Abstract

Sucrose transporter (SUT) is a protein that functions to translocate sucrose from source to sink. Protein activity is determined by the accumulation of sucrose in sugarcane stem. The level of sucrose accumulation is also influenced by the activity of  sucrose phosphat synthase (SPS) and neutral invertase (NI) in conjunction with the synthesis and hydrolysis of sucrose. This aim of this study is to determine the sucrose transporter protein expression in Genetically Modified Product (GMP) of sugarcane.Genetically Modified Product GMP sugarcane events 1, 2, 3, 4, 5, 18, 20 and non GMP were used as materials. The analysis was performed by Western Blot method to observe the presence of SUT protein expression, stem sucrose content by resorcinol method to observe the translocation of sucrose, the SPS enzyme activity and NI that play a role in determining the accumulation of sucrose.The results showed that (1) an increase in SUT protein content in GMP plants events 2 and 18 with a protein band that is thicker than the non GMP plants, (2) SPS and invertase activity at all events GMP crop plants to increase compared to non GMP. Sucrose content of the stem at all events GMP plant to increase with age segment. The older age segments, the higher the sucrose  content. Abstract

Aquaponic is considered one of alternative culture system to improve production and quality of leafy vegetables by combining soil less culture and fish farming. Utilization of fish manure contained in waste water provides nutrient sources for growing plants. Three kinds of leafy vegetables including water spinach (Ipomoea aquatica F.), green coral lettuce (Lactuca sativa L.), and bok choi (Brassica rapa L.) were grown using both aquaponic and hydroponic system at the Green House in Serut village, Panti District, Jember Regency, from April to June 2019. Productions of each leafy vegetable were evaluated on both culture systems. Water spinach represented 29.12% higher production in aquaponic than hydroponic system, shown by significant value in plant height, number of leaves, root volume, and leaf fresh weight. Otherwise, bok choi production displayed 17.03% increased in hydroponic than aquaponic system. However, leaf production of lettuce was not significant between hydroponic and aquaponic system. Lettuce production in aquaponic only 1.90% lowest than hydroponic system indicated that lettuce was appropriate to cultivate in both farming systems. Abstract

Calcium in one of the important minerals that are needed by human body as much as 1200 mg calciums/ day/ person. Every day humans consume calcium as much as 254 mg/ day/ person and it causes people suffering ostheoporosis and other growth disorders. Therefore human needs to consume the source of calcium, one of them is vegetables. Vegetables in one of the source of calcium that is cheap, obtainable and consumable. Lettuce (Lactuca sativa L.) is one of the vegetable with high caclsium but development of lettuce cultivation is still limited.. The efford to increase the quantity and quality of lettuce production can be done with hydroponics technology. Hydroponics technology enables efficient lettuce cultivation. The innovation adding of CaCl2 concentration in hydroponic nutrients is expected to increase the lettuce production. The purpose of this research was to determine the concentration of CaCl2 on the hydroponic nutrients of wick system on the production of Lollo Rossa lettuce (Lactuca sativa L.). The research was conducted at the Green House, Faculty of Agriculture, University of Jember, started in August to Noverber 2016. The research used Completely Randomized Design (CRD) with single factor, which was concentration of CaCl2 on the hydroponic nutrients (C) and it be combined with Lollo Rossa varieties of lettuce (Lactuca sativa L.) Abstract

Induction of somatic embryogenesis in sugarcane requires the composition Plant Growth Hormone (PGH) appropriate. Utilizing of PGH (2,4-D and kinetin) is expected to induce sugarcane somatic embryogenesis. The purpose of this study was to obtain the concentration of 2,4-D and kinetin that effective for the multiplication of sugarcane var. NXI 1-3 through somatic embryogenesis. This study consists of four stages: callus induction, callus proliferation, regeneration of shoots and encapsulation. The plant material used is a spindle leaf sugarcane var. NXI 1-3. Callus induction used 2,4-D with different concentration (2 ppm, 3 ppm and 4 ppm). Callus proliferation used 2,4-D with concentration 1 ppm and 2 ppm. Regeneration of shoots used kinetin 0,5 ppm. The results are showed that the optimal induction of embryogenic callus on medium MS + sucrose 30 g / L + CH 300 ppm + 300 ppm PVP + 2,4-D 4 ppm as indicated by the high percentage of explants forming embryogenic callus that is equal to 40% and the respective concentration 2 ppm and 3 ppm is 33,3% and 37,5%. In proliferation stage, the development callus optimal on medium MS + sucrose 30 g / L + CH 300pm + PVP 300 ppm + 2,4-D 2 ppm and formulations for regeneration shoot on medium MS + sucrose 30 g / L + kinetin 0.5 ppm. Abstract

UPT Agrotechnopark as a partner of the PPUPIK program, has revenue-generating for developing ornamental plant commodities, especially orchids. Training orchid cultivation has been conducted to initiate society by learning about orchids for broadening development and orchid commercialization through empowering entrepreneurship-based programs, especially in Jember Regency. The orchid lovers’ community in Jember Regency has been chosen as a target in this community program. The orchid lovers’ communities have a high potential to develop orchid cultivation since they already have basic competency and experience for practicing orchid cultivation. These communities are easy to be motivated to play a role in orchid commercialization. Community service programs for orchid cultivation training need to be supported by people who already know about cultivation activities, especially orchid plants. This encourages the service team to choose the target community for orchid lovers in Jember Regency to develop products to increase the commercialization of orchid products so that the results are maximized. The targets that will be carried out are training about in vitro orchid cultivation, marketing strategies, crossing techniques, and orchid acclimatization. Community service activities in the form of orchid training can positively contribute to the community in increasing understanding of propagation techniques, orchid marketing strategies, and additional skills in orchid crossing techniques and acclimatization techniques. Training activities can be a provision for the community to play a role in increasing the commercialization of orchids, especially in Jember Regency. --- UPT Agrotechnopark sebagai mitra pelaksana Perguruan Tinggi Program Pengembangan Usaha Produk Intelektual Kampus (PPUPIK) memiliki revenue generating berupa pengembangan komoditi tanaman hias yaitu anggrek. Salah satu kegiatan yang dilakukan yakni melalui pelatihan anggrek. Kegiatan pelatihan anggrek  bertujuan untuk menginisiasi masyarakat untuk lebih mengenal anggrek sehingga dapat memperluas pengembangan dan komersialisasi anggrek melalui pemberdayaan masyarakat berbasis wirausaha terutama di Kabupaten Jember. Masyarakat pecinta anggrek di Kabupaten Jember dipilih sebagai target pada program pengabdian karena mempunyai potensi tinggi untuk pengembangan budidaya anggrek ditinjau dari segi minat dan wawasan pengetahuan tentang anggrek sehingga mudah untuk dibangun jiwa kewirausahaannya khususnya untuk komersialisasi anggrek. Program pengabdian untuk pelatihan budidaya anggrek sangat perlu didukung oleh masyarakat yang telah memiliki pengetahuan dasar kegiatan budidaya, khususnya tanaman anggrek. Hal ini mendorong tim pengabdi untuk memilih sasaran masyarakat pecinta anggrek di Kabupaten Jember dalam upaya pengembangan produk untuk meningkatkan komersialisasi produk anggrek agar hasilnya lebih maksimal.  Target yang akan dilakukan adalah pelatihan budidaya anggrek secara in vitro, strategi pemasaran, teknik persilangan dan aklimatisasi anggrek. Kegiatan pengabdian dalam bentuk pelatihan anggrek dapat memberikan kontribusi positif bagi masyarakat dalam meningkatkan pemahaman terhadap teknik perbanyakan, strategi pemasaran anggrek, dan tambahan ketrampilan tentang teknik persilangan dan teknik aklimatisasi anggrek. Kegiatan pelatihan dapat menjadi bekal bagi masyarakat untuk ikut berperan dalam upaya peningkatan  komersialisasi anggrek khususnya di Kabupaten Jember.

Abstract

Abstract Socialization and hydroponic business practices have been carried out through a Posdaya established by the Jember University Service Institute in Karang Anom Hamlet in Serut village, Panti District, Jember Regency. Serut village was chosen as a target in the partner village development program because it has high potential for the development of hydroponic vegetables in terms of location, community awareness and independence and has an entrepreneurial spirit. On the other hand, the results of observations also found that some residents in Karang Anom Hamlet in Serut Village, Panti District have a fish farming business that has been running for the past few years. Efforts of integrated cultivation of vegetables and fish through the Aquaponics method are expected to produce 2 products at once in a one-time production process, so that products that will benefit the local community and the community at large to meet family food and nutrition are expected. and skills in managing integrated fish and vegetable cultivation in one production process. Therefore, adequate facilities and infrastructure are needed so that the partner groups can utilize aquaponics technology. The results of the activity showed that there was a positive response from the members of the partner group which was shown by the enthusiasm of the participants in participating in a series of training from start to finish. The product has been obtained in one aquaponic production per month will produce 50 kg of catfish and 40 kg of vegetables. For 3 units of fish ponds, 150 kg of catfish and 120 kg of vegetables will be produced within 3 months. Keywords: Aquaponics, Catfish, Vegetables, Hydroponics

Abstract

Akuaponik adalah integrasi dari sistem akuakultur dan hidroponik yang memanfaatkan kotoran ikan sebagai sumber hara tanaman. Input bagi tanaman hanya bergantung pada ketersediaan hara pada kolam ikan, sehingga kebutuhan hara terkadang masih kurang terpenuhi. Inovasi yang dapat diterapkan untuk mengatasi masalah tersebut yaitu dengan pemberian pupuk daun dan zat pengatur tumbuh. Penelitian ini dilakukan untuk mengetahui interaksi konsentrasi pupuk daun dan auksin yang terbaik pada tanaman pakcoy menggunakan sistem budidaya akuaponik rakit apung. Penelitian ini dilakukan menggunakan rancangan acak lengkap (RAL) faktorial dengan 2 faktor. Faktor pertama yaitu konsentrasi pupuk daun sebesar 1 g/L, 2 g/L, dan 3 g/L dan faktor kedua yaitu auksin auksin sebesar 0 ml/L, 1 ml/L, 2 ml/L, dan 3 ml/L. Data yang diperoleh selanjutnya akan dianalisis dengan menggunakan Analysis of Variance (ANOVA) apabila terdapat perlakuan yang berbeda nyata maka akan dilakukan uji beda Duncans Multiple Range Test (DMRT) pada taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa perlakuan konsentrasi pupuk daun dan auksin berpengaruh nyata pada variabel bobot basah dan berpengaruh nyata pada variable tinggi tanaman, jumlah daun, bobot kering dan laju pertumbuhan. Perlakuan konsentrasi pupuk daun 1 g/L dan auksin 3 ml/L merupakan perlakuan terbaik yang dapat meningkatkan hasil  hingga 54,55% dibandingkan dengan budidaya akuaponik tanpa pemberian pupuk daun dan zat pengatur tumbuh.

Abstract

Pembentukan Somatik Embriogenesis (SE) porang secara tidak langsung memiliki proses yang lambat, selama ini yang digunakan adalah induksi secara langsung karena dapat mempercepat penyediaan bibit tetapi pengetahuan tentang induksi SE secara tidak langsung sangat dibutuhkan untuk mengetahui tahapan perkembangannya. Kami menyajikan pengetahuan terkait interaksi hormon 2,4-D dan NAA terhadap induksi kalus, histologi dan scanning elektron microscop (SEM) kalus embriogenik tanaman porang (Amorphophallus muelleri). Tujuan penelitian ini adalah guna mengetahui interaksi pengaruh 2,4-D dan NAA dalam proses induksi SE dan mempelajari tahapan perubahan embrio serta perkembangan struktur embrio tanaman porang. Penelitian ini telah dilangsungkan selama 6 bulan sejak Mei hingga Oktober 2021. Tempat penelitian berada di Laboratorium Kultur Jaringan Agronomi Fakultas Pertanian Universitas Jember. Penelitian ini mengunakan Rancangan Acak Lengkap (RAL) 2 Faktor dengan 3 kali ulangan. Faktor pertama ialah hormon 2,4-D dengan konsentrasi 0 ppm, 1 ppm, 2 ppm, dan faktor kedua yaitu hormon NAA dengan konsentrasi 0,5 ppm, 1 ppm, 1,5 ppm, 2 ppm, 2,5 ppm, kemudian data yang didapatkan dilakukan analisis deskriptif  dan ANOVA. Hasil penelitian menunjukan perlakuan hormon 1 ppm 2,4-D dan 1,5 ppm NAA menghasilkan kalus embriogenik dan pada hasil histologi dan scaning electron mikroskopy (SEM) memperlihatkan tahapan embrio somatik porang meliputi tahapan proembrio, embrio globular, skutelar dan kolioptilar, lalu tahap koleoptilar berkembang ke tahap primodial tunas. Pada konsentrasi 1 ppm 2,4-D dan 1,5 ppm NAA terbentuk proembrio dan embrio globular yang memiliki permukaan proembrio yang  halus.

Abstract

Sosialisasi dan praktek usaha hidroponik sudah dilakukan melalui posdaya yang dibentuk oleh Lembaga Pengabdian Universitas Jember di Dusun Karang Anom desa Serut Kecamatan Panti Kabupaten Jember. Desa Serut dipilih sebagai target pada program pengembangan desa mitra karena mempunyai potensi tinggi untuk pengembangan sayuran hidroponik ditinjau dari lokasi, kepedulian dan kemandirian masyarakatnya serta mempunyai jiwa kewirausahaan. Disisi lain, hasil observasi ditemukan pula sebagian penduduk di Dusun Karang Anom desa Serut Kecamatan Panti memiliki usaha budidaya ikan yang sudah berjalan beberapa tahun terakhir. Upaya Budidaya terpadu sayuran dan ikan melalui metode Akuaponik diharapkan menghasilkan 2 produk sekaligus dalam satu kali proses produksi, sehingga akan diperoleh produk yang bermanfaat bagi masyarakat setempat dan masyarakat secara luas untuk penenuhan pangan dan gizi keluarga.Target yang diharapkan adalah agar anggota kelompok mitra memiliki kemampuan dan ketrampilan dalam mengelola budidaya terpadu ikan dan sayuran dalam satu kali proses produksi. Oleh karena itu diperlukan sarana dan prasarana yang memadai sehingga kelompok mitra dapat memanfaatkan teknologi akuaponik. Hasil kegiatan menunjukkan bahwa terjadi respon positif dari anggota kelompok mitra yang ditunjukkan oleh antusias peserta dalam mengikuti serangkaian pelatihan dari awal sampai selesai. Telah diperoleh hasil produk dalam satu kali produksi akuaponik per bulan akan menghasilkan ikan lele sebanyak 50 kg dan sayuran 40 kg. Untuk 3 unit kolam ikan akan dihasilkan ikan lele 150 kg dan 120 kg sayuran dalam waktu 3 bulan

Abstract

The increase of soybean (Glycine max) production, particularly in the drought-stricken areas, can be done by improving cultivation techniques. Trichoderma sp. fungus is expected to increase the efficiency of the root function of soybean plants in absorbing water, especially in drought conditions. This experiment aimed to examine the effect of drought stress levels with different Trichoderma sp. applications on the growth and yield of soybeans. The experiment was carried out in a plastic house using a two-factorials Completely Randomized Design (CRD). The first factor was the administration of Trichoderma harzianum with four dosage levels, namely T0 (0 grams), T1 (20 grams), T2 (40 grams), and T3 (60 grams). The second factor was three drought stress levels, namely C1 (40%KL), C2 (60%KL), and C3 (80%KL). Data analysis employed the analysis of variance and follow-up test of DMRT at 5% level if there was an interaction between the two treatment factors. Observation variables included plant height, root length, number of flowers, number of productive branches, proline, root wet weight, the weight of biomass, number of effective root nodules, relative growth rate, number of pods, number of pithy pods, number of empty pods, pod weight, and seed weight. The results implied that the effect of the tested drought stress level was still tolerant at 60% KL with the application of Trichoderma sp. at a dose of 40 grams. There was an interaction between the effect of dosage of Trichoderma sp. and drought stress level on plant height and root length and proline content and biomass weight of soybean plants at 81 DAP. Peningkatan produksi kedelai utamanya pada lahan tercekam kekeringan dapat dilakukan melalui perbaikan teknik budidaya. Pemanfaatanjamur Trichodermasp. diharapkan dapat meningkatkan efisiensi fungsi perakaran tanaman kedelai dalam menyerap air khususnya pada kondisi kekeringan. Percobaan ini bertujuan untuk mengkaji pengaruh tingkat cekaman kekeringan dengan aplikasi Trichodermasp. yang berbeda terhadap pertumbuhan dan hasil tanaman kedelai. Percobaan dilakukan di rumah plastik dengan menggunakan Rancangan Acak Lengkap (RAL) faktorial dengan dua faktor, faktor pertama pemberian dosis Trichoderma harzianum dengan empat taraf: T0 (0 gram), T1 (20 gram), T2 (40 gram), dan T3 (60 gram) dan faktor kedua tingkat cekaman kekeringan dengan tiga taraf: C1 (40%KL), C2 (60%KL) dan C3 (80%KL).  Analisa data menggunakan analisis sidik ragam dan uji lanjut DMRT Taraf 5% jika terdapat interaksi pada dua faktor perlakuan. Variabel pengamatan meliputi tinggi tanaman, panjang akar, jumlah bunga, jumlah cabang produktif, prolin, berat basah akar, berat biomassa, jumlah bintil akar efektif, laju pertumbuhan relatif, jumlah polong, jumlah polong bernas, jumlah polong hampa, berat polong dan berat biji. Hasil percobaan menunjukkan pengaruh tingkat cekaman kekeringan yang diujikan masih toleran pada 60% KL dengan aplikasi Trichodermasp.  pada dosis 40gram. Terdapat interaksi antara pengaruh dosisTrichodermasp. dan cekaman kekeringan terhadap tinggi tanaman dan panjang akar serta kandungan prolin dan berat biomassa tanaman kedelai pada umur 81 HST.
