Abstract

Diaporthe vexans (syn: Phomopsis vexans) is a major postharvest pathogen causing Phomopsis blight and fruit rot in eggplant (Solanum melongena), leading to significant economic losses in Indonesia. This study aimed to identify the causal agent of eggplant fruit rot, evaluate its virulence and host range among some Solanaceous crops, and determine the optimal synthetic media for mycelial growth and sporulation. Fungal isolates were obtained from symptomatic eggplants at Kemang Wholesale Market, Bogor, and identified through morphological and molecular analysis amplifying the ITS region. Virulence was tested using mycelial plug and conidial suspension methods, while host range assays were conducted on tomato, chili, black nightshade, and cucumber. Isolate V2 was identified as putatively belonging to D. vexans, showing 98% sequence identity with D. vexans from the Philippines and submitted to the GenBank with accession number PV936605.1. Pathogenicity tests indicated that the apical and middle regions of eggplant fruits tended to be more susceptible to infection than the basal region. Host range evaluations revealed that while D. vexans is primarily asssociated with eggplant but was capable of infecting other Solananeous hosts under artificial inoculation. For laboratory assays, potato dextrose agar (PDA) supported the highest radial growth (79.03 mm), but cornmeal agar (CMA) was the effective for stimulating dense sporulation (4.9 x 106 spores mL⁻¹). These results indicate that D. vexans poses a cross-contamination risk during postharvest storage and that among the media evaluated, CMA was the most effective medium for inoculum production of isolate V2. Abstract

Mung bean is Indonesia's important legume crop after soybean and peanut, and it can be grown as a primary crop or an intercrop. The purpose of this study was to compare disease development and yield on mung bean fields grown under a spreading and dibbling cropping system in Kedungwringin Village, Jatilawang District, Banyumas Regency, Central Java. Data was collected from farmers through interviews and weekly disease observation in six mung bean fields. The type of disease, disease incidence, and severity were also determined using data obtained directly from the field. The study discovered five fungi and virus: powdery mildew (Erysiphe sp.), stem rot (Sclerotium rolfsii), leaf rust (Uromyces sp.), leaf spot (Cercospora sp.), and yellow mosaic disease (Begomovirus). The incidence of such diseases was ranging from 1% up to 85%, with the yellow mosaic disease having a very low incidence. There is no significant difference of disease intensity on both system. Leaf spot was the major disease with severity increasing gradually from 11.11% to 43.17%. Several insect pests and predators were discovered in fields. Considering the higher pod production in response to the disease occurrence, the dibble cropping system is recommended as the better mung bean farming strategy.

Biodiversitas 20: 2937-2942. Tobacco mosaic virus (TMV) is a newly emerging virus infecting cucumbers in Java, Indonesia. The basic characters of the TMV isolated from cucumber need further study to investigate its differences with that from tobacco. Thus, the research aimed to study the character of both isolates based on their biological, symptomatology and nucleic acid of coat protein (CP) gene properties. The TMV isolates from both cucumber and tobacco were able to infect similar indicator plants with differing symptom expressions, especially on eggplant. Homology of nucleotide and amino acid of coat protein gene among isolates were about 90.3% and 91.0%, and homology to other isolates was about 87.6 to 93.8% and 89.3 to 96.8%, respectively. There were 17 amino acid differences in the CP gene which is presumed to differentiate those two isolates. Phylogenetic analysis CP gene sequences compared with corresponding isolates in GenBank showed the two isolates separated in different clades. The cucumber isolate from Kediri is closely related to tomato isolate from China in clade I, while tobacco isolate from Jember closely related to tobacco isolate from China in clade IIindicating the existence of two differ TMV variants. It will increase the difficulty to manage TMV in the fields.
