Abstract

The mangrove ecosystem area in the coastal area of Tatengesan Village, Pusomaen District, Southeast Minahasa Regency has been developed as a mangrove ecotourism area which has been managed by Village-Owned Enterprises (BUMD) since January 2019 with assistance from the Mandiri Rural National Community Empowerment Program (PNPM) team. The development of this mangrove ecotourism activity was stopped for a moment due to the impact of the COVID-19 pandemic, but currently, COVID-19 cases are considered endemic so this mangrove ecotourism activity will be developed again. This research aims to formulate some strategies for the sustainable use of the mangrove ecosystem on the Tatengesan coast, Southeast Minahasa Regency. This research lasted for 5 months, from September 2022 - February 2023. This research used descriptive qualitative methods with survey technique, i.e., in-depth direct interviews with resource persons (respondents). The formulation of priority strategies (key success factors) was analyzed using S.W.O.T and strategy choice analysis. This research produced 9 (nine) priority strategies as academic texts, which can be recommended to governments related to the marine and fisheries sector as elements of public policy-making officials. Abstrak Kawasan ekosistem mangrove di wilayah pesisir Desa Tatengesan, Kecamatan Pusomaen, Kabupaten Minahasa Tenggara dikembangkan sebagai kawasan ekowisata mangrove yang dikelola oleh Badan Usaha Milik Desa (BUMD) sejak Januari 2019 dengan pendampingan oleh tim Program Nasional Pemberdayaan Masyarakat (PNPM) Mandiri Perdesaan.  Kegiatan ekowisata mangrove ini pengembangannya terhenti sejenak dikarenakan adanya dampak pandemi Covid-19, namun saat ini kasus Covid-19 sudah dianggap endemi sehingga kegiatan ekowisata mangrove ini akan dikembangkan kembali.  Tujuan penelitian ini yaitu merumuskan strategi pemanfaatan berkelanjutan ekosistem mangrove ke arah berkelanjutan di pesisir Tatengesan Kabupaten Minahasa Tenggara.  Riset ini berlangsung selama 5 bulan, dari September 2022 - Februari 2023.  Riset ini menggunakan metode kualitatif deskriptif dengan teknik survei, yaitu wawancara langsung secara mendalam (in-depth interview) kepada narasumber (responden).  Perumusan strategi prioritas (faktor kunci keberhasilan) di analisis dengan menggunakan S.W.O.T dan analisis pilihan strategi.  Penelitian ini menghasilkan 9 (sembilan) strategi prioritas sebagai naskah akademik, yang dapat direkomendasikan kepada pemerintah terkait pada sektor kelautan dan perikanan sebagai unsur pejabat pembuat kebijakan publik. Abstract

This research aims to assess the status of snapper and grouper fisheries management in the Sangihe Islands district using an ecosystem approach (EAFM) in the institutional domain. The research method uses observation and interview methods (questionnaires). Data collection used semi-structured interview (SSI) techniques. As a result of the assessment of 6 (six) institutional indicators, there are 2 indicators with a value of 1 (less, red flag model), namely indicators of compliance with the principles of responsible fisheries and indicators of fisheries management plans. 3 indicators each: decision-making mechanism indicators, indicators of the level of synergy of fisheries management policies & institutions, and stakeholder capacity indicators) with a value of 2 (medium, yellow model flag). Only the indicator for the completeness of the rules in fisheries management has a value of 3 (good, green model flag). The average score for the 6 indicators is 1.76 while the composite value is 58.53. This value shows that the application of the EAFM institutional domain in the management of snapper and grouper fisheries in the Sangihe Islands district is in the medium category (yellow model flag). Abstrak Penelitian ini bertujuan untuk menilai  status pengelolaan perikanan kakap dan kerapu di kabupaten Kepulauan Sangihe dengan pendekatan ekosistem (EAFM) pada domain kelembagaan. Metode penelitian menggunakan metode observasi dan wawancara (kuisioner). Pengumpulan data menggunakan teknik semi structured interview (SSI).  Hasil penilaian dari 6 (enam) indikator kelembagaan, ada 2 indikator yang nilai-nya 1 (kurang, flag model merah) yaitu  indikator kepatuhan terhadap prinsip-prinsip perikanan yang bertanggung jawab dan indikator rencana pengelolaan  perikanan. 3 indikator masing-masing : indikator mekanisme pengambilan  keputusan, indikator tingkat sinergisitas kebijakan & kelembagaan pengelolaan perikanan, indikator kapasitas pemangku  kepentingan) bernilai 2 (sedang, flag model kuning). Hanya indikator kelengkapan aturan main dalam  pengelolaan perikanan yang bernilai 3 (baik, flag model hijau).  Nilai skor rerata 6 indikator adalah 1,76 sedangkan  nilai komposit-nya 58,53. Nilai ini menunjukkan bahwa penerapan domain kelembagaan EAFM pada pengelolaan perikanan kakap dan kerapu di kabupaten Kepulauan Sangihe kategori sedang (flag model kuning). Abstract

This research aims to determine the potential for marine tourism in East Likupang Waters, determine the suitability of developing marine tourism in East Likupang Waters, and recommend marine tourism management, collecting primary data through observation and in-depth interviews directly with the community. Secondary data through studies of research results, scientific publications, the Minahasa Regency Maritime and Fisheries Service, and the North Minahasa Regency Maritime and Fisheries Service. Biophysical data is as follows. Observation of coral conditions using Line Intercept Transect (LIT). b) Observation of fish using Underwater Fish Visual Census (UVC). Water quality using a water quality checker (Horiba brand). Apart from that, data analysis uses a matrix of suitability for marine tourism areas, and beach tourism and provides weighting. The results of this research show that the potential that exists in East Likupang Waters and its surroundings includes physical and non-physical potential that has the potential and can be developed into a marine tourism attraction. Keywords: Potential, Marin Tourism, East Likupang Waters Abstrak Penelitian ini bertujuan untuk mengetahui potensi wisata bahari di Perairan Likupang Timur, mengetahui kesesuaian pengembangan wisata bahari di Perairan Likupan Timur dan merekomendasikan pengelolaan wisata bahari, pengumpulan data primer melalui observasi dan wawancara mendalam langsung kepada masyarakat. Data sekunder melalui kajian hasil penelitian, publikasi ilmiah, Dinas Kelautan dan Perikanan Kabupaten Minahasa, dan Dinas Kelautan dan Perikanan Kabupaten Minahasa Utara. Data biofisik adalah sebagai berikut. Pengamatan kondisi karang menggunakan Line Intercept Transect (LIT). b) Pengamatan ikan menggunakan Underwater Fish Visual Census (UVC). Kualitas air menggunakan alat pemeriksa kualitas air (merek Horiba). Selain itu analisis data menggunakan matriks kesesuaian kawasan wisata bahari, dan wisata pantai serta memberikan pembobotan. Hasil penelitian menunjukkan bahwa potensi yang ada di Perairan Likupang Timur dan sekitarnya meliputi potensi fisik dan non fisik yang mempunyai potensi dan dapat dikembangkan menjadi daya tarik wisata bahari. Daya dukung dan rekreasi pantai Pantai Surabaya Luas pantai 2000 (m²) 533 orang/hari Pantai Pulisan 2000 (m²) 533 orang/hari c) Pantai Paal 1000 (m²) 267 orang/hari Pantai Kalinaun 3000 (m²) 800 orang/hari, Diketahui persentase tutupan karang di Perairan Likupang Timur sebesar 47,04% dengan nilai indeks kesesuaian wisata bahari sebesar 62 yang berarti kesesuaian kawasan wisata bahari Perairan Likupang Timur termasuk dalam kriteria layak bersyarat. Abstract

North Sulawesi Province, directly bordering the Pacific Ocean, is located in an area with the highest level of tropical cyclone (TC) activity in the world. As a result, the province is vulnerable to the impacts caused by cyclones, including storm surges. The increase in water levels due to this event has the potential to cause coastal flooding. Previous studies in Manado have identified that sea level rise can be detected through residual water level (RWL), making studying the characteristics of RWL in North Sulawesi important. This research focuses on Manado and Melonguane, allowing for a comparison of characteristics. The data used includes tropical cyclone data from the China Meteorological Administration (CMA) and tidal data from the Geospatial Information Agency. The Unified Tidal Analysis and Prediction (UTide) method is used to identify RWL. The analysis was carried out by using a t-test to compare data at the two locations. The results showed that RWL at those locations had significant differences with Melonguane having the higher value between them. Generally, the increase in RWL in Melonguane occurs shortly after the cyclone period, while the RWL in Manado maximum increases 86 hours after TC's first occurrence. Abstract

This study aims to identify the type, size dimension, and composition of nematocysts from the Scleractinia coral, Stylophora subseriata. This study was carried out from March to May 2023. Observations and measurements of nematocyst cells were conducted using an Optika 4083 B3 microscope connected to a computer equipped with Optika View 7 software. S. subseriata has three main types of nematocyst, i.e., type I micro basic p-mastigophore (MpM- I), type II micro basic p-mastigophore (MpM-II) and micro basic b-mastigophore (MbM).  Type MpM-I has an average capsule length of 182.10 µm, capsule width of 38.98 µm, and shaft length of 76.45 µm;  MpM-II has an average capsule length of 166.72 µm, capsule width of 41.60 µm, and shaft length of 108.48 µm; and MbM has an average capsule length of 154.84 µm and capsule width of 30.96 µm. The nematocysts composition consists of MpM-I 26.67 %, MpM-II 23.33 %, and MbM 50.00 %.  The MbM type is nematocyst dominant in this coral species.  Future study is recommended to examine the comparative characteristics of nematocysts from S. subseriata with other species of corals in the same genus to determine the specific nematocyst of each type of coral with the role of their particular types of nematocyst. Studi ini dilakukan dari Maret - Mei 2023. Pengamatan dan pengukuran sel nematosit dilakukan menggunakan mikroskop Optika 4083 B3 yang terhubung dengan komputer yang dilengkapi software Optika View 7. S. subseriata memiliki tiga tipe nematosit utama, yaitu microbasic p-mastigophore tipe I (MpM-I), microbasic p-mastigophore tipe II (MpM-II) dan microbasic b-mastigopore (MbM).  Tipe MpM-I memiliki rata-rata panjang kapsul 182,10 µm, lebar kapsul 38,98 µm, dan panjang tangkai 76,45 µm;  MpM-II memiliki rata-rata panjang kapsul 166,72 µm, lebar kapsul 41,60 µm, dan panjang tangkai 108,48 µm; serta MbM dengan panjang kapsul 154,84 µm dan lebar kapsul 30,96 µm. Komposisi nematositnya terdiri dari MpM-I 26,67 %, MpM-II 23,33 % dan MbM 50.00 %.  Tipe MbM adalah nematosit yang dominan dalam spesies ini.  Penelitian selanjutnya disarankan untuk meneliti perbandingan antara karakteristik nematosit dari S. subseriata dengan jenis karang lainnya dalam satu genus untuk mengetahui nematosit spesifik dari masing-masing jenis karang tersebut sehubungan dengan peran dari tipe-tipe nematosit spesifiknya.       Kata kunci: Minahasa Utara, nematosit, Pesisir Bulutui, Scleractinia, Stylophora subseriata

Abstract

The research aimed to determine the recruitment density value of scleractinian coral juveniles in Tanjung Pisok (Meras and Bahowo) and Siladen Island. The method for selecting observation locations uses purposive sampling and collecting coral juvenile recruitment data using underwater photo transects. The results of this research obtained data on genus, size, and density values ​​for coral juvenile recruitment. The coral genera found at Station I were 19 genera with the largest number being the Porites genus, 32 colonies, Station II 13 genera with the largest number being the Favites genus, 32 colonies, and at Station III 17 genera with the largest number being the Porites genus, 32 colonies. Coral juveniles in this study were more dominant in the 2 cm - 2.9 cm size group, for Station I 63 colonies, Station II 54 colonies and Station III 32 colonies. Based on the results of this research, it shows that the coral recruitment density at Station I with a value of 12.38 colonies/m² is higher than Station II, the coral recruitment density at Station II with a value of 12.23 colonies/m² is higher than Station III with a value of 7.21 colonies/m². Metode pemilihan lokasi pengamatan menggunakan purposive sampling dan metode pengumpulan data rekrutmen juvenil karang menggunakan underwater photo transect. Hasil penelitian ini memperoleh data genus, ukuran dan nilai densitas rekrutmen juvenil karang. Genus karang yang didapat di Stasiun I yaitu 19 genus dengan yang terbanyak genus Porites 32 koloni, Stasiun II 13 genus dengan yang terbanyak genus Favites 32 koloni, dan Stasiun III 17 genus dengan yang terbanyak genus Porites 32 koloni. Juvenil karang dalam penelitian ini lebih dominan pada kelompok ukuran 2 cm - 2.9 cm, untuk Stasiun I 63 koloni, Stasiun II 54 koloni dan Stasiun III 32 koloni. Berdasarkan hasil penelitian ini menunjukkan densitas rekrutmen karang di Stasiun I dengan nilai 12,38 koloni/m² lebih tinggi dari Stasiun II, densitas rekrutmen karang di Stasiun II dengan nilai 12,23 koloni/m² lebih tinggi dari Stasiun III dengan nilai 7,21 koloni/m². Abstract

This study was conducted for three months, starting from March to June 2024. The research method used was a qualitative descriptive method with a survey technique.  Data collection was carried out by collecting primary data and secondary data.  Data analysis to formulate priority strategies for developing mangrove ecotourism at Budo Tourism Village using SWOT analysis (strength, weakness, opportunity, threat) and strategy choice analysis to obtain key success factors (priority strategies). The results of this research obtained 7 (seven) priority strategies that can be used by stakeholders, especially BUMDES managers in Budo Village for the development of sustainable mangrove ecotourism. To create a public policy, it is recommended to the Budo Village government, especially the BUMDES manager, to pay attention and consider academic studies from the Faculty of Fisheries and Marine Sciences, UNSRAT, in the form of 7 (seven) key success factors or priority strategies. Keywords: key success factors, mangrove ecotourism, SWOT, strategic choice analysis   Abstrak   Riset ini dilaksanakan di kawasan ekowisata mangrove Desa Wisata Budo dan berlangsung selama 3 bulan, dimulai dari bulan Maret sampai Juni 2024.  Metode riset yang digunakan yaitu metode deskriptif kualitatif dengan teknik survei.  Pengumpulan data dilakukan dengan mengumpulkan data primer dan data sekunder.  Analisis data untuk merumuskan strategi pengembangan ekowisata mangrove Desa Wisata Budo menggunakan analisis SWOT (strength, weakness, opportunity, threat) dan analisis pilihan strategi untuk memperoleh faktor-faktor kunci keberhasilan (strategi prioritas). Hasil penelitian ini memperoleh 7 (tujuh) urutan strategi prioritas yang dapat digunakan oleh stakeholders, khususnya pengelola BUMDES Desa Budo untuk pengembangan ekowisata mangrove yang berkelanjutan. Dalam rangka membuat suatu kebijakan publik direkomendasikan kepada pemerintah Desa Budo, khususnya pengelola BUMDES untuk memperhatikan dan mempertimbangkan kajian akademik dari Fakultas Perikanan dan Ilmu Kelautan Unsrat berupa 7 (tujuh) faktor-faktor kunci keberhasilan atau strategi prioritas tersebut. Abstract

Semi-aquatic bugs of the family Veliidae (Hemiptera: Heteroptera: Gerromorpha) are primarily associated with freshwater habitats; their occurrence in coastal environments remains poorly documented, particularly in tropical regions. This study reports the first record of Veliidae from the coastal waters of Tasik Ria Beach, North Sulawesi, Indonesia, thereby expanding the known ecological distribution of this family. Specimens were collected from surface-water microhabitats using a sweep net, and key environmental parameters were measured in situ. Morphological identification was based on stereomicroscopic examination of diagnostic characters. Molecular confirmation was obtained through DNA barcoding of the mitochondrial cytochrome c oxidase subunit I (COI) gene, with sequences compared against the GenBank and BOLD databases. Both morphological and molecular analyses confirmed the specimens' taxonomic placement within Veliidae. Their presence in a coastal habitat indicates greater ecological plasticity and habitat tolerance than previously recognized. This finding establishes important baseline data for future research on the distribution, bioecology, and adaptive strategies of semi-aquatic insects in tropical coastal ecosystems.

Abstract

The CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) system is a specific nucleotide sequence in genomes of certain bacteria and archaea that functions as an adaptive immune system against viral infections and foreign genetic elements. However, not all microorganisms possess CRISPR systems; therefore, bioinformatic approaches are required to trace the occurrence of CRISPR-Cas systems. The objective of this study was to detect CRISPR-Cas systems in genomes of Oscillatoria acuminata and Stanieria cyanosphaera. Both species were identified among 2679 operational taxonomic units (OTUs) of microorganisms associated with ascidian Lissoclinum patella originated from Malalayang waters, based on previously reported metagenomic analyses. These species were selected due to their relatively high abundance and the availability of complete genome sequence data in the NCBI database, which were downloaded as FASTA format and analyzed using CRISPRCasFinder software. The results showed that O. acuminata possesses a complete genome sequence of 7,689,443 bp, containing 80 CRISPR arrays, of which two arrays are associated with Cas systems, and a total of 338 spacers. In the other hand, S. cyanosphaera has a complete genome sequence of 5,041,209 bp, with eight CRISPR arrays, two of which are associated with Cas systems, and a total of 37 spacers. The detected CRISPR loci were visualized using CRISPRCasViewer in three display models: linear, circular, and scatter plot. Further studies are required to determine the specific types of CRISPR-Cas systems present in each microbial genome. Keywords: O. acuminata; S. cyanosphaera; bioinformatics; CRISPRCasFinder; CRISPRCasViewer   Abstrak Sistem CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) merupakan susunan sekuens nukleotida spesifik dalam genom bakteri dan arkaea tertentu yang berfungsi sebagai  sistem imun adaptif mikroba terhadap serangan virus dan materi genetik asing. Namun, tidak semua mikroba memiliki sistem CRISPR, sehingga diperlukan pendekatan analisis bioinformatika untuk mendeteksi CRISPR. Tujuan penelitian ini adalah untuk menelusuri keberadaan sistem CRISPR-Cas pada genom Oscillatoria acuminata dan Stanieria cyanosphaera. Kedua 2 jenis mikroba ini ditemukan di antara 2679 operational taxonomic units (OTUs) mikroba yang berasosiasi dengan ascidia Lissoclinum patella dari perairan Malalayang, berdasarkan analisis metagenom yang telah dilaporkan sebelumnya.  Kedua spesies ini dipilih karena terdeteksi cukup melimpah, dan ketersediaan data urutan genom lengkap di NCBI. Data urutan genom diunduh dalam format FASTA dan dianalisis dengan perangkat lunak CRISPRCasFinder. Ternyata O. acuminata memiliki  sekuens genom  lengkap  7.689.443 bp dengan 80 unit  sistem CRISPR, dimana ada 2 unit dengan sistem Cas, serta 338 spacer. Sedangkan S. cyanosphaera memiliki sekuens genom lengkap sekitar 5.041.209 bp dengan 8 unit CRISPR, dan 2 unit dengan sistem Cas serta 37 spacer. Hasil deteksi CRISPR divisualisasikan menggunakan CRISPRCasViewer dalam tiga model tampilan, yaitu Linear, Circular, dan Scatter Plot. Selanjutnya, ke depan perlu penelusuran lebih lanjut untuk menentukan tipe CRISPR-Cas pada setiap genom mikroba tersebut. Abstract

Mangrove forests are key ecosystems in tropical coastal areas that function as environmental stability guards through physical roles (preventing erosion and tsunamis), chemical (absorbing pollutants and storing carbon), and biological (marine biota habitat and source of biodiversity). This study aims to identify the species and determine the dominant species of mangroves and the comparison of tree and sapling composition in the mangrove area of ​​Sonsilo Village, West Likupang District, North Minahasa Regency.  The research was conducted over three months (September–November 2024) using a purposive sampling method to collect data on density, canopy cover, and species frequency. The results revealed seven mangrove species: A. marina, B. gymnorrhiza, C. tagal, R. apiculata, R. mucronata, S. alba, and X. granatum.  Among these, three species (B. gymnorrhiza, R. apiculata, and R. mucronata) were dominant at both tree and sapling levels, exhibiting the highest Importance Value Index (IVI).  Mangrove species diversity in the area is categorized as low to moderate, as indicated by the Shannon-Wiener Diversity Index (H'), reflecting that species richness is not yet fully optimal. Analysis of the Dominance Index (D) and Evenness Index (J') suggests a relatively even distribution of individuals among species.  The study recommends regular monitoring and evaluation, as well as conservation efforts, to maintain the stability of the mangrove ecosystem and support its sustainable use. These findings highlight the importance of biodiversity management in preserving the ecological balance of mangrove habitats. Keywords: Sonsilo Village, mangroves, mangroves composition
