Abstract

Obesity is a condition where there is excessive fat in the body. Weight loss can be accomplished by inhibiting fat absorption through the inhibition of pancreatic lipase activity, a key enzyme in fat metabolism. Natural products from plants contain various phytochemical compounds that can act as pancreatic lipase inhibitors. This research was conducted to determine the lipase inhibitor activity of the methanol extract of Mangifera foetida leaves. Mangifera foetida leaves were extracted and then the total phenolic and flavonoid levels were determined. The extract was tested for its ability to inhibit pancreatic lipase in vitro. The inhibitory activities of the extract and Orlistat were measured using p-nitrophenyl palmitate as a substrate at concentrations of 31.25, 62.5, 125, 250, and 500 µg/mL. The results showed that the extracts of young leaves (YL) and mature leaves (ML) contained phenol and flavonoid compounds. The IC50 values of young leaf extract (YL) and mature leaves (ML) were 45.22 and 35.50 µg/ml, respectively. In conclusion, the Mangifera foetida leaf extract can be promoted as a good source of anti-obesity compounds

Abstract

The fermentation process of kombucha has become a focus of halal product research due to its ethanol content. Processing kombucha into powder form can be an alternative to address the ethanol content in liquid kombucha, as it has undergone a drying process, making it a potential halal-certified functional drink. The objective of this study was to compare the quality of liquid and powdered kombucha. The research was conducted from March to September 2024 at the Biotechnology Laboratory, Muhammadiyah University of Bandung. The method used was the Complete Randomized Design (CRD) method. The parameters analyzed included ethanol content, total BAA (Bacterial Acetic Acid), pH, organic acid content, and sugar content. In the production of powdered kombucha, optimization of maltodextrin and gum arabic usage was also conducted to achieve the best solubility and taste. The best powdered kombucha was obtained with a maltodextrin-to-gum arabic ratio of 3:2. In the research, liquid kombucha contained ethanol at an average of 0.09%. In comparison, no ethanol was detected in the powdered kombucha (0%). Total BAA in powder decreased after the drying process (p<0.05). The pH value of kombucha in liquid form (4.4) also differed significantly from powdered (3.2) (p<0.05). However, the levels of organic acids and sugars in liquid and powdered kombucha did not differ significantly (p>0.05). The sugar content in liquid and powdered kombucha was lower than that in kombucha in general.

Abstract

Many villages face challenges in optimizing their local potential. Sidomulyo Village, Bambanglipuro, Bantul, is one of the villages with abundant natural resources, including diverse plant species that have not been fully utilized. To address this, a community engagement program has been designed to provide training on the utilization of plant diversity in Sidomulyo Village for creating ecoprint batik fabric. The method employed in this community engagement program involves training in ecoprint creation using natural dyes and various local plant leaves. Throughout the program's implementation, the activities have been successful and have yielded positive outcomes. Participants' knowledge about ecoprint has increased significantly, from 20% to 100%. The enthusiasm of the participants in participating in the ecoprint training has been remarkable. Beyond the knowledge enhancement, many participants have shown an interest in further developing ecoprint variations, and some are even keen on establishing ecoprint businesses. Through this program, it is hoped to stimulate the growth of the creative economy by harnessing the natural potential within the surroundings of Sidomulyo Village. All of these efforts contribute to the improvement of the well-being and empowerment of the local community.

Abstract

Abstract. Aji OR, Widyaningrum T, Nabela P. 2025. Bacterial diversity and antibacterial activity of fermented buni fruit (Antidesma bunius) kombucha. Biodiversitas 26: 3781-3790. Kombucha is a fruit-based fermented beverage with a number of health benefits. Fruit-based substrates can enhance their nutritional value and microbial diversity. This study aimed to characterise the bacterial diversity and antibacterial activity of kombucha produced from buni (Antidesma bunius) fruits. Buni kombucha was fermented statically for 7, 14, and 21 days, and bacterial diversity was analysed by 16S rRNA (V3-V4) amplicon sequencing using the SILVA database. Antibacterial activity was evaluated against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus subtilis using the agar well diffusion method. Metagenomic analysis revealed Komagataeibacter (~80%) as the dominant genus, while low-abundance taxa such as Lactobacillus, Bacillus, and Bifidobacterium were detected; trace gut-associated genera (e.g., Bacteroides, Prevotella) likely originated from the pectin-rich substrate. Antibacterial activity increased significantly with fermentation time (p<0.05), with the strongest inhibition after 21 days: 13.83 ± 0.29?mm (S. aureus), 13.70 ± 0.61?mm (P. aeruginosa), 12.23 ± 0.25?mm (B. subtilis), and 10.77 ± 0.70?mm (E. coli). These results demonstrate that buni kombucha harbours a diverse microbiota and exhibits broad-spectrum antibacterial activity, supporting its potential as a postbiotic-rich functional beverage derived from local tropical fruits.

Abstract

Fragaria x ananassa or strawberry has significant economic value worldwide. However, its cultivation faces various challenges. One example was fungal diseases, such as anthracnose, caused by Colletotrichum acutatum. Disease management generally relies heavily on synthetic fungicides, raising concerns about environmental sustainability and human health risks. Therefore, more environmentally friendly alternatives are needed, such as the use of biological agents. This study aims to assess the potential of endophytic fungi from strawberry plants as natural biocontrol agents against Colletotrichum acutatum. The study began with the isolation of endophytic fungi from strawberry tissues and their characterization morphologically. The antagonistic activity of endophytic fungi against colletotrichum acutatum was evaluated using three methods: a dual culture assay to assess direct inhibition, a cell-free filtrate assay to analyze the effect of fungal metabolites, and a volatile organic compound (VOC) assay to inhibit the growth of pathogenic pathogens. In this study, 20 fungal isolates were successfully isolated. A total of 15 showed antagonism against Colletotrichum acutatum. Isolate A1 showed potent inhibition in the dual culture test by effectively antagonizing Colletotrichum acutatum. Isolate D1 produced extracellular compounds that reduced the growth of Colletotrichum acutatum by 52.28% ± 3.21% in the cell-free filtrate assay. At the same time, isolate D3 proved most effective in the VOC assay, suppressing the growth of pathogenic colonies by 71.35% ± 3.37%. Thus, endophytic fungi, especially isolates A1, D1, and D3, have the potential to serve as an environmentally friendly alternative to combat anthracnose disease on strawberries.
