Abstract

Desa Sedayu Lawas merupakan salah satu sentra produksi garam di Lamongan. Potensi tambak garam di desa ini cukup besar hanya saja produksi garam yang ada sebagian besar masih sangat tergantung kondisi cuaca. Salah satu inovasi dalam memproduksi garam rakyat untuk mengatasi masalah cuaca tersebut adalah dengan menggunakan teknologi Prisma Rumah Kaca.  Inovasi ini memanfaatkan rumah kaca dan plastik geomembran dalam proses kristalisasi garam. Tujuan dari penelitian ini adalah untuk menganalisis kualitas garam dari hasil produksi metode Prisma Rumah Kaca di desa Sedayu Lawas. Parameter yang diukur dalam penelitian ini adalah suhu, salinitas, kandungan Mg, kandungan Ca, kandungan NaCl dan kadar air. Hasil penelitian ini menunjukkan kalau kadar salinitas air pada kolam penampungan air muda adalah 23-24 ppt dan pada kolam penampungan air tua adalah 34-42 ppt. Nilai rata-rata NaCl, Mg dan Ca pada air muda secara berturut-turut adalah 115.000 mg/L, 313,6 mg/L dan 202,3 mg/L. Hasil pengujian air tua menunjukkan nilai rata-rata NaCl, Mg dan Ca secara berturut-turut adalah 129.333,3 mg/L, 313,6 mg/L dan 214,3 mg/L. Hasil uji kualitas garam menunjukkan kadar NaCl atas dasar berat basah (adbb) sebesar 87,56%, kadar Mg sebesar 2,15%, kadar Ca sebesar 3,45% dan kadar air sebesar 5,86%. Hasil penelitian ini mengindikasikan kalau kualitas garam yang dihasilkan melalui metode Prisma Rumah Kaca di desa Sedayu Lawas termasuk dalam kategori Kualitas K1 berdasarkan SNI 4435:2017.

Abstract

The ability of plant cellulose degrading bacteria to become a source ofenergy can increase the digestibility of feed by fish. The purpose of this study was toidentify the cellulolytic bacteria from the mangrove ecosystem, Tukak Sadai District,South Bangka Regency. This study was conducted in March until August 2017.Samples were taken from litter, mud and weathered wood and isolated usingCarboxymetyl Cellulosa (CMC) 1% media and found 22 bacterial isolates. Gramstaining results showed that 3 isolates (TSS 2, TSL 6 and TSK 4) were classified asgram positive and 19 other isolates were gram negative. Cellulolytic test results showed6 isolates had the ability to degrade cellulase, namely 3 isolates from mangrove mudsamples (TSS 1, TSL 7, TSL 1), 2 isolates from leaf litter (TSS 4, TSL 2) and 1 isolatefrom weathered wood (TSK 5) . Abstract

Surimi powder is a modification product of frozen surimi blocks produced through drying at a specific temperature and time. Surimi powder has many uses in the diversification of processed fishery products, namely to increase the nutritional and functional value of a product, especially cereal products. This research aims to determine the effect of different drying times on the physicochemical characteristics of the surimi powder of kurisi fish (Nemipterus japonicus). The drying times were 9, 12, and 15 hours at 60oC. The resulting kurisi fish surimi powder was observed for its characteristics, including proximate values, color, and product acceptance and then analyzed using Minitab software version 19. The results analysis of variance showed that the drying time had a significant effect (p < 0.05) on the value of water content, protein content and ash content, brightness (L*), and yellowish color (b*) but had no significant effect (p > 0.05) on the values of a* and ΔE*ab.

Abstract

Arthrospira platensis culture requires macro and micronutrients. The nutrients needed by Arthrospira platensis according to Agustina and Herman (2021) are macronutrients consisting of nitrogen (N), phosphorus (P), potassium (K), sulfur (S), iron (Fe), magnesium (Mg), silicon (Si), and calcium (Ca). Micronutrients consist of zinc (Zn), cobalt (Co), molybdenum (Mo), boron (B), and copper (Cu). Rice-washing wastewater contains macro and micronutrients needed for the growth of Arthrospira platensis. Almost all of the nutrients needed by Arthrospira platensis are found in the wastewater, except for Co. Previous studies have shown that rice-washing wastewater can be used for Arthrospira platensis culture, but the growth rate is still higher than Walne fertilizer 0.5 ml/L. Zarrouk fertilizer as a standard growth medium for Arthrospira platensis shows that this fertilizer has better growth results than Urea and NPK fertilizers. This research was conducted to determine the effect of rice-washing wastewater as a nutrient source for culture media or a combination of Zarrouk fertilizer and rice-washing wastewater so that it can be used to increase the biomass and carotenoid pigments of Arthrospira platensis. This research used a Factorial Completely Randomized Design (CRD) with 12 treatments and 3 replications. Arthrospira platensis quality test was carried out by testing dry biomass and carotenoid content. The ANOVA test results showed that this study's treatment had a significant effect on the carotenoid content and biomass of Arthrospira platensis. The treatment with the highest effect of biomass and carotenoids was obtained in the treatment of 1.5 ml/L of rice-washing wastewater

Abstract

Banyuurip Village has considerable aquaculture potential, one of which is the culture of green mussels. Abundant production and its low price encourage people to do green mussel products processing. In 2019, UMKM Berkah Mandiri was established to increase the selling value of green mussels by innovating several products. However, the production process is still experiencing ups and downs, coupled with inadequate equipment. In addition, the distribution permit (SPP-PIRT) for the products have not been obtained. Therefore, the aim of the Doctoral community service program was to increase the productivity of UMKM Berkah Mandiri through a series of marketing strategy training activities, registration of NIB and SPP-PIRT and procurement of new production and packaging equipment. At the end of the Doctoral community service program, UMKM Berkah Mandiri has successfully obtained the SPP-IRT certificate and has better understanding of good manufacturing practice of products from green mussels. In addition, the crackers have been tested its nutritional content using a proximate test. The results showed that the protein, moisture, fat, ash and carbohydrate contents of Kopang cracker was 5.30%, 13.83%, 0.69%, 2.99% and 77.19%, respectively. The nutritional content list is used to improve product packaging in accordance with BPOM packaging standards.

Abstract

Bacteria are bioremediation agents that are advised to overcome water pollution. This study aims to isolate biosurfactant-producing bacteria for bioremediation from Tanjung Perak Port, Surabaya. The isolates grown in specific media were tested for drop collapse, oil spreading test, and emulsification index. A total of 12 bacterial isolates were isolated from Tanjung Perak seawater contaminated with diesel oil. In the drop collapse test, four isolates had the highest positive score from this test, isolate LU-5, LU-7, LU-9, and LU-11. While in the second test, the oil spreading test, the highest positive score results were obtained in three isolates, isolates LU-2, LU-7, and LU-11. The emulsification index (E24) value showed that isolates LU-7, LU-9, and LU-11 were significantly higher than the others, which was above 10%. This study concludes that in Tanjung Perak, seawater-contaminated oil isolated several bacteria that produce biosurfactants, which have the potential to be developed as bioremediation agents.Keywords: bacteria, bioremediation, biosurfactant, diesel oil

Abstract

Enzim L-asparaginase (E.C 3.5.1.1) adalah protein tetramer yang termasuk dalam kelompok keluarga amidohydrolases homolog. ). L-asparaginase secara selektif menghidrolisis L-aspargin (L-Asn) menjadi amonia (NH3) dan asam aspartat. Penelitian ini bertujuan untuk mendapatkan spesies bakteri penghasil enzim L-asparaginase yang diisolasi dari mangrove Avicennia marina yang berasal dari Pantai Bajul Mati, Malang, Jawa Timur, Indonesia. Penelitian ini dilakukan beberapa tahapan yaitu pengambilan dan preparasi sampel, skrining L-asparaginase, dan identifikasi menggunakan uji mycrobact system. Hasil penelitian menunjukkan bahwa isolat dengan aktivitas enzim L-asparaginase terbesar adalah isolat DAM6 yang berasal dari daun Avicennia marina. Isolat DAM6 adalah bakteri merupakan bakteri gram negatif, berbentuk basil, bentuk koloni oval, tepi koloni tidak rata dan elevansi cembung. Berdasarkan analisis microbact,  isolat DAM6 adalah bakteri spesies Pseudomonas aeruginosa.                                               AbstractThe L-asparaginase enzyme (E.C 3.5.1.1) is a tetramer protein belonging to the family group of amidohydrolases. L-asparaginase selectively hydrolyzes L-aspargin (L-Asn) to ammonia (NH3) and aspartic acid. This study aims to obtain species of L-asparaginase enzyme producing bacteria isolated from Avicennia marina mangroves from Bajul Mati Beach, Malang, East Java, Indonesia. This research carried out several stages such as took and prepared samples, screened L-asparaginase, and identified using the mycrobact system test. The results showed that the isolates with the largest L-asparaginase enzyme activity were DAM6 and it was originated from the leaves of Avicennia marina. DAM6 isolate is a gram-negative with an oval shape, uneven edges, and convex elevation. Based on microbact analysis, DAM6 isolates are Pseudomonas aeruginosa.

This study aimed to isolate and identify protease-producing bacteria isolated from salt-pond soil at the Tuban Regency, East Java, Indonesia. Bacterial communities in the soil samples were firstly isolated from the soil samples by culture-dependent technique on Luria Bertani agar enriched with 5% NaCl. Thereafter, bacterial colonies that grew on the media were purified and screened for their protease production using a skim-milk agar. The bacterial colony which produced protease was further identified using phenotypic (gram staining) and genotypic assays (the 16S rDNA sequence). The result showed that one isolate out of six (isolate C2) obtained from the soil sample was observed to produce a protease enzyme. Based on its 16S rDNA sequence, the isolate was identified as Bacillus amyloliquefaciens strain UBC. These results suggest that B. Amyloliquefaciens strain UBC is a salt-tolerant bacterium (halophilic bacteria) which has the potential to be further developed for protease-producing biological agents.

Abstract

Highlight Research S. platensis has been analyzed for its productivity. Analysis of the productivity of S. platensis on the use of fish culture wastewater media. S. platensis production can be increased by aquaculture wastewater. Aquaculture wastewater enhances the production of cell density, SGR, biomass, protein, and carotenoid of S. platensis. Abstract Increased productivity of Spirulina sp. in the form of high protein, carotenoids, and biomass content can be achieved by improving its nutrient supply. Inorganic fertilizers are nutrient sources, which are generally used in the culture of this organism on laboratory and industrial scale, but there are several drawbacks, including their high costs and limited availability. Several studies have also reported the use of zarrouk fertilizer as a standard culture medium for Spirulina platensis. Therefore, this study aims to determine the effect and the best concentration of fish culture wastewater treatment in Spirulina platensis culture using biomass, protein content, and carotenoid pigments as indicators. A two-factorial completely randomized designs (CRD) was used in this study, where the factors include the dose of organic waste and Zarrouk fertilizer. The microalgae samples, namely S. platensis were cultured using fresh water. This research consists of two factors. The first factor is the dose of organic waste, and the second factor is the dose of Zarrouk fertilizer. The wastewater treatment consisted of 0, 2, 4, and 6 ml/L, while Zarrouk dosages include 0, 0.5, and 1 ml/L. The best results were obtained from the sample treated with 6 ml/L aquaculture wastewater without the addition of Zarrouk. Furthermore, this treatment gave biomass production of 0.781 g/L, 50.441% protein, and 1.246 mg/L carotenoids. Based on the results, S. platensis culture can be carried out using fish culture wastewater without the addition of Zarrouk fertilizer.

Abstract

Diabetes is a major global health challenge, with α-glucosidase inhibitors playing a key role in managing blood glucose levels by delaying carbohydrate digestion and absorption. This study developed an antidiabetic functional drink combining Sargassum extract with cinnamon (Cinnamomum burmannii) and temulawak (Curcuma xanthorrhiza). A Completely Randomized Design (CRD) was used with four treatments: a control (F0) and formulations with 15% (F1), 17.5% (F2), and 20% (F3) Sargassum extract. Data on pH, polyphenols, and α- glucosidase inhibition were analyzed using ANOVA, while sensory attributes were evaluated with nonparametric tests. The addition of Sargassum extract from 15% to 17.5% significantly increased the phenolic content by 15%. However, it did not affect α-glucosidase inhibition. This suggests that not all phenolic compounds in Sargassum contribute to the inhibition of α-glucosidase. The formulation with 20% Sargassum, 15% cinnamon, and 18.5% temulawak extract exhibited the highest phenolic content (315.21 mg GAE/100 ml) and α-glucosidase inhibition (62.27%). The sensory analysis shows that adding up to 20% Sargassum is still acceptable for the panelists, with a hedonic value of 3.34 to 3.43 (neutral). This study discovers that the bitterness of Sargassum at the highest concentration can be mitigated by adding cinnamon and temulawak without reducing the antidiabetic potential. These results contribute to conveying the potential of this drink as a natural anti-diabetic product with good sensory acceptability. Further research is needed to evaluate bioactive interactions and phenolic composition to optimize efficacy.
