Abstract

Tujuan dari penelitian ini untuk mengetahui penambahan bovine serum albumin dan fruktosa dalam pengencer sitrar-kuning telur terhadap kualitas semen cair babi Landrace. Materi penelitian yang digunakan adalah semen segar dari babi jantan yang telah dewasa kelamin 2,5 tahun dan ternak dalam keadaan sehat. Penelitian ini menggunakan metode eksperimen dan rancangan acak lengkap (RAL) yang terdiri dari 5 perlakuan dan 5 ulangan. Perlakuan tersebut terdiri atas P0= S-KT, P1= S-KT + BSA 0,1% + FRUC 1,0%, P2= S-KT + BSA 0,2% + FRUC 1,0%, P3= S-KT + BSA 0,1% + FRUC 1,5%, P4= S-KT + BSA 0,2 + FRUC 1,5%. Variabel penelitian meliputi motilitas, viabilitas, abnormalitas, dan daya tahan hidup spermatozoa. Hasil dari penelitian menunjukan bahwa P2 menghasilkan kualitas spermatozoa yang lebih tinggi secara statistic (P>0,05%) dibandingkan perlakuan lainnya dengan motilitas: 40,00±0,00%, viabilitas 45,10± 0,55%, abnormalitas 9,80 ±0,45%, dan daya tahan hidup spermatozoa 60,00± 0,00%. Simpulan dari penelitian ini penambahan BSA 0,2% dan fruktosa 0,1% dalam pengencer sitrat-kuning telur memberikan respon baik dan cukup efektif dalam mempertahankan kualitas spermatozoa babi Landrace hingga 60 jam penyimpanan.   Kata kunci: Bovine serum albumin, fruktosa, kuning telur, sitrat, spermatozoa

Abstract

Abstrak Penurunan populasi ternak babi sebagai akibat terserang virus ASF dan pemotongan ternak babi dalam jumlah besar pada berbagai kebutuhan sosial-budaya dan ekonomi, telah berakibat langsung pada kelangkaan pasokan ternak babi sekaligus peningkatan harga ternak yang sangat tinggi di Kabupaten SBD. Pemeliharaan ternak babi secara tradisional sulit untuk melakukan peningkatan populasi secara masif dan signifikan. Selain meningkatkan upaya biosekuriti, penerapan teknologi reproduksi berupa inseminasi buatan (IB) pada ternak babi di SBD sangat urgen untuk peningkatan populasi secara mudah dan murah. Metode yang digunakan dalam PKM ini adalah penyuluhan dan pelatihan IB kepada kelompok peternak Harapan Baru sebanyak 25 peserta (mencakup peternak babi, pemuda dan mahasiswa setempat) untuk peningkatan pengetahuan dan keterampilan terkait dengan penerapan IB. Materi penyuluhan mencakup pemeliharaan induk dan pejantan, pengamatan berahi, penampungan semen, aplikasi teknologi IB, membantu kelahiran dan pemeliharaan anak babi secara baik. IB dilakukan pada 2 induk babi yang diketahui sedang berahi. Aplikasi berjalan lancar dan disaksikan oleh seluruh peserta dengan motivasi yang kuat dan antusiasme yang tinggi. Berdasarkan hasil monitoring dan evaluasi, ternak babi yang di-IB dalam kegiatan PKM tersebut telah berhasil bunting dan melahirkan dengan normal anak babi yang sehat dengan jumlah anak masing 4 ekor (dari induk bunting pertama kali) dan 9 ekor (dari induk yang sudah pernah bunting sebelumnya). Kata kunci: babi, pelatihan, inseminasi buatan Abstract The decline in the pig population due to the ASF virus and the large-scale slaughter of pigs for various socio-cultural and economic needs has directly resulted in a shortage of pig supplies and a very high increase in livestock prices in SBD Regency. Traditional pig farming makes it difficult to increase the population massively and significantly. In addition to improving biosecurity efforts, the application of reproductive technology in the form of artificial insemination (AI) in pigs in SBD is very urgent for easy and affordable population increase. The method used in this PKM was AI counseling and training for the Harapan Baru farmer group of 25 participants (including pig farmers, local youth and students) to increase knowledge and skills related to the application of AI. The extension materials included the maintenance of sows and bulls, monitoring estrus, semen collection, application of AI technology, assisting in the birth and proper care of piglets. AI was performed on two sows known to be in estrus. The application went smoothly and was witnessed by all participants with strong motivation and high enthusiasm. Based on monitoring and evaluation results, the pigs in the PKM program successfully became pregnant and gave birth to healthy piglets, with four piglets (from the first-time sow) and nine piglets (from the previously pregnant sow). Abstract

This study aimed to determine the effect of sugarcane juice substitution in a citrate–egg yolk diluent (CE-Y) on the quality of frozen semen of landrace boars. The semen used in this study was collected from four landrace boars aged between 2–2.5 years using the glove gland method. The study employed a Completely Randomized Design with five treatments and four replications, namely T0 = 100% S-KT + 0% sugarcane juice, T1 = 90% S-KT + 10% sugarcane juice, T2 = 80% S-KT + 20% sugarcane juice, T3 = 70% S-KT + 30% sugarcane juice, and T4 = 60% S-KT + 40% sugarcane juice. All treatments were supplemented with 6% glycerol. Semen was evaluated macroscopically and microscopically. After evaluation, a holding time of 2 hours was applied in S-KT diluent with a ratio of 1:1 at a temperature of 24–26°C. The semen was then centrifuged at 3000 rpm for 15 minutes. The pellet was diluted using a base diluent and then distributed into tubes containing the treatment diluent. The semen was packaged in 0.5 mL straws and equilibrated for 2 hours at 3–5°C. After equilibration, the straws were frozen at a distance of 10 cm above the surface of liquid nitrogen for 10 minutes. The straws were then plunged into liquid nitrogen and stored in a liquid nitrogen container. Thawing was carried out by immersing the frozen semen straws in warm water at 37°C for 30 seconds. After dilution and equilibration, sperm quality was evaluated based on motility, viability, and abnormalities. Post-thaw evaluation included motility, viability, abnormalities, and recovery rate. All obtained data were analyzed using SPSS 27.0. The results showed that sugarcane juice substitution had no significant effect (P>0.05) on motility, viability, abnormalities, and RR. It was concluded that the substitution of sugarcane juice with the addition of citrate–egg yolk diluent did not affect the quality of frozen spermatozoa of landrace boars at the stages of post-dilution, post-equilibration, and post-thawing.
