Abstract

The solubility and partition coefficient are essential physicochemical parameters in developing a pharmaceutical dosage form of medicine. In addition, these parameters help to predict the absorption of an active compound in oral or topical dosage forms. Salicylamide, an active ingredient available in oral and topical dosage forms, is a weak acid (pKa 8.2) and is sparingly soluble in water. Meanwhile, its solubility and partition coefficients are influenced by the pH of the environment. The Henderson-Hasselbalch equation is used to predict solubility-pH and partition-pH profiles at various pH solutions. This study aims to determine salicylamide's solubility and partition coefficient in various pH (2–11). Both tests were carried out in various pH buffer solutions (at a concentration of 0.02 M and 0.2 ionic strength) in a water bath shaker at a temperature of 37 ± 0.5 °C. In addition, the salicylamide content was determined using the UV spectrophotometer method at the maximum wavelength at each pH. The results showed that the solubility increased at pH 2–10, while the partition coefficient value decreased. On the other hand, at pH 11, there was an increase in the number of ionized species, but the solubility decreased.

Abstract

To fulfill the consumer expectation, cosmetic industries mostly add glycolic acid as whitening agent to the sunscreen preparation. The aim of this research was to know the effect of glycolic acid addition with the concentration of 8, 10, and 12% w/w on physical stability and effectiveness of sunscreen product containing oxybenzone and octyl methoxycinnamate (2: 7% w/w) in carbomer 940 gel. The physical stability test was done by observing physical appearance, pH, and spreading ability during 60 day storages on zone IV condition (temperature 30±0.5° and RH 70±2%). The efficacy was determinated base on SPF (Sun Protection Factor) value by spectrophotometer. The result of the physical stability test showed that glycolic acid addition affects the physical appearance (consistency) of sunscreen product, but pH and spreading ability are relatively stable. The efficacy test result showed that the addition of glycolic acid has an effect on SPF value of sunscreen product. Product which combined with glycolic acid showed a significant increase of SPF value, compared to control formula. The addition of glycolic acid increased SPF value and alter the effectiveness category from extra protection to maximal protection.

Abstract

Background: Quercetin is a flavonoid compound that has anti-inflammation activity. However, poor stability presents significant problems for the formulation into dosage forms. Microspheres are one of the potential lung delivery systems because of their ability to encapsulate various types of drugs, protect drugs from environmental effects and can release drugs in a sustained release. Objective: In the present study, the microsphere inhalation system of the anti-inflammation drug, quercetin was developed and evaluated to achieving the targeted delivery of these drugs to the lung. Method: The drug-loaded ca-alginate microspheres were prepared by aerosolization ionic gelation technique followed by freeze-drying. Result: The result of this study showed that particle size was less than 2 µm, the yield ranged from 41.33-76.14%, drug loading was less than 6%, entrapment efficiency ranged from 74.153% - 93.805% and flow properties showed that all formula had an excellent flow. Spherical microspheres were demonstrated by formulations containing 1 and 1.5% sodium alginate. A drug release study showed that the highest drug release of 30.649% was from the formulation with 2.5% sodium alginate, and the lowest drug release of 26.625% was from the formulation with 2% sodium alginate. , A stability study at temperatures of 25áµ’C and 40áµ’C for 28 days showed a decrease in drug loading and entrapment efficiency but an increase in particle size. The formulation containing 1.5% sodium alginate showed the optimal formula. Conclusion: These findings indicated that quercetin ca-alginate microspheres are the potential for inhalation to be delivered to the lung.

Abstract

Background: Green Tea Extract (GTE) is a natural antioxidant compound that can protect the skin from photocarcinogenesis (DNA damage due to ultraviolet exposure). GTE has low stability, which needs a delivery system such as Nanostructured Lipid Carriers (NLC) with decyl glucoside (DG) as a natural surfactant that at the right concentration can produce a significantly small particle size which can improve the stability of the NLC. Objective: To determine the effect of DG usage on the characteristics, physical stability, and irritability of NLC-GTE preparation. Methods: NLC-GTE preparation used the High Shear Homogenization (HSH) method with three formulas, which contained DG 2%, 2.5%, and 3% consecutively. Afterwards, the characteristic and physical stability tests were conducted using the thermal cycling method for three cycles with two different temperatures (48 hours/cycle, 2 - 8°C and 40°C). The irritability test used Hen's Egg Test on the Chorioallantoic Membrane (HET-CAM) method. Results: Characteristic test of organoleptic showed that all formulas were white, odorless, and had a semi-solid consistency. However, the pH, particle size, and polydispersity index values from all formulas were within the normal range of values. The physical stability test result showed that 3% DG was the most stable formula. This formula was within the non-irritating range of values in HET-CAM. Conclusion: NLC-GTE with an increased concentration of DG as a surfactant can improve the characteristics and physical stability of the preparation. F3 (3% DG) is the best formula compared to other formulas and indicates non-irritating in the HET-CAM test.

Abstract

Pharmacy Vocational High School (SMK) students are prepared to work immediately after finishing school. Students receive lessons on the basics of pharmaceutical preparation and entrepreneurship. Cosmetics are a pharmaceutical dosage form widely used by the broader community for personal hygiene, appearance improvement, and health functions. The Minister of Health of the Republic of Indonesia No. 1175/Menkes/Per/VIII/2010 concerning cosmetic production permits provides an opportunity for cosmetic products to be produced with simple technology. During the COVID-19 pandemic, which results in a heavy economic burden for the government and society, there are business opportunities for Pharmacy Vocational School students. Community service aims to increase students' knowledge and insight and encourage students in class XI and XII of the Sekesal Pharmacy Vocational School in Surabaya and Kapasari Pharmacy Vocational School in Surabaya to become entrepreneurs in the cosmetics sector. The implementation method is through a webinar that provides material covering products, formulations, stability, and licensing of cosmetic products. Participants had the opportunity to discuss the material provided, fill out evaluations in the form of pre/post-test and make E-posters about cosmetic products. The evaluation results show an increase in participants' knowledge and insight, and participants are interested in producing cosmetic preparations, which are expected to become new business fields that can improve their welfare. --- Siswa Sekolah Menengah Kejuruan (SMK) Farmasi dipersiapkan untuk segera bekerja setelah menamatkan sekolah. Siswa memperoleh pelajaran tentang dasar-dasar pembuatan sediaan farmasi dan kewirausahaan. Kosmetika merupakan salah satu bentuk sediaan farmasi yang banyak digunakan oleh masyarakat luas untuk tujuan kebersihan diri, peningkatan penampilan, dan memiliki fungsi kesehatan. Permenkes RI No 1175/Menkes/Per/VIII/2010 tentang izin produksi kosmetika memberikan peluang produk kosmetika diproduksi dengan teknologi sederhana.  Dalam  masa pandemi covid 19, yang mengakibatkan beban ekonomi berat bagi pemerintah dan masyarakat, terdapat peluang usaha bagi siswa SMK Farmasi. Pengabdian masyarakat bertujuan untuk meningkatkan pengetahuan dan wawasan siswa serta mendorong siswa  kelas XI dan XII SMK Farmasi Sekesal Surabaya dan SMK Farmasi Kapasari Surabaya untuk berwirausaha di bidang kosmetika. Metode pelaksanaan melalui webinar dengan  pemberian materi meliputi produk, formulasi, stabilitas dan perizinan produk kosmetika. Peserta mendapatkan kesempatan berdiskusi tentang materi yang diberikan,  mengerjakan pre/post test, mengisi evaluasi, dan membuat E-poster tentang produk kosmetika. Dari hasil evaluasi diketahui terdapat peningkatan pengetahuan dan wawasan peserta dan peserta tertarik untuk memproduksi sediaan kosmetika yang diharapkan dapat menjadi lahan usaha baru yang dapat meningkatkan kesejahteraannya.

Abstract

This study aims to determine the effect of surfactant concentration on Quercetin SLM as a potential carrier of respiratory diseases, especially ones resulting from oxidative stress injury. Quercetin is a natural antioxidant with high activity. SLM was formed with 5% compritol 888 ATO as a lipid and different concentrations of poloxamer 188 as a surfactant. SLM was produced by a combination of emulsification and sonication involving freeze drying. SLM is characterized by organolepsis, morphology, yield, particle size, drug loading, and entrapment efficiency. The antioxidant activity of quercetin SLMs was tested using the ABTS method. SLMs are characterized as having round and smooth morphology, high yield (F1 88.53%; F2 91.44%; F3 92.87%); particle size (F1 1.81 um; F2 1.90 um; F3 1.94 um); high drug loading (F1 15.96%; F2 13.74%; F3 13.19%); and high entrapment efficiency (F1 96.53%; F2 87.94%; F3 87.48%). Increasing surfactant concentration did not produce a significant difference between formulas. Quercetin SLM showed high antioxidant activity (Quercetin 94.43%; F1 94.35%; F2 94.36%; F3 94.37%). SLM was stable at storage temperatures between 25°C and 40°C. The effect of surfactant can be seen on particle size, drug loading, and entrapment efficiency at 40°C. Results of in vivo deposition study indicated that all SLM formulas were able to deliver quercetin to the lungs. Increasing the concentration of surfactant in Quercetin SLMs made no difference to the lung deposition as confirmed by observations conducted at 1 hour and 4 hours. Quercetin SLM has the potential for lung delivery by inhalation.

Abstract

Langir (Albizia saponaria L.), belonging to the family of the Fabaceae, is a medicinal endemic plant of South Sulawesi, Indonesia. The Albizia genus shows antioxidant, antibacterial, anthelmintic, antidandruff, and anti-inflammatory properties. In this study, total phenolic (TP) and total flavonoid (TF) extracts from the bark of A. saponaria were screened for robust antioxidant activity in order to identify potential sources of new compounds for use in pharmaceutical formulations in the future. TP and TF of the 96% ethanol extract and fractions (hexane, ethyl acetate, butanol, and water) were calculated by the Folin–Ciocalteu and aluminum chloride procedures, respectively. Furthermore, the antioxidant activity was determined by DPPH free radical scavenging method and ABTS assay. Generally, both extract and fractions showed significant radical scavenging activities. Ethyl acetate fraction exhibited more potent radical scavenging activity in the DPPH method (IC50 35.27 ± 2.85 µg/mL) and ABTS assay (IC50 60.04 ± 0.98 µg/mL), followed by 96% ethanol extract, and hexane, butanol, and water fractions. Furthermore, the highest TP (4.50 ± 0.01 mg/g GAE) and TF (3.55 ± 0.04 mg/g QE) were obtained from ethyl acetate fraction. There was a strong correlation between antioxidant activity with TP (DPPH, R2 = 0.6436; ABTS, R2 = 0.7676) and TF content (DPPH, R2 = 0.5533; ABTS, R2 = 0.5961). The extract’s antioxidant properties may be attributable to its higher phenolic and flavonoid content. In summary, the phenolic and flavonoid content of the ethyl acetate fraction indicates its potential utility as a source of antioxidants.

Abstract

Background: The success of nanoemulsion preparation is characterized by characteristics such as small droplet size, polydispersity index (PDI), and % transmittance, which are close to 100%. One of these factors is the type of oil component used. The Simplex Lattice Design (SLD) method can be used to determine the ratio of oil combinations to obtain an optimal nanoemulsion formula. Objective: The application of the Simplex Lattice Design (SLD) method can help researchers speed up the acquisition of optimal formulas without trial and error so that nanoemulsion formulas that meet specifications can be obtained. Methods: This research uses the Simplex Lattice Design (SLD) method with Design of Expert Version 13 software, with an upper limit value for oil (VCO and Palm Oil) of 2.66% and a lower limit value for oil (VCO and Palm Oil) of 0, which then The results of several formulas come out and characterization testing is carried out to get the best formula from the recommendations produced by the software. Results: The results of the Simplex Lattice Design (SLD) showed that oil affected the droplet size and PDI (p <0.05). Six optimal formulas were obtained, and after testing in the laboratory, there was no significant difference between the results of the SLD program and those of the laboratory (Sig. <0.05). Conclusion: This study shows that the Simplex Lattice Design (SLD) method is very effective.

Abstract

In order to increase public understanding and awareness regarding the safety use of drugs and cosmetics, the Community Service Team of the Faculty of Pharmacy, Airlangga University has held community service in Jonggrangan Village, Klaten, Central Java. The community service was carried out on July 31, 2023, with approximately 60 family welfare programs or PKK cadres and health cadres participating through training, presentations, and discussions. The activity began with a pre-test, continued with training, a presentation of the material by three experts, and a discussion, then ended with giving a post-test to the participants. The results of the pre and post-test analysis showed that participants' understanding increased. The results of the analysis of the answers to the 15 questions given showed that the increase in participants' understanding ranged from  2.13-46.02 percent. The impact of these activities on society. The increase in participants' understanding of each topic is not the same depending on the initial understanding. However, the problem is that the level of public understanding after receiving training is still below 50 percent namely on topics about the stages of using cosmetic products and about drug classification. So it is necessary to deepen the material using other strategies (appropriate methods and media).

Abstract

Astaxanthin is a potent antioxidant belonging to carotenoid compounds that is mainly produced from green microalgae Haematococcus pluvialis. Astaxanthin is beneficial for skin health as antiaging agent, but has limitations in its delivery through the skin. Astaxanthin could be formulated in nanostructured lipid carriers to improve its efficacy. The purpose of this study was to evaluate the stability and antiaging effectiveness of astaxanthin-loaded nanostructured lipid carriers (ASX-NLCs) with cetyl palmitate and soybean oil as lipid combinations at several ratios of 100:0, 90:10, 80:20, and 70:30. ASX-NLCs were synthesized using a high-shear homogenization technique. ASX-NLCs were characterized and stability evaluated after storage for 90 days. The antiaging effectiveness of ASX-NLCs was evaluated by in vitro release test using dialysis bag for 8 hours, as well as collagen density and fibroblast count evaluation on UV-induced skin aging mice for 28 days. After storage, all ASX-NLCs did not change significantly in organoleptic, pH, and particle size. However, other parameters including polydispersity index, viscosity, and entrapment efficiency experienced significant changes in some formulas. The release test showed that F4 (70:30) gave the highest cumulative release and was significantly different from F1 (100:0). The collagen density of the groups treated with ASX-NLC F3 (80:20) and F4 (70:30) increased significantly compared to the UVB control group, while the fibroblast count did not differ significantly in all groups. Overall, ASX-NLCs containing cetyl palmitate and soybean oil at ratios of 80:20 and 70:30 could improve the antiaging effect of astaxanthin which might be influenced by its better stability and release.
