Abstract

Pemurnian enzim cathepsin C diperoleh melalui penggunaan colom kromatografi S-Sepharose FF, Sephacryl S-200 HR, Concanavalin A-Agarose, Affi-gel 501 dan DEAE-Sephacel. Dari 160 g ginjal ikan mas diperoleh 0,33 mg enzim cathepsin C, dengan hasil pemurnian 150 kali. Berat molekul cathepsin C adalah 170 kDa yang terdiri dari 5 subunit, dengan berat molekul berkisar antara 10 sampai 20 kDa. Cathepsin C adalah oligomerik protein dan aktif sebagai eksopeptidase (pH 5) maupun endopeptidase (pH 7)

Abstract

The aim of this research was to count the total leukocytes and phagocytosis activity of goldfish seeds treated with probiotics. The test fish used were goldfish seeds measuring 5-8 cm. 500 fish will be taken from the Tateli Fish Cultivation and Development Center (BP3I). This study used a completely randomized design (CRD) with 5 treatments, where each treatment had 3 replications. The treatments used were probiotic bacteria from the intestines of goldfish with different concentrations. The treatments tested consisted of: A: without the addition of probiotics, B: addition of probiotics 1x106 cfu / mL, C: addition of probiotics 1x107 cfu / mL, D: addition of probiotics 1x108 cfu / mL, E: addition of probiotics 1x109 cfu / mL. Each concentration of bacteria that has been prepared is then sprayed on the commercial feed evenly using a sprayer and then dried to room temperature. After drying the feed was coated with 2% egg yolk and dried again at room temperature. Fish are stocked in an aquarium measuring 60x40x40cm3 with a density of 20 fish / aquarium. Each aquarium is equipped with an aerator and a water pump for recirculation. The fish were then given feed that had been added with probiotics at the same dose and frequency of administration as in the acclimatization process. The treatment is given for 6 weeks. At the end of the treatment feeding, fish samples were taken from each experimental unit to measure immune parameters, namely total leukocytes and phagocytic activity. Based on the results of research using the probiotic Lactobacillus sp. which originated from the intestines of goldfish and added to the feed had a significant effect on increasing total leukocytes and phagocytosis activity and the optimal concenration was  1x107 - 1x108 cfu / mL with a duration of 6 weeks of administration

Abstract

The goal of this research was to observe the effects of yeast on the density rate of Alona sp. with various concentrations compared to the density rate in horse manure media. The treatments are as follows, treatment A yeast 30 mg yeast/0.5 L water, treatment B yeast 10 mg yeast/0.5 L water, treatment C yeast 5 mg yeast/0.5 L water, treatment D yeast 1 mg yeast/0.5 L water, treatment E 0.5 L well water, treatment F horse manure media (horse manure 10 gr, soil 50 g in 0.5 L water). The research was carried out for two weeks, and the density calculation was done once a week. The results obtained based on Anova One Way analysis are that yeast with different concentrations does not have a significant effect on the density of Alona sp. Nevertheless, it was concluded that the giving of yeast to Alona sp. is much more efficient and effective compared to horse manure media, because it is more hygienic, easy to obtain and not expensive, also from nutritional value, yeast has a higher value compared to horse manure media. The measuring of water quality parameter during the research was temperature around 26-30°C.

Abstract

Susceptibility of the marine polychaete Ophryotrocha diadema (Polychaeta: Dorvilleidae) towards the neurotoxic methyl mercury (MeHg) contamination was studied in an experimental chamber, which was aimed to assess and compare the susceptibility level of the organism based on its generations (F0, F1, F2, and F3). Seven variables of growth and reproduction aspects were applied as indicators in this study; they were: 1) individual growth, 2) first time the egg laid, 3) number of eggs per individu, 4) number of eggs per egg mass, 5) number of eggs to larva per egg mass, 6) number of mortality per egg mass, and 7) reproductive potential. Observation was conducted on the treatment (MeHg in concentration of 0,00025 ppb) and the control (no MeHg) to each of the generations (F0, F1, F2, and F3). Data obtained were analysed for average and standard deviation. Comparison of susceptibility within the generations was calculated using the variable of reproductive potential. The results showed that there were differences between the treatments and the control for all of the variables. Comparison on the susceptibility of the polychaete within the generations to MeHg contamination was F0<F1=F3<F2. It was concluded that the F2 generation had the highest susceptibility among the others.

Abstract

The purpose of this research was to investigate the population density of Alona sp. on yeast media with different concentrations. The study was conducted using a completely randomized design (CRD) consisting of 5 treatments and 3 replications for 21 days. The treatments given were A: 10 ppm yeast, B: 5 ppm yeast, C: 1 ppm yeast, D: horse manure, and E: well water. The initial stocking density of Alona sp. for each treatment was 15 individuals/300 mL of well water. The water quality parameter measured during the study was temperature. The results of the one-way ANOVA test showed that there were significant differences between treatments on the density level of Alona sp. (p <0.05). Tukey's further test showed that treatment B was the best with a density of Alona sp. as many as 8059 individuals/300 mL of well water. The temperature during the study, in the morning was 26 ± 0.52°C and in the afternoon was 26 ± 0.46°C.

Abstract

This study aimed to 1) determine the best combination between ketapang and banana peels for the propagation of life feed mosquito larvae; 2) determine the culture media that can produce mosquito larvae optimally. The containers used in this study were 24 buckets with a size of 5 litres. Some of the materials used in the study were dried ketapang leaves, goroho banana peels, kepok banana peels and water. This study used a factorial Completely Randomized Design (CRD) consisting of two factors and three replications. The first factor was the difference in the number of ketapang leaves. The second factor was the difference in banana peels types. The first factor included the difference in the number of ketapang leaves, consisting of: Treatment 0 = without ketapang leaves, Treatment 1 = 50 grams of ketapang leaves, Treatment 2 = 75 grams of ketapang leaves, Treatment 3 = 100 grams of ketapang leaves. The second factor included differences in banana peels, which consist of: Treatment 1 = one kepok banana peel, Treatment 2 = one goroho banana peel. The results were different types of banana peels affected the numbers of mosquito larvae, where the best was kepok banana peels; the difference in weight of ketapang leaves did not have a significant effect on the number of mosquito larvae. The container used in this study was a bucket with a volume of 5 L. The banana peels used were goroho banana peels and ripe yellow kapok banana peels. Ketapang leaves were preapared with a weight 0f 0, 50, 75, and 100 grams, respectively. Shoe banana peels and  goroho banana, one skin eachand ketapang leaves 0, 50, 75, and 100 grams, respectively, put together wit the jalah used to wrap between ketapang leavesand spatu banana peels and goroho banana, then put into a container that has been filled wit 3 L of water.

Abstract

AbstractSindulang Satu Village is one of the villages in Tuminting District which has a coastal area and most of the population works as fishermen. The job as a fisherman generates an uncertain income because it depends on the season and is currently pressured by government regulations in overcoming covid-19 and facing the new normal so that the participation of women as breadwinners to cover their household needs.How far their role is in dealing with the family economy is an interesting topic for conducting research. Research Objectives: 1. To analyze the economic condition of fishermen group households in Sindulang Satu Village, Tuminting District, Manado City. 2. To analyze the role of women in the household conomy of fishermen groups during the Covid-19 pandemic and facing the new normal era in Sindulang Satu Village, Tuminting District, Manado City. The method used in this research is the case study method. The data collected in this study are primary data and secondary data. The so-called respondents were obtained by recording all the women / wives of members of the Daseng fishermen group; and 15 people registered as the population. The 15 people who were used as respondents to be visited one by one conducted interviews with a questionnaire guide as a tool that had been previously prepared by means of a census. The data analysis used in this research is quantitative analysis and qualitative analysis.The economic condition of the family of the Daseng fishing group in Sindulang Satu Village includes age, education and family dependents. The women / wives of the Daseng fishermen group are mostly in the productive age range so they are still active in carrying out income-generating activities and jobs, their education is dominated by elementary school graduates, while the dependents of the family have at most 1 - 3 dependents in the family. The role of women / wives in overcoming the family economy of fishermen groups during the Covid-19 pandemic and facing the new normal era, including domestic roles, namely cooking, cleaning and tidying houses, caring for children and helping husbands, active social roles in spiritual activities (mother worship) also participate in arisan and PKK, and the productive role of women / wives of the daseng fishermen group is that they work as a washerman, open a shop and sell business. All the roles of the wife are very supportive of the economy of the Daseng group's fishing family. Keywords: Role of Women, Family Economy, Sindulang Satu AbstrakKelurahan Sindulang Satu merupakan salah satu desa di Kecamatan Tuminting yang memiliki daerah wilayah pesisir serta ada sebagian besar penduduk bekerja sebagai nelayan. Pekerjaan sebagai nelayan tersebut menghasilkan pendapatan yang tidak menentu karena tergantung kepada musim dan pada saat sekarang tertekan dengan peraturan pemerintah dalam mengatasi covid-19 dan menghadapi new normal sehingga peran serta wanita sebagai pencari nafkah untuk menutupi kebutuhan rumah tangga mereka.Seberapa jauh peranan mereka dalam rangka mengatasi perekonomian keluarga merupakan topik yang menarik untuk mengadakan penelitian. Tujuan Penelitian: 1) menganalisis keadaan perekonomian rumah tangga kelompok nelayan di Kelurahan Sindulang Satu Kecamatan Tuminting Kota Manado, dan 2) Menganalisis sejauh mana peran perempuan dalam perekonomian rumah tangga kelompok nelayan dimasa pandemic covid-19 dan menghadapi era new normal di Kelurahan Sindulang Satu Kecamatan Tuminting Kota Manado. Metode yang digunakan dalam penelitian ini adalah metode studi kasus. Data yang dikumpulkan dalam penelitian ini adalah data primer dan data sekunder. Adapun yang disebut sebagai responden diperoleh dengan cara mendata seluruh wanita/istri anggota kelompok nelayanDaseng; dan yangterdaftar berjumlah 15 orang yang disebut sebagai populasi. Ke 15 orang ini yang dijadikan responden untuk dikunjungi satu persatu mengadakan wawancara dengan panduan kuisioner sebagai alat bantuyang telah disiapkan sebelumnya dengan cara sensus. Analisis data yang digunakan dalam penelitian ini adalah analisis kuantitatif dan analisis kualitatif.Keadaan perekonomian keluarga kelompok nelayan Dasengdi Kelurahan Sindulang Satu meliputi keadaan umur, pendidikan dan tanggungan keluarga. Wanita/istri kelompok nelayan Daseng paling banyak dalam range umur produktif sehingga masih aktif dalam melakukan kegiatan dan pekerjaan yang menghasilkan pendapatan, pendidikan mereka didominasi oleh tamatan SD sedangkan tanggungan keluarga paling banyak memiliki 1 – 3 orang tanggungan dalam keluarga. Peranan wanita/istri dalam mengatasi perekonomian keluarga kelompok nelayan dimasa pandemic covid-19 dan menghadapi era new normal, meliputi peran domestik yaitu memasak, membersihkan dan merapikan rumah, mengurus anak dan membantu suami, peran sosial dengan aktif dalam kegiatan kerohanian (ibadah kaum ibu) juga mengikuti arisan dan PKK, dan peran produktif wanita/istri kelompok nelayan daseng ada yang bekerja sebagai tukang cuci, membuka usaha warung dan berjualan. Semua peran istri sangat mendukung perekonomian keluarga nelayan kelompok Daseng. Kata Kunci: Peranan Wanita, Perekonomian Keluarga, Sindulang Satu

Abstract

This study aimed to examine the effect of probiotic Effective Microorganism-4 (EM4) on the growth rate of the natural feed population of Daphnia magna and to determine the right dose to obtain the highest growth. This research was conducted at the Aquaculture Technology Laboratory, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University, Manado. The time of the study was carried out from April to May 2021. The experimental design used was designed according to a 4 x 4 factorial experimental design in Completely Randomized (CRD). The first factor was the dose of EM4 with 3 levels, namely 0.75 mL, 1 mL and 1.25 mL with 1 control, then the second factor is the time difference (weeks) I, II, III, and IV. The results of the study using the Analysis of Variance (ANOVA) using the JMP application showed 1. The dose difference in the density of daphnia on the culture medium was not significantly affected by the difference between the culture medium and chicken manure (value 'Prob>F)' = 0.1411), 2. The difference in density of daphnia on culture medium was very significant by differences in culture time (Value 'Prob>F' = 0.0003), 3. Interaction, The effect of culture medium factors (EM-4) dose and chicken manure on the density of daphnia changed significantly at the time of the change in the level of the time factor (weeks) (Value 'Prob>F' = 0.0439). Further contrast tests showed that the density of daphnia at week-I was significantly different from the density of daphnia at week-II, week-III, and week-IV and the density of daphnia at week-II, week-III and week-IV were not significantly different.

Abstract

This study aimed to analyze the use of fermented rice bran media for the propagation of Alona sp. The method used is an experimental method, using one-way ANOVA, with three treatments, namely fermented rice bran media, unfermented rice bran media, and water. Each treatment was repeated 3 times, so that 9 experimental units were obtained. The observed variable was the density of Alona sp. The results showed that the density of Alona sp. that were cultured on fermented rice bran media was significantly different from that of unfermented rice bran and water.
