Abstract

Abstract

Abstract

Male factors have contributed to at least 50% of all infertility cases worldwide. Numerous factors causing male infertility have been identified, one of which is azoospermia due to genetic defects. The detection of Y-chromosome microdeletion may assist in diagnosing male infertility as well as predicting the success rate of testicular sperm extraction. A man in his mid-thirties visited the Andrology outpatient clinic at Dr. Soetomo General Hospital accompanied by his wife. They have been married for eight years and have had regular unprotected sexual intercourse, but pregnancy has never been achieved. The patient’s semen analyses showed azoospermia in three examinations conducted at different times. Follicle-stimulating hormone (FSH) and luteinizing hormone (LH) were significantly increased, and genetic screening showed microdeletion in the Azoospermia Factor b (AZFb) subregion. Y-chromosome microdeletion is the second most common genetic defect causing azoospermia after Klinefelter syndrome. The AZF region in the distal part of the Y chromosome plays a key role in regulating spermatogenesis. Mutation or loss of any subregions in this factor may affect spermatogenesis, with the worst outcome being azoospermia. Detailed examinations are important to determine the cause of azoospermia, which may assist a physician in choosing the appropriate management for this condition. Infertile men with Y-chromosome microdeletion face challenges in reproducing naturally. They may also need genetic counseling regarding the possibility of passing on this genetic defect to their offspring and information on how to prevent it.

Abstract

Some preparation techniques, either conventional or advanced, have been provided. Advanced technique may overcome the limitations of conventional techniques. Recently, not all fertility clinics in Indonesia are able to provide advanced preparation techniques. Some techniques require expensive equipments and can only be used for intracytoplasmic sperm injection (ICSI). Some fertility clinics use a modified procedure, namely the combination of density gradient centrifugation with a swim-up method for the preparation of the sperm to be used in ART. This study aimed to determine whether the modified density gradient centrifugation, which is density gradient centrifugation followed by a swim-up, is able to yield better results than conventional density gradient centrifugation. This study was a laboratory experimental pre and pos-test control group design. Population was all adult men aged 21-40 years old and the sampling unit was the man donor's semen which fulfilled inclusion criterias, collected during the periode of the study. Sample size was eight. Sperm analysis were done before and after preparation in conventional and modified group. Descriptive comparation analysis have been used. This study obtained NMSR 7.9+5.5 million/ejaculate and recovery rate (RR) 27.66+11.8 %. RR was lower compared to RR obtained conventional DGC method. RR might be lower because in modified DGC samples undergo two steps selection while conventional DGC only one step selection. But conventional DGC samples should be centrifuged twice, therefore sperms might experience more trauma. Lower RR sperm does not exclude the possibility to be used for ART, because still within the required number for all TRBs. In conclusion, modified DGC preparation method obtained lower NMSR and RR, nevertheless harvested sperms can still be used in all kind of ART.

Abstract

The Natuna Sea is one of the seas located in the northern part of Indonesia, which is influenced by several oceanographic climate phenomena, including the El Nino-Southern Oscillation (ENSO) system, such as La Nina and El Nino, which cause vertical temperature dynamics. This study aimed to examine the relationship between La Nina, El Nino, and normal years on the variability of the mixed layer depth in the Natuna Sea. The study utilized temperature data from the World Ocean Database (WOD) and Marine Copernicus for a period of 3 years, including years with La Nina, El Nino, and normal conditions, as well as mixed layer depth (defined by sigma theta) data from Marine Copernicus. the occurrence of La Nina, El Nino and normal years obtained from the Southern Oscillation Index (SOI). The results showed that the correlation of SOI values with the depth of Maximum Mix Layer Depth (MLD) during El Nino and La Nina was weak and very weak during the Normal year. During the La Nina, normal, and El Nino periods, the deepest MLD occurred during the west monsoon season at depths ranging from 7 to 60 meters. The shallowest MLD occurred during the first transition season at depths ranging from 7 to 25 meters. The MLD values during the east monsoon season depths ranging from 7 to 43 meters, and during the second transition season, the MLD ranges from 7 to 44 meters.

Abstract

The Sulawesi Sea is the western entry point of the Indonesian throughflow, which brings water masses from the Pacific Ocean. This study aims to describe the relationship between water mass characteristics and Sound Velocity Profiler (SVP) in the Sulawesi Sea based on Marine Copernicus data from 2022 with a depth of up to 500 meters. Temperature and Salinity data were used to obtain water mass types from the Tpot-S diagram based on Emmery's classification and Sound Velocity Profiler (SVP) based on the Medwin empirical equation. The temperature, salinity, and SVP data were visualized using ODV 5.6.2 software. The data processing results have identified several water mass characteristics that are believed to be influenced by the Eastern North Pacific Central Water (ENPCW) water mass type found at depths ranging from 130 to 318 meters, Western North Pacific Central Water (WNPCW) water mass found at depths ranging from 130 to 500 meters, Eastern South Pacific Central Water (ESPCW) water mass found at depths ranging from 92 to 453 meters, and Western South Pacific Central Water (WSPCW) water mass found at depths ranging from 130 to 500 meters. The ENPCW water mass type on average has a smaller difference in SVP values compared to the WNPCW water mass type. Meanwhile, the ESPCW water mass type on average has a greater difference in SVP values compared to the WSPCW water mass type. The results indicate a positive relationship between temperature and SVP, meaning that the higher the temperature, the higher the SVP. However, there is a negative relationship between salinity and SVP, meaning that the higher the salinity, the lower the SVP.Keywords:Water MassTemperatureSalinitySound VelocitySulawesi Sea
