Abstract

Doktor Mengabdi is a community service program at Universitas Brawijaya that is carried out to improve the welfare of the community. One of the efforts to help the community in completing its packaging is by providing assistance in the form of business equipment.  The purpose of this study is to investigate the difference in income of Small medium enterprises, Lele Sehat (IGF) which has been provided with the help of vacuum frying and spinners machines. Data retrieval was done through interviews and live observations. The data were tested for its normality and further analyzed using a paired t-test. The results showed that there was a difference in income of small-medium enterprises difference between before and after getting tool assistance. Prior income was IDR 3,007,000. It was increased to IDR. 4,097,000. This indicates that providing tools to small-medium enterprises will increase income and indirectly improve their welfare.

Abstract

Inhibition of intestinal α-amylase and α-glucosidase is an important strategy to control post-prandial hyperglycemia associated with diabetes mellitus. In vitro inhibitory effects of crude extracts of seaweed against α-amylase and α-glucosidase were studied. Crude ethyl acetate extracts of Sargassum aquifolium were the stronger inhibitor to α-amylase and α-glucosidase than others. Furthermore, Sargassum aquifolium ethyl acetate extract significantly suppressed the rise in postprandial glucose level after oral administration of glucose in normal rats. The results of this study suggest that the crude Sargassum aquifolium extract may suppress the rise in postprandial hyperglycemia in vivo in part, through inhibition of alpha amylase and glucosidase.

Abstract

The objectives of this study were to evaluate the prebiotic activity based on the change in cell biomass of the probiotic strain after 24-h growth in the presence of Laminaran from Sargassum crassifolium with the methods of laminaran acid extract (LAE) and laminaran modified extract (LME), inulin, or glucose-relative against the change in cell biomass of Escherichia coli FNCC 0091 grown under the same condition. Prebiotic activity was calculated for L. plantarum FNCC 0051 and Bifidobacterium longum FNCC 1081. The results showed that the increasing cell number of L. plantarum was higher in both substrates LAE and LME (0,58 and 2,03log cycle), whereas that of B. longum was lower. The higher prebiotic activity score obtained for L. plantarum and B. longum grown on LME were positive (0,26 and 0,96 log cycle), whereas the lowest score was for L. plantarum and B. longum grown on LAE, which were negative (-0,35 and -0,31 log cycle), but the higher prebiotic activity score was obtained for inulin (4,08 and 4,78 log cycle). It could be concluded that Laminaran Modified Extract (LME) has potential as prebiotic source, but its potential was lower than inulin.

Abstract

Bakteri selulolitik memiliki kemampuan degradasi selulosa dan membuat karbohidrat lebih mudah dicerna bagi ternak. Penelitian ini bertujuan untuk mengidentifikasi bakteri selulolitik di tanah, kayu lapuk dan daun dari mangrove pulau Bangka melalui skrining dan analisis gen 16s rRNA. Penelitian dilakukan dari September 2017 hingga Maret 2018. Isolat TSL7 dan TSS4 dari Mangrove Tukak Sadai dan SLS5 dari Mangrove Sungailiat yang memiliki kemampuan mendegradasi selulosa terbesar berdasarkan hasil skrining menjadi isolat yang diidentifikasi pada gen 16S rRNA untuk diurutkan dan analisis BLAST. Analisis gen bank menunjukkan kedekatan dengan Pseudomonas aeruginosa dengan query cover 37% - 87%. Hasil query cover rendah dihasilkan karena sekuens yang sesuai pendek. Kontaminasi dan perubahan komunitas karena seleksi lingkungan dengan adanya logam berat dapat menjadi penyebab dominasi Pseudomonas aeruginosa. Pseudomonas aeruginosa yang bersifat patogen tidak disarankan untuk diaplikasikan sebagai pendegradasi selulosa pakan ternak.

Abstract

AbstrakL-asparaginase (EC 3.5.1.1) adalah enzim yang menghidrolisis asam amino L-asparagin menjadi amonia dan asam aspartat. Enzim ini mempunyai manfaat utama dalam bidang farmasi dan industri pangan. Enzim L-asparaginase tersebar secara luas pada mikroorganisme. Mikroorganisme yang mempunyai potensi menghasilkan enzim ini adalah mikroorganisme endofit dari tumbuhan mangrove. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri endofit penghasil L-asparaginase dari tumbuhan mangrove Buta-buta (E. agallocha). Skrining dilakukan dengan menggunakan medium selektif untuk mendapatkan bakteri penghasil enzim L-asparaginase. Identifikasi molekuler dilakukan dengan menggunakan analisis filogenetik berdasarkan data sekuen 16S rDNA. Dari hasil penelitian ini didapatkan lima isolat bakteri endofit penghasil enzim L-asparaginase, di mana isolat penghasil L-asparaginase tertinggi diidentifikasi secara molekuler. Hasil identifikasi filogenetik molekuler menunjukkan bahwa isolat kode D.104 teridentifikasi sebagai Enterobacter cloacae. Molecular Identification of L-asparaginase-Producing Endophytic Bacteria Isolated from Mangrove Buta-Buta (Excoecaria agallocha)AbstractL-asparaginase (EC 3.5.1.1) is an enzyme which hydrolyze amino acid L-asparagine to aspartate and ammonia. Two main applications of this enzyme are in the pharmaceutical and food industries. The enzyme is widely distributed on microorganism. A potential source of L-asparaginase-producing bacteria is an endophytic bacteria from mangrove plant. This study aimed to isolate and identify L-asparaginase-producing endophytic bacteria from a mangrove plant, E. agallocha (Buta-buta). A screening was carried out using a selective medium to obtain the L-asparaginase enzyme producing bacteria. Molecular identification was carried out using phylogenetic analysis based on 16S rDNA sequence data. In this study, five isolates of the L-asparaginase-producing endophytic bacteria were obtained. The molecular phylogenetic identification showed that the highest L-asparaginase-producing bacterial isolate (code D.104) was identified as Enterobacter cloacae.

Abstract

The aims of this study were to screen phytochemical and antibacterial potency of methanol extract of Excoecaria agallocha. The phytochemical screening used qualitative analysis and the antibacterial assay determined by diffusion method. The results showed that extracts of leaf, flower, bark, stem and root of Excoecaria agallocha, respectively, contained alkaloid, steroid, flavonoid, tannin, terpenoid and saponin. Methanol extract of all part of Excoecaria agallocha, except the root, inhibited the growth of gram positive bacteria (Staphylococcus aureus ATCC 9144), Meanwhile, the leaf of Excoecaria agallocha extract not only inhibited the growth of gram positive bacteria (Staphylococcus aureus ATCC 9144) but also the growth of gram negative bacteria (Escherichia coli ATCC 8739).

Abstract

Terasi is a food condiment originating from Indonesia which is processed by fermenting shrimp, fish, or a mixture of both. The processing of terasi in Indonesia is still found traditionally so that it will affect the low quality of terasi which is characterized by being contaminated by pathogenic bacteria such as Salmonella. Salmonella is a bacterium that causes foodborne diseases. The purpose of this study was to evaluate the presence of Salmonella in shrimp paste using conventional method and then confirmed with molecular approach. This study used 10 samples consisting modern terasi and five traditional terasi. The isolation of Salmonella spp. carried out by using selective media, and then the isolates were characterized further using biochemical test (TSIA and LIA). The species identification of Salmonella spp. was carried out by multiplex PCR (mPCR). The isolates that were not detected by mPCR were further identified using 16s rDNA sequencing. The isolation results showed that Salmonella isolates were only detected in traditional terasi with a density range of 2.4 x 1052.9 x 108 CFU/g. A total of 20 isolates were characterized biochemically as Salmonella, and out of 17 isolates consistently identified as Salmonella enterica serovar Typhimurium by using mPCR. The rest three isolates were further identified as Citrobacter freundii based on 16s rDNA sequencing with similarity level of 99%. The presence of Salmonella in the shrimp paste indicates that the processing of traditional shrimp paste (terasi) should be evaluated in accordance with good manufacturing process.

Abstract

Acceleration of wood weathering in the mangrove area is possible with the role of cellulolytic bacteria. Cellulolytic bacteria producing cellulase enzymes are needed by the animal feed and agriculture industries. Mangroves in Muntok Subdistrict undergo a process of adaptation to environmental changes due to tin mining, including microorganisms. Exploration of new species and strains of cellulolytic bacteria in miningaffected mangroves is needed to enrich the collection and its potential use for human needs. This study aims to identify cellulolytic bacteria in mangrove weathered wood in Muntok District, West Bangka Regency through isolation, screening, and biochemical characterization. Three sampling locations, namely Sukal Mangrove, Peltim Mangrove, and Tembelok Mangrove result 22 bacterial isolates with 11 isolates showing the ability of cellulose degradation in qualitative tests using Lugol. The biochemical characterization of bacterial isolates with the greatest cellulose degradation was directed towards Citrobacter freundii and Vibrio alginolyticus in samples of Sukal Mangrove and Actinomyces bovis in isolates from Peltim Mangrove.

Abstract

Abstrak Enzim gelatinase merupakan enzim yang berperan penting pada sektor industri pangan dan non pangan. Sumber dari enzim gelatinase dapat berasal dari hewan, tumbuhan dan mikroorganisme. Enzim gelatinase termasuk enzim protease dan fungsi dari enzim gelatinase sendiri adalah sebagai pengurai gelatin. Penelitian ini bertujuan untuk mendapatkan bakteri penghasil enzim gelatinase dari endofit mangrove Sonneratia alba dan untuk mengetahui jenis spesies bakteri penghasil enzim gelatinase dari endofit mangrove Sonneratia alba dari pantai Sendang Biru. Metode penelitian yang digunakan dalam penelitian ini adalah deskriptif eksploratif yang dilakukan dalam dua tahapan. Tahap pertama yaitu isolasi dan skrining bakteri yang menghasilkan enzim gelatinase dari endofit mangrove Sonneratia alba. Tahap kedua yaitu identifikasi bakteri penghasil enzim gelatinase berdasarkan uji microbact system.  Hasil skrinning enzim menandakan bahwa isolat dari daun Sonneratia alba merupakan isolat bakteri yang mampu tumbuh dengan baik dan menghasilkan enzim gelatinase yang paling baik. Bakteri endofit daun pada uji microbact system diketahui sebagai jenis bakteri Paenibacillus alvei dengan hasil oksidase negatif sehingga termasuk bakteri Gram positif sehingga dilakukan dengan uji microbact 12A/E. Karakteristik bakteri mempunyai karakteristik berbentuk bulat dan berwarna ungu.. Kata kunci: gelatinase, enzim, Sonneratia alba, Malang, halal.

Abstract

The need for protease in the industrial field has been increasing. Candidates for producing these enzymes can be isolated from the digestive tract of catfish (Clarias sp.). The purpose of this study was to obtain bacterial isolates that produce proteolytic from the gastrointestinal tract of catfish and determine the effect of different production media on the activity of proteolytic. The first step of this study was isolation, screening, and identification of bacteria. The second step was to test the effect of the media Luria Bertani, trypticase soy broth, and skim milk broth on proteolytic activity. Nineteen isolates were obtained from the screening process of proteolytic bacteria. Isolate no 1, was known as the best isolate in producing enzymes and was known as Bacillus sp. Tests with different growth media gave results that semi-quantitative, nutrient growth media produced the highest activity with a proteolytic index value of 2.09 ± 0.41. In addition, based on quantitative tests, the media Luria Bertani Broth produced the highest specific activity with a value of 36.479 U/mg. The conclusion of this study, Bacillus sp. from the gastrointestinal tract of catfish that cultured on the Luria Bertani Broth medium produced the best activity.
