Abstract

Latar belakang: Golongan darah merupakan informasi yang penting bagi setiap individu, karena menentukan berbagai aspek medis seseorang yang meliputi antara lain keperluan transfusi darah, transplantasi organ dan kehamilan. Keberhasilan dalam berbagai aspek medis tersebut sangat ditentukan oleh kecocokan golongan darah. Hasil survei pendahuluan menunjukkan bahwa Anak-anak dan pengelola  Panti Asuhan Katolik Sonaf Maneka Kupang  banyak yang belum mengetahui manfaat mengetahui golongan darah dan belum memiliki informasi tentang golongan darahnya masing-masing. Tujuan: memberikan pengetahuan kepada anak-anak dan pengelola panti Asuhan Sonaf Manek di Kupang tentang pentingnya mengetahui golongan darah dan memberikan informasi tentang golongan darahnya masing-masing. Metode: Penyuluhan dilakukan oleh tim mahasiswa dan dosen Universitas Muhammadiyah dalam skema Pembangunan Kesehatan Masyarakat Desa (PKMD) dengan metode ceramah yang didukung penggunaan media poster. Evaluasi kegiatan penyuluhan dilakukan dengan kuesioner, sedangkan pemeriksaan golongan darah sistem ABO (A, B, AB dan O) dan Rhesus dilakukan secara langsung oleh mahasiswa Program Diploma Teknologi Laboratorium Medik. Hasil: Hasil analisis yang dilakukan menunjukkan bahwa kegiatan penyuluhan dengan media poster meningkatkan pemahaman peserta akan pentingnya pengetahuan tentang golongan darah. Kesimpulan: hasil pemeriksaan golongan darah menunjukkan bahwa anak-anak dan pengelola panti asuhan yang bergolongan darah A, B, AB, O berturut-turut 23, 43, 7, dan 27% serta seluruhnya memiliki golongan darah Rhesus Positif (Rh+). Kata kunci: media poster, panti asuhan, pemeriksaan golongan darah, pembangunan kesehatan masyarakat desa _______________________________________________________________________________ Abstract Background: Blood type is important information for every individual, as it determines various aspects of a person's medical such as the need for blood transfusions, organ transplants, and pregnancy. The compatibility of blood types largely determines success in various medical aspects. The preliminary survey results show that many children and tenants at the Sonaf Maneka Catholic Orphanage in Kupang did not know the benefits of knowing their blood type and did not yet have information about their respective blood groups. Objective: to provide knowledge to the children of the Sonaf Maneka Orphanage at Kupang about the importance of knowing their blood type and providing information about their respective blood groups. Method: Counseling was carried out by a team of students and lecturers of Muhammadiyah University in the Village Community Health Development (PKMD) scheme with the lecture method supported by media posters. Evaluation of activities carried out by questionnaires, ABO, and Rhesus system blood group examinations were carried out directly by students of the Diploma Program of Medical Laboratory Technology. Result: Results of the analysis showed that the outreach activities with posters increased participants' understanding of the importance of knowledge about blood groups. Conclusion: the results of the blood group examination showed that orphanage tenants with blood types A, B, AB, and O were 23, 43, 7, and 27% respectively and all were Rh-Positive (Rh+). Abstract

obtained from the fermented intestine of Holothuria scabra (sand sea cucumber). Strain HSFI-5 had been reported to be able to produce proteases, which had shown several characteristics of an antithrombotic agent, i.e., fibrinolytic and clot-lysis activities. However, its anticoagulation activity test is yest to be done. This study aimed to determine the anticoagulant activity of the crude protease HSFI-5 in vitro. The study design was a completely randomized design with a sample size of 90 calculated using the Federer formula. The material used was crude protease from B. tequilensis in skim milk broth. Prothrombin time (PT), activated partial thromboplastin time (aPTT), and plasma recalcification time (PRT) were carried out to test the anticoagulant activity. Citrated platelet poor plasma samples were divided into positive control, normal control, direct examination with crude enzyme in volumes of 50 and 100 µL and pre-incubation at 37ºC for 5, 10, and 15 min with crude enzyme volumes of 50 and 100 µL. The data normality was tested with the Kolmogorov-Smirnov test and the different tests were analyzed by one-way ANOVA with the Post hoc LSD test. The results of one-way ANOVA both on PT, aPTT, and PRT examinations showed that there was a significant difference between the treatment groups (p<0.05). The longest results of PT, aPTT, and PRT are positive controls, and the shortest results are normal controls for PT, and 15’ 50 group for aPTT and PRT. It is clear that crude protease B. tequilensis HSFI-5 exhibits anticoagulant as well as thrombolytic action, raising the possibility that it could function as an antithrombotic drug.

Abstract

Diabetes Mellitus (DM) disease is among the top 10 causes of death in the world. It is a metabolic disease causing unusual high blood sugar level in the body. Fortunately, controlling risk factors through early detection can prevent the abnormality. Dian Darat is among villages in Southeast Maluku Regency, where DM screening is rarely carried due to its relatively remote location. This community activity aimed to provide DM disease education through blood sugar screening and counselling for village’s community. Methods included early detection of DM and counselling based on blood sugar level screening results supported by poster media. Screening test results showed that 9 out of 26 respondents (35%) had blood sugar level higher than normal. Based on results of pre- and post-test scores before and after the education, there was an increase in participants’ knowledge regarding DM disease by 62 %. (from average score of 56,15 ± 13,88 to 90,77 ± 9,77, n=26, p ≤ 0.01). Future community services are recommended on similar and more sustainable community service activities by cooperation with other relevant stakeholders to optimize results.

Abstract

The ethyl acetate extract of Bacillus subtilis subsp. subtilis HSFI-9, a bacteria isolated from intestinal fermentation of sea cucumbers (Holothuria scabra) is known to have antimicrobial properties. This research aimed to determine the activity of HSFI-9 extract ointment against Staphylococcus aureus and Candida albicans. Extract ointment is a topical preparation for skin infections made from a mixture of Vaseline Alba and Adeps lanae bases. The extract ointment was prepared into four concentrations of 0.003%, 0.03%, 0.3%, and 1% w/v using HSFI-9 as active ingredient.  Antimicrobial assay was carried out in vitro using the disc diffusion method. The extract ointment was evaluated based on organoleptic, homogeneity, spreadability, adhesion characteristics, and pH tests. The optimal concentration of the extract ointment to inhibit the growth of S. aureus  0.3% (inhibition zone of 11.67±1.26 mm) and against C. albicans is 0.03% (inhibition zone of 10.16±1.50 mm). The activity of the extract ointment was categorized as strong although not as strong as the antibiotic control ointment Mupirocin 2% or Ketoconazole 2%. The extract ointment organoleptic indicated a characteristic odor of ethyl acetate, was yellowish-white ointment, and had a homogeneous and smooth consistency. The extract ointment also had properties such as good spreadability but poor adhesion and tended to have an acidic pH ( 4.5). The HSFI-9 extract can be declared feasible for the development of topical antimicrobials. The ointment still needs to be optimized, especially regarding improving the adhesion characteristic and pH to be safe for the skin and mucosa.

Abstract

Some proteases act as thrombolytic enzymes to destroy the abnormal blood clot in the body called a thrombus, a leading cause of mortality associated with cardiovascular disease (CVD). In addition, the enzyme may also hydrolyze the meat’s proteins into simple amino acids, causing the meat to become tender. The study's aim was to analyze the potential of the crude protease of Bacillus sp. HSFI-9 is an anticoagulant agent and meat tenderizer. The anticoagulant test of crude protease HSFI-9 on venous blood samples was performed by measuring the clotting time compared to control (10% EDTA) by the Lee-White method. Protein profiles of beef, chicken, and tuna were analyzed before and after immersion in the crude protease of Bacillus sp. HSFI-9 in concentration of 30% v/v for 3 hours based on Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE) method. The anticoagulant test screening showed that the crude protease of Bacillus sp.  HSFI-9 was able to prolong the blood clotting time even though its anticoagulant activity was still less than that of 10% EDTA (a commercial anticoagulant agent used as a positive control). The SDS-PAGE results indicated that soaked beef and chicken using crude protease from Bacillus sp. HSFI-9 could hydrolyze the meats’ proteins by denaturing them into smaller molecules, indicated by the appearance of new minor protein bands and the disappearance of major ones. It can be concluded that the crude protease of Bacillus sp. has potential as an anticoagulant agent and a meat tenderizer.

Abstract

Latar Belakang: Desa Sidorejo kelurahan Tambangan kecamatan Mijen merupakan suatu desa yang banyak warganya menanam tanaman buah rambutan (Nephelium lappaceum L.), namun banyak masyarakat yang belum memiliki pengetahuan tentang manfaat tanaman tersebut sebagaibahan Eco-print. Kegiatan pengabdian kepada Masyarakat (PKM) ini merupakan implementasi hasil penelitian tentang pemanfaatan tumbuhan sebagai motif pada eco-print. Tujuan: Untuk mengedukasi masyarakat tentang manfaat daun rambutan sebagai bahan eco-print, sehingga dapat meningkatkan pendapatan Masyarakat. Metode: Kegiatan dilaksanakan di Sidorejo, Kelurahan Tambangan, Mijen Semarang dalam bentuk ceramah dan pelatihan ketrampilan Eco-print. Partisipan pengabdian masyarakat sebanyak 20 orang. Hasil: Partisipan lebih memahami tentang manfaat daun tanaman rambutan sebagai bahan eco-print.  Nilai rata-rata pengetahuan partisipan mengalami peningkatan dari 56,5 poin saat pre-test menjadi 90,25 post-test, mengalami peningkatan rerata skor pengetahuan sebesar 59,74%. Kesimpulan: Edukasi dalam bentuk ceramah dan pelatihan dapat meningkatkan pengetahuan masyarakat tentang manfaat daun tanaman rambutan dalam pembuatan Eco-print. In the previous research, results on the use of plants as motifs in eco-print were presented. This community service activity is an implementation of it. Objective: To educate the community about the benefits of rambutan leaves as an eco-print material, so that it can increase community income. Method: The activity was carried out in Sidorejo, Tambangan Subdistrict, Mijen, Semarang, in the form of lectures and Eco-print skills training. Among the 20 community service participants are present. Result: Participants better understand the benefits of rambutan leaves as an eco-print material. The average knowledge score of participants increased from 56.5 points in the pre-test to 90.25 in the post-test. The average knowledge score has increased by 59.74%. Abstract

Faktor risiko yang mempengaruhi penyakit DM yaitu usia, genetik, jenis kelamin, dan pola makan. Observasi yang telah dilakukan terhadap penduduk di wilayah perumahan Bukit Kencana Jaya, sebagian besar warga telah rutin melalukan pemeriksaan gula darah, namun terdapat pula beberapa warga yang tidak rutin melakukan pemeriksaan gula darah. Tujuan kegiatan ini adalah untuk menyampaikan informasi kepada warga lansia seputar penyakit diabetes melitus mulai dari faktor risiko, gejalah, pencegahan, dna melakukan pemeriksaan gula darah. Penyuluhan dilakukan oleh tim mahasiswa dan dosen Universitas Muhammadiyah dalam skema PKMD (Pembangunan Kesehatan Masyarakat Desa) dengan metode ceramah yang didukung penggunaan media brosur Evaluasi kegiatan penyuluhan dilakukan dengan kuesioner, sedangkan pemeriksaan gula darah dilakukan secara langsung oleh mahasiswa Program Diploma Teknologi Laboratorium Medik. Hasil dari kegiatan yang dilakukan menunjukkan bahwa kegiatan penyuluhan dengan media brosur meningkatkan pemahaman peserta akan pentingnya pengetahuan tentang diabetes melitus. Dari pemeriksaan kadar gula darah yang dilakukan, terdapat 5 orang warga lansia memiliki kadar gula darahnya melebihi batas normal (hiperglikemia) dan sebanyak 48 orang warga lansia yang memiliki kadar gula darah dalam batas normal. Kemudian, dari hasil sosialisasi mengenai diabetes melitus, didapatkan hasil rata-rata nilai pretest sebesar 77,35 dan hasil rata-rata nilai posttest sebesar 97.73, sehingga dapat disimpulkan bahwa warga lansia telah memahami mengenai diabetes melitus.

Abstract

Latar belakang: Hipertensi dapat menjadi penyebab utama gagal jantung, stoke, dan gagal ginjal. Peningkatan tekanan darah sistolik menunjukan resiko yang lebih penting daripada peningkatan tekanan darah diastolik. Kemungkinan seseorang mengalami hipertensi akan semakin tinggi saat usia semakin bertambah. Tujuan: memberikan penjelasan mengenai pencegahan terjadinya Hipertensi dan Hipotensi. Pelaksanaan PKMD di perumahan Batursari Asri Blok B.87 RT.04 RW.05, Desa Batursari, Kecamatan Mranggen, Kabupaten Demak berdasarkan survey belum pernah dipakai oleh pelajar atau mahasiswa dalam melakukan kegiatan PKMD. Metode: Penyuluhan tentang tekanan darah diberikan secara langsung melalui poster. Evaluasi hasil kegiatan melalui pretest dan posttest. Pretest diberikan sebelum penyuluhan dan posttest serta pemeriksaan tekanan darah diberikan setelah penyuluhan. Hasil: Rerata nilai pretest peserta penyuluhan adalah 80 dengan nilai terendah adalah 60 yang diperoleh satu peserta dan tertinggi adalah 90 yang diperoleh dari enam peserta. Setelah diberikan penyuluhan peserta mendapatkan 10 pertanyaan yang sama seperti sebelumnya dan diperoleh nilai rerata posttest adalah 87,5 dengan nilai terendah 70 dan tertinggi adalah 100. Berdasarkan nilai pretest dan posttest menunjukkan terjadinya peningkatan pengetahuan pada peserta setelah diberikan penyuluhan. Kesimpulan: Peserta penyuluhan diperumaha Batursasri Asri mengalami peningkatan pengetahuan tentang resiko dan pencegahan hipertensi. Elevated systolic blood pressure indicates a more important risk than increased diastolic blood pressure. The possibility of someone experiencing hypertension will be higher as age increases. Objective: provide an explanation of the prevention of Hypertension and Hypotension. The implementation of PKMD in Batursari Asri housing RT.04 RW.05, Batursari Village, Mranggen District, Demak Regency based on surveys has never been used by students or students in carrying out PKMD activities. Method: Counseling on blood pressure is given directly through posters. Evaluation of activity results through pretest and posttest. Pretest is given before counseling and a posttest and blood pressure check is given after counseling. Result: The average pretest score of counseling participants was 80 with the lowest score being 60 obtained by one participant and the highest was 90 obtained from six participants. After being given counseling, participants received the same 10 questions as before and obtained the average posttest score was 87.5 with the lowest score of 70 and the highest was 100. Based on pretest and post-test scores, it shows an increase in knowledge in participants after being given counseling. Conclusion: Counseling participants in Batursasri Asri housing experienced increased knowledge about the risks and prevention of hypertension. Keywords: hypertension, counseling, blood pressure

Abstract

The purpose of this service was to provide counseling, training, and technical guidance in three areas of the partnership program (food technology, economy, and health) to potato farmers in the village. In the field of food technology, counseling included how to process potatoes into flour, chips, and sticks. Counseling in the field of economics focused on the importance of entrepreneurship supported by e-commerce in marketing processed potatoes. Health education in the form of video screenings and distribution of posters about the dangers of open defecation (BABS). The results of the activity obtained information that the communitys understanding of how to wash hands properly was good, but needed guidance on healthy toilet standards. Extension activities in the health sector had a positive impact in increasing knowledge about the correct way to defecate. Counseling in the field of food technology and economics encouraged the growth of seven groups for introducing food technology and e-commerce in Gerlang Village.

Abstract

Utilization of glycerol by lipase producing bacteria offers great benefits for fat and oil waste degradation and waterwaste treatment. Nevertheless, there have been lack of reports about the availability of non-pathogenic, lipase producing bacteria, which could naturally degrade glycerol produced from the lipolysis process by lipase. This study reported a newly identified species of rhizobacteria, Alcaligenes javaensis JG3, which is not only able to produce high level of lipase, but also able to degrade glycerol molecules. Identification of strain JG3 was carried out using SEM (Scanning Electron Microscope), BD Phoenix 100 Automated Microbiology System and 16S rRNA gene analysis to determine its taxonomy status. The ability of the strain to metabolize glycerol was investigated both genotypically and phenotypically using degenerate PCR and a glycerol minimal medium. Identification test results showed that strain JG3 belongs to genus Alcaligenes, with the closest relationship with A. faecalis and A. aquatilis (96% nucleotide similarity maximum). Degenerate PCR resulted in a 248-bp sequence showing 93% similarity with glpK of Candidatus Sodalis pierantonius SOPE, a key gene involved in glycerol metabolism. In vitro glycerol utilization test result showed that Alcaligenes sp. JG3 was able to grow on glycerol aerobically, but not anaerobically. It is concluded that Alcaligenes sp. JG3 possesses genes coding for glycerol metabolism and this trait is phenotypically expressed, thus making the strain potential to be used as an effective fat and oil biodegrader.
