Abstract

Tujuan penelitian adalah untuk mengetahui dan menjelaskan pengaruh dari ektrak kulit buah rambutan terhadap adipogenesis jaringan lemak viserall tikus dengan melihat ekspresi igf-1. Tahapan penelitian yang dilakukan adalah ekstraksi kulit buah rambutan, perlakuan pada tikus, isolasi protein lemak viserall, separasi protein dan western blot. Tikus strain wistar jantan dibagi menjadi tikus berat badan normal dan obesitas. Perlakuan yang dilakukan antara lain kontrol, asam elagat, placebo, dan perlakuan ekstrak dengan dosis 5, 10, 15, dan 20 mg/kg berat badan. Hasil menunjukkan bahwa tren penurunan berat badan terjadi pada grup tikus obesitas dengan asupan pakan yang tidak berbeda signifikan (P>0,05). Dosis efektif terlihat pada perlakuan 10 mg/kg dan pada dosis 20mg/kg justru terjadi kenaikan berat badan untuk kedua jenis grup tikus. Profil protein antara tikus normal dan obesitas menunjukkan pola yang hampir sama dengan perbedaan intensitas tiap pita. Ekspresi igf-1 terpaut dengan ekspresi igfbp-1 pada 36,7 kDa dan terjadi pada seluruh perlakuan dengan perbedaan nyata pada kedua grup. Ekstrak kulit buah rambutan dengan dosis efektif 10mg/kg memiliki efek anti-obesitas pada tikus obesitas. Menariknya, ekspresi igf-1 menurun pada perlakuan 20mg/kg berat badan dibandingkan dengan kelompok lain. Hal ini diduga penghambatan obesitas tidak melewati jalur tyrosin kinase pada IR family.

Abstract

ABSTRAK Kambing perah merupakan salah satu komoditas ternak yang berpotensi untuk dikembangkan. Di Indonesia kambing perah yang banyak dikembangkan adalah kambing Peranakan Etawa (PE). Penelitian ini bertujuan untuk mengetahui profil protein spesifik pada berat molekul 30-60 kDa pada yogurt susu kambing PE dengan kultur tunggal dengan menggunakan metode SDS-PAGE. Tahapan dalam penelitian ini adalah pembuatan kurva pertumbuhan, pembuatan starter kultur tunggal, pembuatan yogurt, isolasi protein, separasi protein dan penghitungan berat molekul. Pembuatan yogurt dilakukan dengan tiga ulangan. Kultur bakteri yang digunakan dalam penelitian ini adalah Lactobacillus acidophillus, Lactobacillus bulgaricus dan Streptococcus thermophillus. Hasil separasi protein menunjukkan yogurt dengan kultur LA memiliki protein dengan berat molekul sebesar 31,59 kDa, 48,36 kDa dan 48,94 kDa. Yogurt kultur LB memiliki protein dengan berat molekul sebesar 30 kDa, 37,83 kDa, 50,22 kDa dan 57,12 kDa. Protein yang terlihat pada yogurt kultur ST adalah pada berat molekul 30,02 kDa, 38,67 kDa, 46,18 kDa dan 55, 15 kDa. Kemungkinan protein dari yogurt fermentasi tunggal mempunyai fungsi yang berbeda sebagai anti-hipertensi atau immunomodulator. Kata kunci: Protein, SDS-PAGE, fermentasi tunggal ABSTRACT Dairy goat farm is one of commodity that has potential to be developed. In Indonesia, the most developed dairy goat is PE. The aim of this study is to determine specific protein at 30-60 kDa molecular weights in single culture PE goat’s yoghurt with SDS-PAGE method. Steps of this research are determination of growth curve, making single culture yoghurt starter and yoghurt, protein isolation, protein separation and molecular weight counting. Yoghurt manufactured with triplicates. Bacterial culture used in this study was Lactobacillus acidophilus, Lactobacillus bulgaricus and Streptococcus thermophillus. Protein separation show that yoghurt with culture LA, LB and ST has molecular weight  31,59 kDa, 37,83 kDa and 38,64 kDa. This protein with molecular weight between 30-38 kDa is thought to α-S1 casein or whey protein. We suggest that the protein of yogurt with single fermentation has specific function as anti-hypertension or immunomodulation. Keywords: Protein, SDS-PAGE, single fermentation

Abstract

Penyakit neurodegenerasi adalah penyakit yang menyerang sel otak dan sumsum tulang belakang dengan mematikan selnya. Salah satu penyakit neurodegenerasi adalah HAD yang prevalensinya sekarang meningkat sekitar 37 %. Oleh karena itu, peneliti mengembangkan penelitian untuk mencari obat yang dapat mencegah atau mengobati penyakit ini, salah satunya dalam bidang kultur jaringan dan sel hewan. Tujuan penelitian ini adalah untuk menganalis waktu optimal pertumbuhan kultur neuron otak tikus (Rattus norvegicus) dan tahapan diferensiasi morfologi sel neuron otak tikus. Penelitian ini dilakukan dengan cara mengisolasi sel neuron fetus tikus yang berumur 18-19 hari dan ditumbuhkan secara in vitro (MEM+10%FBS+3%Penstrep). Hasil yang didapatkan adalah pertumbuhan sel neuron yang optimal terdapat pada hari kedelapan. Diferensiasi morfologi sel neuron dimulai pada hari ke-1 dengan morfologi sel berupa neuroblast apolar (berbentuk bulat), hari ke-2 morfologi sel neuron berupa neuroblast bipolar (sudah terbentuk akson dan dendrit), pada hari ke-3 sampai hari ke-14 sel sudah matang, dengan morfologi sel yaitu neuron bipolar, piramidal dan multipolar. Sel neuron yang matang mengalami optimalisasi pada hari ke-10.Kata kunci : diferensiasi, kultur neuron, morfologi neuron, pertumbuhan sel neuron.

Abstract

Penelitian ini bertujuan untuk menganalisa efek pemberian alpha-S2 kasein  susu kambing PE terhadap perubahan morfologi dan histologi membran sinovial tikus Rheumatoid Arthritis (RA). Tahapan penelitian meliputi pembuatan RA menggunakan multidosis CFA (Complete Freud Adjuvant. Kelompok perlakuan dibagi menjadi (C: Kontrol; CM: Kontrol + alpha-S2 kasein; RA: Rheumatois Arthritis; RAM: Rheumatois Arthritis + alpha-S2 kasein). Treatment RA dilakukan  selama 2 bulan dengan dosis 2mg/kg. Analisa morfologi dilakukan dengan skoring perubahan bentuk kaki dan histologi menggunakan metode HE yang divisualisasi menggunakan mikroskop BX53. Hasil penelitian menunjukkan adanya perbaikan morfologi yang ditandai dengan penurunan hasil skor klinis (C = 0; RA = 4; CM = 0; RAM = 1.3). Perbaikan histologi terletak pada penurunan jumlah hiperplasia pada kelompok RA + alpha-S2 kasein menuju kondisi normal dimana penurunan level tersebut mengidikasikan alpha-S2 kasein dapat digunakan sebagai alternatif penyembuhan RA.

Abstract

Rheumatoid arthritis (RA) merupakan salah satu penyakit autoimun yang menyebabkan inflamasi pada jaringan sinovial. Prevalensi RA semakin meningkat setiap tahunnya dan berpotensi menghambat produktivitas. Komposisi mikrobiota merupakan salah satu faktor lingkungan yang mempengaruhi perkembangan penyakit RA. Tujuan penelitian ini adalah untuk mengamati efek pemberian protein CSN1S2 susu dan yogurt peranakan Ethawah terhadap komposisi mikrobiota pada usus tikus rheumatoid arthritis yang diinduksi Complete Freund’s Adjuvant (CFA). Tikus dibagi menjadi dua model yakni kontrol dan RA yang masing – masing diberi perlakukan protein CSN1S2 susu dan yogurt. Feses yang diperoleh lalu dianalisis total koloni bakteri, isolasi bakteri, karakterisasi, perhitungan indeks Simpson, isolasi DNA bakteri dan amplifikasi gen 16S sRNA. Hasil menunjukkan bahwa pemberian protein CSN1S2 pada model hewan mempengaruhi komposisi bakteri dengan peningkatan jumlah koloni bakteri pada kelompok RAM dan RAY. Pada perlakuan kontrol dan RA didominasi oleh kelompok bakteri Lactobacillus berdasarkan analisis gen 16S rRNA. Kelompok Lactobacillus lebih banyak ditemukan pada perlakuan kontrol dibandingkan RA. Lactobacillus berperan dalam mempengaruhi perkembangan penyakit RA, serta mampu menghambat bakteri patogen dari golongan Clostridium dan Enterococcus, ini bergantung pada tingkat spesies bakteri tersebut.

Abstract

The aim of this study was to make animal dyslipidemia models in Sprague Dawley strains induced by a high fat goat diet formula as hypercholesterol feed for two months. The experimental animal used in this study was 30 male rats (Rattus norvegicus strain Sprague Dawley) with an age of 2-3 months with an average body weight of 150g. Animal models are divided into two groups consisting of a control group without additional diet and dyslipidemia group given food consumption goat hypercholesterolemia with high-fat diet formula orally every day for two months. Physiological characteristics of dyslipidemia SD rats had higher body weight, increased food consumption and fecal weight, and decreased water intake and urine volume than the control group. Total cholesterol, triglyceride, and LDL-cholesterol levels increased, while HDL-cholesterol levels did not change in the dyslipidemia rats group compared to the control group. The conclusion of this study indicated that the hypercholesterol diet formula with a high composition of goat fat was successfully induced the SD rats to become dyslipidemia model rat with specific hypercholerol characteristics.

Abstract

Studies reported that kenitu (Chrysophyllum cainito) has the potential as an anti-obesity agent. However, the mechanism underlying that activity remains unknown. On the other hand, targeting PPAR-y has played a beneficial role in regulating obesity. Therefore, this study will explain the anti-obesity potential of natural compounds from C. cainito in modulating obesity through inhibition of PPAR-y. Molecular docking was employed to identify the possibility of several compounds binding with the PPAR-y. Moreover, the stability of the interaction between PPAR-y and each compound was also evaluated using molecular dynamics analysis. The result showed that beta-sitosterol, lupeol, myricitrin, quercitrin, rutin, vitamin A, epicatechin, and chlorogenic acid are worthy of potential as PPAR-y antagonists. In advance, molecular dynamics simulations revealed that chlorogenic acid, quercitrin, and epicatechin were the most stable compounds to interact as an inhibitor of PPAR-y. In conclusion, compounds from star apple have some potential to improve the health status of patients’ obesity through PPAR-y antagonist activity.

Abstract

Cancer is one of the leading causes of death in the world. Rosmarinic acid becomes one of the important polyphenolic compounds. It includes derivatives of two amino acids, caffeic acid from phenylalanine and 3,4-dihydroxyphenyl-lactic acid from tyrosine, secondary metabolites found in many herbal plants from the subfamily Lamiaceae and Boraginaceae. Rosmarinic acid works in inhibiting the occurrence of metastases and tumor formation. In this study, we evaluated rosmarinic acid and its derivatives through study literature. The compounds were downloaded from the PubChem database, while the three-dimensional structure of HIF-1a was retrieved from RCSB PDB. The biological activity was analyzed using the PASS server, and the cytotoxic effect was predicted using CLC-Pred., The pharmacological properties of bioactive compounds were analyzed using SwissADME. The compounds and HIF-1a were prepared using PyRx 0.8 version and Discovery Studio software, docked specifically using Autodock Vina, visualized using Discovery Studio software, and molecular dynamic simulation using CABS-flex 2.0. Rosmarinic acid and its derivative showed an inhibition with the highest affinity value of compound against HIF-1a are control (-6.9 kcal/mol), Rosmarinic acid (-6.4 kcal /mol), and Methyl rosmarinate (-6.3 kcal/mol). Rosmarinic acid could be expected to have activity as an inhibitor modulated at the post-translational level. Comprehensive and further analysis is required in future research based in vivo and in vitro for the development of cancer treatments.

Abstract

Talinum triangulare is a medicinal plant that has been used broadly by the Indonesia community because of its properties. Several studies have proven the components of secondary metabolites in T. triangulare. However, the content of secondary metabolites in each part of T. triangulare is not clearly understood. Therefore, this study aimed to analyse the types of secondary metabolites of leaf, stems, and roots of T. triangulare. The plant from Malang was chosen as the sample source because it is a highland suitable for the life of T. triangulare. The plant from Kediri was chosen as a lowland location. Samples of leaves, stems, and roots of T. triangulare were extracted by maceration method. Phytochemical analyses include phenolics, flavonoids, alkaloids, tannins, saponins, and terpenoids. The absorbance values were measured at different wavelengths. The leaf and stems of T. triangulare contain flavonoid compounds, tannins, phenolics, alkaloids, saponins, and terpenoids with higher levels of secondary metabolites in the leaf. While the roots have the lowest secondary metabolite was only containing steroids. The differential levels and types of secondary metabolites samples obtained from Kediri tend to be lower than samples obtained from Malang. These results indicate that the leaf metabolite secondary of T. triangulare has high potential health benefits for regulating molecular and cellular metabolism.

Abstract

The Indonesian purple rice (IPR), a crossbreeding of black and white rice, is a potential source of ferulic acid. Up to this point, the genomic similarity between the crossbreeding and its parentals has not been determined, particularly in genes involved in ferulic acid biosynthesis. In this present work, we analysed the profile of Oryza sativa caffeic acid-O-methyltransferase (OsCOMT) gene in IPR. The genomic DNA was extracted by CTAB method, then amplified with a specific primer of OsCOMT gene. The amplicon was sequenced by Sanger method and confirmed by geneID NC_029259.1. The IPR showed an amplicon of 708 bp sequence of the OsCOMT gene, indicating the gene involved in ferulic acid biosynthesis. The IPR gene profile is similar to the parentals, but there are variations in point mutations that distinguish it from the parentals. Aspartic acid was changed to Glutamine by mutations at positions 52, 69, and 79. We suggest that IPR has a novel OsCOMT gene variant that is unique as it is the crossbreed.
