Abstract

Kanker adalah penyakit mematikan karena perkembangan sel yang abnormal sehingga tidak terkendali oleh tubuh. Penyakit kanker telah menyerang laki-laki maupun perempuan dengan jenis kanker berbeda, seperti kanker paru-paru, kolorektum, kanker serviks. Pengobatan kanker terdapat beberapa cara salah satunya terapi. Pengobatan ini banyak diimplementasikan di beberapa jenis kanker, termasuk kanker otak, yaitu glioblastoma. Penelitian beberapa tahun terakhir menunjukkan bahwa pendekatan terapi dengan bakteri Salmonella Typhimurium berhasil memproduksi enzim antikanker, dan mensekresikan antigen sekaligus antibodi spesifik kanker untuk mengobati penyakit ini. Penulisan ini bertujuan mengkaji potensi dan efektivitas Salmonella Typhimirium sebagai agen terapi antikanker melalui rekayasa genetika galur gen VN20009 yang dilemahkan. Pengkajian dilakukan secara kualitatif dengan memaparkan secara deskriptif melalui kajian literatur. Hasil kajian menunjukkan bahwa Salmonella Typhimurium merupakan bakteri Gram negatif, fakultatif anaerob, dan bersifat motil. Bakteri ini berikatan secara spesifik pada sel-sel kanker dengan mentransferkan RNA onkogen serta menghambat pertumbuhan sel kanker. Pengembangan bakteri Salmonella menjadi kandidat terapi sebagai strategi terapi utama dalam pengobatan kanker karena spesifik menargetkan sel kanker sehingga menghambat pertumbuhannya dan meningkatkan kelangsungan hidup penderita kanker. Hasil kajian menunjukkan bahwa Salmonella Typhimurium memiliki prospek yang menjanjikan untuk penyembuhan penyakit kanker serta menjadi referensi untuk penelitian selanjutnya dalam pengembangan dan pemanfaatan Salmonella Typhimurium sebagai antikanker.

Abstract

Drought is a threat that affects plant growth and development. Wheat crops are very sensitive to heat and drought stress during flowering and grain development, which negatively affects grain yield and quality. Wheat with drought tolerant properties is a genetically modified plant obtained by inserting a cold shock protein gene from Escherichia coli so that it can increase the ability of wheat to adapt to drought. This literature review aims to provide information about the role of cold shock protein gene insertion from Escherichia coli in wheat plants to produce drought tolerant traits. The results of the literature review showed that the CspA gene from Escherichia coli inserted into DNA could increase gene expression by changing the nucleotides in the DNA sequence to increase G+C. Gene insertion produces recombinant DNA with specific nucleotide changes. Nucleotide changes were carried out to increase the overall G+C amino acid content without changing the existing CspA and CspB protein sequences. These changes played an important role in helping wheat adapt to drought. The leaves on the transgenic wheat showed a decrease in the rate of water loss because the stomata in these plants closed faster than in the ordinary wheat so that the transgenic wheat became more drought tolerant.

Abstract

Diabetes Mellitus (DM) in Indonesia continues to increase from 2013 to 2018. Diabetes Mellitus is associated with the production of the hormone insulin in the cells of the pancreas. An increase in blood sugar levels can stimulate insulin secretion. There are several ways to treat Diabetes Mellitus, namely by using drugs, herbal methods, and insulin injections. In addition, one of the DM treatments that can reduce pain is using insulin patch technology as a transdermal treatment. The manuscript for this publication was made to find out the potential of using insulin patch technology as transdermal drug delivery to treat DM. Using a literature review research method that discusses four focus studies, including 1) the structure of the pancreas gland, 2) the mechanism of insulin hormone production by the pancreas gland, 3) the causes, symptoms, and conditions of patients with DM, and 4) insulin patch technology as a transdermal drug in the treatment of diabetes. DM treatment efforts. In the Langerhans section of the pancreas, some cells function to produce the hormone insulin. The amino acids that make up insulin are encoded by exons formed in insulin cells. Some causes of DM namely genetics, age, the influence of food, lack of physical activity, and an unhealthy lifestyle. Some of the symptoms, namely polyuria, polydipsia, polyphagia, etc. Patients with DM need insulin intake to help balance blood sugar levels, one of which is by using an insulin patch in which there is a microneedle containing insulin n-vinylpyrrolidone (NVP) and the enzyme glucose oxidase. After a literature review, it can be concluded that the insulin patch has excellent potential and promise because during the study tested on rats and pigs obtained good results and was able to lower blood sugar levels, although development is still needed. The manuscript of this publication still has shortcomings, so the author suggested to the next author to be able to multiply the latest sources related to insulin patch. Through the creation of the publication, the manuscript realized that diabetes mellitus is a form of human negligence in maintaining the body that God has designed carefully and regularly.
