Abstract

One alternative to increase production is to maximize the use of biofloc system cultivation technology. However, the problems in biofloc cultivation are; the difficulty of cultivators to replace or alternatively use commercial bacteria to increase production. One of the efforts made is the use of bacillus sp. The aim of the study was to determine the optimal concentration of bacillus sp that could increase the growth of saline tilapia. This study used a completely randomized design (CRD) with four treatments, each repeated three times, so that there were 12 units. This study tested the concentration of Bacillus sp. 0.0028 ml/L, 0.0030 ml/L, 0.0032 ml/L, and 0.0034 ml/L. The results showed that the highest protein retention was 9.91%, fat retention was 2.05% and the body glycogen content of saline tilapia was 0.68%. This shows that the administration of Bacillus sp. in saline tilapia culture media has a real effect.

Abstract

Vannamei shrimp (Litopenaeus vannamei) is a primary aquaculture commodity with an export contribution of up to 85%. However, its production faces challenges due to disease outbreaks, particularly White Spot Syndrome Virus (WSSV). This study aimed to detect the presence of WSSV in post-larvae of vannamei shrimp using the Nested-PCR method in three different hatcheries. The methodology included sample preparation, DNA extraction, DNA amplification through Nested-PCR, electrophoresis, and DNA visualization. The results showed that in the first week of testing, all samples tested negative for WSSV, as no DNA bands appeared at 941bp. However, in the second week, one sample (C5) tested positive for WSSV, indicated by the presence of a DNA band at 941bp, while the other samples remained negative. The prevalence of WSSV infection in this study reached 16.67%. The DNA concentration ranged from 94.5 ng/µL to 166.5 ng/µL, with a purity level between 1.72 and 1.86. These findings suggest that the Nested-PCR method is effective in detecting WSSV presence in vannamei shrimp, providing reliable detection rates.

Abstract

Abstract

Petani kecil menghadapi hambatan serius berupa keterbatasan legalitas lahan, biaya sertifikasi, akses pembiayaan, serta risiko eksklusi pasar akibat regulasi global seperti EUDR.
