Abstract

The aim of this study was to determine the effect of lipid ratio of stearic acid and oleic acid on the physical characteristics as well as the entrapment efficiency of diethylammonium diclofenac with Nanostructure Lipid Carrier (NLC) system. Diethylammonium diclofenac (DETA) is Non-Steroid Anti-Inflamatory Drugs (NSAIDs) that has been widely used in the treatment of osteoarthritis and rheumatoid arthritis. In the formulation of NLC-DETA, three different lipid ratios were used, which the ratio of  stearic acid:oleic acid were 60:40, 70:30, and 80:20, respectively. In this NLC system, DETA served as the active drug, stearic acid as solid lipid, oleic acid as liquid lipid, and Tween 80 as surfactant components. NLC were characterized for organoleptic characteristics, pH, viscosity, particle morphology, particle size and polydispersity index (PI), profiles of Fourier Transform Infra-Red (FTIR) and Differential Thermal Analysis (DTA), and drug entrapment efficiency. The particle shape and morphology were determined by Transmission Electron Microscopy (TEM). The results  showed that the different ratios of oleic acid lipids and stearic acid had no significant effects on the viscosity and entrapment efficiency of NLC-DETA. On the other hand, it affected the pH of all formulas, which were significantly different. Increasing the amount of liquid lipid in the formulations reduced the size of NLC-DETA particles.

Abstract

Coenzyme Q10, often also known as ubiquinone, coenzyme Q10 or Q10, is soluble in lipids and is naturally present in plants, animals and in mitochondria. Coenzyme Q10 functions as an antioxidant that can protect the body from damage caused by free radicals. Hyaluronic acid is known as a hydrophilic polymer derived from polysaccharides which has the ability to increase percutaneous penetration by changing the composition of tightly arranged stratum corneum cells to increase the permeability of the skin. Nanostructured Lipid Carrier is a modification of the SLN system, consisting of a mixture of solid and liquid lipids (oil), stabilized by aqueous surfactant solution, is one method to increase drug penetration through the stratum corneum because it has several advantages. The purpose of this study was to see the effect of adding hyaluronic acid to the characteristics of the Nanostructure Lipid Carrier (NLC) as anti aging. Examination of characteristics including organoletis, pH, particle size and polidispersity index was carried out. The results of organoleptic NLC coenzym Q10-HA examination obtained dark orange color, liquid consistency, lipid efficacy odor and soft texture. The pH measurement results of the preparation ranged from 5.05-5.23. The results of the particle size examination ranged from 267-128 nm and the particle size distribution ranged between 0.308-0.200 Keywords: Coenzym Q10, Hyaluronic acid , NLC

Abstract

Tomato extract was known contains manyantioxidants (Chauhan et al., 2010), such aslycopene, that are easily degraded by UV-B rays. Themechanism lycopene as an antiaging is to reduceskin regeneration and increase the thickness of theepidermis (Sahasrabuddhe, 2011). As anantioxidant, lycopene easily degraded when exposedto light, heat, and oxygen (Chauhan et al., 2011).Drug delivery system based on nanolipid carriersystem, such as: Solid Lipid Nanoparticles (SLN) wasappropriate to stabilize lycopene, as lipophilicsubstance (Wissing and Muller, 2003; Helgason etal., 2009; and Okonogi dan Riangjanapatee, 2014).One of the limitations of SLN is the drug moleculethat has been trapped expulsed easily duringstorage. It is caused of the orderdness of lattice lipidcrystal. Replacement part of solid lipid using liquidlipid (called as nanostructure lipid carriers or NLC)can reduce the orderdness of lattice lipid crystall andit will repaired it (Kaur et al, 2015).In this research have been tried to be compared theability of SLN and NLC to improve the stability ofantioxidant tomato extract. Using 20% total amountof lipid, extract tomato-SLN made from cetyl alcoholas solid lipid and extract tomato-NLC made fromdifference ratio between cetyl alcohol and isopropylmyristate (IPM) such as: 9:1 and 7:3. Tween 80 andKollipor were used as surfactant and co-surfactant.IPM was choose because of its enhancer andemollient effect so easily to apply to the skin(Vadgama et al, 2015).

Abstract

Background: One of the vital variables affecting the stability and the characteristics of the Nanostructured Lipid Carrier (NLC) is the surfactant concentration. Using the two combinations of surfactants can cause higher stability and a better characteristic of NLC. Tween 80 and Span 20 are anionic surfactants whose combination has not been studied for use in NLC systems. Objective: Determine the effect of different surfactant ratios of Tween 80 and Span 20 on the physicochemical characteristics, stability, and irritability of NLC using the High Shear Homogenization (HSH) method. Methods: Four different surfactant ratios were used in the NLC formulation, in which the ratio of Tween 80:Span 20 were 5:5, 6:6, 7:7, and 8:8, respectively. In this NLC system, cetyl palmitate served as solid lipid, medium-chain triglyceride (CrodamolTM) as liquid lipid, Tween 80, and Span 20 as surfactant components. NLC was characterized for organoleptic, viscosity, pH, zeta potential, particle morphology, particle size, and polydispersity index (PDI), then evaluated for stability using the real-time and freeze-thaw method, and irritability effect. Results: The different ratios of Tween 80 and Span 20 had no significant effect on the particle size, PI, and irritation score of the NLC system. On the other hand, it influenced all formulas' pH value, viscosity, zeta potential, and stability. Conclusions: The different ratios of surfactant combination affect the characteristics and stability of the NLC system.

Abstract

Resveratrol is an antioxidant that can be used as anti-aging. Topical use has several problems because solubility in water is low and unstable to light. This study aimed to evaluate the effect of the combination of soy bean oil and oleic acid liquid lipids on the characteristics, penetration, and stability of resveratrol nanostructured lipid carriers (NLC). Nanostructured lipid carriers (NLC) were made with high shear homogeneous technique. To determine the characterization of NLC, diffraction scanning calorimetry, X-ray diffraction, and Fourier transforms infrared spectrophotometry were used. Examination of the morphological form was carried out with a transmission electron microscope. The particle size and polydispersity index examination were measured by the Delsa Nano™ particle size analyzer, while the efficiency of trapping resveratrol in the NLC system was measured by the dialysis membrane method. Furthermore, the penetration depth test on the skin of mice was done by fluorescence microscope method using rhodamine B markers. Physical stability test was performed by examination of particle size and index polydispersity for 30 days. The formula with liquid soy bean oil and oleic acid liquid lipids improved the characteristics including the effectiveness of entrapment and colloidal stability. However, the formula with soy bean oil and oleic acid combination liquid lipids did not provide better penetration into the skin than the use of single liquid lipids. While the formula with soy bean oil and oleic acid liquid lipids proved to improve the physical stability for 30 days.

Abstract

This research has been carried out to study inhibition of eutectic interaction which occurs on compressed binary mixture of ibuprofen-stearic acid by addition of corn starch. Corn starch was mixed into binary mixture of 40:60 (% b/b) ibuprofen-stearic acid with composition 10, 20, and 30% (w/w) and then compressed into tablet form. The physical characterization was conducted using tensile strength test, differential thermal analysis (DTA), powder X-ray diffraction (PXRD), Fourier transform infrared (FT-IR), and scanning electron microscope (SEM). According to tensile strength test, the value was reduced upon addition of 10 and 20% corn starch compared to the compressed binary mixture. Addition 20% corn starch to compressed binary mixture shifted the eutectic diffraction peaks with a decline in intensity during PXRD study. Thermal analysis of compressed binary mixture-20% corn starch exhibited an increase in eutectic point, and FT-IR spectrum was observed no difference compared to the compressed binary mixture. SEM analysis indicated that 20% corn starch addition was able to inhibit sintering phenomena by dispersed on particle’s surfaces. Physical characterizations result in the conclusion that corn starch can inhibit the eutectic interaction between ibuprofen and stearic acid upon compression
