Abstract

Indonesia is one of a country with high biodiversity and genetic resources. One of them is Madura cattle that derived from crossing between exotic cattle, namely zebu cattle with local Indonesian cattle, namely Bali cattle. Branched-chain alpha-keto acid dehydrogenase is an enzyme complex that metabolizes branched chain amino acids, namely valine, leucine and isoleucine. This study aimed to analyze exons 2-3 variation of the BCKDHA gene and its flanking region in Madura cattle. The sequences of it were obtained using DNA sequencing techniques. A total of seven variations were found, one variation located in the flanking region adjacent to the 5' end of exon 2 of the BCKDHA gene. The A392G variation found in exon 3 does not cause any amino acid changes. There is no variation found in exon 2, it indicates that the exon regions of BCKDHA gene are more conserved than introns in the Madura cattle.

Abstract

Suweg (Amorphophallus paeoniifolius (Dennst.) Nicolson) merupakan tumbuhan liar yang tumbuh dan dimanfaatkan oleh masyarakat desa Darupono. Penelitian ini bertujuan untuk mengetahui pemanfaatan tanaman suweg di Desa Darupono, organ tanaman suweg apa saja yang dimanfaatkan, dan potensi tanaman suweg, serta mengetahui cara budidaya tanaman suweg di Desa Darupono. Penelitian ini tergolong penelitian deskriptif eksploratif dengan menggunakan metode survei, wawancara semi terstruktur, dan menggunakan data pendukung yaitu kuesioner. Pengumpulan data dilakukan dengan teknik acak sederhana dan snowball sampling. Penelitian yang dilakukan di Desa Darupono menunjukkan bahwa tanaman suweg memiliki beberapa potensi antara lain mencegah diabetes, antibakteri, dan antikanker, serta dapat diolah menjadi tepung suweg dan bahan dasar pembuatan mie. Tanaman suweg dimanfaatkan oleh masyarakat Desa Darupono sebagai bahan pangan yaitu dengan cara dikukus, direbus, digoreng, dan diolah menjadi emping, com, pote dan sayur daun oblok-oblok suweg . Selain itu, suweg juga digunakan untuk upacara adat tujuh bulanan atau dalam bahasa Jawa disebut mitoni. Masyarakat Desa Darupono membudidayakan suweg dengan menanam umbi-umbian. Tahapan budidaya suweg adalah penyiapan lahan, penyiapan benih, penanaman, pemeliharaan dan pemanenan.

Abstract

The Gumbregan Maheso tradition became the main source of ethnobiological study. The tradition is related to people’s indigenous knowledge in utilizing livestock. The study aims to identify types of animals and the meaning of the gumbregan maheso ceremony in Banyubiru village, Widodaren Sub-district, Ngawi Regency, Indonesia. The research uses a qualitative descriptive method to deeply describe ethnobiological values of the Gumbregan Maheso tradition of the East Javanese people using a purposive sampling technique and snowball sampling. Data is collected through semi-structured interviews with 15 informants, consisting of traditional leaders, farmers, and communities involved in the implementation of the tradition. The research results show that the type of animals used in the Gumbrega Maheso tradition is a buffalo and ingkung chicken to serve alongside offerings, such as polo pendem, ketupat, and lepet. The ceremony is intended to express gratitude for the gift of a healthy and reproducing animal

Abstract

Tanaman Krisan (Chrysanthemum morifolium) merupakan tanaman hias yang memiliki nilai ekonomi tinggi. Pupuk organik cair dari limbah organik mengandung unsur hara cukup tinggi berpotensi dalam meningkatkan pertumbuhan tanaman. Penelitian ini bertujuan untuk mendapatkan konsentrasi pupuk organik cair yang terbaik dari limbah sayur terhadap pertumbuhan krisan. Desain yang digunakan adalah Rangcangan Acak Lengkap (RAL) satu faktor dengan 6 perlakuan dan 4 kali ulangan sehingga terdapat 24 unit dalam ulangan percobaan. Perlakuan pupuk cair berupa 0 ml (K0), 5 ml (K1), 10 ml (K2), 15 ml (K3), 20 ml (K4), dan 25 ml (K5). Parameter pertumbuhan yang diamati adalah tinggi tanaman (cm), jumlah daun (helai), dan panjang daun (cm). Data yang diperoleh dianalisis menggunakan uji ANOVA One-way dengan taraf signifikansi 5%. Hasil penelitian menunjukkan bahwa pupuk organik cair dengan konsentrasi 15 ml/L air menghasilkan tanaman tertinggi dan konsentrasi 5 ml/L air (K1) menghasilkan jumlah daun terbanyak.

Abstract

Kultur jaringan atau bisa disebut juga dengan perbanyakan tanaman secara in vitro, yaitu suatu budidaya tanaman yang dilakukan dalam botol-botol dengan menggunakan media khusus dan alat-alat yang steril. Sistem perbanyakan tanaman dengan kultur jaringan dapat menghasilkan tanaman baru dalam jumlah banyak dan dalam waktu yang singkat. Salah satu tanaman yang sering dilakukan teknik kultur jaringan yaitu tanaman anggrek dalam hal ini anggrek Dendrobium. Dendrobium merupakan salah satu genus anggrek terbesar dari famili Orchidaceae yang memiliki kurang lebih 2.000 spesies. Tingkat keberhasilan dalam kultur jaringan ditentukan oleh banyak faktor. Dalam penelitian ini yang menjadi permasalahan yaitu adanya kontaminasi yang menghambat pertumbuhan eksplan anggrek. Dilakukan penelitian merupakan suatu bentuk tugas kerja praktik yang diwajibkan oleh prodi serta untuk mengetahui teknik perbanyakan anggrek secara in vitro dengan faktor-faktor yang mempengaruhi tingkat keberhasilannya. Jenis penelitian ini yaitu observasi dan eksperimen, yang dilakukan secara langsung di laboratorium UPTD Kebun Dinas Pertanian Kota Semarang. Dilakukan secara urut dari sterilisasi hingga aklimatisasi. Hasilnya terdapat kontaminasi pada eksplan yang ditanam dengan tingkat keberhasilan yang sangat kecil. Tingkat keberhasilan dalam kultur jaringan ditentukan oleh beberapa faktor, seperti halnya pemilihan eksplan, faktor medium, tingkat sterilisasi, dan berbagai penunjang lainnya. Di UPTD Kebun Dinas Pertanian Kota Semarang diasumsikan bahwa faktor yang paling berpengaruh dalam keberhasilan kultur jaringan yaitu sterilisasinya.

Abstract

Children of growing age are susceptible to various diseases because their immune systems are still developing. In many rural areas in Indonesia, including Colo Village, Mount Muria, people still use traditional medicine to treat diseases in children. This study documents the knowledge and conservation efforts of the Colo Village community regarding medicinal plants used to treat children's diseases. Data were collected qualitatively through semi-structured interviews with forty residents from four hamlets. The findings showed that the community used fifty-six plant species to treat various children's diseases. The results showed that the Colo Village community used 56 plant species to treat diseases in children, such as the Zingiberaceae family, such as turmeric (Curcuma domestica Val.), black ginger (Curcuma aeruginoasa Robx), ginger (Zingiber officinale Rosc.), and others. Diseases that can be treated are: cough, fever, worms, colds, mumps, diarrhea, mouth ulcers, loss of appetite, mouth ulcers, convulsions, and wounds. Plant organs utilized include leaves (45%), rhizomes (18%), fruits (14%), tubers (9%), seeds (7%), stems (3%), and flowers (4%). Drug processing is done by pounding (56%), boiling (11%), grating (24%), slicing (7%), and brewing (2%). Most plants are obtained through cultivation (55%), followed by obtaining from nature (29%) and purchasing at the market (16%). The Colo Village community preserves medicinal plants by cultivating them in their yards and gardens and utilizing other traditional methods. Strengthening ethnobotanical knowledge, especially among the younger generation, is key to maintaining local wisdom and biodiversity in this village.

Abstract

Parijoto (Medinilla speciosa) leaves are widely used for their antimicrobial properties. Endophytic fungi from these leaves show potential as entomopathogenic agents against Helicoverpa armigera larvae, a moth pest of cotton and corn.The research activities conducted in the laboratory include the isolation and characterization of endophytic fungi, as well as entomopathogenic testing against H. armigera larvae using a spraying technique with endophytic fungal extracts. In this study, 5 isolates were obtained from leaves and assessed their entomopathogenic properties. The isolate code is EP1, EP2, EP3, EP4, EP5. Bioassays were conducted to evaluate the effectiveness of these fungi in controlling H. armigera larvae. Among the tested fungi, EP5 demonstrated the highest entomopathogenic activity, significantly reducing the survival rate of the larvae. These findings indicate that EP5 holds substantial promise as a biocontrol agent for managing H. armigera populations with mortality percentage 30,78%, offering a sustainable and environmentally friendly alternative to chemical pesticides. The study underscores the potential of utilizing endophytic fungi from parijoto leaves in integrated pest management strategies.
