Abstract

The main objective of this research was to investigate the potential of Lactic Acid Bacterial isolates and their supernatants, isolated from yoghurt and kefir, to control the growth of A. flavus FNCC6109 in chicken feed concentrate. The experiments were conducted both In vitro and directly applied to chicken feed concentrate. The results showed that two Lactic Acid Bacterial were successfully isolated, identified and both showed initial potential to be developed as biocontrol of A. flavus FNCC6109. Only one best isolate (isolate I) was used in the further investigation. Percentage of growth reduction of A. flavus FNCC6109 in the chicken feed concentrate treated with 30%(v/v) of Lactobacillus sp. supernatant was 72% lower than control (chicken feed concentrate without addition of Lactobacillus sp. Abstract

Cananga odorata and Cymbopogon citratus are two plant species producing inhibitory compounds in their essential oils. In our current research, the potential of their essential oils to inhibit the growth of Staphylococcus aureus and Candida albicans were investigated. MIC (Minimum Inhibitory Concentration) values of mixture of those essential oils with a ratio of 1:1 on those microbes were also assayed. Bioassays were conducted on Nutrient Agar (NA) or Potato Dextrose Agar (PDA) medium added with various ratios of the two plant species essential oils. Medium added with sterile distilled water served as control. The data obtained were analyzed quantitatively with SPSS software for Windows version 22. When significant different at p<0.05 was indicated, the analysis was continued with Duncan's multiple range test. The result showed that all treatment of essential oils applied effectively eliminated the growth of S.aureus. C.albicans appeared to be relatively resistant to 100% of C.odorata essential oil. Elimination of C.albicans was observed when the essential oil of C.odorata was combined with that of C.citratus. When compared to control, all ratios of essential oils treatments were statistically significant (p<0.05). The MIC values of essential oil mixture a ratio of 1:1 on S.aureus and C.albicans were 0,1% and 2,6% respectively. Abstract

Carbapenem Resistant Enterobacteriaceae (CRE) is a group of Enterobacteriaceae resistant to Carbapenem antibiotics (Imipenem, Ertapenem, Meropenem, Doripenem). One species of Enterobacteriaceae frequently showing resistant characteristic to Carbapenem is Klebsiella pneumoniae. Early detection methods on the CRE existence (phenotypically or genetically) are urgently needed. The methods should be able to detect carbapenemases or genes encoding these types of enzymes. In the recent years, phenotypic characteristics can be done by applying methods, such as Modified Hodge Test (MHT), Carba Nordmann-Poirel (Carba NP), or Modified Carbapenem Inactivation Method (MCIM). To detect the presence of genes encoding production of carbapenemases, Polymerase Chain Reaction (PCR) is the most reliable method available in the recent years. Abstract

Tanaman cabai (Capsicum annuum L.) merupakan salah satu komoditas hortikultura yang banyak dibudidayakan oleh petani di Indonesia. Kebutuhan cabai setiap tahun mengalami peningkatan, sedangkan produksinya sangat rendah, salah satu penyebab menurunnya produksi cabai adalah adanya gangguan penyakit layu bakteri, yang disebabkan oleh Ralstonia sp. Untuk menanggulanginya, maka perlu dikembangkan metoda biokontrol yang lebih aman dan ramah lingkungan. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi patogen Ralstonia sp. pada tanaman cabai, serta untuk mengetahui efektivitas kultur Bacillus sp. B3 pada percobaan skala rumah kaca dalam menghambat bakteri Ralstonia sp. pada tanaman cabai. Djereng dkk. (2016), melaporkan Bacillus sp. B3 yang diisolasi dari produk CustomBio mampu menghambat Ralstonia sp. secara in vitro. Isolat Ralstonia sp. diisolasi dengan Platting Method menggunakan media selektif Sucrosa Peptone Agar (SPA) kemudian dilakukan uji Postulat Koch dan bakteri yang didapat dilakukan Uji pewarnaan Gram, uji Gula-gula, uji katalase, uji sitrat, dan hasil disesuaikan dengan buku identifikasi bakteri dari Holt et. al. (1994). Rancangan penelitian yang digunakan pada uji skala rumah kaca adalah Rancangan Acak Kelompok (RAK) dengan parameter yang diukur meliputi lebar daun, panjang daun, panjang akar, tinggi batang, dan diameter batang. Data pengukuran dianalisis dengan Analysis of Varian (ANOVA), kemudian dilanjutkan dengan uji Tukey taraf signifikan 5%. Hasil penelitian menunjukkan bahwa bakteri Ralstonia sp. berhasil diisolasi dan diidentifikasi dari tanaman cabai dengan gejala layu bakteri. Isolat Bacillus sp. B3 mampu menghambat penyakit layu bakteri pada tanaman cabai dengan penurunan insiden infeksi sebesar ± 100% (100% survive), relatif terhadap kontrol (pot yang diinokulasi dengan patogen saja) pada percobaan skala rumah kaca selama 4 minggu.

Abstract

Indonesia is a tropical country with an area of coconut plantation of 3.7 Ha. The fruits of this plant have been used as materials in the production of Virgin Coconut Oil (VCO). Such oil is known to contain higher Medium Chain Fatty Acid (MCFA) or Medium Chain Triglycerides (MCTs) than original frying oil. The MCTs of this oil is very useful to avoid obesity, to reduce total cholesterol as well as to function as anti-microbes. These have been considered as a cheap solution to solve health related problems. The main objective of this research was to produce VCO catalyzed by pure culture of Lactobacillus plantarum isolated from a commercial VCO (Vici’O) combined with enzymes extracted from pineapple fruits. The study was conducted at the laboratory of Microbiology, Faculty of Mathematics and Sciences, Udayana University. The study was started with isolation and identification of Lactobacillus sp. from a commercial VCO. The VCO production method combined the two starters of Lactobacillus isolates (at concentrations of 0 to 10% v/v) and pineapple juice with various volumes (0 to 60 mL). In the first stage of the project, 3 potential Lactobacilli isolates (Sp1, Sp2, and Sp3) were successfully isolated and were confirmed to be Lactobacillus sp. In the main experiment, combination of 10% v/v Lactobacillus starter and 60 mL pineapple juice produced the highest VCO yield (43,17% v/v) with improved organoleptic properties, lower water content, lower saphonification, and lower peroxide than that specified by SNI No 7381: 2008.

Abstract

The main objective of research was to investigate the effectiveness of Schleichera oleosa (Lour) Oken crude extract to inhibit the in vitro growth of E. coli. The crude extract of the plant and the E. coli used in this study were obtained by applying the method of maceration in methanol solution and the method of dilution and spread plate on nutrient agar (NA) medium, respectively. The in vitro bioassay of the extract on E. coli was conducted by applying the method of diffusion on NA medium. The results showed that the crude extract of Schleichera oleosa (Lour) Oken was found to be effective to inhibit the growth of E. coli in the in vitro tests. The diameter of inhibition zone was proportional with the extract concentration exposed to the E. coli.

Abstract

Various attempts have been made by the government to improve the quality of teachers and quality ofeducation, such as allowances certification, training or the opportunity to continue to pursue education toMaster degree. However, these efforts have not able to improve the ability of research as mandated in theMinister Regulation PAN No. 16 Year 2009 Clause 11 that concerning the obligation of teachers in scientificpublications. To assist teachers in implementing the mandate, it is necessary to develop a model of coachingso that teachers can improve their competence and /or teacher professionalism in research. In this paper, it isdeveloped model guidance for teachers in improving the professionalism of teachers in research. The methodused in the development of this coaching model is a method of in-house training, the coaching done bysetting their respective schools, where teachers implement instructional programs. This paper is the result ofthe program community service activities are funded by DIKTI in the scheme of science and technology forsociety that was summarized in the form of coaching model through in-house training which consists of threestages approaches, namely scientific, competence, and publications. The results of this development showeda significant increase, approximately 30% on a teacher's ability to perform research of class action.

Abstract

Sucrose, electrolyte, vitamin, as well as probiotic bacterial contents of palm juice (Arrenga pinnata Merr) areuseful for the maintenance of the healthiness of human body. Due to its low glycemic index, palm sugar issafe to consume and has high potential for use as an alternative companion sugar for diabetes mellituspatients. In Taro village, almost 90% of the people there manage palm sugar plantations, but their ability toprocess and distribute their products is still limited. This program of Science and technology for the Societyaims to improve the quality and value of FEs palm juice by applying revitalization and diversificationmethods. Revitalization was conducted by socializing pre and postharvest preparation techniques andprocedures and the outcomes were measured by questionnaires. Diversification methods were conductedthrough training and advisory preparation of isotonic and probiotic drinks, as well as production of sugarcrystal. The results of this socialization showed that there was an increase in average knowledge of farmersfrom 22.57% (before socialization) to 73.87% (after socialization). Product diversification did not onlyincrease the value of FEs palm juice, but also saved energy and time of processing. As the product wassupported by credible results of laboratory tests, this product is currently marketed in some touristdestinations, such as Tegallalang, Gianyar.

Abstract

Youth Scientific Work (KIR) basically aims to trigger curiosity about natural phenomena related to science and technology. KIR can also increase the ability to think critically about natural phenomena and increase creativity that fosters creative ability and critical thinking. The purpose of this assistance is to introduce research methods for groups of teachers so students get better quality coaching. The mentoring method applied is In-House-Training, which is the implementation of mentoring at the relevant school. In this assistance 4 scientific works have been produced by high school students of Dwijendra Denpasar who are ready to be presented in KIR competitions both regionally and nationally.

Abstract

Teh rosella kombucha lokal Bali dibuat dengan memfermentasikan teh rosella dengan bantuan simbiosis bakteri dan yeast. Diketahui bahwa teh kombucha memiliki aktivitas antioksidan dan aktivitas tersebut dipengaruhi oleh mikroorganisme yang terdapat di dalamnya. Tujuan dari penelitian ini untuk mengetahui karakteristik mikroorganisme yang terdapat dalam teh rosella kombucha lokal Bali. Penentuan karakteristik mikroorganisme dalam teh rosella kombucha lokal Bali dilakukan dengan metode pewarnaan, uji sifat biokimia, motility, pengamatan sifat morfologi dan koloni mikroorganisme serta cellular arrangement. Diketahui bahwa dalam teh rosella kombucha lokal Bali terdapat mikrooganisme yang memiliki kemiripan ciri dengan bakteri genus Acetobacter dan yeast genus Schizosaccharomyces, Saccharomyces, Toruluspora dan Zygosaccharomyces.
