, myoglobin, HIF-1α, and Nrf2 level as our parameters. Activities of SOD, CAT, GPx and GSH increased while the levels of MDA, cytoglobin, myoglobin, HIF-1α, and Nrf2 decreased in all IHH groups compared with the AHH group. A biphasic pattern was observed as IHH sessions increased from 14 to 21 or 28. Where the IHH treatment for more than 14 sessions caused a decrease in endogenous antioxidants, but the response to hypoxia and oxidative stress increased. Our findings presented the molecular alterations of cardiac rats exposed to intermittent hypobaric hypoxia.

Abstract

Introduction: Breast cancer is one of the highest causes of death from cancer in women in Indonesia. This is partly due to the resistance of ROS-based therapies such as radiotherapy and chemotherapy. Breast cancer stem cells (cancer stem cells, CSCs) have a role in this resistance mechanism. Previous studies demonstrated the ability of CSC to survive oxidative stress conditions due to rotenone administration. Therefore, in this study an analysis was carried out on the transcription factor NF-kB in breast cancer cells, both CSCs and Non CSCs, related to the role of NF-kB in maintaining the survival of cancer cells under conditions of oxidative stress. Methods: The study was conducted on human breast cancer stem cells (CD24-/CD44+) and non stem cells (CD24-/CD44-) which were given H2O2 at concentrations of 1.1µM, 11µM, and 110µM with control cells not given H2O2. Assessment was carried out on the parameters of NF-kB mRNA expression, and cell viability.  Results: Administration of H2O2 at a concentration of 11µM showed a significant increase in the expression of NFk-B CSCs mRNA compared to non CSCs (p<0.05). As for the viability test results, at all concentrations of H2O2 it appears that CSCs was able to maintain its viability compared to non CSCs which experienced a decrease in viability (p<0.05). Conclusion: In this study, conditions of oxidative stress due to the administration of H2O2 led to an increase in the expression of NF-kB mRNA in CSCs so that cell viability could be maintained.

Abstract

Since its first report in 1994, understanding of the nuclear factor erythroid 2-related factor 2 (Nrf2) has continued to grow. Initially recognized for its role in cellular response to oxidative stress, Nrf2 is now known to be involved in a variety of regulatory process, including metabolic regulation, autophagy, protein homeostasis (proteostasis), and mitochondrial biogenesis. The expression of Nrf2 target genes is highly dependent on stimulus activation and interactions with transcription factors, activators, and repressors. Nrf2 activation serves as a defense mechanism under physiological conditions, but in the context of cancer, it can trigger the development of cancer cells due to its complex roles. Nrf2 is strongly associated with the onset and development of many diseases, including those caused by metabolic disorders and inflammation. Understanding Nrf2's diverse functions offers valuable insights into disease pathogenesis and potential therapeutic approaches. This review explores the pleiotropic role of Nrf2 regulation.
