Abstract

Orchid is one ofthe flowering plants with a very high level of diversity, including the diversity of morphological characters it has. This study aims to determine the diversity of orchids (orchidaceae) seen morphologically in the Ngalian sub-district forest in Semarang city. The method used in this study is cruise method and characterization techniques. Analysis of research data was carried out descriptively and displayed in the form of tables and photos. Orchids in the teak forest cover 940 ha, spread in 4 villages, namely Wonosari village, Gondoriyo village, Podorejo village and Wates village. The results showed that there were 3 types of orchid plants (orchidaceae). The types of orchids include Rhynchostylis retusa (L.) Bi., Rhynchostylis gigantea (Lindl.) Rindl. and Dendrobium crumenatum (L.) Sw. The diversity of morphological characters in each species of orchid such as diversity in leaf color, leafshape, leaftip shape, stem color, root type etc.

Abstract

Indonesia has a strategic geographical area with high biodiversity potential, it is located in the equatorial region which causes Indonesia have a tropical climate. The biodiversity of forests in Indonesia includes ferns. Nail plants can be found with a variety of types in various environments, especially in shady areas such as forests. Ferns are plants that have an important role in the forest ecosystem as a source of germplasm. Besides that, ferns also have the potential as a source of food and medicine for the Penggaron Forest, or who is familiarly called the Penggaron Tourism Area, is one of the natural tourism objects located in Semarang Regency, precisely in Susukan Village, Ungaran District, Semarang Regency. This study aims to identify the species of ferns in the Penggaron Forest. Data collection techniques used in identifying plant ferns are: 1) orientation and exploration of locations; 2) gathering ferns by exploring along the main road to the location of the campsite; 3) record and document the ferns found in the field. The results of the identification of ferns in the Penggaron Forest there are 2 families of ferns consisting of 23 species of plants. The Scizaeaceae family consists of 4 types of plants namely Lygodium flexuosum, Lygodium japonicum, Lygodium palmatum, and Lygodium circinatum. While the Polypodiaceae family consists of 19 types namely Nephrolepis hirsutula, Nephrolepis biserrata, Dryopteris scotii, Adiantum, Adiantum raddianum, Mickelopteris cordata, Pteris ensiformis, Pteris asperula, Pteris biaurita, Pteris vittata, Adiantum, Adiantum raddianum, Mickelopteris cordata, Pteris ensiformis, Pteris asperula, Pteris biaurita, Pteris vittata, Adiantum, Adiantum raddianum, Mickelopteris cordata, Pteris ensiformis, Pteris asperula, Pteris biaurita, Pteris vittata crenata, Tectaria angulata, Tectaria maingayi, Tectaria heracleifolia, Pleocnemia irregularis.

Abstract

Ficus is a genus with the largest number of members of the Moraceae tribe which are often found in Indonesia and has a significant role in the tropics as a keystone species. Ficus has a fairly wide distribution with several members of which F. fistulosa and F. variegata can grow in primary forests to urban areas such as Mount Botol, Mount Halimun Salak National Park and Bogor City. Differences in growing locations allow for differences in genetic diversity in plants so that information needs to be explored and disclosed. This study aims to determine the optimum temperature at the annealing stage and to select primers with the best amplicon as the first step to reveal the genetic diversity of F. fistulosa and F. variegata. The molecular marker used is inter simple sequence repeats (ISSR) which is a molecular marker with a very high level of polymorphism and is quite often used to reveal genetic diversity in plants. A total of four samples of F. fistulosa and F. variegata and 10 ISSR primers were used in this study. Modifications in the amplification stage can be done by changing the temperature used based on certain intervals. A good amplicon is influenced by the optimum temperature conditions at the annealing stage when amplification is carried out. Seven ISSR primers were successfully amplified at an annealing temperature of 52 C, while the other two primers were amplified at an annealing temperature of 53 C. The ISSR 5 primer could not be amplified even when the third annealing temperature modification was carried out, so it was not used in the next study. Nine ISSR primers with known optimum temperatures can be used in the study of embodiment of genetic diversity of Ficus spp. in the Bogor City and Mount Botol areas.

Abstract

Various plants were planted as shade trees and ornamentals in campus of UIN Walisongo Semarang. Many plants were also growing wild in this area. Plant dentification is an important step so that people know about plant diversity and plant potentials. Therefore, this research aimed to provide information on the diversity of plants and their potentials in UIN Walisongo Semarang. Research was conducted by using survey method, identification, documentation, species validation and literature review. Field observations were conducted in Campus I, II and III in Ngaliyan Semarang. This research obtained approximately 231 species, which were classified based on wild or cultivated, their habits, and their potentials description. Many plants also used as teaching material in botanic course.

Abstract

Indonesia is one of a country with high biodiversity and genetic resources. One of them is Madura cattle that derived from crossing between exotic cattle, namely zebu cattle with local Indonesian cattle, namely Bali cattle. Branched-chain alpha-keto acid dehydrogenase is an enzyme complex that metabolizes branched chain amino acids, namely valine, leucine and isoleucine. This study aimed to analyze exons 2-3 variation of the BCKDHA gene and its flanking region in Madura cattle. The sequences of it were obtained using DNA sequencing techniques. A total of seven variations were found, one variation located in the flanking region adjacent to the 5' end of exon 2 of the BCKDHA gene. The A392G variation found in exon 3 does not cause any amino acid changes. There is no variation found in exon 2, it indicates that the exon regions of BCKDHA gene are more conserved than introns in the Madura cattle.

Abstract

Homalomena pexa, a tomentose haired-leaf aroid, was a newly discovered and described plant species in early 2020. This species has currently only been reported from South Tapanuli. A molecular study to provide DNA sequence is essential in this preliminary investigation. This research aimed to characterize the barcode sequence of H. pexa and estimate the species' position in an Araceae phylogenetic tree. This research used ITS sequence to perform DNA barcoding on H. pexa. The sequencing result revealed that 1040 bp nucleotides were effectively amplified. The phylogenetic tree generated using the Neighbor-Joining method and the Kimura 2-parameter revealed that H. pexa clustered with H. atrox and H. humilis, with a bootstrap value of 97%. This investigation provided and demonstrated that ITS sequences could be used to validate and support the proper identification of Araceae species.

Abstract

The Community Service Program (KKN) is a manifestation of the tridharma of higher education in the form of community service. Canggal Village in Temanggung, where the majority of the population works in the agricultural sector, was chosen as the location for the KKN by Walisongo State Islamic University and Gunung Djati State Islamic University in Bandung. One of the main programs carried out is the Islamic Generation Festival, a competition event for TPQ children from two hamlets in the village, with the aim of strengthening brotherhood and improving Islamic character. The competitions held include the call to prayer, memorization of short letters, calligraphy, and quizzes with Islamic themes. This festival not only aims to improve the Islamic character of children but also as a means of fostering friendship between TPQ teachers in various hamlets. As a result, this festival succeeded in gathering more than 80 participants with high enthusiasm, where participants showed increased self-confidence in developing their potential, especially in the field of religion. This activity is expected to become a routine activity that will continue in the future.
