Abstract

Abstract. Rahmi HA, Syukur M, Sukma D, Aisyah SI. 2024. Diversity of quantitative, qualitative, and vitamin C content characteristics in F5 generation of chili pepper Capsicum baccatum species. Biodiversitas 25: 2878-2886. Chili is a popular vegetable worldwide, and Indonesia is one of the largest chili producers. However, the cultivation of Capsicum baccatum species is still uncommon in Indonesia. This study aimed to assess the diversity and heritability of yield and yield quality of F5 populations of C. baccatum. This study was conducted at the Experimental Field of Perumahan Alam Sinar Sari, Bogor. The experiment followed a randomized complete block design (RCBD) using 12 genotypes with three replications and four sample plants per replication. The F5 populations of the following genotypes were used: 4k-6p, 4k-7p, 7m-4q, 2m-2m, 7m-3q, 7m-3m, 7m-2q, 2m-2q, 7m-8m, 7m-1m, Bishop Crown, and Lemon Drop. The observations included quantitative, qualitative character, vitamin C contents, and soluble solids. The results showed that the fruit weight per plant ranged from 37.73 to 94.24 grams, while the number of fruits per plant ranged from 11.46 to 30.95. Fruit length ranged from 2.65 to 6.08 cm, and fruit diameter ranged from 15.70 to 41.43 mm. The weight per fruit ranged from 3.43 to 14.89 grams. Brightness or L* values ranged from 19.06 to 41.93, a* values ranged from 5.62 to 31.49, and b* values ranged from 22.90 to 57.45. The chroma value ranged from 30.87 to 64.20, and the hue value ranged from 46.54 to 80.23. The analysis of vitamin C content showed a range of 12.20 to 17.74 mg/20 g, and the test results of soluble solids ranged from 8.77% to 13.49%. All quantitative characters showed high heritability, indicating a strong genetic influence on these characters. The qualitative observations were analyzed, and the results showed that the 12 C. baccatum chilies had similarity coefficients ranging from 0.0 to 0.5. The similarity analysis resulted in grouping the 12 genotypes of C. baccatum into four main groups.

Abstract

Abstract. Sari YN, Aisyah SI, Sukma D, Syukur M. 2025. Increasing ploidy level of marigold (Tagetes patula Sudamala Barak) using colchicine with acute and chronic methods. Biodiversitas 26: 2726-2734. Marigold (Tagetes sp.) is a popular and potential ornamental plant in Indonesia, especially in Bali, commonly used in religious ceremonies and industry. A significant challenge in the floriculture industry is producing plants with desirable traits, such as larger flowers. Sudamala Barak has dark red flowers with small size (4,5-5,6 cm), but consumers prefer larger ones (8-10 cm). Colchicine is a chemical mutagen that disrupts spindle fiber formation during mitosis, thereby inducing chromosome doubling and resulting in polyploid cells. Tagetes patula is an allotetraploid species (2n = 48). This study aimed to determine the LC50 value and analyze the morphological, histological, and cytological characteristics of Sudamala Barak mutants. The research was conducted from May 2024 to February 2025 using two application methods: acute (seven concentrations) and chronic (four concentrations) based on LC50 (lethal concentration 50%). The results showed that colchicine significantly affected various plant characteristics. The LC50 value was found to be 0.346%. Overall, the chronic method produced greater diversity than the acute method. Colchicine treatment influenced plant height, canopy width, number of primary branches, stem diameter, flowering time, and flower diameter. It also increased the number of stomata and chloroplasts, potentially improving photosynthetic efficiency. In the acute method, the highest ploidy level was euploid-pentaploid (2n = 60) at concentrations of 0.05%; 0.1%; 0.15%; 0.25%; and 0.3%. In contrast, in the chronic method, the highest ploidy level was euploid-autoalooctaploid (2n = 96) at 10× and 20× LC50 dilution concentrations.

Abstract

Abstract. Nurcholis W, Aisyah SI, Komala N, Djamaludin H, Mahendra FR, Putra RP. 2026. The use of colchicine as an elicitor increases the accumulation of phenolics and flavonoids in Java cardamom (Amomum compactum) leaves. Biodiversitas 27 (3): d270326. https://doi.org/10.13057/biodiv/d270326. Java cardamom (Amomum compactum) is a medicinal plant valued for its secondary metabolites. This study examined the effects of colchicine treatment on plant morphology, physiology, and phytochemical content using vegetative buds collected from a wild population in West Java, Indonesia. A total of 80 buds were treated with colchicine at 0% (control), 0.1%, 0.2%, and 0.3% concentrations. Morphological parameters—including plant height, leaf number, and tiller count—were measured at 18 weeks after planting. Total Phenolic Content (TPC), Total Flavonoid Content (TFC), and antioxidant capacity were evaluated using ABTS, DPPH, FRAP, and CUPRAC assays. Colchicine treatment caused dose-dependent reductions in plant height and tiller number, with mild leaf shape alterations, but did not induce significant changes in chlorophyll content or stomatal density. TPC and TFC were significantly increased across all treated groups, with the highest TPC at 0.1% colchicine and the highest TFC and ABTS antioxidant activity at 0.3%. Flow cytometry confirmed that colchicine did not induce polyploidy in A. compactum. Bioinformatic analysis based on Arabidopsis thaliana orthologs revealed upregulation of genes involved in cytokinin signaling (RR17, RAP2.4, GIS2) and phenolic biosynthesis (LAP5, KMD3), supporting a possible hormonal mechanism underlying phytochemical enhancement. The results suggest that colchicine acts as a chemical elicitor, enhancing secondary metabolism without chromosomal duplication. These findings support the potential of colchicine for improving medicinal quality in Java cardamom. Further studies are recommended to optimize treatment parameters and validate gene-level responses directly in A. compactum for targeted phytochemical enhancement.

Abstract

Abstract. Hardiany L, Zahra A, Nafisah AZ, Santika S, Aisyah SI, Dinarty D, Nurcholis W. 2026. Flower color-associated variation in phenolic content and antioxidant capacity in Portulaca grandiflora. Biodiversitas 27 (4): d270413. https://doi.org/10.13057/biodiv/d270413. Phenolic and flavonoid compounds are important plant secondary metabolites contributing to antioxidant defense through radical scavenging and redox-related mechanisms. Portulaca grandiflora is an ornamental species with diverse flower colors, yet information on the relationship between flower color, phytochemical composition, and antioxidant activity is limited. This study evaluated four defined flower color variants—Light Reddish Purple bicolor (LRP-bi), Strong Reddish Purple (SRP), Light Orange Yellow (LOY), and Brilliant Yellow (BY)—to determine Total Phenolic Content (TPC), Total Flavonoid Content (TFC), and antioxidant capacity using CUPRAC, FRAP, ABTS, and DPPH assays. Significant differences were observed among variants. SRP exhibited the highest TPC (3.98 mg GAE g⁻¹ FW) and the strongest reducing capacity, with CUPRAC values approximately 6.8-fold higher than LOY. In contrast, LOY and BY showed the highest TFC (1.15 and 1.09 mg QE g⁻¹ FW, respectively), while LRP-bi demonstrated the highest radical scavenging activity (ABTS and DPPH). Correlation analysis indicated strong positive associations between TPC and reducing power assays (r ≈ 0.87), whereas TFC showed negative relationships with antioxidant responses. These findings indicate that flower color is associated with variation in phenolic composition and antioxidant capacity, with red-purple variants exhibiting stronger reducing potential. Although based on a limited number of variants and in vitro assays, this study highlights flower color as a practical phenotypic indicator of phytochemical variation. This approach may support rapid screening, germplasm evaluation, and selection of ornamental plants with enhanced antioxidant-related traits.
