Abstract

Phenobarbital, a hypnotic and sedative agent, was found to form inclusion complex with hydroxypropyl-β-cyclodextrine at 1:1 molar ratio. The formation of inclusion complex in the solution was detected due to the increased solubility of phenobarbital and in the solid was confirmed by differential scanning calorimetry, X-Ray diffractometry, and infrared spectroscopy. In the solid, inclusion complex was prepared by freezed drying method. The inclusion complex formed was characterized, compared with phenobarbital, hydroxypropyl-β-cyclodextrine, and the physical mixture of phenobarbital and hydroxypropyl-β-cyclodextrine.Analyses using DSC and X-ray diffractometry showed that phenobarbital formed inclusion complex with hydroxypropyl-β-cyclodextrine.Key words : phenobarbital, hydroxypropyl-β-cyclodextrine, inclusion complex

Abstract

Cyclodextrins are cyclic oligosaccharides which have been used extensively to improve the solubility in the formation of inclusion complex. The ability of cyclodextrin to form an inclusion complex has been attributed to factors including size of cyclodextrin cavity to the guest molecule and thermodynamics of their interactions. In this study, complex formation of phenobarbital and hidroxypropyl-β-cyclodextrin in the acetate buffer 0.01 M pH 4.4 and phosphate buffer 0.01 M pH 7.4 all with μ =0.10 and at temperatures of 32.o, 37.o, and 42o±0.5oC were determined. The results showed that phenobarbital formed inclusion complex with hidroxypropyl-β-cyclodextrin. The interaction shown a negative enthalphy (ΔH < 0 ), a spontaneous processes (ΔG < 0) and an increase in the entropy of the system (ΔS > 0).Key words : phenobarbital and hidroxypropyl-β-cyclodextrin, inclusion complex, thermodynamics

Abstract

Aim of this research is to determine physical characteristic of ibuprofen-stearic acid due to compression force. Binary mixture of ibuprofen-stearic acid with weight ratio of 4:6, 5:5, and 6:4 was compressed under various compression force using hydraulic press with 13mm diameter flat punch.Identification of solid state interaction between these two components was performed by Hot Stage Microscopy (HSM). Physical characterization has been studied byDifferential Thermal Analysis (DTA), X-ray Powder Diffraction (XRPD), Scanning Electron Microscopy (SEM), and (Fourier Transform Infrared (FT-IR) spectroscopy. According to hardness measurement, 4:6 weight ratio has the highest tensile strength on 170.6N/cm2 under 19.93kN compression force. Interaction identification by HSM showed single blank line that indicates eutectic formation. Thermal analysis of DTA also revealed eutectic formation upon compression whichthe endotermic peak of ratio of 4:6has the lowest melting temperature of 53.2°C. X-ray diffraction of three peaksgenerally showed that peak intensity decreases as compression force increases, but at particular point it begins to increase again. Sintering phenomenon at the surface of compressed tablet was observed from SEM analysis. FT-IR study confirms the formation of simple eutectic. Key words: ibuprofen, stearic acid, binary mixture, compression force, physical characterization

Abstract

It is known that piper leaves (sirih) from Piper betle Linn has been used traditionally as medicine, for example for mouthwash and wound curing. The usage of hand gels antiseptic preparation was getting increased in the current time as the incidence of desease caused by virus and bacteria like SARS appears. This research was aimed to develop preparation of hand gels from piper leaves extract obtained from extraction with hot water. The formulation of gels were made using 5%, 10%, 15%, 20% and 25% of piper leaves extract in Carbopol gel base. The evaluation of antiseptics activity was carried out using the Replica method. A series of test was done by pouring the gels at hand palm and slightly tap the thumb fingerprint to nutrient agar media. The media was incubated at 37°C for 24 hours, and the number of colonies of microorganism was counted. The antiseptic activity of the gels also compared with two hand gels antiseptic preparation containing ethanol and triclosan as antiseptic agentThe result showed that the piper leaves extract gels had a transparent pale yellow performance. The gel preparation with concentration of 15% extract had activity to reduce the microorganism of skin normal flora up to 50%. Yet the 25% concentration of piper leave extract could eliminate all the microorganism. It was also known that the 15% piper leaves extract gel had no significance different with the ethanol preparation, since the 20% and 25% piper leaves extract gels had similar antiseptic activity to the triclosan preparation.Key words: piper leaves extract, gel, antiseptic activity, replica methods

Abstract

Abstract

The solubility and partition coefficient are essential physicochemical parameters in developing a pharmaceutical dosage form of medicine. In addition, these parameters help to predict the absorption of an active compound in oral or topical dosage forms. Salicylamide, an active ingredient available in oral and topical dosage forms, is a weak acid (pKa 8.2) and is sparingly soluble in water. Meanwhile, its solubility and partition coefficients are influenced by the pH of the environment. The Henderson-Hasselbalch equation is used to predict solubility-pH and partition-pH profiles at various pH solutions. This study aims to determine salicylamide's solubility and partition coefficient in various pH (2–11). Both tests were carried out in various pH buffer solutions (at a concentration of 0.02 M and 0.2 ionic strength) in a water bath shaker at a temperature of 37 ± 0.5 °C. In addition, the salicylamide content was determined using the UV spectrophotometer method at the maximum wavelength at each pH. The results showed that the solubility increased at pH 2–10, while the partition coefficient value decreased. On the other hand, at pH 11, there was an increase in the number of ionized species, but the solubility decreased.
