Abstract

Electrogenic microbes have been exploited in the microbial fuel cell (MFC) system for harvesting bioelectricity. Electrogens are widely distributed in various environments, but the exploration of this microbial group from ecosystems in Indonesia is still limited. This present study aimed to isolate the electrogenic bacteria from sediments of two waterlogged ecosystems (dam and paddy field) using serial dilution into extinction method prior to streak plate method on the solid thioglycollate media supplemented with Fe3+. Electrochemical evaluation was conducted in glucose-fed dual-chamber MFC by using Arduino UNO-based data logger for the accurate monitoring of electricity production in the term of open circuit voltage (OCV). A total of 54 electrogens were successfully isolated from these two ecosystems, ranging from weakness to strongest electrogens (OCV >800 mV) and ranging from microaerophilic, aerotolerant and facultative anaerobes, to obligate anaerobes. This result also suggested that sediment of waterlogged ecosystems rich in electron donor and solid acceptor electron compounds could potentially host electrogenic microbes. The exploration of electrogens from many other waterlogged ecosystems in Indonesia, both natural and anthropic ecosystems, could be conducted to collect genetic resources of novel electrogenic bacteria for the development of MFC technology in Indonesia.

Abstract

This study is aimed to get lipid producing microalgae as feedstock for biofuel production. The microalgae were isolated from 355 collected water samples which  represented many distinct ecosystems such as paddy fields, rivers, agricultural dams, ponds, swampy areas and unique ecosystem of volcano and mud-volcano craters in West- and Central Java, Indonesia. A total of  267 strains of microalgae were isolated from the samples of which  221 strains of them have capability to produce lipid. There were four promising strains that produce lipid between 14.7 – 45.7 percent dry weight in optimal condition that were identified as Chlamydomonas sp. KO-7267 and PK-7195, Chlorella sp. Abstract

Bioremediation, involving biostimulation and/or bioaugmentation, is a promising method to overcome oil spills in Cilacap coastal waters. Cilacap coastal area has high risk on oil pollution. This study investigated the stimulatory effect of nitrogen concentration, bacterial density and the composition of bacterial culture in enhancing oil degradation in this area. The applications of 4 different concentrations of Slow Release Fertilizer (SRF) and 2 different densities of bacterial cells in the form of single (RCO/B/08_008) and mixed culture were employed in microcosm experiments for 28 days. The efficacy of combining bacterial culture and fertilizer application in various concentrations was also tested. Oil degradation, bacterial growth and environmental parameters were monitored periodically during the experiments. The results showed that oil degradation rate was more influenced by nutrient concentration (biostimulation) than bacterial number or culture composition (bioaugmentation) added. The efficacy of biostimulation in degrading oil was better than that of bioaugmentation. Biostimulation increased oil degradation up to 6.4 times higher than the control. The optimum of fertilizer concentration added was 7.5 mg N/g (C:N ratio of 1,000:75), which increased depletion rate both in biostimulation-only and the combination of biostimulation with bioaugmentation up to 6.4 and 7.5 times higher than the control, respectively. It is suggested that bioremediation of oil-contaminated sandy beach in Cilacap would be optimal by employing a combination of Slow Release Fertilizer at concentration having C/N ratio = 1,000 : 75 and RCO/B/08_008 culture at density of 0.5 x 108 cells/mL (100% homology with Alcanivorax sp. TE-9).

Abstract

Open mining activities result in decreased microbial biomass and negatively impact soil fertility. Soil microbes play a key role in the decomposition of soil organic matter and in nutrient cycling through mineralization mediated by the enzymes they produce. The purpose of this study was to analyze soil fertility levels at PT Bukit Asam’s various reclaimed land sites in Muara Enim Regency, South Sumatra, Indonesia, as determined by microbial populations and soil enzyme activity. The research was conducted using an explorative method across PT Bukit Asam’s reclaimed land sites. Soil samples from seven reclamation areas of different ages were analyzed. Our results showed that soil conditions and enzyme activity vary with reclamation age. At KTU, a 12-year-old reclaimed site, urease enzyme activity was 68.83 mg NH₄⁺·g⁻¹ dm·h⁻¹, with a microbial population of 82.64 × 10⁴ CFU·g⁻¹ soil. The highest phosphatase activity, 95.66 mg pNP·g⁻¹ dm·h⁻¹, was observed at the 9-year-old SP702 reclaimed site, with a soil pH of 5.23. Cellulase activity at the 21-year-old Udongan site was 21.51 mg GE·g⁻¹ dm·h⁻¹, with a cellulolytic microbial population of 1.9 × 10⁴ CFU·g⁻¹ soil, higher than at other reclamation sites. Invertase activity at the 15-year-old Tupak site was 24.37 mg GE·g⁻¹ dm·h⁻¹. Soil enzyme activity can serve as an indicator of soil quality and microbial activity, reflecting biochemical transformations in the soil and exhibiting high sensitivity to environmental changes.

Abstract

 The Biological Nitrogen Fixing (BNF) cyanobacteria can reduce atmospheric nitrogen into ammonium. This ability makes BNF cyanobacteria a potential eco-friendly N-source for soil-planted pad-dy. Apart from a few success stories of BNF cyanobacteria applica-tion in the rice field, its role as an ammonium producer is still an open question. There is also a possibility that indeed cyanobacteria biomass which provides nitrogen through the biological decomposing process. This study aimed to analyze the influence of three strains BNF cyanobacteria on paddy grown in the Deep-Water Culture (DWC) hydroponic system. Yoshida Nutrient Solution was used as a growth medium with the application of 0, 45.7, and 91.4 gL-1 of ammonium nitrate (NH4NO3) with five replications. The result showed that inter-action between cyanobacteria and nitrogen significantly influenced the total nitrogen of paddy plant, but not the paddy plant height, the num-ber of tillers, and biomass. Cyanobacterial filaments were found most-ly attached to the paddy root rather than floated in the DWC system. Paddy treated with cyanobacteria had more lateral root than control. The result suggested that cyanobacteria support paddy growth indi-rectly through nitrogen deposition in plant tissue and root development.

Abstract

Returning-back unharvested crop biomass into soil which is intended to maintain soil fertility status is now facing a challenge concerning self-DNA negative impacts related to the application of compost prepared from biomass of the same crop species at certain compost maturity stage. This study aimed at to evaluate the application effects of composts prepared from biomass of the same and different food crop species with different composting periods on the vegetative growth of rice, maize, and string beans and of DNA extracts from the applied composts on seed germination of the studied crops. The experiment applied CRD with 10 treatments [control, k (compost materials): rice (P), maize (J), and string beans (K) biomass, and composting periods: 0, 30, and 90 d]. DNA was isolated by the modified CTAB method. The results showed that application of mature compost supported the vegetative growth of the test crops, while kP30 and kJ30 treatments, stands respectively for the application of 30 days-composted rice and maize biomass, showed inhibitory effect to the vegetative growth of the same species (rice or maize compost applied to rice or maize cropping). However, in vitro self-DNA inhibition effect was inconsistent and therefore considered not occurred in this experiment.
