Abstract

Background: Diphtheria is a re-emerging disease in Indonesia that can be prevented by vaccination. This study evaluates the anti-diphtheria toxoid immunoglobulin G levels among healthy children aged 6 – 7 years old who had not received the second booster immunization for Diphtheria, Tetanus, and Pertussis (DTP) commonly administered at 5 – 7 years old. Methods: This cross-sectional study was conducted in the Senen district of Jakarta, Indonesia. All subjects had been vaccinated with three doses of immunization for DTP during the first year of life, and subsequently classified into a group of children who received the first booster immunization of DTP at 18 – 24 months old and who had not received it yet. Antibody against Diphtheria toxoids within the sera samples was assessed by commercial Anti-Diphtheria toxoid IgG Enzyme-Linked Immunosorbent Assay and classified in four groups: no protection (< 0.01 IU/ml), uncertain (0.01 – 0.09 IU/ml), full protection (0.10 – 1 IU/ml) and long-term protection (>1 IU/ml). Results: Eighty-nine children were included in this study; only 71 subjects (79.7%) had received the first DTP booster at 18–24 months old. The specific humoral immunity against diphtheria was observed among 57 children (64%). Notably, among the 89 subjects, only two subjects not receiving the first DTP booster still had antibody protection for diphtheria (p=0.001, OR 27.5, 95% CI: 5.71 – 132.42). Conclusion: The first diphtheria booster vaccination at 18 – 24 months old is crucial to protect pre-school children against diphtheria.

Abstract

Lactobacillus species are particularly noteworthy due to their ability to synthesize a variety of antimicrobial substances, including organic acids and bacteriocins. Lactiplantibacillus plantarum SU-KC1a, a member of the genus of Lactobacillus, had been previously isolated from human breast milk by our group. A preliminary in silico investigation had identified that Lpb. plantarum SU-KC1a possessed genes responsible for plantaricin, which is a type of bacteriocin produced by Lpb. plantarum strains. This study therefore aimed to assess the antibacterial activity of Lpb. plantarum SU-KC1a against pathogenic Listeria monocytogenes ATCC-7644 and to induce the biosynthesis of plantaricin by Lpb. plantarum SU-KC1a through co-culture with L. monocytogenes ATCC-7644 as the inducer strain. Our results showed that the cell-free supernatants from both co-culture and monoculture of Lpb. plantarum SU-KC1a exhibited a modest antibacterial activity against L. monocytogenes ATCC-7644. However, it was further determined that the inhibitory effect was not due to the production of plantaricin. Instead, it was primary attributed to the production of organic acids, which decreased the pH and inhibited the growth of L. monocytogenes ATCC-7644. In conclusion, our study demonstrated that Lpb. plantarum SU-KC1a could exert a modest antibacterial activity towards L. monocytogenes ATCC-7644, but not through the induction of plantaricin.

Abstract

Antibiotic resistance is increasingly becoming a global health threat, prompting the search for alternative treatments from natural compounds. Capsicin is a bioactive component of Capsicum species, exhibiting antimicrobial, antifungal, anticancer, and anti-inflammatory properties. Probiotics such as Bifidobacterium breve are also known for their immunomodulatory and antimicrobial effects. This study explored the synergistic potential of capsaicin and B. breve combination. Capsaicin was extracted from Capsicum frutescens using grinding and solvent methods, and its concentration was measured by spectrophotometry at 650 nm. The capsaicin extract was added to B. breve BS2-PB3 cultures and tested against Methicillin-resistant Staphylococcus aureus (MRSA). The results showed a significant increase in clear zone diameter with capsaicin supplementation compared to individual controls, indicating synergistic potential in enhancing the antimicrobial activity.Bahasa Indonesia Abstract:Resistensi antibiotik semakin menjadi ancaman kesehatan global, sehingga mendorong pencarian alternatif pengobatan dari senyawa alami. Capsaicin merupakan komponen bioaktif dari spesies Capsicum yang menunjukkan sifat antimikroba, antifungal, antikanker dan anti-inflamasi. Probiotik seperti Bifidobacterium breve juga dikenal karena efek imunomodulator dan antimikrobanya. Penelitian ini mengeksplorasi potensi sinergis kombinasi capsaicin dari ekstrak cabai rawit dan B. breve strain BS2-PB3. Capsaisin diekstraksi dari Capsicum frutescens menggunakan metode penggilingan dan pelarut, dimana konsentrasinya diukur dengan spektrofotometri pada 650 nm. Ekstrak cabai yang berisikan capsaicin ditambahkan ke kultur B. breve BS2-PB3 dan diuji terhadap Methicillin-resistant Staphylococcus aureus (MRSA). Hasil menunjukkan peningkatan signifikan pada diameter zona bening ketika MRSA dipaparkan terhadap kombinasi B. breve dengan suplementasi ekstrak cabai dibandingkan perlakuan-perlakuan individual. Hal tersebut menunjukkan potensi sinergis antara kultur probiotik dengan suplementasi capsaicin dalam meningkatkan aktivitas antimikroba.

Abstract

Probiotics are a group of bacteria with health benefits, which are widely used in various nutritional products. Various efforts are made to effectively increase growth of probiotics, including adding supplemental nutrition, such as protein. In this study, a series of tests were conducted (optical density, colony forming unit, and pH test) to evaluate the effect of collagen hydrolysate supplementation, obtained from tuna waste, on the growth of Bacillus subtilis G8, a candidate of probiotic bacteria which have been isolated from the Japanese traditional food natto. The results demonstrated positive impacts of collagen hydrolysate supplementation on cell density and colony-forming units, as well as on the reduction of pH of bacterial growth medium. Concentration of 5% was observed as the most optimum concentration to support the bacterial growth, while the most acidic pH of bacterial growth medium is observed at 3% supplementation. In conclusion, supplementation of collagen hydrolysate derived from tuna waste supported the growth of B. subtilis G8 as well as reduced the pH of its growth media. Bahasa Indonesia Abstract: Probiotik merupakan sekelompok bakteri yang memiliki manfaat kesehatan dan banyak digunakan dalam berbagai produk nutrisi. Beragam upaya telah dilakukan untuk secara efektif meningkatkan pertumbuhan probiotik, salah satunya melalui penambahan nutrisi tambahan seperti protein. Dalam penelitian ini, serangkaian uji dilakukan, meliputi uji kerapatan optik, unit pembentuk koloni (CFU), dan pengukuran pH, untuk mengevaluasi pengaruh suplementasi kolagen hidrolisat yang diperoleh dari limbah ikan tuna terhadap pertumbuhan Bacillus subtilis G8, yakni kandidat bakteri probiotik yang diisolasi dari makanan tradisional Jepang, natto. Hasil penelitian menunjukkan bahwa suplementasi kolagen hidrolisat memberikan dampak positif terhadap kerapatan sel dan jumlah koloni, serta menyebabkan penurunan pH medium pertumbuhan bakteri. Konsentrasi 5% diketahui sebagai konsentrasi yang paling optimal dalam mendukung pertumbuhan bakteri, sedangkan pH terendah pada medium pertumbuhan tercatat pada konsentrasi suplementasi sebesar 3%. Dengan demikian, dapat disimpulkan bahwa suplementasi kolagen hidrolisat yang berasal dari limbah ikan tuna mampu mendukung pertumbuhan B. subtilis G8 serta menurunkan pH media pertumbuhannya.

Abstract

Hepatocellular carcinoma (HCC) ranks sixth as the most common cancer and fourth as the most common cause of cancer-related death globally. The standard treatment for advanced HCC is by prescribing sorafenib, a tyrosine kinase inhibitor. Despite its moderate efficacy and concerning side effects, there is no better alternative to sorafenib to treat HCC. However, a new combination of atezolizumab (an inhibitor of PD-L1) and bevacizumab (an inhibitor of vascular endothelial growth factor), has shown a potential to surpass the efficacy of sorafenib. This review was written to provide an insight into pharmacodynamics of sorafenib and atezolizumab plus bevacizumab, effectiveness of sorafenib and the one of atezolizumab plus bevacizumab, utilization of atezolizumab plus bevacizumab in the clinical practice, as well as to argue that this combination can replace sorafenib as the standard palliative treatment for HCC.

Abstract

Glossina morsitans (G. morsitans), commonly known as tsetse fly, have caused public health concerns throughout the years. G. morsitans is the vector for Trypanosoma brucei (T. brucei), the parasite responsible for causing the deadly African sleeping disease (African trypanosomiasis). Researchers have searched for ways to contain this disease, but to little avail. Fortunately, new advances in sequencing methods have given researchers a new opportunity to win the war against the disease. The whole-genome sequence of G. morsitans provides essential data regarding involved genes that transmits T. brucei to humans. Information about those unique genes facilitates researchers to create new methods to prevent G. morsitans from becoming the vector of T. brucei, enabling the containment of this disease. With this, we review the unique genes in the G. morsitans genome, such as those that contribute to blood-feeding ability, establish a relationship with symbionts, and G. morsitans unique sensory genes, with an expectation that it would enhance our knowledge of G. morsitans as the vector for parasites causing African trypanosomiasis.

Abstract

Cancer treatments have developed over the years. A particular improvement is the utilization of oncolytic viruses to treat cancers. Oncolytic viruses are one of the immunotherapeutic tools that potentially could provide good results and benefits to the patients. Oncolytic viruses could mediate antitumor effects. Indeed, the connection between viral infections and cancer treatment have been reported historically. It is known that oncolytic viruses prefer to infect cancer cells rather than normal cells, resulting in the presentation of tumor-associated antigens to the immune system, boosting immunological activity in the tumor microenvironment, as well as assisting in the expression of inflammatory and immunomodulatory cytokines. Oncolytic viruses are a novel regimen in the cancer therapy, in which knowledge and technology of utilizing oncolytic viruses to treat cancer are still evolving. Importantly, clinical trials demonstrated that the viruses were well tolerated by cancer patients. Considering its potency and prospect, oncolytic viral treatments could be a useful additional tool for cancer therapy.

Abstract

Cancer is one of the deadliest form of diseases in humans, with the annual deaths ranging in the millions. Conventional treatments including chemotherapy, radiotherapy, or surgery has their limitations, including common off-target and non-specific effects. Ribonucleic acid interference (RNAi) offers a new strategy for treating cancer by silencing specific genes to prevent gene expression. This review highlights the application of RNAi-based approach in targeting cancer, discusses its potential advantages and limitations, summarizes the existing clinical trials and provides a greater understanding of RNAi-based therapy in cancer.

Abstract

Background: Cow’s milk allergy is relatively common among Indonesian infantswho are not exclusively breastfed. Despite extensive hydrolyzed formula and aminoacid-based formula are the recommended nutrition for infants with moderate-tosevere illness, not all parents could provide those specialized formulas, presumablydue to the high cost, low availability and poor palatability. In that case, soy-basedinfant formula could be an alternative nutrition. However, it was unknown whetherIndonesian mothers were aware on that alternative nutrition. Objective: We therefore assessed the knowledge levels of mothers who participated in an online platform of PrimaKu on the usefulness of soy-based infant formula to support growthand development of children with cow’s milk allergy. Methods: The study subjectsappeared to have sufficient levels of knowledge on the usefulness of soy-based infant formula for children with cow’s milk allergy. Results: In addition, by increasing their knowledge on iron and fiber fortification in soy-based infant formula, e.g.,through regular academic presentations, mothers could be more selective in choosing alternative nutrition for children with cow’s milk allergy. Conclusion: Our studysuggested that Indonesian mothers who were active in the online platform of Primaku had sufficient levels of knowledge on the usefulness of soy-based protein formanaging children with cow’s milk allergy.

Abstract

Mikrobiota susu kambing dapat mencakup bakteri asam laktat (BAL), khususnya Lactobacillus spp., yang berpotensi sebagai kandidat probiotik dan dapat dipergunakan dalam berbagai produk fermentasi. Pada penelitian ini, dilakukan isolasi dan karakterisasi BAL, khususnya Lactobacillus spp., dari susu kambing yang berasal dari peternakan lokal. Pada awalnya, didapatkan 25 isolat BAL dari susu kambing lokal. Semua isolat dikarakterisasi sesuai Bergey’s Manual of Systematics Archaea and Bacteria, yang dimulai dengan pewarnaan Gram, endospora dan ketahanan asam. Selanjutnya dilakukan uji aktivitas biokimia (uji aktivitas katalase, uji aktivitas hemolitik, serta uji fermentasi karbohidrat) dan uji ketahanan isolat terhadap berbagai konsentrasi NaCl, pH dan suhu. Berdasarkan hasil yang diperoleh, isolat B4 dan B6 dipilih untuk diidentifikasi lebih lanjut. Kedua isolat menunjukkan kemiripan karakteristik dengan Lactobacillus spp., yaitu berbentuk basil atau kokobasil, Gram positif, tidak membentuk spora, tidak tahan asam, katalase negatif, tidak memfermentasi manitol, sensitif terhadap konsentrasi NaCl tinggi, serta tumbuh optimum pada pH 5.5-6.2 dan pada suhu 37-45°C. Identifikasi kedua isolat berdasarkan sekuens 16S rRNA menunjukkan bahwa isolat B4 adalah Lacticaseibacillus paracasei dan isolat B6 adalah Limosilactobacillus fermentum. Sebagai kesimpulan, penelitian ini menunjukkan bahwa susu kambing peternakan lokal dapat menjadi sumber alternatif untuk mendapatkan isolat Lactobacillus spp.
