Abstract

This study aims to determine the effect of Colocasia esculenta L on the growth and motility of zebrafish (Danio rerio) larvae after induction of rotenone as a stunting model. The Zebrafish have divided 3 groups i.e Normal (embryonic medium without rotenone induction), Negative Control (embryonic medium with rotenone induction), and Treatment Groups (embryonic medium with rotenone induction and Colocasia esculenta extracts, dose 30-60-90 ppm). The stunting model is designed by induction of 12.5 ppb rotenone to Zebrafish larvae. The growth of the larvae was observed by using a microscope to measure the body and head length. Evaluation of motility was carried out by recording the movement of the fish for 1 minute, and the pace was analyzed by using Video Tracker. The evaluations were observed at 3, 6, 9 dpf (day post-fertilization). Colocasia Esculenta Extract (CEE) groups have higher body lengths compared to the Negative Control Group. The lengths were 125.4 µm for CEE30, 119.4 µm for CEE60, and 121.4 µm CEE90 while the length for the Negative Control Group was 118.3 µm at 9 dpf. However, there were no different values for the ratio of head length to body length of all groups. The ratio was 1:6 at 3 dpf, and 1:4 at 6 and 9 dpf. Moreover, the motility was higher in CEE groups compared to the Negative Control at 3, 6, 9 dpf. The body length and motility of Zebrafish larvae higher compare to the Negative Control Group showing the potential of Colocasia esculenta extract on growth and enhancement of motoric activity.

Abstract

The Indonesian people empirically use Sidaguri (Sida rhombifolia L.) in medicine, such as anti-hyperuricemia. Herbal-based treatment is currently much preferred. Drug metabolism, in general, mainly occurs in the liver, so the possibility of damage to this organ is high. This study aims to determine the acute toxicity category of 96% ethanol extract of sidaguri based on the LD50 value, changes in body weight, changes in organ weight, and macro pathology in the organs of male DDY mice in vivo. The method used in this research was experimental with a fixed dose design following the Indonesian Food and Drug Authority regulations regarding guidelines for in vivo non-clinical toxicity tests. The test group was divided into six dose groups (standard, 50, 300, 2000, 5000, and 15000 mg/kg Body of weight). Observations were made for 24 hours. Observations were continued for 14 days on death parameters, toxicity symptoms, body weight, and relative organ weights using five main organs (heart, lungs, liver, spleen, and kidneys, and pathological examination). The acute toxicity test results showed no death in all dose treatment groups. Macropathological analysis did not show abnormalities in organs in all groups of mice. LD50 value is more than 15000 mg/kg. Sidaguri 96% ethanol extract is safe and Practically non-toxic.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui apakah ekstrak etanol 95% daun teh putih (Camellia sinensis) secara peroral meningkatkan aktivitas motorik mencit sebagai indikator efek antidepresan. Uji dilakukan menggunakan metode Rotarod selama 30 menit untuk mengukur kemampuan mencit dalam mempertahankan diri di atas roda yang berputar. Perlakuan diberikan dengan variasi dosis, dan hasil pengamatan berupa waktu ketahanan serta jumlah jatuh dianalisis menggunakan uji ANOVA. Hasil analisis waktu ketahanan menunjukkan bahwa nilai p (0,000) < 0,005 menunjukkan bahwa ada perbedaan signifikan antar kelompok uji. Analisis terhadap jumlah jatuh mencit juga menunjukkan hasil signifikan dengan p (0,002) < 0,005, sehingga hipotesis alternatif diterima. Hasil menunjukkan bahwa ekstrak etanol 95% daun teh putih memiliki efek antidepresan dengan meningkatkan aktivitas motorik mencit pada dosis 400 mg/KgBB. Temuan ini menunjukkan bahwa daun teh putih berpotensi sebagai sumber alami kandidat antidepresan melalui mekanisme peningkatan aktivitas motorik hewan uji.
