Abstract

Senyawa pektin dapat menyebabkan jus jeruk tampak keruh dan berkabut. Klarifikasi dengan enzim pektinase dapat dilakukan untuk menghilangkan kekeruhan tersebut. Enzim pektinase diisolasi dari bakteri pektinolitik yang diperoleh dari limbah kulit jeruk. Tujuan dari penelitian ini yaitu mengisolasi dan mengkarakterisasi bakteri pektinolitik dari limbah kulit jeruk, menentukan karakter enzim pektinase (pH optimum, suhu optimum, KM dan Vmaks) dan mengetahui pengaruh enzim pektinase dalam klarifikasi jus jeruk manis. Hasil pewarnaan Gram dan pewarnaan endospora menunjukkan bahwa semua isolat bakteri yang didapatkan termasuk bakteri Gram negative dan tidak menghasilkan spora. Dari sepuluh isolat bakteri dipilih tiga isolat yang menghasilkan enzim yang terbaik dalam proses klarifikasi jus jeruk manis yaitu isolat 2, 3 dan 9. Isolat dipilih berdasarkan pada nilai viskositas yang rendah, nilai total padatan terlarut yang tinggi dan nilai transmitansi yang tinggi. Karakter enzim ditentukan berdasarkan pH optimum, suhu optimum, kestabilan pH dan suhu, nilai KM dan Vmaks. Isolat 2, 3 dan 9 optimum pada pH 4,0, Isolat 2 optimum pada suhu 450C, Isolat 3 dan 9 optimum pada suhu 500C. Isolat 2, 3 dan 9 stabil pada kisaran pH 3,0-4,0 dan suhu 30-500C. Nilai KM isolat 2, 3 dan 9 berturut-turut yaitu 0,0246; 0,0199; 0,0170 mg/ml, sedangkan nilai Vmaks berturut-turut yaitu 0,0402; 0,0393; 0,0386 U/ml.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh variasi perlakuan pendahuluan (daun segar utuh, daun pemeraman, daun kering angin giling kasar (15 mesh), dan daun kering angin giling halus (60 mesh)) terhadap karakteristik mutu minyak atsiri daun jeruk purut yang meliputi rendemen, berat jenis, putaran optik, indeks bias, viskositas, dan kelarutan dalam alkohol 70% serta kandungan dan kadar senyawa aktif minyak atsiri daun jeruk purut terpilih. Hasil penelitian yang diperoleh menunjukkan bahwa minyak atsiri daun jeruk purut dengan perlakuan pemeraman memiliki rendemen tertinggi sebesar 0,867%. Berat jenis, viskositas, indeks bias, putaran optik, dan kelarutan alkohol 70% berturut-turut sebesar 0,837-0,845 g/ml, 0,016-0,023 N.m/s2, 1,450-1,453 dan 1:3-1:4. Senyawa aktif minyak atsiri daun jeruk purut (Citrus hystrix DC) terpilih yaitu Citronellal 64,15%, Beta-Citronelol 10,71%, Linalool 5,54%, dan Trans-Caryophyllene 5,31% yang didapat dari hasil destilasi daun jeruk purut dengan perlakuan pendahuluan pemeraman.

Abstract

Kenteng Village Puntukrejo is one of the industrial centers ofprocessed sweet potato in Karanganyar Regency. Processed purple sweetpotato products produced include bakpia, wingko, and ice cream.Generally, the problems that occur in the sweet potatoes processing arethe equipment used is not in accordance with the standards for the foodindustry, the limited amount of equipment, hygiene and sanitation duringprocessing is not considered so as to bring the risk of contamination to theend product, as well as stagnant business conditions because the absenceof product diversification and limited marketing area. To solve theproblem through this program of dedication will be done (1) Introductionof appropriate technology for the processing of sweet potato; (2) Trainingof food industry management; (3) Introduction of processing technologyfor 'cassava leaf chips' as an effort to diversify products; (4) IT-basedmarketing training; (5) as well as facilitation of testing of nutrient contenton the end product. Through this dedication activity has been doneIntroduction of processing technology for 'cassava leaf chips' as the effortof product diversification. To improve product quality and productioncapacity has been given the equipment in the form of oven, large capacitymixer, wingko molding, freezer, analytical scales, stainless steel basin, andstainless steel table. Food industry management training in the form ofGood Manufacturing Practices (GMP) training has been given to ensurequality and safe the products. It also provides IT-based marketing trainingto improve product marketing. The results of laboratory analysis, purplesweet potato bakpia has water content 33.29%, mineral content 1.51%, fatcontent 11.91%, protein content 9.34%, carbohydrate content 43.95%, andantioxidant activity 6.20%. Purple sweet potato wingko has water content34.47%, mineral content 1.32%, fat content 11.70%, protein content 3.06%,and carbohydrate content 49.45%. Abstract

The aim of this research was to study the binding ability of viable and non-viable of Lactobacillus paracasei SNP-2 to aflatoxin B1 (AFB1) in phosphate buffer saline (PBS) at pH 7.3. Bacterial cells were grown in MRS broth at 37 °C for 24 h, and then centrifuged at 1,800g[a1]  for 20 min at 10 °C to get the pellet. Pellet was suspended in PBS pH 7.3 until the cell concentration was about 1010 CFU/ml. Viable cells, the heated, and acid-killed cells were evaluated their ability to bind AFB1 in PBS pH 7.3. Stability of the L. paracasei SNP-2/AFB1 complexes was evaluated by determining the amount of the released AFB1 to the PBS following five times washing. The results showed that AFB1 binding ability to heat-and acid-kill bacteria were higher than that of by viable cells. More than 70% of bound AFB1 was released from viable bacteria after five times washing. However, the heated and acid-killed cell treatments significantly increased the complex stability of bacteria-AFB1

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui pengaruh lama fermentasi moromi terhadapviskositas, kadar protein terlarut, aktivitas antioksidan, dan sensori dari kecap bungkil wijen putih sangrai dannonsangrai, dan untuk mengetahui perlakuan yang menghasilkan kecap dengan kualitas terbaik dari parameterpengujian yang dilakukan. Perancangan Penelitian menggunakan Rancangan Acak Lengkap (RAL) yaitu lamafermentasi 0, 2, 4, 6, dan 8 minggu kecap bungkil wijen putih sangrai dan nonsangrai. Hasil penelitianmenunjukkan bahwa pengaruh lama fermentasi moromi dari 0 sampai 8 minggu memberikan perbedaan yangnyata pada 5% terhadap viskositas, kadar protein, antioksidan kecap bungkil wijen sangrai dannonsangrai. Namun untuk sensori tidak begitu memberikan perbedaan yang nyata. Hasil yang didapat viskositasterbesar pada fermentasi 4 minggu sebesar 977 cP, kadar protein terlarut pada fermentasi 6 minggu sebesar 3,03% dan 2,97 %, aktivitas antioksidan pada fermentasi moromi 6 minggu sebesar 6,96%/mg dan 6,02%/mg.Perlakuan lama fermentasi moromi 6 minggu dipilih sebagai perlakuan lama fermentasi moromi yangmenghasilkan kecap yang terbaik. Untuk perlakuan penyangraian, pada penelitian ini memberikan pengaruh,dimana pada kecap bungkil wijen putih sangrai memberikan viskositas, dan aktivitas antioksidan yang lebihtinggi dibandingkan tanpa penyangraian

Abstract

Pasteurized milk stored at cool temperatures generally have a relatively short shelf life. Antibacterialactivity of noni juice in maintaining the quality of pasteurized goat's milk stored at cold temperatures wasinvestigated. Characteristics of pasteurized goat's milk examined were organoleptic performances, microbialgrowth (total plate count and psychotrophic bacteria), chemical characteristics (protein, pH and acid total) withthe addition of noni juice at variation concentrations) (0%, 5%, 7.5%, 10%, and 12.5%) and times of storage(0.5, 10, 15 and 20 storage days). The results showed that the concentration of noni juice addition of 0 % -7.5 %on the parameters of colour, flavour, taste and homogenity and still were acceptable by consumers. Theinhibitory effects of TPC amount of growth were shown in 12.5% (2.8x101-3.3x101cfu/ml), and this inhibitoryeffects weren’t significantly different between each other of concentrations) (α> 0.05). The low decrease ofprotein content was indicated at concentrations of 7.5 % and still with the value of under SNI up to 20 days ofstorage (from 3.48 to 3.19%). The lowest decrease of pH and the increase acid total titration were shown atconcentrations of 5% with the values of pH (6.40 to 6.17) and acid total (0.20 %-0.22%). Based on organoleptic,microbial and chemical characteristics, it can be concluded that the best concentration of noni juice than that theothers can be added to the pasteurized goat milk during storage was 5%.

Abstract

Tujuan dari penelitian ini untuk mengetahui pengaruh konsentrasi tepung uwi putih kulit cokelat terhadap nilai pH, viskositas, kadar asam laktat, karakteristik sensori, dan total bakteri probiotik pada minuman sinbiotik uwi putih kulit cokelat. Penelitian ini terdiri dari dua tahap utama yaitu pembutatan tepung uwi putih kulit cokelat dan pembuatan minuman sinbiotik uwi putih kulit cokelat. Rancangan percobaan adalah Rancangan Acak Lengkap (RAL) dengan satu faktor yaitu kombinasi tepung uwi putih kulit cokelat. Untuk melihat beda nyata antar perlakuan dianalisis secara statistik menggunakan oneway ANOVA pada taraf α 0,05 dilanjutkan dengan menggunakan uji Duncan Multiple Range Test (DMRT) pada taraf α 0,05. Perbedaan konsentrasi tepung uwi putih kulit cokelat memberikan pengaruh yang berbeda terhadap nilai pH, nilai viskositas, kadar asam laktat, nilai sensori, dan total bakteri probiotik. Nilai pH terendah (3,68), kadar asam laktat tertinggi (0,60%), dan total bakteri probiotik terbesar (9,39 log cfu/ml) didapatkan dari penambahan 2,5% tepung uwi putih kulit cokelat dan 7,5% susu skim. Nilai viskositas terbesar didapatkan dari penambahan 10% tepung uwi putih kulit cokelat dan 0% susu skim yaitu 10,92mPas. Berdasarkan analisis sensori penambahan tepung uwi putih kulit cokelat 7,5% dan susu skim 2,5% memiliki nilai sensori paling baik diantara keempat sampel lainnya berdasarkan parameter warna (2,78), kekentalan (2,41) dan overall (2,85).

Abstract

Curing is a method that can be used to maintain the quality of beef. The curing method uses nitrate and nitrite compounds, which have useful roles in the curing process. However, those compounds have a side effect on the body if consumed continuously, which causes cancer disease (carcinogenic). This harmful carcinogenic property can be controlled by the addition of ascorbic acid. Ascorbic acid is commonly found in several fruits, one of which is red guava (Psidium guajava L). Therefore, this study aims to find out the most optimal addition of red guava extract in maintaining the beef qualities with curing treatment on the parameters of water content, pH, whc, texture, color, ascorbic acid, and nitrite residue. This study was conducted using a Completely Randomized Design with the various concentration of red guava extract (0%-5%) and observed at soaking times of 0, 2, 4, and 6 days. The analysis was performed by twice of sample repetition and analysis repetition. The results of the analysis were statistically analyzed by One Way ANOVA continued with the DMRT test if the results were significantly different (α = 0.05). Based on the results of the study, the higher of concentration in red guava extract, the pH, water content, whc, b* color, a* color, nitrite residue were smaller while the hardness, springiness, cohesiveness, L* color, a* color, the ascorbic acid content in curing were higher. The chosen treatment due to the test parameter in this study was the addition of 4% concentration in red guava extract in the curing process with 2 days soaking times.
